ABSTRACTAdditive Manufacturing (AM) processes have the capability to promote a more rational use of resources, reduce generated waste, as well as repair damaged components to extend their lifespan. With the aim of contributing to a more efficient and environmentally friendlier manufacturing process, this research work investigates the energy consumption of the Laser Directed Energy Deposition process (DED-LB) through the introduction of new parameters in its characterisation, which are energy and mass efficiency. First, the energy efficiency of the laser generator itself has been characterised. Based on the results obtained, it was possible to conclude that pulsed laser operation strategies allow a reduction in consumption of up to 9% compared to a continuous laser operation mode. Second, a design of experiments was carried out to relate the energy and mass efficiency of the deposition process to the input parameters: laser power, feed rate, and mass rate. The parameter that most influences the energy efficiency of the process is the mass rate, where high rates are advisable to maximise its value. On the other hand, it is concluded that an increase in the mass rate or a reduction in the feed rate results in an increase in the mass efficiency

We study a model problem describing vibrational resonance by means of a high-order averaging technique based on so-called word series. With the technique applied here, the tasks of constructing the averaged system and the associated change of variables are divided into two parts. It is first necessary to build recursively a set of so-called word basis functions and, after that, all the required manipulations involve only scalar coefficients that are computed by means of simple recursions. As distinct from the situation with other approaches, with word-series, high-order averaged systems may be derived without having to compute the associated change of variables. In the system considered here, the construction of high-order averaged systems makes it possible to obtain very precise approximations to the true dynamics.

In prostate cancer (PCa), androgens upregulate tumorigenesis, whereas in benign tissue, the revival of androgen receptor (AR) signaling suppresses aggressive behaviors, suggesting therapeutic potential. Dogs, natural PCa models, often lack AR in PCa. We restored AR in dog PCa to investigate resultant characteristics. Three AR-null canine PCa lines (1508, Leo, 1258) were transfected with canine wild-type AR and treated with dihydrotestosterone (DHT). In 1508, AR restoration decreased clonogenicity (p = 0.03), viability (p = 0.004), migration (p = 0.03), invasion (p = 0.01), and increased expression of the tumor suppressor NKX3.1, an AR transcriptional target (p = 0.001). In Leo, AR decreased clonogenicity (p = 0.04) and the expression of another AR transcriptional target FOLH1 (p < 0.001) and increased the expression of NKX3.1 (p = 0.01). In 1258, AR increased migration (p = 0.006) and invasion (p = 0.03). Epithelial-mesenchymal transition (EMT) marker (Vimentin, N-cadherin, SNAIL1) expression increased with AR restoration in Leo and 1258 but not 1508; siRNA vimentin knockdown abrogated AR-induced 1258 migration only. Overall, 1508 showed AR-mediated tumor suppression; AR affected proliferation in Leo but not migration or invasion; and EMT and AR regulated migration and invasion in 1258 but not proliferation. This study highlights the heterogeneous nature of PCa in dogs and cell line-specific effects of AR abrogation on aggressive behaviors.

