Abstract: Planetary systems in mean motion resonances hold a special place among the planetary population. They allow us to study planet formation in great detail as dissipative processes are thought to have played an important role in their existence. Additionally, planetary masses in bright resonant systems may be independently measured both by radial velocities (RVs) and transit timing variations (TTVs). In principle, they also allow us to quickly determine the inclination of all planets in the system, as for the system to be stable, they are likely all in coplanar orbits. To describe the full dynamical state of the system, we also need the stellar obliquity that provides the orbital alignment of a planet with respect to the spin of their host star and can be measured thanks to the Rossiter-McLaughlin effect. It was recently discovered that HD 110067 harbours a system of six sub-Neptunes in resonant chain orbits. We here analyze an ESPRESSO high-resolution spectroscopic time series of HD 110067 during the transit of planet c. We find the orbit of HD 110067 c to be well aligned with sky projected obliquity $\lambda =6^{+24}_{-26}$ deg. This result is indicative that the current architecture of the system has been reached through convergent migration without any major disruptive events. Finally, we report transit-timing variation in this system as we find a significant offset of 19 $\pm$ 4 minutes in the center of the transit compared to the published ephemeris., Comment: Accepted to A&A

Abstract: The diffusion of emergent pathogens in grapevine nursery is increasingly compromising the sanitary quality of the propagation material. The complexity of the problem lies on the plurality of plant diseases involved and on the high environmental and economic impact of the conventional methods applied to comply the phytosanitary standards. By a critical point analysis, the study suggests the development of nursery strategies focused on the sanitary quality of the production and on its long-term environmental sustainability.

