Understanding how to pharmacologically manipulate amyloids is an essential step towards managing dozens of amyloid-based human diseases. Here, we present a pipeline that yields small molecule antagonists of amyloidogenesis. Screens for small molecules that perturb the development of the nematode C. elegans revealed 24 distinct small molecule scaffolds that crystalize on the non-luminal face of the pharyngeal cuticle. Consistent with the amyloid-like nature of the pharyngeal cuticle, 25% of these scaffolds are known to bind human amyloids with nanomolar affinity. The growing crystals preferentially disrupt the amyloid-like material within the cuticle but leave its chitin-based matrix relatively intact. We screened a collection of drugs and natural products for those that suppress crystal formation and found 45 distinct scaffolds, 33% of which are known amyloid busters. Screening for pharmacological suppressors of crystal formation therefore represents an inexpensive high-throughput in vivo approach that enriches for small molecules with amyloid-busting potential.

Roughly 4.5 million species of ecdysozoan animals repeatedly shed their old cuticle and construct a new one underneath to accommodate growth. How cuticles are constructed is not well understood. Here, we systematically mine gene expression datasets to uncover the spatiotemporal blueprint for how the chitin-based pharyngeal cuticle of the nematode Caenorhabditis elegans is built. We demonstrate that the blueprint correctly predicts expression patterns and functional relevance to cuticle development. We find that as larvae prepare to molt, catabolic enzymes are upregulated and the genes that encode chitin synthase, chitin cross-linkers, and homologs of amyloid regulators subsequently peak in expression. 48% of the gene products secreted during the molt are predicted to be intrinsically disordered proteins (IDPs), many of which belong to four distinct families that are expressed in overlapping waves. These include the IDPAs, IDPBs, and IDPCs that are introduced for the first time here. We find that all four families have sequence properties known to drive phase separation and show in vitro phase separation for one of these proteins. This systematic analysis reveals the massive contribution that IDPs make to the cuticle and highlights how reversibly phase-separating materials may facilitate cuticle disassembly and reassembly during the molt.

Background Mitochondrial dysfunction is one of the leading causes of neurological disorders in humans. Mitochondrial perturbations lead to adaptive mechanisms that include HIF-1 stabilization, though the consequences of increased levels of HIF-1 following mitochondrial stress remain poorly understood. Results Using Caenorhabditis elegans, we show that a hif-1 loss-of-function mutation confers resistance towards the mitochondrial toxin ethidium bromide (EtBr) and suppresses EtBr-induced production of ROS. In mammals, the PD-related gene DJ-1 is known to act as a redox sensor to confer protection against antioxidants and mitochondrial inhibitors. A deletion mutant of the C. elegans homolog djr-1.1 also showed increased resistance to EtBr. Furthermore, our data implicates p38 MAP kinase as an indispensable factor for survival against mitochondrial stress in both hif-1 and djr-1.1 mutants. Conclusions We propose that EtBr-induced HIF-1 activates pathways that are antagonistic in conferring protection against EtBr toxicity and that blocking HIF-1 activity may promote survival in cells with compromised mitochondrial function.
