This article reviews the geographical distribution, ethnomedicinal applications, and phytochemistry of the genus Monanthotaxis Baill, tribe Uvariae of the family Annonaceae. The reviewed works of literature were collected from various electronic databases including Google Scholar, PubMed, Science Direct, The International Plant Names Index (IPNI), and Research Gate. During this review, ninety-eight species of the genus Monanthotaxis were found to be widely distributed in tropical Africa. Some of those species are used in folkloric medicine by various communities to manage diseases and disease conditions such as fever, vomiting, headache, stomach-ache, malaria, helminthiasis, and hysteria. In the past 44 years (1979 to 2023), one hundred and nineteen secondary metabolites with different biomedical potentials have been reported from this genus. The reported compounds are categorised into flavonoids, alkaloids, terpenoids, polyoxygenated cyclohexane, and cyclohexene derivatives, benzyl derivatives, cinnamic acid derivatives, and stilbenoids. Toussaintia orientalis Verdc. (Annonaceae) is a medicinal plant species endemic to Tanzania. It is classified under the International Union for Conservation of Nature (IUCN) as an extremely endangered species threatened with extinction. The review covers the phytochemistry of this plant species and the pharmacological properties of the compounds obtained therefrom. The chemistry of this plant species entails natural products with different structures including uncommon cinnamoyl tetraketide derivatives, aristolactam alkaloids, and flavonoids. The review identifies 27 compounds belonging to different subclasses of natural products obtained from this species for the past fourteen years (2010–2024). These compounds are discussed along with other 13 related natural products. T. orientalis derived compounds exhibit varied potential pharmacological applications as antibacterial, anticancer, anti‐inflammatory, antiviral, and neuroprotective agents. Some of the reported compounds displayed pharmacological properties corroborating the use of this plant species in traditional medicine. This review provides baseline data as one comprehensive compilation that will ignite interest and guide future research and development of therapeutic agents inspired by the chemodiversity presented by this plant species while at the same time attracting the attention of plant conservationists to initiate efforts to conserve this highly endangered biomedical treasure. The reported investigations were aimed at establishing anticancer potential of the leaf extracts from Croton dictyophlebodes (CDE), C. kilwae (CKE) and C. longipedicellatus ssp austrotanzanicus (CLE). In vitro cytotoxicity assays of the extracts were performed against human laryngeal cancer cell line (HEp-2) and human breast cancer cell line (MCF-7), while in vivo toxicological studies were done using Swiss albino mice (Mus musculus) following the Organization for Economic Cooperation and Development guideline number 423 of 2001. Results for in vitro assays revealed that CDE and CLE induced cytotoxicity to MCF-7 and HEp-2 cell lines at EC50 values of 86 and 63 μg/mL, respectively, while CKE was inactive to HEp-2 cells at 100 μg/mL. For in vivo studies, mice were given calculated doses based on globally harmonized standards and monitored for 14 days to observe signs of toxicity. Haematological analysis was also carried out to obtain information on changes in blood conditions. The findings showed that LD50 for CKE and CDE were > 2000 mg/kg, while for CLE was 175 mg/kg body weight (b. w.). The oral administration of CDE and CKE did not reveal significant changes in haematological parameters of treated mice, while CLE provoked some. Besides changes in life styles and improper dietary patterns, the rapidly increasing burden of diabetes mellitus type 2 is correlated with antidiabetic agents currently in clinical use, which are costly, less effective, and present undesirable side effects. Natural products from plants are promising alternative sources for developing more effective and cost friendly antidiabetic agents. This study therefore investigated the antidiabetic potential of Vitex mombassae fruits extract (VMFE) in streptozotocin-nicotinamide induced diabetic Wistar rat model. Oral acute toxicity test was conducted in male Wistar rat using up and down method by following OECD Guideline 423 (2002). No clinical signs of toxicity were observed at 300 and 2000 mg/kg body weight (b.w.) doses used in the assays. The LD50 value was estimated to be ˃ 2000 mg/kg b.w. In evaluation of antidiabetic activity, diabetic Wistar rats were administered