Abstract: The ExoClock project has been created with the aim of increasing the efficiency of the Ariel mission. It will achieve this by continuously monitoring and updating the ephemerides of Ariel candidates over an extended period, in order to produce a consistent catalogue of reliable and precise ephemerides. This work presents a homogenous catalogue of updated ephemerides for 450 planets, generated by the integration of $\sim$18000 data points from multiple sources. These sources include observations from ground-based telescopes (ExoClock network and ETD), mid-time values from the literature and light-curves from space telescopes (Kepler/K2 and TESS). With all the above, we manage to collect observations for half of the post-discovery years (median), with data that have a median uncertainty less than one minute. In comparison with literature, the ephemerides generated by the project are more precise and less biased. More than 40\% of the initial literature ephemerides had to be updated to reach the goals of the project, as they were either of low precision or drifting. Moreover, the integrated approach of the project enables both the monitoring of the majority of the Ariel candidates (95\%), and also the identification of missing data. The dedicated ExoClock network effectively supports this task by contributing additional observations when a gap in the data is identified. These results highlight the need for continuous monitoring to increase the observing coverage of the candidate planets. Finally, the extended observing coverage of planets allows us to detect trends (TTVs - Transit Timing Variations) for a sample of 19 planets. Abstract: The ExoClock project is an inclusive, integrated, and interactive platform that was developed to monitor the ephemerides of the Ariel targets to increase the mission efficiency. The project makes the best use of all available resources, i.e., observations from ground telescopes, mid-time values from the literature and finally, observations from space instruments. Currently, the ExoClock network includes 280 participants with telescopes capable of observing 85\% of the currently known Ariel candidate targets. This work includes the results of $\sim$1600 observations obtained up to the 31st of December 2020 from the ExoClock network. These data in combination with $\sim$2350 mid-time values collected from the literature are used to update the ephemerides of 180 planets. The analysis shows that 40\% of the updated ephemerides will have an impact on future scheduling as either they have a significantly improved precision, or they have revealed biases in the old ephemerides. With the new observations, the observing coverage and rate for half of the planets in the sample has been doubled or more. Finally, from a population perspective, we identify that the differences in the 2028 predictions between the old and the new ephemerides have an STD that is double what is expected from gaussian uncertainties. These findings have implications for planning future observations, where we will need to account for drifts potentially greater than the prediction uncertainties. Abstract: Recent publications have argued that there are potentially serious consequences for researchers in recognising distinct genera in the terminal fusarioid clade of the family Nectriaceae. Thus, an alternate hypothesis, namely a very broad concept of the genus Fusarium was proposed. In doing so, however, a significant body of data that supports distinct genera in Nectriaceae based on morphology, biology, and phylogeny is disregarded. A DNA phylogeny based on 19 orthologous protein-coding genes was presented to support a very broad concept of Fusarium at the F1 node in Nectriaceae. Here, we demonstrate that re-analyses of this dataset show that all 19 genes support the F3 node that represents Fusarium sensu stricto as defined by F. sambucinum (sexual morph synonym Gibberella pulicaris). The backbone of the phylogeny is resolved by the concatenated alignment, but only six of the 19 genes fully support the F1 node, representing the broad circumscription of Fusarium. Furthermore, a re-analysis of the concatenated dataset revealed alternate topologies in different phylogenetic algorithms, highlighting the deep divergence and unresolved placement of various Nectriaceae lineages proposed as members of Fusarium. Species of Fusarium s. str. are characterised by Gibberella sexual morphs, asexual morphs with thin- or thick-walled macroconidia that have variously shaped apical and basal cells, and trichothecene mycotoxin production, which separates them from other fusarioid genera. Here we show that the Wollenweber concept of Fusarium presently accounts for 20 segregate genera with clear-cut synapomorphic traits, and that fusarioid macroconidia represent a character that has been gained or lost multiple times throughout Nectriaceae. Thus, the very broad circumscription of Fusarium is blurry and without apparent synapomorphies, and does not include all genera with fusarium-like macroconidia, which are spread throughout Nectriaceae (e.g., Cosmosporella, Macroconia, Microcera). In this study four new genera are introduced, along with 18 new species and 16 new combinations. These names convey information about relationships, morphology, and ecological preference that would otherwise be lost in a broader definition of Fusarium. To assist users to correctly identify fusarioid genera and species, we introduce a new online identification database, Fusarioid-ID, accessible at www.fusarium.org. The database comprises partial sequences from multiple genes commonly used to identify fusarioid taxa (act1, CaM, his3, rpb1, rpb2, tef1, tub2, ITS, and LSU). In this paper, we also present a nomenclator of names that have been introduced in Fusarium up to January 2021 as well as their current status, types, and diagnostic DNA barcode data. In this study, researchers from 46 countries, representing taxonomists, plant pathologists, medical mycologists, quarantine officials, regulatory agencies, and students, strongly support the application and use of a more precisely delimited Fusarium (= Gibberella) concept to accommodate taxa from the robust monophyletic node F3 on the basis of a well-defined and unique combination of morphological and biochemical features. This F3 node includes, among others, species of the F. fujikuroi, F. incarnatum-equiseti, F. oxysporum, and F. sambucinum species complexes, but not species of Bisifusarium [F. dimerum species complex (SC)], Cyanonectria (F. buxicola SC), Geejayessia (F. staphyleae SC), Neocosmospora (F. solani SC) or Rectifusarium (F. ventricosum SC). Abstract: Planetary systems in mean motion resonances hold a special place among the planetary population. They allow us to study planet formation in great detail as dissipative processes are thought to have played an important role in their existence. Additionally, planetary masses in bright resonant systems can be independently measured via both radial velocities and transit timing variations. In principle, they also allow us to quickly determine the inclination of all planets in the system since, for the system to be stable, they are likely all in coplanar orbits. To describe the full dynamical state of the system, we also need the stellar obliquity, which provides the orbital alignment of a planet with respect to the spin of its host star and can be measured thanks to the Rossiter–McLaughlin effect. It was recently discovered that HD 110067 harbors a system of six sub-Neptunes in resonant chain orbits. We here analyze an ESPRESSO high-resolution spectroscopic time series of HD 110067 during the transit of planet c. We find the orbit of HD 110067 c to be well aligned, with a sky-projected obliquity of λ =6+24-26 deg. This result indicates that the current architecture of the system was reached through convergent migration without any major disruptive events. Finally, we report transit-timing variation in this system as we find a significant offset of 19 ± 4 min in the center of the transit compared to the published ephemeris. [ABSTRACT FROM AUTHOR]

