Remote ischemic preconditioning (RIPC) can evoke cardioprotection following ischemia/reperfusion and this may depend on the anesthetic used. We tested whether 1) extracellular vesicles (EVs) isolated from humans undergoing RIPC protect cardiomyoblasts against hypoxia-induced apoptosis and 2) this effect is altered by cardiomyoblast exposure to isoflurane or propofol. EVs were isolated before and 60 min after RIPC or Sham from ten patients undergoing coronary artery bypass graft surgery with isoflurane anesthesia and quantified by Nanoparticle Tracking Analysis. Following EV-treatment for 6 hours under exposure of isoflurane or propofol, rat H9c2 cardiomyoblasts were cultured for 18 hours in normoxic or hypoxic atmospheres. Apoptosis was detected by flow cytometry. Serum nanoparticle concentrations in patients had increased sixty minutes after RIPC compared to Sham (2.5x\(10^{11}\)\(\pm\)4.9x\(10^{10}\) nanoparticles/ml; Sham: 1.2x\(10^{11}\)\(\pm\)2.0x\(10^{10}\); p = 0.04). Extracellular vesicles (EVs) mediate targeted cellular interactions in normal and pathophysiological conditions and are increasingly recognised as potential biomarkers, therapeutic agents and drug delivery vehicles. Based on their size and biogenesis, EVs are classified as exosomes, microvesicles and apoptotic bodies. Due to overlapping size ranges and the lack of specific markers, these classes cannot yet be distinguished experimentally. Currently, it is a major challenge in the field to define robust and sensitive technological platforms being suitable to resolve EV heterogeneity, especially for small EVs (sEVs) with diameters below 200 nm, i.e. smaller microvesicles and exosomes. Most conventional flow cytometers are not suitable for the detection of particles being smaller than 300 nm, and the poor availability of defined reference materials hampers the validation of sEV analysis protocols. Following initial reports that imaging flow cytometry (IFCM) can be used for the characterisation of larger EVs, we aimed to investigate its usability for the characterisation of sEVs. This study set out to identify optimal sample preparation and instrument settings that would demonstrate the utility of this technology for the detection of single sEVs. By using CD63eGFP-labelled sEVs as a biological reference material, we were able to define and optimise IFCM acquisition and analysis parameters on an Amnis ImageStreamX MkII instrument for the detection of single sEVs. In addition, using antibody-labelling approaches, we show that IFCM facilitates robust detection of different EV and sEV subpopulations in isolated EVs, as well as unprocessed EV-containing samples. Our results indicate that fluorescently labelled sEVs as biological reference material are highly useful for the optimisation of fluorescence-based methods for sEV analysis. Finally, we propose that IFCM will help to significantly increase our ability to assess EV heterogeneity in a rigorous and reproducible manner, and facilitate the identification of specific subsets of sEVs as useful biomarkers in various diseases. Die Homöostase des Immunsystems und aller Blutzellen des menschlichen Körpers erfolgt durch die tägliche Produktion von Milliarden spezialisierter Blutzellen. Der Ablauf und die Regulation der Hämatopoese sind bis heute unvollständig verstanden. In den letzten Jahren wurden unterschiedliche neue Verwandtschaftsbeziehungen in der humanen Hämatopoese beschrieben. Im revidierten Modell befindet sich erythrozytäres sowie eosinophiles/basophiles Potential in den erythro-myeloischen Progenitoren (EMP). Im lympho-myeloischen Ast wird lymphatisches Potential im multilymphatischen Progenitor (MLP) und Neutrophilen- und Makrophagen-Potential im Granulozyten-Makrophagen-Progenitor (GMP) realisiert. Insgesamt wurde Makrophagen-Potential in drei distinkten Vorläufern, dem MLP, GMP und EMP, beobachtet. Im Kontrast zu anderen Modellen besitzen beide Äste des revidierten Modells distinkte myeloische Potentiale, sodass der Ursprung von myeloischen Zelltypen in Frage gestellt wird. Für die weitere Entschlüsselung der humanen Hämatopoese wurde die Bildung von dendritischen Zellen (DC), welche aus vier heterogenen Subtypen [cDC1, cDC2, pDC und von Monozyten abgeleiteten DC (MoDC)] bestehen, untersucht. Das Ziel dieses Projektes war es, den Ursprung der DC-Subtypen im revidierten Modell der humanen Hämatopoese zu identifizieren, die Herkunft von Makrophagen aus mehreren Linien zu validieren und mögliche Makrophagen-Subtypen zu klassifizieren. Für die Generierung von DC Subtypen, Makrophagen, Granulozyten und NK-/B-Zellen aus humanen CD34+ Zellen wurde zunächst ein geeignetes Kultursystem entwickelt. Über die funktionellen und molekularen Eigenschaften der in vitro generierten Zellen konnte die Bildung von DC-Subtypen und Makrophagen nachgewiesen werden. Indem das Differenzierungspotential von individuell abgelegten hämatopoetischen Stamm- und Vorläuferzellen im etablierten Kultursystem analysiert wurde, konnte das cDC1- und pDC-Potential einzig in Vorläufern mit lymphatischem Potential bzw. lymphatischer Herkunft beschrieben werden. Im