with 100, 200 and 300 mg/kg b. w. of VMFE. VMFE resulting into significant lowering of the levels of fasting blood glucose (p < 0.05) and increased of level of high-density lipoprotein-cholesterol (HDLC), levels of total cholesterol (TC), triglyceride (TG) and low-density lipoprotein-cholesterol (LDL-C) were reduced significantly (p < 0.05). Therefore, this study provides scientific evidence for antidiabetic activities of V. mombassae fruits extracts, and validates its traditional use in the management of diabetes mellitus type 2. Extreme environments such as soda lakes are potential sources of microbes with biotechnological applications in different sectors. This study aimed at isolation, characterization and investigation of antibacterial potential of actinobacteria from Momela Soda Lakes, at Arusha National Park in Tanzania. One hundred and twenty (120) isolates were recovered from soil and water samples using the dilution plate technique. The isolates were morphologically and biochemically characterized, and further, screened for antimicrobial activity by disc diffusion method as well as the micro dilution technique. Cytotoxic effects were determined using the brine shrimp lethality test. Results showed that, all 120 isolates were Gram-positive rod-coccus shaped. Forty-four out of them showed antibacterial activity against Staphylococcus aureus and Escherichia coli. The Streptomyces (101TI) and Dietzia (56BI) strains exhibited exceptionally higher antibacterial activity compared to the rest with inhibition zones of 16.25 and 21.00 mm, respectively. These two strains were toxic against brine shrimplarvae. Microbacterium (5LI), Hoyosella (113BI), Streptomyces (62BI), Dietzia (117SI), Hoyosella (37SI) and Microbacterium (3BI) strains had low antibacterial and cytotoxic activities. Chemical compositions and antimicrobial activities of twenty-two Aloe vera branded commercial products in Tanzania, a case of marketed soaps, creams, lotions, petroleum jelly, toothpastes and hair conditioner products in Dar es Salaam, were investigated. Chemical compositions were analysed using gas chromatography-mass spectrometry (GC-MS) whereas antimicrobial activities were evaluated using agar dilution method against four bacteria species, namely Staphylococcus aureus, Escherichia coli, Klebsiella pneumoneae and Salmonella typhi) and two fungal species Candida albicans and Cryptococcus neoformans. The GC-MS analysis revealed the presence of seven non-polar constituents, namely methyl palmitate, 9-octadecenoic acid methyl ester, methyl stearate, tetratetracontane, hexacosane, and pentacosane and methyl tetradecanoate as the most common ingredients among the products. Eleven compounds were detected in both the commercial products and reference A. vera extracts. The commercial products AVC5 and AVL3 inhibited the growth of E. coli and S. typhi at minimum inhibitory concentrations (MICs) of 7.5 and 12.5% (v/v), respectively, whereas AVC2 and AVC5 inhibited the growth of C. albicans and C. neoformans at 5.0% (v/v). AVC6 had 7.5 and 15.0% (v/v) MICs for C. neoformans and C. albicans, respectively. Callosobruchus chinensis causes damage to the Phaseolus vulgaris seeds. Traditionally, Dioscorea sansibarensis serves as a medicinal plant. Naturally, D. sansibarensis has toxins that protect against herbivores and the surrounding invasive plants in its natural habitat. Phytochemical analysis by thin-layer chromatography (TLC) and laboratory experiments was carried out to determine the activity of D. sansibarensis leaves, bulbils, and yams powders on antioviposition and inhibition of the F1 emergence of C. chinensis. Bioassay data were subjected to nonparametric (Kolmogorov–Smirnov) statistical analysis and a generalized linear model at P ≤ 0.05. Statistically, the powders had an antioviposition activity of 34.3% (R2 = 0.343). A recommendable activity on antioviposition was displayed by the yams powder; treatment by 0.8 g of yams powder had a Wald Chi-Square value of 1.291, P = 0.26. Inhibition of F1 emergence was significantly attained by the yams powder; the treatment by 0.6 g of yams powder had a Wald Chi-Square value of 7.72, P = 0.01. Statistically, the bulbils powder displayed low antioviposition and inhibition of F1 emergence. Observations on the TLC exposed compounds with similar Rf values; saponin with an Rf value of 0.72 was portrayed in the leaves, bulbils, and yams. A terpenoid and a flavonoid with Rf values of 0.37 and 0.71, respectively, were observed in bulbils and yams but absent in leaves. A terpenoid with an Rf value of 0.49 was visualized in leaves and bulbils but not in the yams powder. The study concluded