Abstract: Phomopsis cane and leaf spot (PCLS), known in Europe as “excoriose,” is an important fungal disease of grapevines caused by Diaporthe spp., and most often by Diaporthe ampelina (synonym Phomopsis viticola). PCLS is re-emerging worldwide, likely due to climate change, changes in the management of downy mildew from calendar- to risk-based criteria that eliminate early-season (unnecessary) sprays, and the progressive reduction in the application of broad-spectrum fungicides. In this study, a mechanistic model for D. ampelina infection was developed based on published information. The model accounts for the following processes: (i) overwintering and maturation of pycnidia on affected canes; (ii) dispersal of alpha conidia to shoots and leaves; (iii) infection; and (iv) onset of disease symptoms. The model uses weather and host phenology to predict infection periods and disease progress during the season. Model output was validated against 11 independent PCLS epidemics that occurred in Italy (4 vineyards in 2019 and 2020) and Montenegro (3 vineyards in 2020). The model accurately predicted PCLS disease progress, with a concordance correlation coefficient (CCC) = 0.925 between observed and predicted data. A ROC analysis (AUROC>0.7) confirmed the ability of the model to predict the infection periods leading to an increase in PCLS severity in the field, indicating that growers could use the model to perform risk-based fungicide applications.

Abstract: Recent publications have argued that there are potentially serious consequences for researchers in recognising distinct genera in the terminal fusarioid clade of the family Nectriaceae. Thus, an alternate hypothesis, namely a very broad concept of the genus Fusarium was proposed. In doing so, however, a significant body of data that supports distinct genera in Nectriaceae based on morphology, biology, and phylogeny is disregarded. A DNA phylogeny based on 19 orthologous protein-coding genes was presented to support a very broad concept of Fusarium at the F1 node in Nectriaceae. Here, we demonstrate that re-analyses of this dataset show that all 19 genes support the F3 node that represents Fusarium sensu stricto as defined by F. sambucinum (sexual morph synonym Gibberella pulicaris). The backbone of the phylogeny is resolved by the concatenated alignment, but only six of the 19 genes fully support the F1 node, representing the broad circumscription of Fusarium. Furthermore, a re-analysis of the concatenated dataset revealed alternate topologies in different phylogenetic algorithms, highlighting the deep divergence and unresolved placement of various Nectriaceae lineages proposed as members of Fusarium. Species of Fusarium s. str. are characterised by Gibberella sexual morphs, asexual morphs with thin- or thick-walled macroconidia that have variously shaped apical and basal cells, and trichothecene mycotoxin production, which separates them from other fusarioid genera. Here we show that the Wollenweber concept of Fusarium presently accounts for 20 segregate genera with clear-cut synapomorphic traits, and that fusarioid macroconidia represent a character that has been gained or lost multiple times throughout Nectriaceae. Thus, the very broad circumscription of Fusarium is blurry and without apparent synapomorphies, and does not include all genera with fusarium-like macroconidia, which are spread throughout Nectriaceae (e.g., Cosmosporella, Macroconia, Microcera). Abstract: Recent publications have argued that there are potentially serious consequences for researchers in recognising distinct genera in the terminal fusarioid clade of the family Nectriaceae. Thus, an alternate hypothesis, namely a very broad concept of the genus Fusarium was proposed. In doing so, however, a significant body of data that supports distinct genera in Nectriaceae based on morphology, biology, and phylogeny is disregarded. A DNA phylogeny based on 19 orthologous protein-coding genes was presented to support a very broad concept of Fusarium at the F1 node in Nectriaceae. Here, we demonstrate that re-analyses of this dataset show that all 19 genes support the F3 node that represents Fusarium sensu stricto as defined by F. sambucinum (sexual morph synonym Gibberella pulicaris). The backbone of the phylogeny is resolved by the concatenated alignment, but only six of the 19 genes fully support the F1 node, representing the broad circumscription of Fusarium. Furthermore, a re-analysis of the concatenated dataset revealed alternate topologies in different phylogenetic algorithms, highlighting the deep divergence and unresolved placement of various Nectriaceae lineages proposed as members of Fusarium. Species of Fusarium s. str. are characterised by Gibberella sexual morphs, asexual morphs with thin- or thick-walled macroconidia that have variously shaped apical and basal cells, and trichothecene mycotoxin production, which separates them from other fusarioid genera. Here we show that the Wollenweber concept of Fusarium presently accounts for 20 segregate genera with clear-cut synapomorphic traits, and that fusarioid macroconidia represent a character that has been gained or lost multiple times throughout Nectriaceae. Abstract: Botryosphaeria dieback (BD) is an important trunk disease affecting grapevines. Several Lasiodiplodia species have been shown to be involved in BD affecting the perennial organs of grapevine, mainly causing cankers. (R)-(-)-mellein and tyrosol, two well-known fungal phytotoxins, were isolated from the organic extract of culture filtrate of Lasiodiplodia laeliocattleyae (syn. egyptiacae), which had been isolated from grapevines affected by BD in Brazil. This increases knowledge of the secondary metabolites produced by Lasiodiplodia species, confirming that (R)-(-)-mellein is a toxin typically produced by Botryosphaericeae species.

