The Younger Dryas stadial was characterised by a rapid shift towards cold-climate conditions in the North Atlantic realm during the last deglaciation. While some climate parameters including atmospheric temperature and glacier extent are widely studied, empirical constraints on permafrost temperature and snow thickness are limited. To address this, we present a regional dataset of cryogenic cave carbonates (CCCs) from three caves in Great Britain that formed at temperatures between −2 and 0 °C. Our CCC record indicates that these permafrost temperatures persisted for most of the Younger Dryas. By combining ground temperatures with surface temperatures from high-resolution ground-truthed model simulations, we demonstrate that ground temperatures were approximately 6.6 ± 2.3 °C warmer than the mean annual air temperature. Our results suggest that the observed temperature offset between permafrost and the atmosphere can be explained by an average snow thickness between 0.2 and 0.9 m, which persisted for 233 ± 54 d per year. By identifying modern analogues from climate reanalysis data, we demonstrate that the inferred temperature and snow cover characteristics for the British Isles during the Younger Dryas are best explained by extreme temperature seasonality, comparable to continental parts of today's Arctic Archipelago. Such a climate for the British Isles necessitates a winter sea ice margin at approximately 45° N in the North Atlantic Ocean.

IntroductionMicroalgae provide a sustainable basis for protein-rich food production. However, human data concerning microalgae ingestion, subsequent postprandial amino acid (AA) availability and associated metabolic responses are minimal.ObjectivesWe investigated ingesting Arthrospira sp. (spirulina;SPR), and Chlorella sp. (chlorella; in ‘whole cell’ [WCC] and ‘split cell’ [SCC] forms, the latter proposed to improve digestibility), compared with a high-quality animal derived protein source (milk; MLK).Subjects/methodsTen participants (age; 21 ± 1y, BMI; 25 ± 1 kg·m−2) completed a randomised, crossover, double-blind study, partaking in 4 counterbalanced (for order) experimental visits. At each visit participants ingested SPR, WCC, SCC or MLK drinks containing 20 g protein and 75 g carbohydrate. Arterialised venous blood samples, indirect calorimetry and visual analogue scales were assessed postabsorptive, and throughout a 5 h postprandial period to measure AA, glucose, insulin and uric acid concentrations, whole-body energy expenditure and appetite scores, respectively.ResultsProtein ingestion increased plasma AA concentrations (p 0.05) were detected between conditions for postprandial glucose or insulin concentrations, whole-body energy expenditure or appetite scores, but serum uric acid concentrations increased (p < 0.05) following microalgae ingestion only.ConclusionOur data imply that microalgae can present a bioavailable source of protein for human nutrition, however, challenges remain, requiring species selection and/or biomass processing to overcome.

BackgroundAutoimmune encephalitis (AIE) comprises a group of rare, immune system-mediated conditions. Clinical manifestations among children are not well-characterized, and there are challenges in testing and diagnosis. This can result in treatment delays, which has been found to correlate with poorer long-term outcomes. This challenge is exacerbated by the scarcity of epidemiological reporting of AIE. The objective of this systematic literature review (SLR) was to identify studies reporting epidemiological data on AIE in children.MethodsMEDLINE, Embase, the Cochrane Library, and the University of York Centre for Reviews and Dissemination (CRD) were searched in May 2023 for studies reporting on the epidemiology of AIE in children. These were supplemented with additional searches of conference proceedings, gray literature, and the reference lists of identified SLRs. Quality of studies was assessed using a modified version of the Joanna Briggs Institute (JBI) Checklist for Prevalence Studies.ResultsForty-three publications reporting on 41 unique studies were included. Nine studies reported incidence estimates of different subtypes of AIE, with only one reporting the incidence of overall AIE in children ≤ 18 years, estimated at 1.54 per million children per year in the Netherlands. Three studies reported the incidence of pediatric N-methyl-D-aspartate receptor (NMDAR)-AIE [in United Kingdom (UK), Hong Kong, and Denmark]. The other studies reported incidence data for selected populations.ConclusionThis SLR highlights a paucity of epidemiology data for AIE in children, which is likely reflective of difficulties in testing and diagnosis. There is a clear need for further research and awareness of these challenges in clinical practice to avoid treatment delays and improve patient outcomes. A deeper understanding of the epidemiology of AIE will help determine the worldwide burden of disease and inform research, health policies and clinical decision-making.

