15. Cell wall disassembly is delayed by rhamnogalacturonate lyase gene silencing: potential role in fruit firmness

Abstract

Strawberry fruits greatly reduce their quality due to softening during ripening with economically important losses. Texture changes of fleshy fruits during ripening are mainly due to middle lamellae dissolution, cell-to-cell adhesion losses and wall weakening of parenchyma cells by the coordinated action of several cell wall enzymes. Pectin degradation has been proven a key factor in strawberry softening by functional analysis of several pectinase genes (polygalacturonase, pectate lyase and -galactosidase). The complexity and highly dynamic nature of pectins remains a challenge to fully elucidate structure-function relationships of pectins. In this work, we present the functional analysis of two independent strawberry transgenic lines with more than 95% silencing of a rhamnogalacturonate lyase gene (FaRGLyase1). Firmness of ripe fruit was significantly higher in both transgenic lines than in the control. Cell walls from these fruits were extracted and analyzed by glycan microarray profiling. This high‐throughput technique allows a wide screening of cell-wall glycan occurrence based on the detection of specific cell wall oligosaccharide epitopes by monoclonal antibodies and reveals profiles which can be used as potential fingerprints specific for a singular organ and/or developmental stage. Our microarray results showed that the silencing of FaRGLyase1 reduced degradation of several rhamnogalacturonan-I related epitopes, as expected. Additionally, comparison of transgenic cell walls from ripe fruits with those extracted from control fruits at different developmental stages (green, white and red) by hierarchical clustering, demonstrated a higher similarity of transgenic fruit cell walls with the control cell walls from fruits at the white stage. 16. Downregulation of NAC transcription factors modifies cell wall composition and increases strawberry fruit firmness

Abstract

The strawberry is a soft fruit with a very short post-harvest shelf life. The changes in texture during fruit ripening are mainly due to the dissolution of the middle lamellae, reducing cell-to-cell adhesion, and the weakening of parenchymal cell walls as result of the action of cell wall modifying enzymes. At present, no master regulator of this process has been discovered yet. NAC transcription factors have been involved in numerous physiological processes, including fruit ripening. In strawberry, the NAC family comprises more than 110 genes, and at least 6 of them are expressed during fruit development. In this research, we performed a functional analysis of two ripening-related NAC genes, FaNAC2 and FaNAC3, in Fragaria x ananassa Duch. cv. Chandler. Several RNAi transgenic lines showing low FaNAC2 or FaNAC3 mRNA levels in fruit were obtained through Agrobacterium-mediated transformation. These lines produced fruits significantly firmer than control at the ripe stage, being the increase in firmness higher in FaNAC2 silenced plants. Cell walls were extracted from ripe transgenic fruits and characterized by ELISA and Epitope Detection Chromatography (EDC), using monoclonal antibodies against different polysaccharide epitopes. FaNAC2 transgenic lines showed more extensive changes than FaNAC3; these modifications involved increased amounts of demethylated pectins (LM19) in water and CDTA fractions and an alteration of the lateral branches of RG-I, decreasing the amount of arabinan epitopes and increasing galactan epitopes detected by LM6 and LM5, respectively. The amount of arabinogalactan proteins recognized by the JIM13 antibody was also affected, decreasing in the Na2CO3 fraction and increasing in the 4M KOH and cellulase fraction of the transgenic lines.The results obtained indicate that NAC genes could be involved in the regulation of cell wall disassembly associated to strawberry fruit softening. Abstract

