Na busca de novos hits para o desenvolvimento de fármacos, o núcleo 8-hidroxiquinolina merece destaque por permitir diversas modificações estruturais levando à formação de derivados bioativos. O objetivo deste trabalho é planejar, sintetizar e caracterizar novos derivados da 8-hidroxiquinolina e avaliar a atividade antifúngica contra Candida sp. e fungos dermatófitos, além da atividade antibacteriana contra Mycobacterium tuberculosis. Além disso, o mecanismo de ação antimicrobiana dos derivados mais promissores é investigado. O presente trabalho contribui com quatro Capítulos para a área de desenvolvimento de novos agentes antimicrobianos, sendo dois Capítulos já publicados na forma de artigos científicos (um artigo de revisão e um experimental) e outros dois Capítulos referentes a artigos experimentais que serão submetidos. Quatro rotas sintéticas, de duas a seis etapas, foram utilizadas para a obtenção dos novos compostos. Para a síntese da série de derivados 8-metoxi ou 8-hidroxiquinolina-5-cloro-7-arilamina, foi necessário a proteção da posição 8 do clioquinol, seguida da reação de Buchwald-Hartwig e, por fim, desproteção da posição 8. Para a síntese de sulfonamidas na posição 5, a 8-hidroxiquinolina sofreu reação de clorossulfonação, seguida da substituição por amina apropriada. A estratégia de hibridação molecular foi aplicada para o planejamento de sulfonilureias na posição 5 e derivados 7-pirimidiniltriazólicos. O derivado contendo uma sulfonilureia na posição 5 foi preparado através de metilação da 8-hidroxiquinolina, clorossulfonação, reação com amina apropriada, hidrólise, formação de sulfonil isocianato e reação com amina apropriada. A síntese de um derivado 7-pirimidiniltriazólico iniciou com a proteção do clioquinol, seguida da reação de Buchwald-Hartwig, seguida de substituição nucleofílica aromática para a formação do intermediário azido, seguida de click chemistry e, por fim, desproteção da posição 8. Esse trabalho contribui para a área de química orgânica, através da padronização de duas metodologias de síntese para a obtenção de sulfonilureias derivadas de 8-hidroxiquinolina. Além disso, até onde sabemos, a síntese de derivados de 8-hidroxiquinolina, utilizando a reação de Buchwald-Hartwig, é inédita. Ao todo, 30 produtos finais foram sintetizados, purificados e caracterizados, apresentando rendimentos globais variando de 5 a 81%. Dentre os compostos sintetizados, 5h (derivado 7-pirimidinilamino) foi o mais ativo contra células fúngicas (CIM: 4 μg/mL), mas não apresentou atividade contra M. tuberculosis na maior concentração testada. Este derivado apresentou baixa citotoxicidade em leucócitos e baixa irritabilidade em modelos ex vivo. Além disso, o mecanismo de ação antifúngica de 5h pode estar relacionado à sua capacidade quelante. Três derivados 7-arilamino substituídos foram os mais promissores na avaliação anti- M. tuberculosis (CIM: 10 μg/mL), além de serem identificados como inibidores da enoil-(proteína transportadora de acil) redutase de M. tuberculosis (MtInhA), com CI50 na faixa micromolar baixa. Até onde sabemos, esse é o primeiro estudo que reporta esse alvo molecular microbiano para 8-hidroxiquinolinas. Sulfonamidas N-substituídas na posição 5 contendo uma cadeia propílica também apresentaram resultados interessantes contra M. tuberculosis, mas não foram inibidoras de MtInhA. Os resultados deste estudo confirmam que os derivados de 8-hidroxiquinolina são potenciais novos hits para o desenvolvimento de novos antimicrobianos. Além disso, esse trabalho contribui para a identificação de um novo alvo molecular para 8-hidroxiquinolinas antimicrobianas. In the search of novel hits for drug development, the 8-hydroxyquinoline core deserves to be highlighted for allowing several structural changes leading to bioactive derivatives. The purpose of this study is to design, synthesize and characterize 8-hydroxyquinoline derivatives and to evaluate their antifungal activity against Candida sp. and dermatophytes, in addition to antibacterial activity against Mycobacterium tuberculosis. Furthermore, the mechanism of antimicrobial action of the most active derivatives is investigated. This work contributes with four Chapters for the field of development of novel antimicrobial agents, being two Chapters already published in the form of scientific articles (one review article and one experimental article) and two other Chapters referring to experimental articles that will be submitted. Four synthetic routes, of two to six steps, were used to obtain the novel compounds. For the synthesis of 8-methoxy or 8-hydroxyquinoline-5-choro-7-arilamine derivatives, it was necessary to protect position 8 of the clioquinol, followed by Buchwald-Hartwig reaction and, finally, deprotection of the 8-position. For the synthesis of 5-sulfonamides, chlorosulfonation of the 8-hydroxyquinoline and substitution by an appropriate amine were carried out. The molecular hybridization strategy was applied to design the 5-sulfonylureia and 7-pyrimidinyltriazole compounds. The derivative containing a sulfonylurea at position 5 was prepared by methylation of 8-hydroxyquinoline, chlorosulfonation, reaction with an appropriate amine, hydrolysis, sulfonyl isocianate formation, and reaction with appropriate amine. The synthesis of the 7-pyrimidinyltriazole derivative started with protection of the 8-hydroxyl, followed by Buchwald-Hartwig reaction, nucleophilic aromatic substitution (to obtain the azido intermediate), click chemistry and, finally, deprotection of position 8. This work contributes to the field of organic chemistry, through the development of two synthesis methodologies for obtaining sulfonylureas derived from 8-hydroxyquinoline. Furthermore, to the best of our knowledge, the synthesis of 8-hydroxyquinoline derivatives using the Buchwald-Hartwig reaction is unprecedented. 30 final products were synthesized, purified and characterized, with overall yields ranging from 5 to 81%. Among all the synthesized compounds, 5h (a 7-pyrimidinylamino derivative) was the most active against fungal cells (MIC: 4 μg/mL), but it showed no activity against M. tuberculosis in the highest tested concentration. This derivative has presented low cytotoxicity in leukocytes and low irritability in ex vivo models. Moreover, the mechanism of antifungal action of 5h may be related to its chelating capacity. Three substituted 7-arilamino derivatives were the most promising compounds in the anti-TB evaluation (MIC: 10 μg/mL), in addition to being identified as Mycobacterium tuberculosis enoyl-acyl carrier protein reductase (MtInhA) inhibitors, with IC50 in the low micromolar range. To the best of our knowledge, it is the first study that reports this microbial molecular target for 8-hydroxyquinolines. Sulfonamides in position 5 substituted by a propyl chain also presented interesting results against M. tuberculosis, without inhibitory activity against MtInhA. The results of this study confirm that the 8-hydroxyquinoline derivatives are potential novel hits for the development of new antimicrobials. Moreover, this work contributes for the identification of a new molecular target for antimicrobial 8-hydroxyquinolines.

