The Bur Ni Geureudong volcanic complex is a potential area for geothermal development and utility. This volcanic complex is situated in northern Sumatra, Aceh Province, Indonesia. Geothermal potential in this area is manifested by several hot springs discharged in the Pinto Rime Gayo (PRG), Simpang Balek (SB), Uning Bertih (UB), and Silih Nara (SN) springs. All springs have a neutral pH and a surface temperature ranges from 34.10 to 67.11°C. A geochemical investigation was conducted on the hot springs to understand the hydrochemistry of the geothermal fluid and estimate the reservoir temperature of the geothermal area. This investigation was an exploration study of geothermal resources in the area. Hot waters in the PRG, SB, UB, and SN springs were collected and analyzed in May 2018 to identify the cations and anions contained. Piper and Cl-SO4-HCO3 diagrams have been plotted to characterize the geothermal fluid. Na/K geothermometer has been applied to calculate the temperature of the geothermal reservoir. Isotope analysis has also been conducted to estimate the origin of the fluid. This research discovered that the major element of the water demonstrates the various geothermal water types. The PRG spring contains Na-K-Cl water, whereas the SN spring contains diluted Cl-HCO3 water with mixed cation concentrations of Ca, Mg, Na, and K. The waters in SB and UB are a mixed form of Cl-HCO3-SO4 waters. According to the δD and δ18O relationship, the genesis of geothermal fluids is hydrothermal fluids of meteoric water. Based on the Na/K geothermometer, the reservoir temperature of the Bur Ni Geureudong geothermal system is a range of 171-289°C which is indicated as a high enthalpy system (>225°C). It can be concluded the Bur Ni Geureudong geothermal region is recognized by this study as a promising source of renewable energy. Plant viruses are major restrictive pathogens to cucurbits production in Jordan. During field surveys conducted in September 2022 in the main cucurbit growing areas (desert area, Jordan Valley, and highlands), virus-like symptoms such as vein clearing, mosaic patterns, interveinal chlorosis, and fruit malformation were observed, in the presence of high whitefly populations (Bemisia tabaci MEAM1, Mdanat et al., 2022). A total of 80 leaf samples from different species [48 cucumber (Cucumis sativus), 11 squash (Cucurbita pepo), 14 melon (Cucumis melo) and 7 watermelon (Citrullus lanatus)], including 70 symptomatic and 10 asymptomatic samples, were collected for further investigations. Total RNA and DNA were extracted using RNeasy and DNeasy plant mini kit (QIAGEN), respectively, and molecular detection against an array of cucurbit-infecting viruses was conducted using protocols available in the literature (Suppl. Table). Squash vein yellowing virus (SqVYV; Potyviridae, Ipomovirus) was detected in combination with other cucurbit-infecting viruses, in 10 of 70 symptomatic samples with primers (SqYVV-v4762; SqYVV-c5512) targeting a portion of the cylindrical inclusion (CI) gene (Hernandez et al., 2021), including 3 cucumber, 4 squash and 3 melon samples, however, it was not detected in watermelon. Among other RNA viruses, cucumber green mottle mosaic virus, cucurbit yellow stunting disorder virus, cucurbit aphid-borne yellows virus, cucumber vein yellowing virus, and cucurbit chlorotic yellows virus were detected in 9, 34, 7, 18, and 23 samples, respectively. The DNA viruses, squash leaf curl virus and watermelon chlorotic stunt virus, were detected in 36 and 30 samples. None of the detected viruses were present in asymptomatic samples. All 80 samples were negative for watermelon mosaic virus and tomato leaf curl New Delhi virus. Cloning (pGEM T-Easy Vector; Promega), sequencing, and BLASTn analysis of 4 CI-specific cloned amplicons (~ 0.75 kb, GenBank Acc no. PP908660-PP908663) confirmed the identification of SqVYV, with highest BLASTn identity of ~91% and ~99% to isolates ESF3 from USA (MW584342) and SqVYV-IL from Israel (KT721735), respectively. To reconstruct the complete genome sequence of the SqVYV isolate from Jordan, total RNA from a pool of squash and melon plants was used to construct a cDNA library with the Illumina DNA Prep kit, which was sequenced on a NextSeq2000 instrument as paired reads (2x150 bp) at Leibniz Institute DSMZ, to generate 18,723,252 total reads. Bioinformatic analysis in Geneious (Biomatters) resulted in the assembly of a single genome sequence of 9,831 nt (GenBank Acc. no. PP916052), covered by 83,995 reads, with mean coverage of 1,197 (Geneious mapper, 10% maximum mismatches per read). The complete genome sequence and the deduced polyprotein sequence shared over 99% identity with SqVYV-IL from Israel. Mechanical inoculation of 10 cucumber (cv. Giant Global) and 10 squash (cv. Lebanese) plants with inoculum from infected cucumber, resulted in vein clearing and mosaic symptoms, after three weeks from inoculation, while no symptoms were observed in the six negative controls. Symptomatic plants were confirmed by PCR to be infected by SqVYV as described earlier. In this study, we report the occurrence of SqVYV for the first time in cucurbit crops in Jordan. SqVYV has been reported to cause large economic losses in cucurbits in USA and Israel, posing a major threat to watermelon growers (Adkins et al., 2007; 2008 a & b; 2013; Reingold et al., 2016; Webster et al., 2013). Our findings should encourage further studies on the incidence and prevalence of SqVYV in cucurbit and non-cucurbit crops, including weeds, to understand its epidemiology in Jordan, including its natural host range.