To identify novel genes associated with ALS, we undertook two lines of investigation. We carried out a genome-wide association study comparing 20,806 ALS cases and 59,804 controls. Independently, we performed a rare variant burden analysis comparing 1,138 index familial ALS cases and 19,494 controls. Through both approaches, we identified kinesin family member 5A (KIF5A) as a novel gene associated with ALS. Interestingly, mutations predominantly in the N-terminal motor domain of KIF5A are causative for two neurodegenerative diseases: hereditary spastic paraplegia (SPG10) and Charcot-Marie-Tooth type 2 (CMT2). In contrast, ALS-associated mutations are primarily located at the C-terminal cargo-binding tail domain and patients harboring loss-of-function mutations displayed an extended survival relative to typical ALS cases. Taken together, these results broaden the phenotype spectrum resulting from mutations in KIF5A and strengthen the role of cytoskeletal defects in the pathogenesis of ALS. Using a large-scale genome-wide association study and exome sequencing, we identified KIF5A as a novel gene associated with ALS. Our data broaden the phenotype resulting from mutations in KIF5A and highlight the importance of cytoskeletal defects in the pathogenesis of ALS.

Abstract

To analyze outcome after thymectomy in patients with myasthenia gravis (MG).Thirty-five patients with MG underwent surgery in our service between June 1987 and June 1998. Ten had associated thymomas. Preoperative Osserman classification showed 2 at level I, 20 at level IIA, 11 at level IIB and 2 at level III. Extended thymectomy through a medial sternotomy was performed in all.Postoperative complications developed in three patients (1 medullary aplasia, 1 postoperative reintubation, 1 myasthenic crisis). Mean follow-up was 89 months, with 22.8% achieving complete remission and 97.1% reporting improvements. The results were similar in the 10 patients with thymomas (20% full remission and 90% showing improvement). By DeFilippi classification, 22.8% were in class 1, 22.8% in class 2, 51.4% in class 3 and 2.8% in class 4. By Osserman classification, 9 were in the same category before and after surgery, 12 had improved one level, 10 had improved 2 levels, 3 had improved 3 levels and 1 patient had improved 4 levels.Thymectomy is an appropriate therapeutic procedure in the multidisciplinary treatment of patients with MG and it is the approach of choice for patients with associated thymomas. The intra- and post-operative complication rate is low and the rate of clinical improvement is high.

Abstract

This paper shows the interaction among the algebraic-analytical data fields theory, the complete descriptions that may be truncated for practical visual tasks and the different level of processing in the early visual pathway, in order to establish a measure of the representation of similarity between images. Our work is based on the approach of modeling the representational capabilities of systems of receptive fields found in early mammalian vision. It is well known that a representation scheme with a metric in the space of representation induce, in a natural way, a similarity measure between images. Abstract

Under stress conditions (darkness, nitrogen starvation, high ammonium concentrations, glutamine synthetase and glutamate synthase inhibition) glutamate dehydrogenase aminating activity levels of Chlamydomonas cells varied inversely to those of glutamine synthetase. Nitrogen and carbon sources also influenced glutamate dehydrogenase levels in Chlamydomonas, the highest values being found in cells cultured mixotrophically with ammonium, under which conditions glutamate dehydrogenase and glutamine synthetase levels were likewise inversely related. These facts, together with the analysis of internal fluctuations of ammonium, 2-oxoglutarate, and the amino acid pool as well as the variations of certain enzymes involved in carbon metabolism indicate that glutamate dehydrogenase aminating activity is adaptative, being involved in the maintenance of intracellular levels of L-glutamate when they cannot be maintained by the GS-GOGAT cycle, and probably more connected with carbon than nitrogen metabolism. Abstract

