Abstract

We present results of a Hubble Space Telescope (HST) UBVI-band study of star clusters in tidal tails, using new WFC3 and ACS imaging to complement existing WFPC2 data. We survey 12 tidal tails across seven merging systems, deriving ages and masses for 425 star cluster candidates (SCCs). The stacked mass distribution across all systems follows a power law of the form $dN/dM \propto M^{\beta}$, with $\beta = -2.02 \pm 0.15$, consistent with what is seen in other star forming environments. GALEX and Swift UV imaging provide star formation rates (SFRs) for our tidal tails, which when compared with ages and masses of our SCCs, allows for a determination of the cluster formation efficiency (CFE). We find the CFE increases with increasing SFR surface density, matching the theoretical model. We confirm this fit down at SFR densities lower than previously measured (log $\Sigma_\text{SFR} \: (\text{M}_\odot \: \text{yr}^{-1} \: \text{kpc}^{-2}) \approx -4.2$), as related to the CFE. We determine the half-light radii for a refined sample of 57 SCCs with our HST WFC3 and ACS imaging, and calculate their dynamical age, finding the majority of them to be gravitationally bound. We also provide evidence of only low-mass ($< 10^4 \: \text{M}_\odot$) cluster formation in our nearest galaxy, NGC 1487, consistent with the theory that this system is a dwarf merger., Comment: 26 pages, 27 figures. Abstract

BACKGROUND: Diffuse midline gliomas (DMG) are highly invasive brain tumors with rare survival beyond two years past diagnosis and limited understanding of the mechanism behind tumor invasion. Previous reports demonstrate upregulation of the protein ID1 with H3K27M and ACVR1 mutations in DMG, but this has not been confirmed in human tumors or therapeutically targeted. METHODS: Whole exome, RNA, and ChIP-sequencing was performed on the ID1 locus in DMG tissue. Scratch-assay migration and transwell invasion assays of cultured cells were performed following shRNA-mediated ID1-knockdown. In vitro and in vivo genetic and pharmacologic [cannabidiol (CBD)] inhibition of ID1 on DMG tumor growth was assessed. Patient-reported CBD dosing information was collected. RESULTS: Increased ID1 expression in human DMG and in utero electroporation (IUE) murine tumors is associated with H3K27M mutation and brainstem location. ChIP-sequencing indicates ID1 regulatory regions are epigenetically active in human H3K27M-DMG tumors and prenatal pontine cells. Higher ID1-expressing astrocyte-like DMG cells share a transcriptional program with oligo/astrocyte-precursor cells (OAPCs) from the developing human brain and demonstrate upregulation of the migration regulatory protein SPARCL1. Genetic and pharmacologic (CBD) suppression of ID1 decreases tumor cell invasion/migration and tumor growth in H3.3/H3.1K27M PPK-IUE and human DIPGXIIIP* in vivo models of pHGG. The effect of CBD on cell proliferation appears to be non-ID1 mediated. Finally, we collected patient-reported CBD treatment data, finding that a clinical trial to standardize dosing may be beneficial. CONCLUSIONS: H3K27M-mediated re-activation of ID1 in DMG results in a SPARCL1+ migratory transcriptional program that is therapeutically targetable with CBD.