Abstract Studies of extreme endurance have suggested that there is an alimentary limit to energy intake (EI) of ∼2.5 × resting metabolic rate (RMR). To gain further insight, this study aimed to simultaneously measure EI, total energy expenditure (TEE) body mass and muscle mass in a large cohort of males and females of varying ages during a transatlantic rowing race. Forty‐nine competitors (m = 32, f = 17; age 24–67 years; time at sea 46 ± 7 days) in the 2020 and 2021 Talisker Whisky Atlantic Challenge rowed 12–18 hday−1 for ∼3000 miles. Monosaccharides such as L-arabinose (Ara) and D-tagatose (derived from D-galactose, Gal) are low-calorie sweeteners associated with improved glycaemic and insulin control compared to disaccharides such as sucrose. However, alternative sources and better sugar-sugar separation methods are needed to improve the sustainability and economics of their production. Here, these sugars were obtained from purified and ultrafiltered compression screw pressate (CSP) of thermo-mechanical pulping of softwood (Pinus radiata) and orange peels (OPs). Ba-substituted zeolite X (BaX) molecular sieves showed superior separation performance of Ara from other sugars compared to conventional Ca-form ion exchange resin. To facilitate subsequent separation of sugars, OP hydrolysates were fermented to leave just Ara and Gal, while OP pectin was hydrolysed to generate a Gal-rich mixture. Overall, BaX has good potential for separating Ara from Ara-rich hydrolysates containing several different sugars. It is also suited for separating Ara and another monosaccharide such as Gal or Xyl in the absence of other sugars.

9. Red clays indicate sub-aerial exposure of the Rio Grande Rise during the Eocene volcanic episode

Abstract Autonomous underwater vehicle (AUV) mapping of the western Rio Grande Rise (RGR), South Atlantic, and subsequent exploration and photography of horizontal lava flows exposed in near vertical, faulted escarpments, showed occurrences of red clays/weathered volcanic tops trapped between successive alkaline lava flows. These red clays indicate a hiatus in successive volcanic eruptions. Here, we report detailed mineralogical, geochemical, and rock magnetic characteristics of one such distinct red clay dredged from ~ 650 m water depth in the western RGR. The mineral constituents of the red clay are kaolinite, magnetite, oxidized magnetite (/maghemite), hematite, and goethite, with biogenic calcite and halite occupying voids or precipitated on the surface of the red clay. The chemical index of alteration (CIA) has a value of 93, showing that red clay is a product of extreme chemical weathering of the lava flows. The alkaline volcanic rocks recovered from nearby show an age of ~ 44 Ma, indicating an Eocene age for the volcanism. We show that the red clays are a product of sub-aerial chemical weathering of these Eocene volcanic rocks, in a warm-wet climate, before the thermal subsidence of the RGR to its modern-day bathymetric depth.

Abstract Background The decline in postabsorptive and postprandial muscle protein fractional synthesis rates (FSR) does not quantitatively account for muscle atrophy during uncomplicated, short‐term disuse, when atrophy rates are the highest. We sought to determine whether 2 days of unilateral knee immobilization affects mixed muscle protein fractional breakdown rates (FBR) during postabsorptive and simulated postprandial conditions. Methods Twenty‐three healthy, male participants (age: 22 ± 1 year; height: 179 ± 1 cm; body mass: 73.4 ± 1.5 kg; body mass index 22.8 ± 0.5 kg·m−2) took part in this randomized, controlled study. After 48 h of unilateral knee immobilization, primed continuous intravenous l‐[15N]‐phenylalanine and l‐[ring‐2H5]‐phenylalanine infusions were used for parallel determinations of FBR and FSR, respectively, in a postabsorptive (saline infusion; FAST) or simulated postprandial state (67.5 mg·kg body mass−1·h−1 amino acid infusion; FED). Bilateral m. vastus lateralis biopsies from the control (CON) and immobilized (IMM) legs, and arterialized‐venous blood samples, were collected throughout. Abstract Metabolic stable isotope labeling with heavy water followed by liquid chromatography coupled with mass spectrometry (LC-MS) is a powerful tool for in vivo protein turnover studies. Several algorithms and tools have been developed to determine the turnover rates of peptides and proteins from time-course stable isotope labeling experiments. The availability of benchmark mass spectrometry data is crucial to compare and validate the effectiveness of newly developed techniques and algorithms. In this work, we report a heavy water-labeled LC-MS dataset from the murine liver for protein turnover rate analysis. The dataset contains eighteen mass spectral data with their corresponding database search results from nine different labeling durations and quantification outputs from d2ome+ software. The dataset also contains eight mass spectral data from two-dimensional fractionation experiments on unlabeled samples.