Unterschied dazu korrelierte das Makrophagen-, MoDC- und cDC2 Potential sowohl mit lymphatischem als auch mit neutrophilem Potential. Daraus ließ sich schlussfolgern, dass cDC1 und pDC selektiv aus dem MLP entstehen, wohingegen cDC2, Makrophagen und MoDC aus beiden Linien, dem MLP und GMP, gebildet werden. Um die Frage zu klären, ob die myeloischen Zellen beider Linien unterschiedliche Eigenschaften besitzen, wurden die Expressionsprofile von MLP Makrophagen, MLP MoDC, GMP-Makrophagen und GMP-MoDC zueinander verglichen. Dabei konnten MLP und GMP-spezifisch überexprimierte Gensignaturen identifiziert werden, sodass die Zellen linienabhängige Transkriptionsunterschiede besitzen. In diesem Rahmen wurden CD35 und CD89 als mögliche Marker für die Diskriminierung von MLP- und GMP abgeleiteten Monozyten, Makrophagen und MoDC in vitro identifiziert. Zudem wurde der Einfluss der Notch- und Wnt Signalwege auf die Linienentwicklung in vitro getestet. Eine DLL1-Aktivierung des Notch-Signalweges induzierte die Bildung von cDC1 auf Kosten aller anderen Zelltypen. CHIR99021, ein Wnt-Signalweg-Aktivator, führte zum Anstieg der cDC1- und Makrophagen Bildung und zur Reduktion der pDC- und MoDC-Generierung. Folglich konnten der Notch- und Wnt-Signalweg als Regulatoren der humanen DC-Subtypen-Entwicklung identifiziert werden. Abschließend lässt sich zusammenfassen, dass die Erkenntnisse dieser Arbeit die Bildung unterschiedlicher Makrophagen/MoDC aus mehreren Linien sowie die selektive Herkunft von cDC1 und pDC aus dem lymphatischen Ast aufzeigen und somit zu einem besseren Verständnis der humanen Hämatopoese beitragen., Homeostasis within the human immune and blood system is maintained through the daily production of billions of blood cells. The developmental course and regulation of the hematopoiesis remains to this day not fully understood. In recent years different novel lineage relationships have been described in human hematopoiesis. In the revised model erythroid as well as eosinophil/basophil potential lies within the erythro-myeloid progenitor (EMP) pool. Inside the lympho-myeloid branch, the multilymphoid progenitor (MLP) retains lymphoid potentials whereas neutrophil potential resides in the granulocyte-macrophage progenitor (GMP). In total, macrophage potential was observed in three different lineages, the MLP, GMP and EMP. In contrast to other models both branches of the revised model have distinct myeloid potentials, whereby the origin of myeloid cell types in general is questioned. For further analysis of the human hematopoiesis the origin and relations of dendritic cells (DC), which can be divided into four subtypes [cDC1, cDC2, pDC and monocyte-related DC (moDC)], were analyzed. The aim of the project was to identify the origin of DC subsets in the revised model of human hematopoiesis, validate the multi-lineage generation of macrophages and classify potential macrophage subsets. At first, a culture system for the generation of DC subsets, macrophages, granulocytes and NK/B cells from CD34+ cells was developed. The generation of cDC1, cDC2, pDC, moDC and macrophages could be verified by the functional and molecular properties of the in vitro generated cells. By analyzing the differentiation potential of individually deposited hematopoietic stem and progenitor cells in the developed in vitro culture system, cDC1 and pDC potential was exclusively detected in progenitors with lymphoid potential or origin. In contrast, macrophage, moDC and cDC2 potential correlated with lymphoid potential in the MLP as well as with neutrophil potential in the GMP. Based on these observations, the DC subsets cDC1 and pDC originate exclusively from the MLP whereas macrophages, moDC and cDC2 are formed from both lineages, the MLP and GMP. To address whether myeloid cells derived from the MLP and GMP lineage have different properties, the transcriptional profiles of MLP-macrophages, MLP-moDC, GMP-macrophages and GMP-moDC were compared. Here, MLP-specific and GMP-specific gene signatures were identified, indicating lineage-dependent transcriptional differences between these populations. In this context CD35 and CD89 were described in vitro as potential markers for separating MLP- and GMP-derived monocytes, macrophages and moDC. Moreover the effect of the Notch and Wnt signaling pathway on DC subset generation was analyzed in vitro. A DLL1-dependent activation of the Notch pathway promoted the cDC1 formation at the expense of all other detected cell types. Supplying CHIR990221, a Wnt signaling agonist, to the culture caused an increase of the cDC1 and macrophage formation as well as a reduction of the pDC and moDC generation. Hence, the Notch and Wnt signaling pathway were identified as regulators of the DC subset development in humane hematopoiesis. In conclusion, the findings of this project show the formation of different macrophage/moDC from multiple lineages as well as an exclusive origin of cDC1 and pDC in the lymphoid branch. By this means the project contributes to a better understanding of human hematopoiesis, particularly with respect to DC and macrophage development.