that the D. sansibarensis yams and leaves powders are viable for application by the farmers in the protection of stored legumes against attack by C. chinensis. Chitosan (CS, molecular weight 20.2 kDa, degree of deacylation (DD) 73.31%) was successfully obtained by deacetylation of chitin extracted from shrimp (Litopenaeus vannamei) shell wastes. The encapsulation of the bioactive natural product, panchovillin (PANV), isolated from Erythrina schliebenii, on a chitosan-tripolyphosphate (CS/TPP) nano-framework was achieved by ionotropic gelation. Characterization of pure CS, CS/TPP and PANV-CS/TPP nanocomposites was performed by FTIR, SEM and XRD. The molecular weight of chitosan and the thermal stability of the materials were determined by MALDI-TOF-MS and simultaneous thermal analyzer (STA)/DTG, respectively. The respective encapsulation efficiency and loading capacity of the PANV were found to be 70% and 0.36%. The in vitro release studies showed an initial burst of 42% of PANV in the first six hours. This was followed by a slow and sustained release up to 72 h. The in vivo antimycobacterial activities of both PANV and PANV-CS/TPP nanocomposite against Mycobacterium indicus pranii (MIP) using Galleria mellonella larvae as an in vivo infection model are reported in this paper. Aflatoxin M1(AFM1) contamination in raw milk from household cows fed with sunflower seedcakes or sunflower-based seedcake feeds was determined in 37 milk samples collected randomly from different locations in Singida region, Tanzania. Aflatoxin B1(AFB1) contamination in sunflower-based seedcake feed was determined in 20 feed samples collected from the same household dairy farmers. The samples were analysed by RP-HPLC using fluorescent detection after immunoaffinity column clean-up. Recoveries were 88.0% and 94.5%, while the limits of detection (LOD) were 0.026 ng mL−1and 0.364 ng g−1for AFM1and AFB1, respectively. Of the analysed cow’s milk samples, 83.8% (31/37) contained AFM1, with levels ranging from LOD to 2.007 ng mL−1, exceeding both the European Commission (EC) and Tanzania Food and Drug Authority (TFDA) limit of 0.05 ng mL−1. Of the contaminated samples, 16.1% exceeded the Codex Alimentarius limit of 0.5 ng mL−1. AFB1was present in 65% (13/20) of the feed samples with levels ranging from LOD to 20.47 ng g−1, 61.53% exceeding the TFDA and EC maximum limits of 5 ng g−1for complete dairy animal feed. Clinical applications of many small molecules are limited due to poor solubility and lack of controlled release besides lack of other desirable properties. Experimental and computational studies have reported on the therapeutic potential of polyamidoamine (PAMAM) dendrimers as solubility enhancers in pre-clinical and clinical settings. Besides formulation strategies, factors such as pH, PAMAM dendrimer generation, PAMAM dendrimer concentration, nature of the PAMAM core, special ligand and surface modifications of PAMAM dendrimer have an influence on drug solubility and other recommendable pharmacological properties. This review, therefore, compiles the recently reported applications of PAMAM dendrimers in pre-clinical and clinical uses as enhancers of solubility and other desirable properties such as sustained and controlled release, bioavailability, bio-distribution, toxicity reduction or enhancement, and targeted delivery of small molecules with emphasis on cancer treatment. Parkinson's disease (PD) is a movement disorder resulting from the loss of dopaminergic neurons over time. While there is no cure for PD, available conventional therapies aid to manage the motor symptoms. Natural products (NPs) derived from plants are among the most potent alternative therapies for PD. This study explored the neuroprotective potential of selected cinnamoyl derivatives namely toussaintine A (1), E-toussaintine E (2), asperphenamate (3) and julocrotine (4) against PD indicators using rotenone-challenged Drosophila melanogaster and in silico models. The compounds were first assessed for their toxicity preceding treatment experiments. Adult flies (aged 1-4 days) were exposed to varying concentrations of the compounds for 7 days. During the experiment, the mortality of flies was observed, and the lethal concentration (LC 50 ) of each tested compound was determined. The LC 50 values were found to be 50.1, 55.6, 513.5, and 101.0 µM for compounds 1, 2, 3, and 4, respectively. For seven days, we exposed flies to 500 µM of rotenone and co-fed with a chosen dose of 40 µM of each test compound in the diet. Using a negative geotaxis test, rotenone-challenged flies exhibited compromised climbing ability in comparison to control flies, the condition