Abstract: The control of grapevine trunk diseases (GTDs) represents a big challenge for grapevine nurseries due to the high infection risks on the propagating material. Plant material disinfection before storage and the grafting practice are both potential factors able to affect the future GTDs development in young vines in field. In this framework, this research is aimed to evaluate the microbiome (especially taking into account GTDs pathogens) in the propagation material undergone through different disinfection treatments, and to compare the impact of three grafting methods on the formation of an active and functional vascular system. To assess the first aim, the microbiome of the scion (cv. Glera) and rootstock (cv. Kober 5BB) (by DNA sequencing technologies and intelligent computing) was preliminary analyzed just before grafting, in order to detect differences on the fungal and bacterial community prior treatment. For the second aim, three methods of grafting, (i) Omega graft, (ii) Whip and Tongue graft and (iii) Full cleft graft, were compared by observing xylogenesis and vascular tissue formation at the grafting point, in order to detect the factors potentially influencing the vine physiology. Early results showed a distinct profile in fungi and bacteria for both rootstock and scion. In rootstock, a higher abundance of GTDs agents was detected, especially in the bark. The anatomical observation on grapevine cuttings, immediately after callusing, revealed differences between all the three grafting methods: the full cleft graft showed the most complete and homogenous callus, which is the ideal condition to avoid the presence of necrosis, potentially colonized by the GTDs pathogens.

Abstract: Among grapevine trunk diseases-related fungi, the basidiomycete Fomitiporia mediterranea (Fmed) is responsible for wood-decay in Vitis vinifera, showing an intermediate behaviour between brownand white-rot fungi. Nowadays, it is widely accepted that, in brown-rot fungi, wood decay is the result of synergic enzymatic and non-enzymatic degrading action. In the first one, enzymatic action, laccases and manganese peroxidases (MnP) play a major role in catalysing lignin degradation, splitting C?-C?, ?-aryl, C1-C? and aromatic C-C bonds. In the second one, non- enzymatic action, the lignin/cellulose degradation is attributed to the non-enzymatic Fenton/Haber Weiss pathway, generating hydroxyl radicals within the wood cell, also thanks to Fe oxidation. Because low-molecular weight (LMW) iron-binding compounds mediate this wood degradation, the mechanism has been named "chelator-mediated Fenton" (CMF) reaction. Since it is still not clear if Fmed has both mechanisms of lignin/cellulose degradation (enzymatic and CMF reactions), these two pathways were evaluated screening 13 Fmed strains, assaying: i) laccase and MnP activity; ii) the strains capability to produce LMW iron-binding compounds, ferrous-iron (Fe2+) accumulation and the consequent production of hydroxyl radicals.

29. Innovative delivery of Cu(II) ions by a nano-structured hydroxyapatite: potential application in planta to enhance the sustainable control of Plasmopara viticola