15. Validating the efficacy and value proposition of mental fitness vocal biomarkers in a psychiatric population: prospective cohort study

BackgroundThe utility of vocal biomarkers for mental health assessment has gained increasing attention. This study aims to further this line of research by introducing a novel vocal scoring system designed to provide mental fitness tracking insights to users in real-world settings.MethodsA prospective cohort study with 104 outpatient psychiatric participants was conducted to validate the “Mental Fitness Vocal Biomarker” (MFVB) score. The MFVB score was derived from eight vocal features, selected based on literature review. Participants’ mental health symptom severity was assessed using the M3 Checklist, which serves as a transdiagnostic tool for measuring depression, anxiety, post-traumatic stress disorder, and bipolar symptoms.ResultsThe MFVB demonstrated an ability to stratify individuals by their risk of elevated mental health symptom severity. Continuous observation enhanced the MFVB’s efficacy, with risk ratios improving from 1.53 (1.09-2.14, p=0.0138) for single 30-second voice samples to 2.00 (1.21-3.30, p=0.0068) for data aggregated over two weeks. A higher risk ratio of 8.50 (2.31-31.25, p=0.0013) was observed in participants who used the MFVB 5-6 times per week, underscoring the utility of frequent and continuous observation. Participant feedback confirmed the user-friendliness of the application and its perceived benefits.ConclusionsThe MFVB is a promising tool for objective mental health tracking in real-world conditions, with potential to be a cost-effective, scalable, and privacy-preserving adjunct to traditional psychiatric assessments. User feedback suggests that vocal biomarkers can offer personalized insights and support clinical therapy and other beneficial activities that are associated with improved mental health risks and outcomes.

Abstract Heavy water metabolic labeling followed by liquid chromatography coupled with mass spectrometry is a powerful high throughput technique for measuring the turnover rates of individual proteins in vivo. The turnover rate is obtained from the exponential decay modeling of the depletion of the monoisotopic relative isotope abundance. We provide theoretical formulas for the time course dynamics of six mass isotopomers and use the formulas to introduce a method that utilizes partial isotope profiles, only two mass isotopomers, to compute protein turnover rate. The use of partial isotope profiles alleviates the interferences from co-eluting contaminants in complex proteome mixtures and improves the accuracy of the estimation of label enrichment. In five different datasets, the technique consistently doubles the number of peptides with high goodness-of-fit characteristics of the turnover rate model. We also introduce a software tool, d2ome+, which automates the protein turnover estimation from partial isotope profiles.

The Chinese government promotes the Belt and Road Initiative (BRI) as a global strategy for regional integration and infrastructure investment. With a projected US$1 trillion commitment from Chinese financial institutions, and at least 138 countries participating, the BRI is attracting intense debate. Yet most analysis to date focuses on broad drivers, risks, and opportunities, largely considered to be emanating from a coherent policy imposed by Beijing. In this special issue, we instead examine the BRI as a relational, contested process - a bundle of intertwined discourses, policies, and projects that sometimes align but are sometimes contradictory. We move beyond policy-level, macro-economic, and classic geopolitical analysis to study China's global investments "from the ground". Our case studies reveal the BRI to be dynamic and unstable, rhetorically appropriated for different purposes that sometimes but do not always coalesce as a coherent geopolitical and geoeconomic strategy. The papers in this special issue provide one of the first collections of deep empirical work on the BRI and a useful approach for grounding China's role in globalization in the critical contexts of complex local realities.

