6. Protocells Featuring Membrane-Bound and Dynamic Membraneless Organelles

Abstract

Living cells, especially eukaryotic ones, use multicompartmentalization to regulate intra- and extracellular activities, featuring membrane-bound and membraneless organelles. These structures govern numerous biological and chemical processes spatially and temporally. Synthetic cell models, primarily utilizing lipidic and polymeric vesicles, have been developed to carry out cascade reactions within their compartments. However, these reconstructions often segregate membrane-bound and membraneless organelles, neglecting their collaborative role in cellular regulation. To address this, we propose a structural design incorporating microfluidic-produced liposomes housing synthetic membrane-bound organelles made from self-assembled poly(ethylene glycol)-block-poly(trimethylene carbonate) nanovesicles and synthetic membraneless organelles formed via temperature-sensitive elastin-like polypeptide phase separation. This architecture mirrors natural cellular organization, facilitating a detailed examination of the interactions for a comprehensive understanding of cellular dynamics.

Abstract

Nowadays, about half of hospital-acquired infections are due to medical devices implantation. In this context, we have developed two types of antimicrobial coatings adapted to the biomedical field. The first one is based on peptide composed of an anchoring sequence, an antimicrobial sequence and a pathogen-specific cleavage site and grafted on the substrate. The antimicrobial site will be released only in the presence of the pathogen through the use of the cleavage site. Despite of the success of peptide grafting, some parameters must be optimized in order to obtain an antimicrobial effect. The second antimicrobial coating concept is based on the layer-by-layer technique by using poly(L-arginine) (PAR) and hyaluronic acid (HA). The effect of the size of PAR chains on the antimicrobial character of the coating was investigated and it is proven that only films composed with PAR of 30 residues present an antibacterial effect. Moreover HA is the only polyanion leading to such antimicrobial multilayer. It is also demonstrated that this antimicrobial properties is maintained when other cationic homopolypeptides are used in association with HA in layer-by-layer films., De nos jours, la moitié des infections nosocomiales sont liées à la pose de dispositifs médicaux. Dans ce contexte, nous avons développé deux types de revêtements antimicrobiens adaptés au domaine biomédical. Le premier s'appuie sur le greffage d’un peptide comprenant une séquence d’accroche, une séquence antimicrobienne et un site de clivage spécifique d'un pathogène. La séquence antimicrobienne sera alors libérée uniquement en présence du pathogène. Malgré la réussite de l’accroche du peptide, certains paramètres restent encore à optimiser afin d’obtenir un effet antimicrobien. Le deuxième revêtement s’appuie sur la conception d’un film antimicrobien « couche-par-couche » avec l’utilisation de poly(L-arginine) (PAR) et d’acide hyaluronique (HA). L’influence de la taille des chaînes de PAR a été étudiée et seul le film construit à partir de PAR de 30 résidus possède un effet antibactérien. Avec cette PAR, nous avons démontré que HA est le seul polyanion conduisant à des propriétés antibactériennes. Ces propriétés antimicrobiennes sont maintenues lorsque d’autres homopolypeptides cationiques sont associés à HA.

Abstract

Ultracentrifugated compact polyelectrolyte complexes (uCoPECs) represent a new class of materials that are obtained by ultracentrifugation of solutions of polyanion/polycation complexes in the presence of salt. In the present study, two polysaccharides, chitosan and alginate, were used to form such complexes, thus providing a solid material uniquely composed of polysaccharides. The conditions for obtaining the uCoPEC material were optimized: the optimal salt concentration and polysaccharide concentrations were assessed, and the ultracentrifugation speed proved to be a key parameter to obtain compact and homogeneous materials. The Young's modulus, E, of the material was of the order of 12 MPa, which is the highest E value measured for a uCoPEC. The material contained nanometer-sized crystals of chitosan as indicated by X-ray diffraction. Most strikingly, this material proves to be totally cell- and bacteria-resistant. Abstract

International audience; Coatings with antimicrobial properties are garnering interest to prevent implant-associated infections. Recently, we showed that poly(arginine)/hyaluronic acid (PAR/HA) multilayers built with PAR chains constituted from 30 arginine residues (PAR30) have strong antimicrobial properties through a contact-killing mechanism. This property is due to the ability of PAR30 chains, when associated with HA, to diffuse in the multilayer. Here, we investigate the effect of the nature of the polyanion on the antimicrobial activity of (PAR30/polyanion) multilayers. Four polysaccharides, one polypeptide, and one synthetic polyelectrolyte are investigated. Surprisingly, only HA leads to films with antimicrobial character. We relate this property to the strong diffusion capacity of PAR30 chains in (PAR30/HA) multilayers compared to their diffusion ability in the other (PAR30/polyanion) films. Through isothermal microcalorimetry experiments, we also demonstrate that interactions in solutions of PAR30 chains with the different polyanions are characterized by a negative reaction enthalpy for all of the investigated polyanions except for HA, where the enthalpy of reaction is positive. Moreover, the molecular weight of HA is not a key parameter for the diffusion ability of PAR chains or for the bioactivity of the film. These results constitute an important step toward the establishment of rules to design contact-killing antimicrobial polyelectrolyte multilayers