Abstract: Downy mildew caused by Plasmopara viticola is probably the most serious disease affecting grapevine (Vitis vinifera L.) and is capable of causing consistent yield losses. In organic viticulture, the only acceptable and effective means to control the disease is by applications of copper-based fungicides. However, the use of copper in agriculture is expected to be further restricted by European countries because of its critical ecotoxicological and phytotoxicological profile. Research on ways to reduce the effective amounts of copper by developing innovative formulations as well as optimization of the distribution and persistence of copper-based pesticides for downy mildew control appears to be a promising approach. The present research investigated the delivery properties of biomimetic synthetic hydroxyapatite (HA) to enhance the biological activity of Cu(II) ions. To this aim, four Cu(II) compounds were formulated with the innovative HA component and applied in an in vitro antifungal assay against Botrytis cinerea, a common grapevine pathogen suitable for in vitro activity tests, and finally in in-planta efficacy assays against P. viticola under greenhouse conditions. The in vitro results highlighted a different inhibition activity for each Cu(II) compound and indicated a different interaction between the cupric compounds and HA, potentially related to different delivery mechanisms of Cu(II) from HA. Under greenhouse conditions, further findings on the biological activity of the applied formulations were gained, especially on the efficacy of various concentrations of HA in the formulations, on the influence of dose variation of the formulation and the treatment efficiency, and persistence under rain-washing effect. The present study revealed promising findings on the formulation based on the HA particles and the soluble Cu(II) compound, which resulted in reduced disease severity and incidence in all the experimental conditions, including the lower Cu(II) dosage and the rain-washing effect. This suggests that coformulation of the three insoluble Cu(II) compounds with HA might significantly enhance the adsorption and release of Cu(II) ions by HA particles.

Abstract: [EN] Since the early 1990s, grapevine trunk diseases (GTDs) have posed threats for viticulture. Esca complex, Eutypa- and Botryosphaeria-diebacks, mostly detected in adult vineyards, are currently responsible for considerable economic losses in the main vine-growing areas of the world. Other GTDs, such as Petri- (Esca complex) and Black-foot diseases, are emerging problems in grapevine nurseries (resulting in grafting failures and/or loss of saleable plants) and in young vineyards. The impacts of GTDs in modern viticulture depend on several factors, some related to their complexity, and others linked to host plant characteristics, changes in vineyard management and to the scarcity of simple tools for their control. For these reasons control of GTDs remains difficult, also depending on knowledge transfer from research to field and vice versa. This paper outlines the main preventive and curative techniques currently applied, scientifically tested or not that have resulted from the outcomes of "Winetwork", a European Union funded project with special emphasis on the promising and innovative approaches.

In this study we explore the occurrence, diversity and pathogenicity of Diaporthe spp. associated with Vitis vinifera in major grape production areas of Europe and Israel, focusing on nurseries and vineyards. Surveys were conducted in Croatia, Czech Republic, France, Hungary, Israel, Italy, Spain and the UK. A total of 175 Diaporthe strains were isolated from asymptomatic and symptomatic shoots, branches and trunks. A multi-locus phylogeny was established based on five genomic loci (ITS, tef1, cal, his3 and tub2), and the morphological characters of the isolates were determined. Preliminary pathogenicity tests were performed on green grapevine shoots with representative isolates. The most commonly isolated species were D. eres and D. ampelina. Four new Diaporthe species described here as D. bohemiae, D. celeris, D. hispaniae and D. hungariae were found associated with affected vines. Pathogenicity tests revealed D. baccae, D. celeris, D. hispaniae and D. hungariae as pathogens of grapevines. No symptoms were caused by D. bohemiae. This study represents the first report of D. ambigua and D. baccae on grapevines in Europe. The present study improves our understanding of the species associated with several disease symptoms on V. vinifera plants, and provides useful information for effective disease management.

Abstract: Poplar short rotation coppice represents a very good opportunity for the production of bioenergy as a strategy against global changes. In recent years, numerous plantations in central Italy showed worrying stump mortality of uncertain cause. Investigations have therefore been conducted to determine the etiology and symptomatology of this phenomenon. The external symptoms begin with leaf yellowing and sometimes necrosis, microphyllia, and reduced shoots growth, followed by withering and death of the stump. Upon sectioning of the stump, we detected browning of internal tissues of roots and crown, gradually deepening in. In severe cases, we observed wood decay in the central part of the stump, eventually deepening into the root system. Cultures from the edge of necrotic tissue on PDA yielded isolates of the same fungus. Sequence analysis of the Internal Transcriber Spacer region (ITS1-5.8S-ITS2) and of the ?-tubulin gene showed that the isolates belong to the genus Cryptosphaeria. Species identification is still ongoing. Eventually, wood decay organisms belonging to the genera Pholiota and Collibia colonized the tissues as secondary invaders. Future goals will be to identify the species of the pathogen, define the disease predisposing factors and identify possible control measures.