Abstract

Childhood posterior fossa group A ependymomas (PFAs) have limited treatment options and bear dismal prognoses compared to group B ependymomas (PFBs). PFAs overexpress the oncohistone-like protein EZHIP (enhancer of Zeste homologs inhibitory protein), causing global reduction of repressive histone H3 lysine 27 trimethylation (H3K27me3), similar to the oncohistone H3K27M. Integrated metabolic analyses in patient-derived cells and tumors, single-cell RNA sequencing of tumors, and noninvasive metabolic imaging in patients demonstrated enhanced glycolysis and tricarboxylic acid (TCA) cycle metabolism in PFAs. Furthermore, high glycolytic gene expression in PFAs was associated with a poor outcome. PFAs demonstrated high EZHIP expression associated with poor prognosis and elevated activating mark histone H3 lysine 27 acetylation (H3K27ac). Genomic H3K27ac was enriched in PFAs at key glycolytic and TCA cycle–related genes including hexokinase-2 and pyruvate dehydrogenase. Similarly, mouse neuronal stem cells (NSCs) expressing wild-type EZHIP (EZHIP-WT) versus catalytically attenuated EZHIP-M406K demonstrated H3K27ac enrichment at hexokinase-2 and pyruvate dehydrogenase, accompanied by enhanced glycolysis and TCA cycle metabolism. AMPKα-2, a key component of the metabolic regulator AMP-activated protein kinase (AMPK), also showed H3K27ac enrichment in PFAs and EZHIP-WT NSCs. The AMPK activator metformin lowered EZHIP protein concentrations, increased H3K27me3, suppressed TCA cycle metabolism, and showed therapeutic efficacy in vitro and in vivo in patient-derived PFA xenografts in mice. Our data indicate that PFAs and EZHIP-WT–expressing NSCs are characterized by enhanced glycolysis and TCA cycle metabolism. Repurposing the antidiabetic drug metformin lowered pathogenic EZHIP, increased H3K27me3, and suppressed tumor growth, suggesting that targeting integrated metabolic/epigenetic pathways is a potential therapeutic strategy for treating childhood ependymomas.

4. Population Growth, Energy Use, and the Implications for the Search for Extraterrestrial Intelligence

Abstract

Von Hoerner (1975) examined the effects of human population growth and predicted agricultural, environmental, and other problems from observed growth rate trends. Using straightforward calculations, VH75 predicted the "doomsday" years for these scenarios (2020-2050), when we as a species should run out of space or food, or induce catastrophic anthropogenic climate change through thermodynamically unavoidable direct heating of the planet. Now that over four decades have passed, in this paper we update VH75. We perform similar calculations as that work, with improved data and trends in population growth, food production, energy use, and climate change. For many of the impacts noted in VH75 our work amounts to pushing the "doomsday" horizon back to the 2300s-2400s (or much further for population-driven interstellar colonization). This is largely attributable to using worldwide data that exhibit smaller growth rates of population and energy use in the last few decades. While population-related catastrophes appear less likely than in VH75, our continued growth in energy use provides insight into possible future issues. We find that, if historic trends continue, direct heating of the Earth will be a substantial contributor to climate change by about 2260, regardless of the energy source used, coincident with our transition to a Kardashev type-I civilization. We also determine that either an increase of Earth's global mean temperature of 12K will occur or an unreasonably high fraction of the planet will need to be covered by solar collectors by about 2400 to keep pace with our growth in energy use. We further discuss the implications in terms of interstellar expansion, the transition to type II and III civilizations, SETI, and the Fermi Paradox. Abstract

Human influence on the biosphere has been evident at least since the development of widespread agriculture, and some stratigraphers have suggested that the activities of modern civilization indicate a geological epoch transition. The study of the anthropocene as a geological epoch, and its implication for the future of energy-intensive civilizations, is an emerging transdisciplinary field in which astrobiology can play a leading role. Habitability research of Earth, Mars, and exoplanets examines extreme cases relevant for understanding climate change as a planetary process. Energy-intensive civilizations will also face thermodynamic limits to growth, which provides an important constraint for estimating the longevity of human civilization and guiding the search for extraterrestrial intelligence. We recommend that missions concepts such as LUVOIR, HabEx, and OST be pursued in order to make significant progress toward understanding the future evolution of life on our planet and the possible evolution of technological, energy-intensive life elsewhere in the universe., Comment: A white paper on "Astrobiology Science Strategy" submitted to the NAS