that was reversed by the action of studied compounds. Rotenone exposure also elevated malondialdehyde levels in the brain tissues, as measured by lipid peroxidation, when compared to control flies. In flies exposed to rotenone and co-fed with the compounds, this effect was lessened. In flies exposed to rotenone, mRNA levels of antioxidant enzymes such as superoxide dismutase and catalase were raised but were normalized in flies treated with the investigated compounds. Moreover, in-silico studies examined the inhibitory ability of compounds 1-4 against selected PD molecular targets, revealing the strong power of toussaintine A (1) against Adenosine receptor 2 (A2AR) and monoamine oxidase B. Thus, our findings suggest that cinnamoyl derivatives have neuroprotective potential via reducing the oxidative burden and improving locomotor ability after toxin invectives. Luteolin is a flavonoid obtained from different plant species. It is known for its versatile biological activities. However, the beneficial effects of luteolin have been limited to small concentrations as a result of poor water solubility. This study aimed at investigating the hydrophobic interaction and hydration of luteolin towards the improvement of its solubility when used as a drug. We report the aggregation properties of luteolin in water by varying the number of monomers using atomistic molecular dynamics simulation. Results show that the equilibrium structure of luteolin occurs in an aggregated state with different structural arrangements. As the monomers size increase, the antiparallel flipped conformation dominates over T-shaped antiparallel, T-shaped parallel, and antiparallel conformations. The formation of intramolecular hydrogen bonding of 0.19 nm between the keto-enol groups results in hydrophobic characteristics. A larger cluster exhibits slow hydrogen bond dynamics for luteolin-luteolin than luteolin-water interaction. Water structure at large cluster size exhibited slow dynamics and low self-diffusion of luteolin. The existence of hydrophobic π-π and hydrogen bonds between luteolin molecules drives strong self-aggregation resulting in poor water solubility. Three new oxygenated cyclohexene derivatives, pandensenol D - F (1-3), two new flavanoids, pandensone A and B (4-5), and seven known compounds (6-12) were isolated from the methanol extract of the leaves of Uvaria pandensis Verdc. (Annonaceae). The structures were characterized by NMR spectroscopic and mass spectrometric analyses. The isolated metabolites were evaluated for their antibacterial activity against the Gram-positive bacteria Bacillus subtilis and Staphylococcus epidermidis, the Gram-negative bacteria Enterococcus raffinosus, Escherichia coli, Paraburkholderia caledonica, Pectobacterium carotovorum and Pseudomonas putida, and for cytotoxicity against the MCF-7 human breast cancer cell line. Out of the tested compounds, pandensenol D (1) and (6',7'-dihydro-8'α,9'β-dihydroxy)-3-farnesylindole (12) showed weak activity, whereas (8'α,9'β-dihydroxy)-3-farnesylindole (11) strong activity against B. subtilis. The new isoflavonoid kirkinone A (1) and biflavonoid kirkinone B (2) along with six known compounds (3-8) were isolated from the methanolic extract of the root bark of Ochna kirkii. The compounds were identified by NMR spectroscopic and mass spectrometric analyses. Out of the eight isolated natural products, calodenin B (4) and lophirone A (6) showed significant antibacterial activity against the Gram-positive bacterium Bacillus subtilis with MIC values of 2.2 and 28 μM, and cytotoxicity against the MCF-7 human breast cancer cell line with EC 50 values of 219.3 and 19.2 μM, respectively. The methanolic crude extract of the root bark exhibited cytotoxicity at EC 50 8.4 μg/mL. The isolated secondary metabolites and the crude extract were generally inactive against the Gram-negative Escherichia coli (MIC ≥400 μg/mL). Parkinson's disease (PD) is a movement disorder associated with the progressive loss of dopaminergic neurons (DA). PD treatment remains unsatisfactory as the current synthetic drugs in clinical use relies on managing only motor symptoms. This study investigated antioxidant potentials of selected compounds namely, 5,6,7,4'-tetramethoxyflavone (1), 6-hydroxy-2,3,4,4'-tetramethoxychalcone (2), 6-methoxyhamiltone A (3), diosquinone (4) and toussantine D (5) against rotenone (6) induced PD in Drosophila melanogaster. Toxicity of these compounds was conducted by monitoring flies' survival for seven days and determining the lethal concentrations (LC 50 ). Whereas compound 1 had LC 50 value of 91.3 μM within three days, compounds 2, 3, 4, and 5 had LC 50 values