21. Evaluation of freezing state of sandstone using ultrasonic time-frequency characteristics

Common problems in engineering projects that involve artificial ground freezing of soil or rock include inadequate thickness, strength and continuity of artificial frozen walls. It is difficult to evaluate the freezing state using only a few thermometer holes at fixed positions or with other existing approaches. Here we report a novel experimental design that investigates changes in ultrasonic properties (received waveform, wave velocity Vp, wave amplitude, frequency spectrum, centroid frequency fc, kurtosis of the frequency spectrum KFS, and quality factor Q) measured during upward freezing, compared with those during uniform freezing, in order to determine the freezing state in 150 mm cubic blocks of Ardingly sandstone. Water content, porosity and density were estimated during upward freezing to ascertain water migration and changes of porosity and density at different stages. The period of receiving the wave increased substantially and coda waves changed from loose to compact during both upward and uniform freezing. The trend of increasing Vp can be divided into three stages during uniform freezing. During upward freezing, Vp increased more or less uniformly. The frequency spectrum could be used as a convenient and rapid method to identify different freezing states of sandstone (unfrozen, upward frozen, and uniformly frozen). The continuous changes in reflection coefficient rφ, refraction coefficient tφ and acoustic impedance field are the major reason for larger reflection and refraction during upward freezing compared with uniform freezing. Wave velocity Vp, wave amplitude Ah, centroid frequency fc and quality factor Q were adopted as ultrasonic parameters to evaluate quantitatively the temperature T of uniformly frozen sandstone, and their application within a radar chart is recommended. Determination of Vp provides a convenient method to evaluate the freezing state and calculate the cryofront height and frozen section thickness of upward frozen sandstone, with accuracies of 73.37%–99.23%.

Abstract Pronounced glacial and interglacial climate cycles characterized northern ecosystems during the Pleistocene. Our understanding of the resultant community transformations and past ecological interactions strongly depends on the taxa found in fossil assemblages. Here, we present a shotgun metagenomic analysis of sedimentary ancient DNA (sedaDNA) to infer past ecosystem‐wide biotic composition (from viruses to megaherbivores) from the Middle and Late Pleistocene at the Batagay megaslump, East Siberia. The shotgun DNA records of past vegetation composition largely agree with pollen and plant metabarcoding data from the same samples. Interglacial ecosystems at Batagay attributed to Marine Isotope Stage (MIS) 17 and MIS 7 were characterized by forested vegetation (Pinus, Betula, Alnus) and open grassland. The microbial and fungal communities indicate strong activity related to soil decomposition, especially during MIS17. The local landscape likely featured more open, herb‐dominated areas, and the vegetation mosaic supported birds and small omnivorous mammals. Parts of the area were intermittently/partially flooded as suggested by the presence of water‐dependent taxa. During MIS 3, the sampled ecosystems are identified as cold‐temperate, periodically flooded grassland. Diverse megafauna (Mammuthus, Equus, Coelodonta) coexisted with small mammals (rodents). The MIS 2 ecosystems existed under harsher conditions, as suggested by the presence of cold‐adapted herbaceous taxa. Typical Pleistocene megafauna still inhabited the area. The new approach, in which shotgun sequencing is supported by metabarcoding and pollen data, enables the investigation of community composition changes across a broad range of taxonomic groups and inferences about trophic interactions and aspects of soil microbial ecology.