7. A Tale of Two Tails: Exploring Stellar Populations in the Tidal Tails of NGC 3256

Abstract

We have developed an observing program using deep, multiband imaging to probe the chaotic regions of tidal tails in search of an underlying stellar population, using NGC 3256's 400 Myr twin tidal tails as a case study. These tails have different colours of $u - g = 1.05 \pm 0.07$ and $r - i = 0.13 \pm 0.07$ for NGC 3256W, and $u - g = 1.26 \pm 0.07$ and $r - i = 0.26 \pm 0.07$ for NGC 3256E, indicating different stellar populations. These colours correspond to simple stellar population ages of $288^{+11}_{-54}$ Myr and $841^{+125}_{-157}$ Myr for NGC 3256W and NGC 3256E, respectively, suggesting NGC 3256W's diffuse light is dominated by stars formed after the interaction, while light in NGC 3256E is primarily from stars that originated in the host galaxy. Using a mixed stellar population model, we break our diffuse light into two populations: one at 10 Gyr, representing stars pulled from the host galaxies, and a younger component, whose age is determined by fitting the model to the data. We find similar ages for the young populations of both tails, ($195^{-13}_{+0}$ and $170^{-70}_{+44}$ Myr for NGC 3256W and NGC 3256E, respectively), but a larger percentage of mass in the 10 Gyr population for NGC 3256E ($98^{+1}_{-3}\%$ vs $90^{+5}_{-6}\%$). Additionally, we detect 31 star cluster candidates in NGC 3256W and 19 in NGC 2356E, with median ages of 141 Myr and 91 Myr, respectively. Abstract

Nearby Type III (galaxy-spanning) Kardashev supercivilizations would have high mid-infrared (MIR) luminosities. We have used the Wide-field Infrared Survey Explorer (WISE) to survey ~$1 \times 10^5$ galaxies for extreme MIR emission, $10^3$ times more galaxies than the only previous such search. We have calibrated the WISE All-sky Catalog pipeline products to improve its photometry for extended sources. We present 563 extended sources with $|b| \ge 10$ and red MIR colors, having visually vetted them to remove artifacts. No galaxies in our sample host an alien civilization reprocessing more than 85% of its starlight into the MIR, and only 50 galaxies, including Arp 220, have MIR luminosities consistent with >50% reprocessing. Ninety of these (likely) extragalactic sources have little literature presence; in most cases they are likely barely resolved galaxies or pairs of galaxies undergoing large amounts of star formation. Five are new to science and deserve further study. The Be star 48 Librae sits within a MIR nebula, and we suggest that it may be creating dust. WISE, 2MASS, and Spitzer imagery shows that IRAS 04287+6444 is consistent with a previously unnoticed, heavily extinguished cluster of young stellar objects. We identify five "passive" (i.e. red) spiral galaxies with unusually high MIR and low NUV luminosity. We search a set of optically "dark" HI galaxies for MIR emission, and find none. Abstract

Pediatric and adult high-grade gliomas (HGGs) frequently harbor PDGFR[alpha] alterations. We hypothesized that cotreatment with everolimus may improve the efficacy of dasatinib in PDGFR[alpha]-driven glioma through combinatorial synergism and increased tumor accumulation of dasatinib. We performed dose-response, synergism, P-glycoprotein inhibition, and pharmacokinetic studies in in vitro and in vivo human and mouse models of HGG. Six patients with recurrent PDGFR[alpha]-driven glioma were treated with dasatinib and everolimus. We found that dasatinib effectively inhibited the proliferation of mouse and human primary HGG cells with a variety of PDGFR[alpha] alterations. Dasatinib exhibited synergy with everolimus in the treatment of HGG cells at low nanomolar concentrations of both agents, with a reduction in mTOR signaling that persisted after dasatinib treatment alone. Prolonged exposure to everolimus significantly improved the CNS retention of dasatinib and extended the survival of PPK tumor-bearing mice (mutant TP53, mutant PDGFR[alpha], H3K27M). Six pediatric patients with glioma tolerated this combination without significant adverse events, and 4 patients with recurrent disease (n = 4) had a median overall survival of 8.5 months. Abstract

The peculiar spiral NGC 2782 is the result of a minor merger with a mass ratio ~4:1 occurring ~200 Myr ago. This merger produced a molecular and HI rich, optically bright Eastern tail and an HI-rich, optically faint Western tail. Non-detection of CO in the Western Tail by Braine et al. (2001) suggested that star formation had not yet begun. However, deep UBVR and H-alpha narrowband images show evidence of recent star formation in the Western tail, though it lacks massive star clusters and cluster complexes. Using Herschel PACS spectroscopy, we discover 158 micron [CII] emission at the location of the three most luminous H-alpha sources in the Eastern tail, but not at the location of the even brighter H-alpha source in the Western tail. The Western tail is found to have a normal star formation efficiency (SFE), but the Eastern tail has a low SFE. The lack of CO and [CII] emission suggests the Western tail HII region may have a low carbon abundance and be undergoing its first star formation. The Western tail is more efficient at forming stars, but lacks massive clusters. We propose that the low SFE in the Eastern tail may be due to its formation as a splash region where gas heating is important even though it has sufficient molecular and neutral gas to make massive star clusters. The Western tail, which has lower gas surface density and does not form high mass star clusters, is a tidally formed region where gravitational compression likely enhances star formation., Comment: Accepted to Astrophysical Journal