The involvement of three NAD(P)+-L-glutamate dehydrogenase (GDH; EC 1.4.1.3) isoenzymes, named GDH1, GDH2 and GDH3, in the carbon and nitrogen metabolism of the green alga Chlamydomonas reinhardtii 6145c has been investigated under different environmental and stress conditions. GDH1 activity decreased, but GDH2 and GDH3 activities increased with the age of cultures. When the extracellular ammonium concentration was high, only GDH1 activity increased with growth whereas GDH2 and GDH3 remained unchanged. In the presence of L-methionine-D,L-sulfoximine (MSX), an inhibitor of L-glutamine synthetase (GS), a significant increase of GDH1 and a slight decrease in GDH3 activity was observed, whereas GDH2 did not change. A significant increase in the intracellular 2-oxoglutarate was also found upon addition of azaserine, an inhibitor of L-glutamate synthase (GOGAT) activity. However, no significant changes in GDH isoenzyme activities were observed after addition of azaserine or azaserine plus MSX, except an induction of GDH3 in the latter case. Moreover, in the presence of ethoxyzolamide (ETZ), an inhibitor of carbonic anhydrase activity, an induction of total GDH activity, mainly due to an increase in GDH1 and to a minor extent in GDH2, was observed in cells under low CO2 (0.03%). In the dark, cells showed an increase in GDH1 activity, but when acetate was present GDH1 activity was repressed. All these results taken together suggest a relationship between GDH1 and nitrogen assimilation, whereas GDH2 and GDH3 seem to be involved in the production of 2-oxoglutarate to fuel the tricarboxylic acid (TCA) cycle. Abstract

A cDNA clone encoding a putative dihydroflavonol 4-reductase gene has been isolated from a strawberry (Fragaria x ananassa cv Chandler) DNA subtractive library. Northern analysis showed that the corresponding gene is predominantly expressed in fruit, where it is first detected during elongation (green stages) and then declines and sharply increases when the initial fruit ripening events occur, at the time of initiation of anthocyanin accumulation. The transcript can be induced in unripe green fruit by removing the achenes, and this induction can be partially inhibited by treatment of de-achened fruit with naphthylacetic acid, indicating that the expression of this gene is under hormonal control. We propose that the putative dihydroflavonol 4-reductase gene in strawberry plays a main role in the biosynthesis of anthocyanin during color development at the late stages of fruit ripening; during the first stages the expression of this gene could be related to the accumulation of condensed tannins.

Abstract

Introduction: The objective of this retrospective study was to describe the short- and long-term patterns of IVIg use, safety, and response to treatment in patients with chronic inflammatory demyelinating polyneuropathy (CIDP). Methods: Response to therapy was defined as an improvement of 1 point on the modified Rankin score at short- and mid-term visits. Patient status at long term was classified as remission, stability, or non-responder. Results: Eighty-six patients were included; 60.5% responded at short term and 54.6% at mid-term. At long term, 25.6% of patients were in remission, 65.1% were stable, and 9.3% were non-responders. The only variable associated with remission was a better response during the first 6 months of follow-up. Conclusions: A significant percentage of patients did not require any additional drugs in the long term. This suggests that treatment effect or disease outcome may be stable over time, and treatment regimens should therefore be individualized to avoid overtreatment. Muscle Nerve48: 870-876, 2013

Abstract

A strawberry (Fragaria x ananassa cv. Chandler) fruit cDNA (Fahyprp -cDNA) and its corresponding gene (Fahyprp) showing sequence homology to higher plant hyprp genes have been isolated. The cDNA contains an open reading frame encoding a 16 kDa protein with 156 amino acids. The peptide has an amino-terminal signal sequence, a repetitive proline-rich sequence, and a cysteine-rich carboxy-terminal region homologous to other HyPRP proteins. Northern blot and QRT-PCR analysis have shown that the strawberry transcript is specifically expressed in fruit, not being detected in other plant tissues. " In situ " hybridization and immunolocalization studies have indicated that the Fahyprp gene is strongly expressed in achene sclerenchyma cells, in the vascular and receptacle cells of immature green fruit and in the vascular cells of mature red fruits. The achenes removal from unripe green fruits induced the expression of this Fahyprp gene. This induction was reverted by treatment of deachened fruit with the auxin NAA, supporting the idea that Fahyprp gene expression is regulated by auxins. Furthermore, the HyPRP protein has been localized in parenchymatic cells of immature fruits associated to structures containing condensed tannins. The results are discussed supporting a putative role of this protein in the anchoring of polymeric polyphenols in the strawberry fruit during growth and ripening.
