The study was conducted to evaluate the antioxidant and antipyretic activity of Adansonia digitata (African Baobab) fruit, leaf and bark extracts. Antioxidant activity of plant part extracts were determined by free radical Diphenylpicrylhydrazyl (DPPH) assay while the antipyretic activity was assessed using experimental animals. Antioxidant activity of leaf extract showed a value of 13.40±0.77, bark extract 29.23±1.69 while fruit extract gave a value of 39.21±2.26. For the Antipyretic activity results, the average temperature of Albino Wister rats was 21.65±0.96 oC before fever inducement and 30.64±1.37 oC after inducement. Following the administration of the leaf extract (400 mg/kg and 800 mg/kg), bark extract (400 mg/kg and 800 mg/kg) and fruit extract (400 mg/kg and 800 mg/kg), the body temperature of the albino rats fell to25.96±1.16 oC vs. 25.28±1.13 oC, 26.06±1.17 oC vs. 25.02±1.12 oC, and 25.78±1.15 oC vs. 24.30±1.09 oC respectively. The results showed that the fruit extract has a higher antioxidant and antipyretic activity than bark and leaf extracts.

Microdochium nivale is one of the most harmful fungal diseases, causing colossal yield losses and deteriorating grain quality. Wheat genotypes from the world collection of the N.I. Vavilov Institute (VIR) were evaluated for fifty years to investigate their resistance to biotic stress factors (M. nivale). Between 350 to 1085 of winter wheat genotypes were investigated annually. Ten out of fifty years were identified as rot epiphytotics (1978, 1986, 1989, 1990, 1993, 1998, 2001, 2003, 2005 and 2021). The wheat collection was investigated by following the VIR methodological requirements and CMEA unified classification of Triticum aestivum L. The field investigations were carried out in the early spring during fixed-route observations and data collection was included on the spread and development degree of the disease, followed by microbiological and microscopic pathogen identifications. The observations revealed that the primary reason for pink snow mold to infect the wheat crops was abiotic stress factors, such as thawed soil covered in snow that increased the soil temperature by 1.0–4.6 °C above normal. Under these conditions, the plants kept growing, quickly exhausting their carbohydrate and protein resources, thus weakening their immune systems, which made them an easy target for different infections, mainly cryophilic fungi, predominantly Microdochium nivale in the Moscow region. In some years, the joint effect of abiotic and biotic stresses caused crop failure, warranting the replanting of the spring wheat. The investigated wheat genotypes exhibited variable resistance to pink snow mold. The genotypes Mironovskaya 808 (k-43920) from Ukraine;l Nemchinovskaya 846 (k-56861), from Russia; Novobanatka (k-51761) from Yugoslavia; Liwilla (k-57580) from Poland; Zdar (UH 7050) from the Czech Republic; Maris Plowman (k-57944) from the United Kingdom; Pokal (k-56827) from Austria; Hvede Sarah (k-56289) from Denmark; Moldova 83 (k-59750) from Romania; Compal (k-57585) from Germany; Linna (k-45889) from Finland and Kehra (k-34228) from Estonia determined the sources, stability and tolerance to be used in advanced breeding programs.

In the light of Vavilov’s Law, grain traumatization in the standing crop of wheat and other crops due to the enzyme stage of enzyme-mycotic seed depletion (EMSD) was confirmed, the parameters of open and hidden harmfulness were detected, and a scale of plant resistance to such traumatization was developed. The current study demonstrates that pathogen contamination in grains occurs before harvesting and its degree is determined by favorable humidity and temperature conditions and by the open and hidden grain traumatization due to the enzyme stage of EMSD, i.e., the grain’s hydrolytic enzymes providing a growth substrate for a fungal spread that is later substituted by pathogen enzymes leading to grain spoiling and self-warming. The most common technique to preserve grain quality is to support a moisture level that prevents further spreading of the fungi. The grains that are contaminated with very low temperature and humidity levels facilitate the germinability and high quality of the grain. The new ways to withstand EMSD should, first of all, include a selection of activities. Using biological, biochemical and physical (X-ray) methods, genetic sources of resistance towards EMSD were found in the VIR world collection that is recommended for further selection. These sources have become a basis for the varieties, such as Moskovskaya 39, Ilot (winter wheat), Gremme and Gremme 2U (hulless spelt), Alcoran (winter spelt) and Kanysh (spring wheat).

