In this study, the genetic diversity of Donax vittatus across the Iberian Peninsula was investigated using four mitochondrial (COI, Cytb, 16S F and M types) and three nuclear (H8, 18S and 285) genes. These same molecular markers were also sequenced in D. semistriatus and D variegatus to address the phylogenetic relationships of the species of the genus Donax common along the European coasts. Our results showed high haplotype diversity in combination with a low nucleotide diversity and a star-shaped network with a predominant haplotype, indicating a recent population expansion for the examined sampling sites of D. vittatus. Furthermore, analyses of population differentiation performed with COI mitochondrial marker, including global Far estimation and pairwise Far values, indicated the non-existence of significant genetic structure in D. vittatus of Northwest Iberian populations. Because these localities show a high genetic similarity, we suggest that D. vittatus could be a potentially alternative exploitable resource, as complement to the D. trunculus fisheries, whose natural stocks have decreased dramatically in some areas. Morphological uniformity in geographically widespread species may cause genetically distinct entities to pass unnoticed if they can only be detected by molecular approaches. The importance of uncovering such cryptic diversity is prompted by the need to understand the putative adaptive potential of populations along species ranges and to manage biodiversity conservation efforts. In this study, we aim to assess cryptic intraspecific genetic diversity and taxonomic status of the widely distributed intertidal mussel Mytilus galloprovincialis, along Atlantic southwestern (SW) Iberian, Atlantic northwestern (NW) Moroccan and Mediterranean Tunisian shores. By using mitochondrial (16S restriction-fragment length polymorphism) and nuclear (polyphenolic adhesive protein gene, Glu-5') markers, we discovered a more complex taxonomic diversity of M. galloprovincialis than previously known. Both Atlantic and Mediterranean haplogroups of M. galloprovincialis were detected along Atlantic SW Iberian shores along with M. galloprovincialis/edulis hybrids (92.2% Atlantic, 3.9% Mediterranean and 3.9% hybrids). In contrast, NW Moroccan populations consisted solely of Atlantic M. galloprovincialis. The Mediterranean populations did not include M. galloprovincialis/ edulis hybrids, but both Atlantic (58%) and Mediterranean (42%) lineages were detected. Divergent selection between coastlines and/or indirect larval dispersal by human activities may be the drivers of this geographically structured genetic diversity. A biometric, nutritional and sensory analysis of raw and cooked mussels comparing Mytilus sp. from the north-west coast of Portugal and Spain (Minho and Galicia, respectively) and the new offshore production site of Armona (Algarve, south Portugal) was carried out. In addition, multiple factorial analysis was performed to explore potential relationships between sensory attributes and nutritional content properties between the different mussels. Results showed that, at similar times of sale, biometrics of mussels from Armona and Vigo were similar and bigger than the remaining. Nonetheless, despite some similarities in proximate composition, mussels presented differences in lipid classes, fatty acid content and free amino acids profiles. These differences were not fully reflected in the sensory assessment by the panel, which were able to distinguish different production sites in raw specimens but displayed problems in discrimination these in cooked mussels. Eukaryotic cells typically contain numerous mitochondria, each with multiple copies of their own genome, the mtDNA. Uniparental transmission of mitochondria, usually via the mother, prevents the mixing of mtDNA from different individuals. While on the one hand, this should resolve the potential for selection for fast-replicating mtDNA variants that reduce organismal fitness, maternal inheritance will, in theory, come with another set of problems that are specifically relevant to males. Maternal inheritance implies that the mitochondrial genome is never transmitted through males, and thus selection can target only the mtDNA sequence when carried by females. A consequence is that mtDNA mutations that confer male-biased phenotypic expression will be prone to evade selection, and accumulate. Here, we review the evidence from the ecological, evolutionary and medical literature for male specificity of mtDNA mutations affecting fertility, health and ageing. While such effects have been discovered experimentally in the laboratory, their relevance to natural populations—including the human population—remains unclear. We suggest that the existence of male expression-biased mtDNA mutations is likely to be a broad phenomenon, but that these mutations remain cryptic owing to the presence of counter-adapted nuclear compensatory modifier mutations, which offset their deleterious effects.
