12. Synthesis and evaluation of a new polymeric scale inhibitor with multiple scale inhibition properties.

Abstract

In this article, the monomers epoxy succinic acid (ESA) and maleic acid ethanolamine (PTA) were prepared, and then the terpolymer scale inhibitor P(ESA-AA-PTA) was synthesized by aqueous solution polymerization using ESA, PTA, and acrylic acid (AA) as monomers. The structures of the synthesized monomers and polymers were characterized by FTIR. The optimum synthesis conditions of the terpolymer were obtained by static scale inhibition experiment, and the effects of the amount of scale inhibitor, test temperature, test time, and solution pH on the inhibition performance of the four scales, CaCO3, CaSO4, BaSO4, and SrSO4, were discussed. The results showed that the best CaCO3 scale inhibition efficiency was 85% when the molar ratio of ESA:AA:PTA was 1:1:0.5, the initiator dosage was 10 wt%, the polymerization time was 3.5 hours and the polymerization temperature was 95 °C. In the optimum water environment, the scale inhibition efficiency of CaCO3 scale and CaSO4 scale reached 100%, BaSO4 scale could reach 75% and SrSO4 scale could reach 54%. P(ESA-AA-PTA) had better scale inhibition properties than the majority of commercially available oilfield scale inhibitors. SEM and XRD results showed that the addition of P(ESA-AA-PTA) scale inhibitor hindered the growth of crystals and made the microscopic morphology of scale samples changed. [ABSTRACT FROM AUTHOR]

Abstract

Objective: A lack of relevant research on Lycium barbarum polysaccharide‐glycoprotein (LBP) application in oral diseases. Here, we focused on the effect of LBP on osteogenic differentiation of human periodontal ligament stem cells (hPDLSCs) and periodontitis bone loss. Methods: Human periodontal ligament stem cells (hPDLSCs) were isolated and identified by flow cytometry. Alkaline phosphatase (ALP) activity, Alizarin Red staining, and combined qPCR and Western blot analyses were performed to elucidate the effects of LBP on the osteogenic potential of hPDLSCs. In vivo experiments were performed with the treatment of LBP in rat periodontal model. MicroCT scanning and histological analysis were conducted to evaluate osteogenesis in situ. Results: Human periodontal ligament stem cells (hPDLSCs) were successfully isolated and identified with CD90, CD29, and CD45. LBP enhanced hPDLSCs proliferation and migration and promoted RUNX2, ALP, Collagen I, and Osteocalcin expression through activating the ERK1/2 signaling pathway in vitro. The inflammatory factors, including interleukin 6 (IL‐6) and interleukin 8 (IL‐8) were reduced after LBP treatment. Alveolar bone resorption was significantly decreased in the LBP‐treated groups in vivo, and osteoclast was markedly decreased by LBP application. Conclusion: LBP promoted hPDLSC osteogenesis by targeting the ERK1/2 signaling pathway and reverse bone loss by reducing inflammation. Abstract

The impact of hyperbaric oxygen (HBO) on the healing of bone tissues around implants was studied using an animal model.A total of 32 beagle dogs were selected and randomly divided into the HBO group and the blank group. The dogs in the HBO group were subjected to 90 minutes of HBO therapy. X-ray, cone-beam computerized tomography imaging, implant stability quotient (ISQ) values, histological observation, quantitative analysis of bone histomorphometry, and hematoxylin-eosin (HE) and Masson staining were evaluated.In this study, at 4 weeks after the surgery, the mean ISQ value in the HBO group was higher than that in the blank group, and the difference had statistical significance (P0.05). At week 4 and 8, the mean values of bone ingrowth fraction (BIC%) and the percentage of bone area (BA%) in the HBO group were both higher than those of the blank group. Decalcified paraffin sections were stained with HE and Masson staining showed that the bone tissue around the implant in the HBO group has more osteoblasts than control group, and many irregular marrow cavities and Haversian bone plates were observed in the new bone tissue.This study showed that after implantation, early osteogenesis was better in the HBO group than in the blank group. On one hand, the healing time of the bone tissue around the implants was reduced.

35. Role of Inflammatory Factors in Regulation of Osteogenesis in Tissue-Engineered Bone

Abstract

It was traditionally considered that the inhibition of inflammatory reaction is necessary for osteogenesis, but the latest issue argued inflammation is unavoidable in the process of bone trauma, and physiological inflammatory reaction is essential to achieve bone formation. Tissue-engineered bone graft is not only associated with osteoblast-related cells; the inflammatory reaction is the initial physiological process, mainly with neutrophil infiltration, which secretes MCP-1, IL-8, and other chemokines and further promotes dendritic cells, lymphocytes, and mononuclear macrophages to move in. The activation pathways of macrophages have a direct effect on the outcome of the inflammatory reaction and the healing, which are divided into the classical approach (M1) and the alternative approach (M2). The M1 pathway secretes IL-1 beta, IL-6, TNF-α, and other pro-inflammatory factors, while the M2 pathway secretes arginase, IL-1Ra, IL-4, and other anti-inflammatory cytokines, also with bone-healing-related growth factors, which promote homing of bone mesenchymal stem cells (bMSCs).

42. Targeting Discoidin Domain Receptor 2 for the Development of Disease-Modifying Osteoarthritis Drugs

Abstract

One of the most pressing issues in osteoarthritis (OA) research is the development of disease-modifying OA drugs (DMOADs), as currently there are no such drugs available. The paucity of suitable DMOADs is mostly due to the lack of approved ideal therapeutic targets necessary for the development of these drugs. However, based on recent discoveries from our laboratory and other independent laboratories, it is indicated that a cell surface receptor tyrosine kinase for collagen type II, discoidin domain receptor 2 (DDR2), may be an ideal therapeutic target for the development of DMOADs. In this article, we review the current status of research in understanding roles of DDR2 in the development of OA.

Abstract

OBJECTIVE: To compare the protein profiles between decapeptide-treated and untreated planktonic cells of Streptococcus mutans (S. mutans) by differential proteomic analysis to determine and identify the key proteins. METHODS: In our previous study, we investigated decapeptide (KKVVFKVKFK–NH(2)), which was a novel adenosine monophosphate. Compared with other oral pathogens tested, decapeptide had a preferential antibacterial activity against S. mutans. It also inhibited S. mutans biofilm formation and reduced the one-day developed biofilm. In the present study, we first synthesized decapeptide, and then compared the protein profiles between decapeptide-treated and untreated planktonic cells of S. mutans by two-dimensional gel electrophoresis and matrix-assisted laser desorption ionization time-of-flight mass spectrometry. We also verified different expressions of key protein enolase in the protein level. RESULTS: The results showed that decapeptide altered the protein expression of planktonic S. mutans. These proteins were functionally involved in carbohydrate degradation by glycolysis, protein folding, conjunction, transport, translation, adenosine triphosphate binding, protein binding, sequence-specific DNA binding, transcription factor activity, and two-component response regulator activity. Western blot results showed that enolase protein expression decreased obviously in decapeptide-treated cells of S. mutans. CONCLUSION: The protein expression of S. mutans significantly changed after synthetic antimicrobial decapeptide treatment, suggesting that decapeptide may present a preferential effect on oral caries by changing the expression of certain key proteins, such as enolase protein.