ABSTRACT Mycobacterium tuberculosis whole-genome sequencing (WGS) is a powerful tool as it can provide data on population diversity, drug resistance, disease transmission, and mixed infections. Successful WGS is still reliant on high concentrations of DNA obtained through M. tuberculosis culture. Microfluidics technology plays a valuable role in single-cell research but has not yet been assessed as a bacterial enrichment strategy for culture-free WGS of M. tuberculosis. In a proof-of-principle study, we evaluated the use of Capture-XT, a microfluidic lab-on-chip cleanup and pathogen concentration platform to enrich M. tuberculosis bacilli from clinical sputum specimens for downstream DNA extraction and WGS. Three of the four (75%) samples processed by the microfluidics application passed the library preparation quality control, compared to only one of the four (25%) samples not enriched by the microfluidics M. tuberculosis capture application. WGS data were of sufficient quality, with mapping depth of ≥25× and 9 to 27% of reads mapping to the reference genome. These results suggest that microfluidics-based M. tuberculosis cell capture might be a promising method for M. tuberculosis enrichment in clinical sputum samples, which could facilitate culture-free M. tuberculosis WGS. IMPORTANCE Diagnosis of tuberculosis is effective using molecular methods; however, a comprehensive characterization of the resistance profile of Mycobacterium tuberculosis often requires culturing and phenotypic drug susceptibility testing or culturing followed by whole-genome sequencing (WGS). The phenotypic route can take anywhere from 1 to >3 months to result, by which point the patient may have acquired additional drug resistance. The WGS route is a very attractive option; however, culturing is the rate-limiting step. In this original article, we provide proof-of-principle evidence that microfluidics-based cell capture can be used on high-bacillary-load clinical samples for culture-free WGS.

Background Many individuals hospitalised with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection experience post-acute sequelae of SARS-CoV-2 infection (PASC), sometimes referred to as “long COVID”. Our objective was to conduct a systematic literature review and meta-analysis to identify PASC-associated symptoms in previously hospitalised patients and determine the frequency and temporal nature of PASC. Methods Searches of MEDLINE, Embase, Cochrane Library (2019–2021), World Health Organization International Clinical Trials Registry Platform and reference lists were performed from November to December 2021. Articles were assessed by two reviewers against eligibility criteria and a risk of bias tool. Symptom data were synthesised by random effects meta-analyses. Results Of 6942 records, 52 studies with at least 100 patients were analysed; ∼70% were Europe-based studies. Most data were from the first wave of the pandemic. PASC symptoms were analysed from 28 days after hospital discharge. At 1–4 months post-acute SARS-CoV-2 infection, the most frequent individual symptoms were fatigue (29.3% (95% CI 20.1–40.6%)) and dyspnoea (19.6% (95% CI 12.8–28.7%)). Many patients experienced at least one symptom at 4–8 months (73.1% (95% CI 44.2–90.3%)) and 8–12 months (75.0% (95% CI 56.4–87.4%)). Conclusions A wide spectrum of persistent PASC-associated symptoms were reported over the 1-year follow-up period in a significant proportion of participants. Further research is needed to better define PASC duration and determine whether factors such as disease severity, vaccination and treatments have an impact on PASC.

Abstract At the Trans‐Atlantic Geotraverse hydrothermal field, metalliferous sediments cover extinct hydrothermal mounds and the surrounding seafloor. Here, we report the morphological, mineralogical and geochemical processes that deposit these sediments, remobilize their metals, and affect their preservation. We found that the initial sediment metal tenor is controlled by physical transport of hydrothermal material from its source, followed by diagenetic redistribution and potentially diffuse fluid flow after high‐temperature hydrothermal activity has ceased. We distinguished three different environments: (a) proximal metalliferous sediments on top of extinct mounds are mainly derived from oxidative weathering of primary sulfide structures and are predominantly composed of Fe oxyhydroxides with low contents of Cu, Co, and Zn; metal enrichments in specific layers are likely related to upward flow of low‐temperature hydrothermal fluids; (b) medial distant metalliferous sediments found at the base of the mounds, deposited by mass transport, contain cm‐thick layers of unsorted sulfide sands with high base metal contents (e.g., up to 28% Cu); these buried sulfides continue to undergo dissolution, resulting in metal release into porewaters; (c) distal metalliferous sediments, found in depositional basins a few hundreds of meters from the extinct mounds, include fining‐upwards sequences of thin sulfide sand layers with Fe oxyhydroxides and were deposited by recurrent turbiditic flows. Dissolved metals (e.g., Cu2+ and Mn2+) diffuse upwards under reducing conditions and precipitate within the sediment. Hence, when using hydrothermal sediments to construct reliable geochronological records of hydrothermal activity, distance from source, local seafloor morphology, mass‐transport and depositional, and diagenetic modification should all be considered.