Introdution Depression is ranked third highest among disorders responsible for global disease burden. Preventive strategies are more cost effective than treatment alone. Depression is biological disease having serious psychological and social consequences comprehensive program based on biopsychsocial model can curtail its mortality and morbidity. Method Pubmed search was made to study about prevention of depression. Result Different studies suggested food, exercise relaxation and social support system to prevent depression. At biological level food is the first item. It should include omega 3, antioxidant, vitamin D3, B6, B12, folic acid, fiber. Foods with anti inflammatory effects too are recommended. Moderate exercise has biological effects mediated by endorphins. It is advised to avoid proinflammatory foods, refined sugar, smoking, and addiction. Whole some sexual life has anti anxiety and antidepressant effects. At psychological level high risk population is screened. Susceptible are taught cognitive restructuring and coping strategies for stress. At social level greater integration with family and friend can help the person. Disscussion Essential ingredients like omega 3 is derived from fish, nuts seed oils. Fruit and vegetable and beans are rich in antioxidant, vitamins and fiber. Healthy food maintains hippocampal volume, which gets shrunken by unhealthy food. Increased levels of homocystine due to inflammation can decrease monoamine neurotransmitters. Anti-inflammatory food include turmeric and garlic. Sex increase blood levels of oxytocin during arousal dopamine serotonin and endorphins at orgasm. Cotton (Gossypium hirsutum L.) is of great economic importance as a cultivated crop in many parts of the world. In addition to being a pillar of the textile industry, cotton and its byproducts are used for livestock feed, seed oil, and other products. Bacillus thuringiensis crystal toxin (Bt) expression in cotton provides effective protection against chewing insects but does not defend plants from piercing/sucking insect pests. With the aim to create transgenic plants with resistance against piercing/sucking pests, we used Agrobacterium-mediated genetic transformation of cotton cultivar Coker 312 to express the Allium sativum leaf agglutinin (ASLA) gene from the phloem-specific rolC promoter. The ASLA transgene was stably inherited and showed Mendelian segregation in the T1 generation. Transgenic lines, expressing the ASLA gene, showed explicit resistance against major sap-sucking pests. Green peach aphid (Myzus persicae Sulzer) choice assays showed that 75% of aphids preferred untransformed cotton plants relative to those expressing the ASLA gene. In detached leaf bioassays, plants expressing ASLA caused 82% aphid mortality and 44-53% reduction in fecundity. Clip cage bioassays with whiteflies (Bemisia tabaci Gennadius) showed 74-82% mortality and 44-60% decrease in fecundity due to ASLA gene expression. In whole plant bioassays, whiteflies showed 77% mortality and a 54% decrease in fecundity on ASLA transgenics. Importantly, we did not observe a negative effect of the ASLA gene on ladybugs (Coccinella septempunctata) that consumed these whiteflies. Together, our findings demonstrate the potential of ASLA-transgenic cotton for providing protection against two devastating insect pests, whiteflies and aphids. Improving the low productivity levels of native cattle breeds in smallholder farming systems is a pressing concern in Pakistan. Crossbreeding high milk-yielding holstein friesian (HF) breed with the adaptability and heat tolerance of Sahiwal cattle has resulted in offspring that are well-suited to local conditions and exhibit improved milk yield. The exploration of how desirable traits in crossbred dairy cattle are selected has not yet been investigated. This study aims to provide the first overview of the selective pressures on the genome of crossbred dairy cattle in Pakistan. A total of eighty-one crossbred, thirty-two HF and twenty-four Sahiwal cattle were genotyped, and additional SNP genotype data for HF and Sahiwal were collected from a public database to equate the sample size in each group. Within-breed selection signatures in crossbreds were investigated using the integrated haplotype score. Crossbreds were also compared to each of their parental breeds to discover between-population signatures of selection using two approaches: cross-population extended haplotype homozygosity and fixation index. We identified several overlapping genes associated with production, immunity, and adaptation traits, including U6, TMEM41B, B4GALT7, 5S_rRNA, RBM27, POU4F3, NSD1, PRELID1, RGS14, SLC34A1, TMED9, B4GALT7, OR2AK3, OR2T16, OR2T60, OR2L3, and CTNNA1. Coronary artery disease (CAD) remains a significant global health concern with considerable high morbidity and mortality and its development is influenced by various genetic and environmental factors. Proprotein convertase