2. Cladobotric acids A-F, new cytotoxic polyketides from a New Zealand Cladobotryum sp.

Abstract

Cladobotric acids A−F (1−6), fungal-derived polyketides, were isolated from the fermentation broth of a New Zealand Cladobotryum species. Structures were determined by extensive spectral analysis and X-ray crystallography, and the polyketide origin of 1−6 was concluded from feeding experiments with 13C-labeled precursors. The observed folding pattern for the polyketide chain is unusual for fungi. Cladobotric acids A−F (1−6) exhibited notable cytotoxicity against the murine P388 leukemia cell line and were also active against Bacillus subtilis and Candida albicans.

18. Communesins G and H, New Alkaloids from the Psychrotolerant Fungus Penicillium rivulum

Abstract

Communesins G (1) and H (2), two new alkaloids, have been isolated from the psychrotolerant, new species Penicillium rivulum Frisvad. The compounds were isolated by high-speed countercurrent chromatography and preparative HPLC using UV-guided fractionation. The structures were determined from 1D and 2D NMR techniques and HRESIMS. In contrast to all other known communesins, 1 and 2 were found inactive in antimicrobial, antiviral, and anticancer assays.

Abstract

Fermentation of Pseudeurotium zonatum led to the isolation of the known cytochalasin G (1) and three new cytochalasins, X, Y, and Z (2−4). These four compounds are the only naturally occurring cytochalasins reported to date that contain an indole-substituted perhydroisoindol-1-one fused with an 11-membered macrocycle.

23. Cortamidine Oxide, a Novel Disulfide Metabolite from the New Zealand Basidiomycete (Mushroom) Cortinarius Species

Abstract

Three disulfide metabolites were isolated from the fruiting bodies of the basidiomycete (mushroom) Cortinarius sp., collected in the Catlins, New Zealand. The structures of these compounds were determined as the unsymmetrical disulfide cortamidine oxide (1), 2,2‘-dithiobis(pyridine N-oxide) (2), and the symmetrical disulfide 3. Both 1 and 2 showed significant antimicrobial activity and cytotoxicity. 2,2‘-Dithiobis(pyridine N-oxide) (2) and the symmetrical disulfide 3 are assumed to be artifacts of the isolation procedure.

Abstract

Aqueous extracts from the African plant Myrianthus holstii potently inhibited the infection of the T-lymphoblastoid cell line, CEM-SS, by human immunodeficiency virus-1RF (HIV-1RF). The active constituent, M. holstii lectin (MHL), was purified by LH-20 column chromatography and reversed phase HPLC. MHL, a 9284-Da cysteine-rich protein, was characterized by amino acid analysis, N-terminal sequencing, ESIMS, and matrix-assisted laser-desorption ionization−time-of-flight mass spectrometry. Pure MHL had anti-HIV activity, with an EC50 value of 150 nM. Delaying the addition of MHL for up to 8 h after initial exposure of CEM-SS cells to virus did not result in loss of the antiviral activity; however, if addition of the compound was delayed for 16 h or more, there was a marked decrease in the antiviral activity. MHL bound to a virus-free, soluble form of the viral envelope protein gp120 but did not inhibit the subsequent binding to a cell-free, soluble form of the cellular receptor CD4.

Abstract

The structure, stereochemistry, and conformation of theonellapeptolide IIIe (1), a new 36-membered ring cyclic peptolide from the New Zealand deep-water sponge Lamellomorpha strongylata, is described. The sequence of the cytotoxic peptolide was determined through a combination of NMR and MS−MS techniques and confirmed by X-ray crystal structure analysis, which, with chiral HPLC, established the absolute stereochemistry.

29. Hirsutide, a Cyclic Tetrapeptide from a Spider-Derived Entomopathogenic Fungus, Hirsutella sp.

Abstract

The entomopathogenic fungus Hirsutella sp., isolated from an infected spider, was found to produce the new cyclotetrapeptide hirsutide (1), cyclo-(l-NMe-Phe-l-Phe-l-NMe-Phe-l-Val), along with the known cytochalasin Q (2), using a cytotoxicity-guided isolation procedure. The structure of 1 was elucidated using one- and two-dimensional NMR experiments, mass spectrometry, and Marfey's method for analyzing the configuration of the amino acids.

Abstract

Three novel thiodiketopiperazine derivatives, T988 A, B, and C (1−3), were isolated from a Tilachlidium sp. Their structures were elucidated by spectroscopic analyses using various 1D and 2D NMR techniques. All the compounds showed potent cytotoxicity against cultured P388 leukemia cells.

The crude extract of a New Zealand ascidian displayed antitumor activity. Bioassay-directed fractionation yielded a novel alkaloid, coproverdine (1). The structure of 1 was assigned on the basis of detailed spectroscopic analysis. Coproverdine (1) was responsible for the antitumor activity of the crude extract.