Background: The coronavirus disease-2019 (COVID-19) pandemic significantly affected management of cardiovascular disease around the world. The effect of the pandemic on volume of cardiovascular diagnostic procedures is not known. Objectives: This study sought to evaluate the effects of the early phase of the COVID-19 pandemic on cardiovascular diagnostic procedures and safety practices in Asia. Methods: The International Atomic Energy Agency conducted a worldwide survey to assess changes in cardiovascular procedure volume and safety practices caused by COVID-19. Testing volumes were reported for March 2020 and April 2020 and were compared to those from March 2019. Data from 180 centers across 33 Asian countries were grouped into 4 subregions for comparison. Results: Procedure volumes decreased by 47% from March 2019 to March 2020, showing recovery from March 2020 to April 2020 in Eastern Asia, particularly in China. The majority of centers cancelled outpatient activities and increased time per study. Practice changes included implementing physical distancing and restricting visitors. Although COVID testing was not commonly performed, it was conducted in one-third of facilities in Eastern Asia. The most severe reductions in procedure volumes were observed in lower-income countries, where volumes decreased 81% from March 2019 to April 2020. Conclusions: The COVID-19 pandemic in Asia caused significant reductions in cardiovascular diagnostic procedures, particularly in low-income countries. Further studies on effects of COVID-19 on cardiovascular outcomes and changes in care delivery are warranted.

ABSTRACT INTRODUCTIONIn recent years, there has been growing acknowledgment of problematic unprofessional behaviours such as bullying in the health workforce. AIMThe purpose of this research is to investigate how bullying manifests in general practice, responses to bullying and the impact of this behaviour. METHODSQualitative research using semi-structured interviews or focus groups was conducted. Twenty-four doctors working in the scope of general practice in New Zealand participated: 21 by interview and three by focus group. Participants had been accused of bullying or subject to bullying. RESULTSResults are reported in three categories: people who have experienced being bullied, people accused of bullying, and the cycle of bullying. Participants experienced a range of negative behaviours and had varied responses to bullying, yet the impact of the behaviour was the same. There was considerable overlap in experiences of participants experiencing and accused of bullying, and these categories were not static. DISCUSSIONBullying has a substantial negative impact on the general practice workforce. Responses to bullying often fail to consider the complexities of the behaviour and are premised on simple notions of bully and victim, failing to consider the wider work environment.

The Wnt signalling pathway plays key roles in cell proliferation, differentiation and fate decisions in embryonic development and maintenance of adult tissues, and the twelve Armadillo (ARM) repeat-containing protein β-catenin acts as the signal transducer in this pathway. Here we investigate the interaction between β-catenin’s ARM repeat domain and the intrinsically disordered protein adenomatous polyposis coli (APC). APC is a giant multivalent scaffold that brings together the different components of the so-called “β-catenin destruction complex”, which drives β-catenin degradation via the ubiquitin-proteasome pathway. Mutations and truncations in APC, resulting in loss of APC function and hence elevated β-catenin levels and upregulation of Wnt signalling, are associated with numerous cancers including colorectal carcinomas. APC has a long intrinsically disordered region (IDR) that contains a series of 15-residue and 20-residue binding regions for β-catenin. Here we explore the multivalent nature of the interaction of β-catenin with the highest affinity APC repeat, both at equilibrium and under kinetic conditions. We use a combination of single-site substitutions, deletions and insertions to dissect the mechanism of molecular recognition and the roles of the three β-catenin-binding subdomains of APC.