Wound healing in the mouth has its challenges due to masticatory movements and the presence of bacteria that can inhibit its process. The aim of this study was to analyze the contents of red algae ( Gracilaria verrucosa ) from Indonesia, and its potential as a wound-healing agent for oral wounds using animal model. Red algae content was determined by phytochemical tests and gas chromatography-mass spectroscopy (GC-MS). The wound was made by making an incision on the gingival mucosa of Rattus norvegicus and the parameters assessed were bleeding time, number of fibroblast cells, and time of wound closure. Three doses of G. verrucosa gel were used (2.5%, 5%, and 10%) and the gels were applied twice a day, at 6:00 and 18:00. Application was carried out topically by applying 0.1 ml of gel to the incision wound using a 1 mL syringe. Our phytochemical test indicated that the G. verrucosa contained alkaloids, steroids, flavonoids, and phenols. The dominant contains of the G. verrucosa were glycerol (36.81%), hexadecenoic acid (20.74%), and cholesterol (7.4%). The hemostasis test showed that the 2.5% G. verrucosa extract gel had the shortest bleeding time, 33.98±2.66 seconds. On the seventh day of the initial proliferation phase, the number of fibroblasts was not significantly different among groups. On day 14, the number of fibroblasts was only significantly different between 10% G. verrucosa and untreated group ( p =0.007). On day 28, however, both 5% and 10% G. verrucosa were significantly higher compared to untreated group, both had p =0.010. Daily clinical examination showed that animals that were given 2.5% and 5% of G. verrucosa extract gel experienced wound closure on day 10. Animals treated with 10% of extract gel, all wounds healed on day 9. Synthesis of silver nanoparticles-chitosan composite particles sphere (AgNPs-chi-spheres) has been completed and its characterization was fulfilled by UV-vis spectroscopy, Fourier transform infrared (FT-IR) spectroscopy, X-ray diffraction (XRD), scanning electron microscopy (SEM), and zetasizer nano. UV-vis spectroscopy characterization showed that AgNPs-chi-spheres gave optimum absorption at a wavelength of 410 nm. The XRD spectra showed that the structure of AgNPs-chi-spheres were crystalline and spherical. Characterization by SEM showed that AgNPs-chi-spheres, with the addition of 20% of NaOH, resulted in the lowest average particle sizes of 46.91 nm. EDX analysis also showed that AgNPs-chi-spheres, with the addition of a 20% NaOH concentration, produced particles with regular spheres, a smooth and relatively nonporous structure. The analysis using zetasizer nano showed that the zeta potential value and the polydispersity index value of the AgNPs-chi-sphere tended to increase with an increased NaOH concentration. The results of the microbial activity screening showed that the AgNP-chi-Spheres with highest concentration of NaOH, produced the highest inhibition zone diameters against S. aureus , E. coli , and C. albicans , with inhibition zone diameters of 19.5, 18.56, and 12.25 nm, respectively.

In a simple and instant procedure for detecting fish freshness, a hydrogel and hydrophilic pectin matrix membrane was used successfully as an optical pH sensor by immobilizing the chromoionophore ETH 5294 (CI), which is very selective and sensitive for the membrane. The Pe/CI optical pH sensor exhibited excellent linearity between pH 5 and pH 9, with a sensor response time of 5 min and reproducibility of 1.49% relative standard deviation (RSD). The sensor showed response stability for 15 days and a response reduction of 8.6%. The sensor's capability was demonstrated by the detection of fish freshness for 17 days at 4 °C.

Objective: Traditionally vascular rings in infants and children are treated through an open thoracotomy. Recently, thoracoscopic surgery has been used for these complex procedures. This study reports our early experience with thoracoscopic division of vascular rings and evaluates the efficacy and safety of this approach., Material and Methods: Patients who underwent thoracoscopic division of vascular rings at King Khalid University Hospital, Riyadh, Saudi Arabia, from December 2004 to January 2006 are included. Their data were carefully analyzed looking at demographics, clinical presentation, diagnostic modality, type of the anomaly, operative details, complications, and outcome., Results: A total of 9 patients underwent thoracoscopic division of vascular rings. Age at surgery ranged between 2 and 108 months (mean, 24 months). Weight varied between 5.3 and 32 kg (mean, 10.3 kg). All patients were symptomatic. Computed tomographic scan was diagnostic and accurately defined the type of anomaly in all the patients. Four patients had a right aortic arch with an aberrant left subclavian artery and left ductus/ligamentum arteriosum, 2 had double aortic arches, and 3 had a right aberrant subclavian artery. One patient developed right-sided pneumothorax on the contralateral site, and another one developed apnea 12 hours after surgery, requiring mechanical ventilation. There was no mortality. Operative time ranged between 50 and 145 minutes, the mean being 107 minutes. The average hospital stay was 4 days. Five patients had their preoperative symptoms completely resolved, and the rest are showing steady improvement. The average follow-up period is 6 months., Conclusion: Our early experience indicates that thoracoscopic division of vascular rings is safe and effective. Because it takes away the need for thoracotomy, it is likely that it can result in less postoperative pain and rapid convalescence. It also prevents the ill effects of thoracotomy and gives good cosmetic results.

By oxidation-reduction assay, the viability of Mycobacterium tuberculosis in sputum specimens was evaluated. The technique is based on the Alamar Blue and Malachite Green dyes, which change their color in response to M. tuberculosis growth. The method is simple, permits visual reading of results, and is applicable for laboratories with limited resources.
