Abstract

Nannopus palustris Brady, 1880 is a free-living widely distributed harpacticoid copepod, which has been formerly assumed to be a single, cosmopolitan but highly variable species. We compared several geographically distant N. palustris populations in terms of their morphology and genetics. Populations from the White Sea (WS), the North Sea (NS), the Black Sea (BS) and two sympatric morphs from South Carolina, USA (SC notched and SC straight morphs), were considered. The NS, BS and to a lesser extent SC notched specimens were morphologically similar and partly coincided to the ‘canonical’ description of the species. By contrast, WS population showed remarkable anatomical and morphometric peculiarities that correspond to some earlier descriptions. Genetic analyses of mitochondrial (cytochrome b) and nuclear (28S rDNA) genes demonstrated the significant distinctness among WS, both SC and (NS+BS) populations, the latter two being genetically indistinguishable. Concordance between mitochondrial and nuclear gene trees and morphological data supports that N. palustris is in fact composed of several pseudo-sibling species, which are genetically and morphologically divergent. Neither correlation between genetic divergence and geographical distance nor significant intrapopulation diversity was found for these species. Taxonomic status, distribution and phylogenetic relationships of the species within the Nannopus genus need to be reconsidered. A further subdivision of species complexes might have important implications for the analysis of biodiversity of benthic copepods and consequently for the interpretation of their (species-specific) ecological function.

Abstract

Six postglacial lakes were studied along both sides of the Kamchatka central mountain range, Russia. Pairs of local morphotypes of species of Arctic charrs, Salvelinus spp., have previously been described from the southernmost lakes while the fish fauna of the four northernmost lakes was studied here for the first time. Phenotypic data support the division of Kamchatkan lacustrine charrs into two groups according to the number of gill rakers and pyloric caeca, as well as snout-dorsal and snout-ventral distances (MANOVA, P < 0·001). These groups respectively correspond to phenotypes commonly referred to as Salvelinus malma and Salvelinus taranetzi. To clarify the identity of these groups, D-loop and cytochrome b ( cytb) region sequences were analysed. Haplotype network analysis of mtDNA shows the salmonids inhabiting four lakes on the south and north are phylogenetically close to either Beringian S. malma or to S. taranetzi from the Chukotka and Kolyma River basins (the mean ± s.e. pairwise per cent sequence divergence is 0·006 ± 0·001). Phenotype-genotype discordance suggests that mitochondrial introgression between species has occurred in the two smallest lakes (<0·5 km2) in the central part of the peninsula. Identical haplotypes of D-loop and cytb regions were found for the populations of S. taranetzi from the most distant southern and northern lakes, indicating all lakes were colonized by both species simultaneously after the last glacial maximum. Salvelinus taranetzi may have colonized the Kamchatka peninsula from one or both of two different source regions: the Arctic Beringia and the northern coast of the Okhotsk Sea. Abstract

Temperature acclimation of poikilotherms entails metabolic rearrangements provided through the variation of enzyme properties. However, in most cases the underlying molecular mechanisms resulting in structural changes of enzymes are obscure. The present study reports that acclimation to low (5°C) and high (18°С) temperatures causes differential expression of alternative forms of the LDH-A gene in white skeletal muscle of weatherfish Misgurnus fossilis. Two isoforms of LDH-A mRNA were isolated and characterised: a short (1332 bp) and long (1550 bp) form, which differ in the length of 3'UTR, despite identical ORF lengths. These two mRNAs have 44 nucleotide point mismatches along the sequence resulting in three amino acid mismatches (Gly214Val; Val304Ile and Asp312Glu) between protein products from short and long forms of mRNA. Correspondingly, LDH-A subunit synthesized from short form of mRNA has been denoted as S-subunit, whereas the other one is L-subunit. It is expected that the L-subunit is more aliphatic due to properties of the mismatched amino acids. According to molecular modelling of M.fossilis LDH-A the Val304Ile mismatch is located in the subunit contacts of the tetramer, whereas the remaining two surround the contact area, which should manifest in kinetic and thermodynamic properties of the assembled tetramer. In warm acclimated fish the relative ratio between short and long isoforms of the LDH-A mRNA expression is around 5:1, whereas in cold acclimated fish expression of long mRNA is reduced to almost zero. This indicates that at low temperature the pool of overall tetrameric LDH-A is more homogeneous in terms of subunit composition. The temperature acclimation pattern of proportional pooling of subunits with different kinetic and thermodynamic properties of the tetrameric enzyme could result in the fine tuning of properties of overall skeletal LDH-A, which is in line with previously observed kinetic and thermodynamic differences between cold and wa

Abstract

The article presents a study which evaluates the genetic variation in the mitochondrial control region of bowhead whales in the Western Arctic. Study shows that there were no significant differences detected in spatial and temporal comparisons along Alaska's North Slope. However, there was a significant difference between autumn and spring whales from St. Lawrence Island. Moreover, it was concluded that the genetic variation of bowhead whales is established from mitochondrial cytochrome B gene sequences amplified from ancient DNA.