Glaciation during the late Middle Pleistocene is widely recognized across continental northwest Europe, but its extent and palaeoenvironmental significance in the British Isles are disputed. Although glaciogenic sediments at Wolston, Warwickshire, in the English West Midlands, have been used to define the stratotype of the Wolstonian Stage, their age has been variably assigned between marine isotope stages (MIS) 12 and 6. Here we present sedimentological and stratigraphical observations from five sites across the English West Midlands whose chronology is constrained by new luminescence ages from glaciofluvial sediments, supplemented by cosmogenic 36Cl exposure dating of erratic boulders. The ages suggest that between 199 ± 5 and 147 ± 2.5 ka the British Ice Sheet advanced into the English West Midlands as far south as Moreton-in-Marsh, Gloucestershire. This advance is assigned to the Moreton Stadial of the Late Wolstonian Substage. Dating of the glaciation to this substage allows correlation of the Moreton Stadial glacial deposits in the English West Midlands with those of the Drenthe Stadial during the Late Saalian Substage across continental northwest Europe.

Summary: A recent report on screening in the UK proposed that the responsibility for recommendations on population and targeted screening programmes should be held by one new integrated advisory body. There is no wide international consensus on the definition of targeted screening. Our review identified and compared the defining components of screening terms: targeted, population, selective, and cascade screening, and case finding. Definitions of targeted screening and population screening were clearly demarcated by the eligible population; targeted and selective screening were found to be conceptually interchangeable; cascade screening, whilst conceptually similar to targeted screening across several components, was only used within the context of genetic diseases. There was little consensus between different definitions of case finding. These comparisons contributed to an updated definition of targeted screening. Considerable overlap between definition components across terms implies that a broad range of disease areas may fall into the remit of the new advisory body.

Automated monitoring of freeze-thaw cycles and fracture propagation in mountain rockwalls is needed to provide early warning about rockfall hazards. Conventional geoelectrical methods such as electrical resistivity tomography (ERT) are limited by large and variable ohmic contact resistances, requiring galvanic coupling with metal electrodes inserted into holes drilled into rock, and which can be loosened by rock weathering. We report a novel experimental methodology that combined capacitive resistivity imaging (CRI), ERT, and microseismic event recording to monitor freeze-thaw of six blocks of hard and soft limestones under conditions simulating an active layer above permafrost and seasonally frozen rock in a nonpermafrost environment. Our results demonstrate that the CRI method is highly sensitive to freeze-thaw processes; it yields property information equivalent to that obtained with conventional ERT and offers a viable route for nongalvanic long-term geoelectrical monitoring, extending the benefits of the methodology to soft/hard rock environments. Contact impedances achieved with CRI are less affected by seasonal temperature changes, the aggregate state of the pore water (liquid or frozen), and the presence of low-porosity rock with high matrix resistivities than those achieved with ERT. Microseismic monitoring has the advantage over acoustic emissions that events were recorded in relevant field distances of meters to decameters from cracking events. For the first time we recorded about 1000 microcracking events and clustered them in four groups according to frequency and waveform. Compared to previous studies, mainly on ice-cracking in glaciers, the groups are attributed to single- or multiple-stage cracking events such as crack coalescence.

ABSTRACT INTRODUCTIONMass masking is emerging as a key non-pharmaceutical intervention for reducing community spread of COVID-19. However, although hand washing, social distancing and bubble living have been widely adopted by the ‘team of 5 million', mass masking has not been socialised to the general population. AIMTo identify factors associated with face masking in New Zealand during COVID-19 Alert Level 4 lockdown to inform strategies to socialise and support mass masking. METHODSA quantitative online survey conducted in New Zealand during April 2020 invited residents aged ≥18 years to complete a questionnaire. Questions about face masking were included in the survey. The sample was drawn from a commissioned research panel survey, with boosted sampling for Māori and Pacific participants. Responses were weighted to reflect the New Zealand population for all analyses. RESULTSA total of 1015 individuals participated. Self-reported beliefs were strongly related to behaviours, with respondents viewing face masking measures as ‘somewhat' or ‘very' effective in preventing them from contracting COVID-19 more likely to report having worn a face mask than respondents who viewed them as ‘not at all' effective. The strongest barriers to face mask use included beliefs that there was a mask shortage and that the needs of others were greater than their own. DISCUSSIONHighlighting the efficacy of and dispelling myths about the relative efficacy of mask types and socialising people to the purpose of mass masking will contribute to community protective actions of mask wearing in the New Zealand response to COVID-19.