BackgroundThe upper-airway microbiome is involved in asthma exacerbations despite inhaled corticosteroid (ICS) treatment. Although human genetics regulates microbiome composition, its influence on asthma-related airway bacteria remains unknown.ObjectiveWe sought to identify genes and biological pathways regulating airway-microbiome traits involved in asthma exacerbations and ICS response.MethodsSaliva, nasal, and pharyngeal samples from 257 European patients with asthma were analyzed. The association of 6,296,951 genetic variants with exacerbation-related microbiome traits despite ICS treatment was tested through microbiome genome-wide association studies. Variants with 1 × 10-4 -6 were examined in gene-set enrichment analyses. Significant results were sought for replication in 114 African American and 158 Latino children with and without asthma. ICS-response-associated single nucleotide polymorphisms reported in the literature were evaluated as microbiome quantitative trait loci. Multiple comparisons were adjusted by the false discovery rate.ResultsGenes associated with exacerbation-related airway-microbiome traits were enriched in asthma comorbidities development (ie, reflux esophagitis, obesity, and smoking), and were likely regulated by trichostatin A and the nuclear factor-κB, the glucocorticosteroid receptor, and CCAAT/enhancer-binding protein transcription factors (7.8 × 10-13 ≤ false discovery rate ≤ 0.022). Enrichment in smoking, trichostatin A, nuclear factor-κB, and glucocorticosteroid receptor were replicated in the saliva samples from diverse populations (4.42 × 10-9 ≤ P ≤ .008). The ICS-response-associated single nucleotide polymorphisms rs5995653 (APOBEC3B-APOBEC3C), rs6467778 (TRIM24), and rs5752429 (TPST2) were identified as microbiome quantitative trait loci of Streptococcus, Tannerella, and Campylobacter in the upper airway (0.027 ≤ false discovery rate ≤ 0.050).ConclusionsGenes associated with asthma exacerbation-related microbiome traits might influence asthma comorbidities. We reinforced the therapeutic interest of trichostatin A, nuclear factor-κB, the glucocorticosteroid receptor, and CCAAT/enhancer-binding protein in asthma exacerbations.

Abstract: Manta and devil rays (Mobulids) face several immediate threats, including incidental capture in industrial tropical tuna fisheries. As a result, efforts have emerged to avoid or mitigate Mobulid bycatch in these fisheries. However, many mitigation efforts fail to incorporate fisher expertise from the outset, potentially leading to interventions that are not viable. Here, we combine survey and focus group data to synthesize knowledge of Mobulid bycatch and mitigation ideas in Eastern Pacific Ocean purse seine fisheries. Primary obstacles for mitigating Mobulid bycatch, according to respondents, are: (1) an inability to sight Mobulids before capture, (2) the lack of specific equipment on board, and (3) the difficulty of releasing large individuals; we suggest that the latter two can be addressed by simple operational modifications. We also find that Mobulids are most likely to be sighted by fishers after capture, suggesting that this is an important time in the fishing operation for bycatch mitigation interventions that ensure Mobulids survive capture. To address this, we share creative ideas brought by fishers for avoidance of Mobulids. This study provides a model of how to incorporate stakeholder input in the design of bycatch technology in large-scale fisheries and could inform similar efforts around the world.

Melanagromyza sojae, the soybean stem fly (Zehntner) (Diptera: Agromyzidae), is a new pest in soybean and chickpea crops in South America. The objective of this study was to determine population fluctuations of M. sojae in both crops using adults caught in yellow sticky traps and by sampling injured plants. Additionally, an update of its geographical distribution in Argentina is provided. Adults of this species were collected for multiple years and during the cycles of both soybean and chickpea crops, on volunteer soybean plants, and during periods when the crops were not present. Injuries caused by M. sojae occurred throughout the growing period of chickpea and the reproductive stages of soybean. The highest infestation level was registered in chickpea crops. Regarding the distribution of M. sojae, we have reported Medicago sativa, Vigna radiata, Mellilothus sp., Helianthus annuus, Carthamus tinctorius, Heliotropium sp., Glandularia sp., and Parthenium sp. for the first time as new host plants in Argentina. The presence of M. sojae in Chaco province (Argentina) was also reported for the first time. Finally, the highest percentage of infestation registered in chickpea crops shows that M. sojae is a pest in chickpea-growing areas in Argentina where this pest is widely distributed. It is of vital importance to develop effective control methods, to research and recommend efficient management strategies and control methods for M. sojae in chickpea.