of 87.2, 58.0, 64.0 and > 1000 μM, respectively on the seventh day of the experiment. We exposed flies (1-4 days old) to 500 μM rotenone and co-treated with different doses of the test compounds in the diet for seven days at final concentrations of 11.0, 43.6 and 87.2 μM for compounds 2 and 3. The concentrations used for compound 4 were 8.0, 32.0 and 64.0 μM, while 250, 500 and 1000 μM were used for compound 5. Rotenone fed flies showed impaired climbing ability compared to control flies, the phenotype that was rescued by the treatment of tested phytochemicals. Rotenone toxicity also increased malondialdehyde levels assayed by lipid peroxidation in the brain tissues relative to control flies. This effect was reduced in flies exposed to rotenone and co-treated with the phytochemicals. Moreover, expression levels of mRNA of antioxidant enzymes; superoxide dismutase and catalase were elevated in flies exposed to rotenone and normalized in flies that were co-treated with tested compounds. Besides compound 1, this study provides overall evidence that the tested flavonoids and polyketides ameliorated the rotenone provoked neurotoxicity in D. melanogaster by battling the induced oxidative stress in brain cells including DA neurons and hence rescue the locomotor behaviour deficits., (Copyright © 2020 Elsevier B.V. The stem bark and root bark extracts of Croton dictyophlebodes (Euphorbiaceae) yielded seven undescribed ent-clerodanes: 15,16-epoxy-17,12(S)-olide-ent-cleroda-1,3,13(16),14-tetraen-18-oic acid methyl ester (crotodictyo A), 3β,4β:15,16-diepoxy-ent-cleroda-13(16),14-dien-20-al (crotodictyo B), 3β,4β:15,16-diepoxy-ent-cleroda-13(16),14-dien-19,20-dioic acid (crotodictyo C), 3β,4β:15,16-diepoxy-ent-cleroda-13(16),14-dien-20,19-olide (crotodictyo D), 3β,4β:15,16-diepoxy-20,12(R)-olide ent-cleroda-13(16),14-dien-19-oic acid methyl ester (crotodictyo E), 15,16-epoxy-ent-cleroda-3,13(16),14-trien-12-oxo-18-oic acid (crotodictyo F) and 15,16-epoxy-ent-cleroda-1,3,13(16),14-tetraen-12-oxo-18-oic acid (crotodictyo G), in addition to 15,16-epoxy- ent-cleroda-3,13(16),14-trien-12-oxo-18-oic acid methyl ester (crotodictyo H), reported previously as a synthetic derivative, and acetyl aleuritolic acid. The root extract yielded two ent-trachylobanes, ent-trachylobane-18,19-diol, the undescribed ent-trachylobane-2α,19-diol, along with ent-kaur-16-en-19-oic acid and 2-methoxybenzyl benzoate. Phytochemical investigations of ethanol root bark and stem bark extracts of Cleistochlamys kirkii (Benth.) Oliv. (Annonaceae) yielded a new benzopyranyl cadinane-type sesquiterpene (cleistonol, 1 ) alongside 12 known compounds ( 2 - 13 ). The structures of the isolated compounds were established from NMR spectroscopic and mass spectrometric analyses. Structures of compounds 5 and 10 were further confirmed by single crystal X-ray crystallographic analyses, which also established their absolute stereochemical configuration. The ethanolic crude extract of C. kirkii root bark gave 72% inhibition against the chloroquine-sensitive 3D7-strain malaria parasite Plasmodium falciparum at 0.01 μg/mL. The isolated metabolites dichamanetin, ( E )-acetylmelodorinol, and cleistenolide showed IC 50 = 9.3, 7.6 and 15.2 μM, respectively, against P. falciparum 3D7. Both the crude extract and the isolated compounds exhibited cytotoxicity against the triple-negative, aggressive breast cancer cell line, MDA-MB-231, with IC 50 = 42.0 μg/mL (crude extract) and 9.6-30.7 μM (isolated compounds). Our findings demonstrate the potential applicability of C. kirkii as a source of antimalarial and anticancer agents.

A total of 61 samples comprising sunflower seeds (40) and unrefined sunflower oils (21) samples collected randomly from Singida, Tanzania were analysed by Reverse Phase-high performance liquid chromatography (RP-HPLC). 15% (6/40) of the seed samples were contaminated with aflatoxin B 1 ranging from limit of detection (LOD) to 218 ng g -1 with three of them exceeding the European Commission/European Union (EC/EU) and Tanzania Bureau of Standards (TBS)/Tanzania Food and Drug Authority (TFDA) maximum limits of 2 ng g -1 for AFB 1 in oilseeds. The levels of total aflatoxins (AFT) in seeds ranged from LOD to 243 ng g -1 . Other aflatoxins, except AFG 2 , were also detected. For the unrefined sunflower oils, the levels of AFB 1 ranged from LOD to 2.56 ng mL -1 . About 80.9% (17/21) of the analysed oil samples contained AFB 1 of which 17.65% (3/17) exceeded the EC/EU and TBS/TFDA maximum limits of 2 ng mL -1 . Other aflatoxins were also detected in the oils. The measured levels indicate there is a need for food quality education among food processors.