This study investigates acoustic voice and speech features as biomarkers for acute decompensated heart failure (ADHF), a serious escalation of heart failure symptoms including breathlessness and fatigue. ADHF-related systemic fluid accumulation in the lungs and laryngeal tissues is hypothesized to affect phonation and respiration for speech. A set of daily spoken recordings from 52 patients undergoing inpatient ADHF treatment was analyzed to identify voice and speech biomarkers for ADHF and to examine the trajectory of biomarkers during treatment. Results indicated that speakers produce more stable phonation, a more creaky voice, faster speech rates, and longer phrases after ADHF treatment compared to their pre-treatment voices. This project builds on work to develop a method of monitoring ADHF using speech biomarkers and presents a more detailed understanding of relevant voice and speech features.

49. Particle Image Velocimetry Evaluation of Hemodynamics Proximal to the Kissing Stent Configuration in the Aorto-Iliac Bifurcation

Purpose: The kissing stent (KS) method is low-risk compared with open surgery techniques. It is often used to treat aorto-iliac occlusive disease (AIOD). Deployment of the KS geometry has a high technical success rate. However, stent patency reduces in the first 5 years potentially due to deleterious flow behavior. Potentially harmful hemodynamics due to the KS were investigated in vitro.Methodology: A compliant phantom of the aorto-iliac bifurcation was manufactured. Two surrogate stent-grafts were deployed into the phantom in the KS configuration to investigate effects of the presence of the stents, including the compliance mismatch they cause, on the hemodynamics proximal and distal to the KS. The investigation used pulsatile flow through a flow circuit to simulate abdominal aortic flow. Particle image velocimetry (PIV) was used to quantify the hemodynamics.Results: PIV identified peak proximal and distal velocity in vitrowas 0.71 and 1.40m·s−1, respectively, which were within physiological ranges. Throughout systole, flow appeared normal and undisturbed. A lumen wall collapse in the sagittal plane formed during late systole and continued to early diastole proximal to the aorto-iliac bifurcation, distal to the inlet stent position. The wall collapse led to disturbed flow proximal to the stented region in early diastole producing potential recirculation zones and abnormal flow patterns.Conclusion: The normal systolic flow behavior indicates the KS configuration is unlikely to cause an inflammatory response of the arterial walls. The collapse has not been previously identified and may potentially cause long-term patency reduction. It requires further investigation.Clinical Impact The role of this article is to provide further insight into the haemodynamic behavior through a stented aorto-iliac artery. The results of this investigation will improve the understanding of the effects that using the kissing stent method may have on a patient and help to identify high risk regions that may require more detailed monitoring. This paper also develops the in vitro modelling techniques that will enable further research that cannot be carried out within patients.

While obesity blunts the ability of muscle to mount a protein synthetic response to an amino acid infusion in older adults, it is unclear if this insensitivity to nutrition persists long term and in response to complete foods is unknown. To address this, young (2 months old) and old (17–20 months old) Brown Norway rats were randomized to receive either chow or a 12 wk diet of calorie-dense human foods. At wk 10 drinking water was supplemented with 2% heavy water, followed 2 weeks later by a flooding dose of [2H5]-phenylalanine and an oral leucine bolus, allowing the short and long-term effects of age and diet on muscle protein synthesis rates to be determined. The experimental diet increased energy intake in both young (7.4 ± 0.9%) and old (18.2 ± 1.8%) animals (P < 0.01), but only led to significant increases in body weight in the former (young: 10.2 ± 3.0% (P < 0.05) and old: 3.1 ± 5.1% (NS) vs. age-matched controls). Notably, energy expenditure in response to the cafeteria diet was increased in old animals only (chow: 5.1 ± 0.4; cafe: 8.2 ± 1.6 kcal.kg b.w−1.h−1; P < 0.05). Gastrocnemius protein fractional synthetic rates in response to either an acute leucine bolus or two weeks of feeding were equivalent across groups irrespective of age or diet. Rodents in old age appear capable of preventing weight gain in response to a calorie-dense diet by increasing energy expenditure while maintaining the anabolic sensitivity of muscle to nutrition; the mechanisms of which could have important implications for the aging obese human.