Introduction: Congenital diaphragmatic hernia (CDH) remains a therapeutic challenge. The surgical classification recommended by the Congenital Diaphragmatic Hernia study group (CDHSG), based on the size of the defect, is used for staging in reference centres. Larger defects are associated with poorer outcomes. Our aim was to describe and compare the morbidity at hospital discharge of newborns who underwent surgical correction of CDH at the Juan P. Garrahan, according to the surgical staging of the defect proposed by the CDHSG. Material and methods: The study included patients with CDH admitted to the Juan P. Garrahan Hospital between 2012 and 2020, and we analysed the distribution, morbidity and mortality associated with the size of the defect. We carried out a descriptive analysis, calculating measures of central tendency and dispersion, and bivariate and multivariate analyses. Results: A total of 230 patients with CDH were admitted and 158 underwent surgery. Objectives To validate and test the generalisability of the SASKit-ML pipeline, a prepublished feature selection and machine learning pipeline for the prediction of health deterioration after a stroke or pancreatic adenocarcinoma event, by using it to identify biomarkers of health deterioration in chronic disease.Design This is a validation study using a predefined protocol applied to multiple publicly available datasets, including longitudinal data from cohorts with type 2 diabetes (T2D), inflammatory bowel disease (IBD), rheumatoid arthritis (RA) and various cancers. The datasets were chosen to mimic as closely as possible the SASKit cohort, a prospective, longitudinal cohort study.Data sources Public data were used from the T2D (77 patients with potential pre-diabetes and 18 controls) and IBD (49 patients with IBD and 12 controls) branches of the Human Microbiome Project (HMP), RA Map (RA-MAP, 92 patients with RA, 22 controls) and The Cancer Genome Atlas (TCGA, 16 cancers).Methods Data integration steps were performed in accordance with the prepublished study protocol, generating features to predict disease outcomes using 10-fold cross-validated random survival forests.Outcome measures Health deterioration was assessed using disease-specific clinical markers and endpoints across different cohorts. In the HMP-T2D cohort, the worsening of glycated haemoglobin (HbA1c) levels (5.7% or more HbA1c in the blood), fasting plasma glucose (at least 100 mg/dL) and oral glucose tolerance test (at least 140) results were considered. For the HMP-IBD cohort, a worsening by at least 3 points of a disease-specific severity measure, the Simple Clinical Colitis Activity Index or Harvey-Bradshaw Index indicated an event. For the RA-MAP cohort, the outcome was defined as the worsening of the Disease Activity Score 28 or Simple Disease Activity Index by at least five points, or the worsening of the Health Assessment Questionnaire score or an increase in the number of swollen/tender joints were evaluated. Finally, the outcome for all TCGA datasets was the progression-free interval.Results Models for the prediction of health deterioration in T2D, IBD, RA and 16 cancers were produced. The T2D (C-index of 0.633 and Integrated Brier Score (IBS) of 0.107) and the RA (C-index of 0.654 and IBS of 0.150) models were modestly predictive. The IBD model was uninformative. TCGA models tended towards modest predictive power.Conclusions The SASKit-ML pipeline produces informative and useful features with the power to predict health deterioration in a variety of diseases and cancers however, this performance is disease-dependent.

Cat domestication likely initiated as a symbiotic relationship between wildcats (Felis silvestris subspecies) and the peoples of developing agrarian societies in the Fertile Crescent. As humans transitioned from hunter-gatherers to farmers ~12,000 years ago, bold wildcats likely capitalized on increased prey density (i.e., rodents). Humans benefited from the cats' predation on these vermin. To refine the site(s) of cat domestication, over 1000 random-bred cats of primarily Eurasian descent were genotyped for single-nucleotide variants and short tandem repeats. The overall cat population structure suggested a single worldwide population with significant isolation by the distance of peripheral subpopulations. The cat population heterozygosity decreased as genetic distance from the proposed cat progenitor's (F.s. lybica) natural habitat increased. Domestic cat origins are focused in the eastern Mediterranean Basin, spreading to nearby islands, and southernly via the Levantine coast into the Nile Valley. Cat population diversity supports the migration patterns of humans and other symbiotic species.