13. Smooth and Starburst Tidal Tails in the GEMS and GOODS Fields

Abstract

GEMS and GOODS fields were examined to z~1.4 for galaxy interactions and mergers. The basic morphologies are familiar: antennae with long tidal tails, tidal dwarfs, and merged cores; M51-type galaxies with disk spirals and tidal arm companions; early-type galaxies with diffuse plumes; equal-mass grazing-collisions; and thick J-shaped tails beaded with star formation and double cores. One type is not common locally and is apparently a loose assemblage of smaller galaxies. Photometric measurements were made of the tails and clumps, and physical sizes were determined assuming photometric redshifts. Antennae tails are a factor of ~3 smaller in GEMS and GOODS systems compared to local antennae; their disks are a factor of ~2 smaller than locally. Collisions among early type galaxies generally show no fine structure in their tails, indicating that stellar debris is usually not unstable. One exception has a 5x10**9 Msun smooth red clump that could be a pure stellar condensation. Most tidal dwarfs are blue and probably form by gravitational instabilities in the gas. One tidal dwarf looks like it existed previously and was incorporated into the arm tip by tidal forces. The star-forming regions in tidal arms are 10 to 1000 times more massive than star complexes in local galaxies, although their separations are about the same. If they all form by gravitational instabilities, then the gaseous velocity dispersions in interacting galaxies have to be larger than in local galaxies by a factor of ~5 or more; the gas column densities have to be larger by the square of this factor., Comment: 24 pages plus 18 figures, ApJ, vol 664, July 20, 2007

Abstract

Background: Pediatric and adult high-grade glioma (HGG) frequently harbor PDGFRA alterations. Treatment of PDGFRA-driven HGG with targeted agents, such as the tyrosine kinase inhibitor dasatinib, has failed in the clinic. We hypothesized that co-treatment with everolimus may improve the efficacy of dasatinib in PDGFRA-driven glioma through combinatorial synergism and increased tumor accumulation of dasatinib. Methods: Dose response, synergism studies, P-gp inhibition and pharmacokinetic studies were performed on in vitro and in vivo human and mouse models of HGG. De novo tumors were generated in mice using intra-uterine electroporation (IUE) by injecting PB plasmids of TP53 mutation, PDGFRA mutation and H3K27M mutation (PPK) in the lateral vertical of mice embryos. Two children with recurrent PDGFRA-driven HGG were treated with dasatinib and everolimus with correlate CSF analysis. Results: Dasatinib effectively inhibited the proliferation of mouse and human primary HGG cells with a dose-dependent reduction of PDGFRA and pPDGFRA. Dasatinib exhibited synergy with everolimus in the treatment of HGG cells at low nanomolar concentrations of both agents. Prolonged exposure to everolimus significantly improved the CNS retention of dasatinib and extended survival of PPK tumor bearing mice. Two children with recurrent PDGFRA-driven HGG treated with dasatinib and everolimus survived six months and nine months after progression. A paired CSF sample from the patient with PDGFRA-amplified HGG showed 50% increase in CSF dasatinib levels after the addition of everolimus. Conclusion: Efficacy of dasatinib treatment of PDGFRA-driven HGG is improved with everolimus and suggests a promising route for improving targeted therapy for this patient population. Abstract

Despite beliefs that race is declining in importance within U.S. society, analysis of census data from the southwestern United States indicates continuing residential segregation, even for higher income Blacks. Higher income Hispanics found it easier to leave segregated neighborhoods because Anglos were much less likely to move when Hispanics moved in. (Author/IS)