subtilisin/kexin type 9 (PCSK9) is a vital regulator of low-density lipoprotein receptor (LDLR) metabolism, directly impacting serum cholesterol levels. However, its role in development of CAD is not fully understood. The aim of this study was to assess the association between the level of PCSK9 and coronary lesion severity in patients with CAD. A case-control study using consecutive sampling was conducted among CAD patients at H. Adam Malik General Hospital and Murni Teguh Memorial Hospital, Medan, Indonesia. A total of 200 CAD patients were divided into two groups based on the SYNTAX score: control (score ≤22, n=100) and case (score >22, n=100). Plasma PCSK9 levels were measured from venous blood using quantitative sandwich enzyme immunoassay. The Chi-squared test was used to analyze the data. Our data suggested that PCSK9 level was associated with coronary lesion severity ( p <0.001) of which high PCSK9 level was associated with severe coronary lesion. We also found that hypertension ( p <0.001), smoking ( p =0.072), diabetes ( p <0.001), dyslipidemia ( p <0.001), obesity ( p =0.023), and family history ( p =0.001) were associated with lesion severity. Using the receiver operating characteristic (ROC) curve analysis, the cut-off 70.35 ng/mL of PCSK9 had sensitivity 75% and specificity 78% to predict severe coronary lesion. Linkage disequilibrium (LD) affects genomic studies accuracy. High-density genotyping platforms identify SNPs across animal genomes, increasing LD evaluation resolution for accurate analysis. This study aimed to evaluate the decay and magnitude of LD in a cohort of 81 crossbred dairy cattle using the GGP&#95;HDv3&#95;C Bead Chip. After quality control, 116,710 Single Nucleotide Polymorphisms (SNPs) across 2520.241 Mb of autosomes were retained. LD extent was assessed between autosomal SNPs within a 10 Mb range using the r 2 statistics. LD value declined as inter-marker distance increased. The average r 2 value was 0.24 for SNP pairs < 10 kb apart, decreasing to 0.13 for 50-100 kb distances. Minor allele frequency (MAF) and sample size significantly impact LD. Lower MAF thresholds result in smaller r 2 values, while higher thresholds show increased r 2 values. Additionally, smaller sample sizes exhibit higher average r 2 values, especially for larger physical distance intervals (> 50 kb) between SNP pairs. Effective population size and inbreeding coefficient were 150 and 0.028 for the present generation, indicating a decrease in genetic diversity over time. These findings imply that the utilization of high-density SNP panels and customized/breed-specific SNP panels represent a highly favorable approach for conducting genome-wide association studies (GWAS) and implementing genomic selection (GS) in the Bos indicus cattle breeds, whose genomes are still largely unexplored. Yellow mosaic disease (YMD) is one of the major devastating constraints to soybean production in Pakistan. In the present study, we report the identification of resistant soybean germplasm and a novel mutation linked with disease susceptibility. Diverse soybean germplasm were screened to identify YMD-resistant lines under natural field conditions during 2016-2020. The severity of YMD was recorded based on symptoms and was grouped according to the disease rating scale, which ranges from 0 to 5, and named as highly resistant (HR), moderately resistant (MR), resistant (R), susceptible (S), moderately susceptible (MS), and highly susceptible (HS), respectively. A HR plant named "NBG-SG Soybean" was identified, which showed stable resistance for 5 years (2016-2020) at the experimental field of the National Institute for Biotechnology and Genetic Engineering (NIBGE), Faisalabad, Pakistan, a location that is a hot spot area for virus infection. HS soybean germplasm were also identified as NBG-47 (PI628963), NBG-117 (PI548655), SPS-C1 (PI553045), SPS-C9 (PI639187), and cv. NARC-2021. The YMD adversely affected the yield and a significant difference was found in the potential yield of NBG-SG-soybean (3.46 ± 0.13 a t/ha) with HS soybean germplasm NARC-2021 (0.44 ± 0.01 c t/ha) and NBG-117 (1.12 ± 0.01 d t/ha), respectively. The YMD incidence was also measured each year (2016-2020) and data showed a significant difference in the percent disease incidence in the year 2016 and 2018 and a decrease after 2019 when resistant lines were planted. The resistance in NBG-SG soybean was further confirmed by testing for an already known mutation (SNP at 149 th position) for YMD in the Glyma.18G025100 gene of soybean. The susceptible soybean germplasm in the field was found positive for the said mutation. Moreover, an ortholog of the CYR-1 viral resistance gene from black gram was identified in soybean as Glyma.13G194500 , which has a novel deletion (28bp/90bp) in the 5`UTR of susceptible germplasm. The characterized soybean lines from this study will assist in starting soybean breeding programs for YMD resistance.