Studies of the impact of nitrification inhibitors (NIs), specifically DMPP and DMPSA, on N2O emissions during “hot moments” have produced conflicting results regarding their effectiveness after rewetting. This study aimed to clarify the effectiveness of NIs in reducing N2O emissions by assessing residual DMP concentration and its influence on ammonia-oxidizing bacteria (AOB) in two pot experiments using calcareous (Soil C, Calcic Haploxerept) and acidic soils (Soil A, Dystric Xerochrepts). Fertilizer treatments included urea (U), DMPP, and DMPSA. The experiments were divided into Phase I (water application to dry period, 44 days) and Phase II (rewetting from days 101 to 121). In both phases for Soil C, total N2O emissions were reduced by 88% and 90% for DMPP and DMPSA, respectively, compared with U alone. While in Phase I, the efficacy of NIs was linked to the regulation of AOB populations, in Phase II this group was not affected by NIs, suggesting that nitrification may not be the predominant process after rewetting. In Soil A, higher concentrations of DMP from DMPP were maintained compared to Soil C at the end of each phase. Despite this, NIs had no significant effect due to low nitrification rates and limited amoA gene abundance, indicating unfavorable conditions for nitrifiers. The study highlights the need to optimize NIs to reduce N2O emissions and improve nitrogen efficiency, while understanding their interactions with the soil. This knowledge is necessary in order to design fertilization strategies that improve the sustainability of agriculture under climate change.

Lignocellulosic biochar has become a research hotspot recently due to its versatility in several applications for environmental remediation. It is a sustainable material, which has been proven to offer high performance in applications such as effluent treatment (through the adsorption of organic and inorganic compounds), CO2 capture, air cleaning or silage agent. Typically, a chemical activation is applied to biochars after the thermochemical process, to increase the porosity of this solid in terms of active surface area as well as pore size and distribution, to improve its performance as adsorbent. Activation with KOH has been proved to achieve well developed pore structure and active functional groups on the biochar surface. Excellent adsorption results for the removal of different pollutants have been reported with biochars produced by the mentioned method. Focusing on a potential scale and industrialization of the lignocellulosic biochar production process, the combination of the thermochemical and chemical activation processes in one step could be interesting in terms of simplicity and time saving. In this work, activated biochars were prepared using a lignocellulosic agricultural waste (apple pruning) as precursor and KOH as a chemical activator, by two different processes: a single and a two-step route, to compare the properties of the biochars generated by both processes. The obtained materials were characterized in terms of proximal analysis, active surface area, pore size and pore distribution. Furthermore, adsorption capacity ABCs was tested and compared.

Abstract Numerous policy and international frameworks consider that “destructive fishing” hampers efforts to reach sustainability goals. Though ubiquitous, “destructive fishing” is undefined and therefore currently immeasurable. Here we propose a definition developed through expert consultation: “Destructive fishing is any fishing practice that causes irrecoverable habitat degradation, or which causes significant adverse environmental impacts, results in long‐term declines in target or nontarget species beyond biologically safe limits and has negative livelihood impacts.” We show strong stakeholder support for a definition, consensus on many biological and ecological dimensions, and no clustering of respondents from different sectors. Our consensus definition is a significant step toward defining sustainable fisheries goals and will help interpret and implement global political commitments which utilize the term “destructive fishing.” Our definition and results will help reinforce the Food and Agricultural Organization's Code of Conduct and meaningfully support member countries to prohibit destructive fishing practices.