Flexibilidade e fun??o s?o propriedades relacionadas no estudo da din?mica proteica. A flexibilidade reflete no potencial conformacional das prote?nas e, deste modo, nas suas funcionalidades. A presen?a de intera??es entre complexos prote?na-ligante e prote?na-prote?na, de substratos e altera??es ambientais, podem alterar a plasticidade proteica, atuando desde o rearranjo das cadeias laterais de amino?cidos at? o desdobramento/dobramento de grandes motivos estruturais. Com o intuito de avaliar os efeitos da flexibilidade em sistemas proteicos, definimos como alvo do nosso trabalho a enzima 2-trans-enoil-ACP (CoA) redutase de Mycobacterium tuberculosis (Mtb), ou MtInhA, a qual apresenta-se como um homotetr?mero em solu??o. Apesar de a sua estrutura quatern?ria ser a forma biologicamente ativa, estudos computacionais simulam a MtInhA como um mon?mero, justificando a independ?ncia dos s?tios ativos devido ?s suas dist?ncias, assim como, o custo computacional. Entretanto, j? foram descritas diferen?as na flexibilidade entre as formas tetram?rica e monom?rica, as quais impactam no tamanho da cavidade de liga??o da enzima. Desta forma, o objetivo deste trabalho foi construir um modelo de simula??o monom?rico da MtInhA que apresente as caracter?sticas conformacionais da estrutura quatern?ria da enzima. Para espelhar a influ?ncia das subunidades e obter comportamento similar ? prote?na na forma nativa, aplicamos diferentes constantes de for?a na estrutura monom?rica, para restringir movimento dos res?duos que comp?em a cavidade de liga??o do substrato. A partir da compara??o entre os sitemas monom?rico com restri??es, tetram?rico e monom?rico sem restri??es, observamos que as simula??es com constantes de for?a aplicadas apresentam um comportamento similar ? prote?na nativa. N?o obstante, essas semelhan?as s?o mais proeminentes nas al?as A e B, as quais s?o regi?es importantes para a modula??o da flexibilidade do s?tio ativo. Portanto, a partir destes resultados apresentamos um modelo de simula??o monom?rico da MtInhA, que possui caracter?sticas conformacionais da estrutura biologicamente ativa. Assim, os dados obtidos neste trabalho poder?o ser aplicados para a gera??o de modelos flex?veis mais confi?veis para docagem molecular, e tamb?m para a realiza??o de simula??es por din?mica molecular mais longas e com um menor custo computacional. Flexibility and function are related properties in the study of protein dynamics. Flexibility reflects in the conformational potential of proteins and thus in their functionalities. The presence of interactions between protein-ligands and protein-protein complexes, substrates and environmental changes, can alter protein plasticity, acting from the rearrangement of the side chains of amino acids to the unfolding/folding of large structural motifs. In order to evaluate the effects of flexibility in protein systems, we defined as the target of our work the enzyme 2-trans-enoyl-ACP (CoA) reductase of Mycobacterium tuberculosis (Mtb), or MtInhA, which presents as a homothernamer in solution. Although its quaternary structure is the biologically active form, computational studies simulate MtInhA as a monomer, justifying the independence of active sites due to their distances, as well as computational cost. However, differences in flexibility between the tetrameric and monomeric forms have already been described, which impact in the size of the enzyme binding cavity. Thus, the objective of this work was to construct a monomeric simulation model of MtInhA that presents the conformational characteristics of the quaternary structure of the enzyme. To mirror the influence of the subunits and obtain behavior similar to the protein in the native form, we applied different force constants in the monomeric structure, to restrict movement of the residues that composes the substrate binding cavity. From the comparison between monomeric systems with restrictions, tetrameric and monomer without restrictions, we observed that the simulations with force constants applied present a behavior similar to the native protein. Nevertheless, these similarities are more prominent in the A and B loops, which are important regions for modulating the flexibility of the active site. Therefore, from these results we present a monomeric simulation model of MtInhA, which has conformational characteristics of the biologically active structure. Thus, the data obtained in this work can be applied to the generation of more reliable flexible models for molecular production, and also for the performance of simulations by molecular dynamics longer and with a lower computational cost.
