Abstract

The five prime end of the large nuclear ribosomal DNA (rDNA) of the coding region of Fusarium strains were analyzed to determine the genetic relationship of the trichothecene-producing microbe. Sequence analysis of the DNA at the five prime end of the large nuclear rDNA indicated a correlation between trichothecene production and the phylogenetic placement of a large number of Fusarium species. Furthermore, the correlation between trichothecene production and rDNA sequence indicated the need for a revision of the systematics of the Fusarium strains.

Abstract

Background The fungal genus Aspergillus is of critical importance to humankind. Species include those with industrial applications, important pathogens of humans, animals and crops, a source of potent carcinogenic contaminants of food, and an important genetic model. The genome sequences of eight aspergilli have already been explored to investigate aspects of fungal biology, raising questions about evolution and specialization within this genus. Results We have generated genome sequences for ten novel, highly diverse Aspergillus species and compared these in detail to sister and more distant genera. Comparative studies of key aspects of fungal biology, including primary and secondary metabolism, stress response, biomass degradation, and signal transduction, revealed both conservation and diversity among the species. Observed genomic differences were validated with experimental studies. This revealed several highlights, such as the potential for sex in asexual species, organic acid production genes being a key feature of black aspergilli, alternative approaches for degrading plant biomass, and indications for the genetic basis of stress response. A genome-wide phylogenetic analysis demonstrated in detail the relationship of the newly genome sequenced species with other aspergilli. Conclusions Many aspects of biological differences between fungal species cannot be explained by current knowledge obtained from genome sequences. The comparative genomics and experimental study, presented here, allows for the first time a genus-wide view of the biological diversity of the aspergilli and in many, but not all, cases linked genome differences to phenotype. Insights gained could be exploited for biotechnological and medical applications of fungi.

Abstract

A study was conducted to identify 77 black aspergilli isolated from grape berries collected in a 2-year survey in 16 vineyards throughout Italy, using amplified fragment length polymorphisms (AFLPs) and genomic DNA sequences. The results showed that ochratoxin is produced by A. tubingensis and it may be responsible for the ochratoxin contamination of wine in Italy, when it is together with A. carbonarius and A. niger.

Abstract

[Synopsis] Fusarium proliferatum is a worldwide pathogen of several agriculturally important plants. The species belongs to Liseola section of the Fusarium genus and its teleomorph belongs to the Gibberella fujikuroi complex, where at least six different mating populations (MPs), which represent different biological species, have been recognized. Strains of F. proliferatum belong to MPs C and D and they can produce a broad range of toxins such as fumonisin B_1, (FB_1), beauvericin (BEA) and fusaproliferin (FUP). In order to study the intraspecific variability of this species, 73 strains of F. proliferatum isolated from various plants (asparagus, maize, rice, tomato and wheat) and different geographic origin, were characterized for their ability to produce toxins (FB_1, BEA, and FUP), and their belonging to a specific MP. All strains here reported but the isolates from rice belonged to MP D, while all the isolates from rice belonged to MP C. Toxigenic data confirmed significant differences among MPs with respect to secondary metabolism. Fumonisin B_1, and FUP were produced only by D MP, whereas BEA was produced by members of both MPs. Moreover, in order to study the genetic variability and to detect molecular markers to differentiate isolates from different hosts, enomic DNA was analyzed by RAPDs using decamer oligonucleotides of arbitrary base composition. Preliminary data showed that genomic variability between toxigenic isolates of MPs C and D was found by using a specific primer and RAPDs also revealed an intraspecific polymorphism among MP D isolates, mostly according with their original host plants. Moreover, we report here data of a previous DNA comparison by RAPDs, between F. proliferatum and the closely related species F. verticillioides, from which interspecific polymorphic markers were found.

23. Biotyping of some toxigenic Fusarium species by the yeast killer system

Abstract

Sixty selected killer yeasts were tested against 44 isolates of Fusarium belonging to 5 different species (14 isolates of F. sporotrichioides, 10 of F. chlamydosporum, 10 of F. tricinctum, 6 of F. solani and 4 of F. poae). Based on the system used, it was possible to observe markedly different biotypes both among and within Fusarium species. In particular, F. sporotrichioides, F. poae, F. chlamydosporum, and F. tricinctum showed 8, 4, 6, 6 different biotypes, respectively, whereas no susceptibility was found in F. solani. The results obtained suggest that the yeast killer system may be utilized in the evaluation of the biodiversity, as well as in the biological control and for epidemiological purposes.