Oceanic whitetip shark (Carcharhinus longimanus) is an important top predator in pelagic ecosystems currently classified as globally Critically Endangered by the International Union for the Conservation of Nature. This species is incidentally caught by fisheries targeting highly migratory tunas and billfishes throughout the Indian Ocean. Understanding the temporal, spatial and environmental factors influencing the capture of this species is essential to reduce incidental catches. In this study, we used generalized additive models to analyze the spatio-temporal distributions of the juvenile oceanic whitetip shark catches and the environmental conditions in the western Indian Ocean using observer data from 2010 to 2020 of the European Union and associated flags purse seine fishery. We found sea surface temperature and nitrate concentration to be the most important environmental variables predicting the probability of catching an oceanic whitetip shark. A higher probability of capture was predicted in areas where sea surface temperature was below 24°C and with low nitrate concentrations close to zero and intermediate values (1.5-2.5 mmol.m-3). We also found a higher probability of capture in sets on fish aggregating devices than in sets on free schools of tuna. The Kenya and Somalia basin was identified to have higher probabilities of capture during the summer monsoon (June to September) when upwelling of deep cold waters occurs. We provide the first prediction maps of capture probabilities and insights into the environmental preferences of oceanic whitetip shark in the western Indian Ocean. However, the causal mechanisms behind these insights should be explored in future studies before they can be used to design spatial management and conservation strategies, such as time-area closures, for bycatch avoidance.

Abstract Natural geochemical markers in the otolith of yellowfin tuna (Thunnus albacares) were used to establish nursery-specific signatures for investigating the origin of fish captured in the western Atlantic Ocean (WAO). Two classes of chemical markers (trace elements, stable isotopes) were used to first establish nursery-specific signatures of age-0 yellowfin tuna from four primary production zones in the Atlantic Ocean: Gulf of Mexico, Caribbean Sea, Cape Verde, and Gulf of Guinea. Next, mixture and individual assignment methods were applied to predict the origin of sub-adult and adult yellowfin tuna from two regions in the WAO (Gulf of Mexico, Mid Atlantic Bight) by relating otolith core signatures (corresponding to age-0 period) to baseline signatures of age-0 fish from each nursery. Significant numbers of migrants from Caribbean Sea and eastern Atlantic Ocean (EAO) production zones (Gulf of Guinea, Cape Verde) were detected in the WAO, suggesting that fisheries in this region were subsidized by outside spawning/nursery areas. Contributions from local production (Gulf of Mexico) were also evident in samples from both WAO fisheries, but highly variable from year to year. High levels of mixing by yellowfin tuna from the different production zones and pronounced interannual trends in nursery-specific contribution rates in the WAO emphasize the complex and dynamic nature of this species’ stock structure and population connectivity. Given that geographic shifts in distribution across national or political boundaries leads to governance and management challenges, this study highlights the need for temporally resolved estimates of nursery origin to refine assessment models and promote the sustainable harvest of this species.

Introducción: Casi 800 muertes de mujeres en forma diaria ocurren por causas prevenibles asociadas a complicaciones durante o después del embarazo o el parto, equivaliendo dos muertes maternas por minuto. Objetivo: Desarrollar un proceso de atención de enfermería a gestante que cursó con hipertiroidismo empleando las taxonomías diagnósticas, resultados e intervenciones de enfermería con enfoque de la teoría de Orem. Metodología: Estudio de caso a mujer gestante atendida en hospital de segundo nivel de atención, se empleó la guía de valoración de Orem desarrollando las etapas: valoración, diagnóstico, planeación, ejecución y evaluación, se diseñaron y ejecutaron planes de cuidados individualizados. Se protegió la privacidad omitiendo identidad, respetando los derechos y bienestar de la participante. Resultados: El factor condicionante de riesgo que comprometía la salud del binomio madre-hijo, dio paso a la planificación del cuidado encaminado al proceso de maternidad ineficaz y autogestión ineficaz de la salud, estableciendo intervenciones como cuidados del embarazo de alto riesgo y enseñanza del proceso de enfermedad y medicamentos prescritos, se obtuvo mejora en el estado de salud de la paciente, lo que permitió su alta y un adecuado manejo del embarazo de alto riesgo. Conclusión: El proceso de atención de enfermería expuso el contexto en el que ingresa la mujer gestante con patología conocida no controlada, lo que comprometía el bienestar materno-fetal. Se requiere educación por parte de enfermería para fomentar la salud materna preconcepcional para futuros embarazos.
