Abstract Background National legislative guidelines for sleep and rest are lacking in the Canadian licensed childcare sector. No review of Canadian legislation for licensed childcare facilities has focused on sleep. This paper provides a review of the Canadian provincial and territorial legislative landscape, regarding sleep, rest, and naps in licensed childcare centers. Methods Childcare statutes and regulations for each province and territory were identified and downloaded on a particular date. Statutes and regulations were reviewed focusing on sections articulating licensed childcare facility mandates governing sleep, rest, naps, and sleep equipment. An excel file was used to facilitate systematic data retrieval and comparisons across provinces and territories. Two authors developed and discussed themes that summarized data from the documents. Results No statutes indicated recommendations for sleep, rest, or naps. Only one regulation defined rest (Alberta). Our analysis of regulations identified four themes representing sleep, rest, and naps: programming (general programming, daily programming); space (dedicated space, amount of space, age-specific space); equipment (developmental appropriateness, acceptable sleep equipment, age-specific equipment); and safety (staffing during sleep/rest, sleep position, sleep monitoring, sleep equipment safety, prohibited practices). In Canada, minimal regulatory consistency is evident in required sleep programming, space, acceptability of sleep equipment, and sleep safety considerations. Most jurisdictions’ regulations indicated necessity for developmentally appropriate rest or sleep areas and equipment, in particular for infants, but there was minimal consistency in defining infant age groups. Conclusions Although we identified themes related to sleep across regulations, childcare regulations differ in their definitions of infants and specifications for children’s sleep and rest in licensed Canadian childcare facilities. Without adequate definitions in legislative components of appropriate sleep duration linked to children’s developmental stages, childcare facilities lack guidance to support healthy sleep for children in their care. Future research can examine translation of healthy sleep guidelines into government legislation and mandates for sleep, rest, and naps among young children in licensed childcare.

Abstract We study the classical dynamics of spinning particles using scattering amplitudes and eikonal exponentiation. We show that observables are determined by a simple algorithm. A wealth of complexity arises in perturbation theory as positions, momenta and spins must be iteratively corrected at each order. Even though we restrict ourselves to one-loop computations at quadratic order in spin, nevertheless we encounter and resolve a number of subtle effects. Finally, we clarify the links between our work and various other eikonal approaches to spinning observables.

Abstract Mycobacterial glycolipids are important cell envelope structures that drive host-pathogen interactions. Arguably, the most important are lipoarabinomannan (LAM) and its precursor, lipomannan (LM), which are trafficked from the bacterium to the host via unknown mechanisms. Arabinomannan is thought to be a capsular derivative of these molecules, lacking a lipid anchor. However, the mechanism by which this material is generated has yet to be elucidated. Here, we describe the identification of a glycoside hydrolase family 76 enzyme that we term LamH (Rv0365c in Mycobacterium tuberculosis) which specifically cleaves α−1,6-mannoside linkages within LM and LAM, driving its export to the capsule releasing its phosphatidyl-myo-inositol mannoside lipid anchor. Unexpectedly, we found that the catalytic activity of this enzyme is important for efficient exit from stationary phase cultures, potentially implicating arabinomannan as a signal for growth phase transition. Finally, we demonstrate that LamH is important for M. tuberculosis survival in macrophages.

Abstract We construct classical curvature spinors in topologically massive gauge theory and topologically massive gravity, expressed in terms of massive three-particle amplitudes. We show that when the amplitudes double copy, the curvature spinors associated with particular solutions satisfy the Cotton double copy, the three-dimensional cousin of the Weyl double copy. Furthermore, we show that under certain circumstances the Cotton double copy can be derived via a dimensional reduction of the Weyl double copy.

Abstract We study the variance in the measurement of observables during scattering events, as computed using amplitudes. The classical regime, characterised by negligible uncertainty, emerges as a consequence of an infinite set of relationships among multileg, multiloop amplitudes in a momentum-transfer expansion. We discuss two non-trivial examples in detail: the six-point tree and the five-point one-loop amplitudes in scalar QED. We interpret these relationships in terms of a coherent exponentiation of radiative effects in the classical limit which generalises the eikonal formula, and show how to recover the impulse, including radiation reaction, from this generalised eikonal. Finally, we incorporate the physics of spin into our framework.

Many types of cancer are currently treated using a combination of chemotherapeutics, but unfortunately, this strategy is considerably limited by severe side effects. The current development of nanocarriers enables the use of multiple drugs anchored on one unique platform thus enhancing the initiated therapeutic effect and minimizing the possibility of drug resistance. In this context, a graphene‐oxide‐based 2D nanoplatform is developed, which is functionalized using highly branched polyethylene‐glycol and a multimodal set of two drugs with various mechanisms of action, namely Pt‐based complex (a Pt(IV) prodrugs based on cisplatin) and doxorubicin (DOX). We performed in vitro 2D screening on two cancer cell lines, namely glioblastoma and osteosarcoma, that were selected as models of two aggressive tumors that remain a massive challenge in oncology. The therapeutic effect of the developed nano‐platform is higher at lower concentrations (15 μm of Pt‐drug, 0.6 μm DOX) compared to the impact of the free drugs. This indicates a possible positive effect of the accumulation and transport of the drugs using this nanoplatform. Results obtained on 3D cell models using MG63 osteosarcoma cells uncovered an understandable lowered diffusion profile of the developed nanoplatforms, compared to the application of free drugs.

Abstract We explore topologically massive gauge theories using the covariant colour kinematics duality recently introduced in [1]. We show that the massive bi-adjoint scalar field is simply related to topologically massive gauge theory by the duality, and that enacting the same duality on the gauge theory produces topologically massive gravity coupled to a scalar or, equivalently, an antisymmetric field. We also show that different choices for the replacement of the colour structure constants with kinematic structure constants lead to different theories, including a topologically massive generalisation of Born-Infeld theory.

Abstract Rare genetic diseases affect 5–8% of the population but are often undiagnosed or misdiagnosed. Electronic health records (EHR) contain large amounts of data, which provide opportunities for analysing and mining. Data analysis in the form of visualisation and statistical testing, was performed on a database containing deidentified health records of 1.28 million patients across 3 major hospitals in Singapore, in a bid to improve the diagnostic process for patients who are living with an undiagnosed rare disease, specifically focusing on Fabry Disease and Familial Hypercholesterolaemia (FH). On a baseline of 4 patients, we identified 2 additional patients with potential diagnosis of Fabry disease, suggesting a potential 50% increase in diagnosis. Similarly, we identified > 12,000 individuals who fulfil the clinical and laboratory criteria for FH but had not been diagnosed previously. This proof-of-concept study showed that it is possible to perform mining on EHR data albeit with some challenges and limitations.

BackgroundIn recent years, social media have emerged as important spaces for commercial marketing of health tests, which can be used for the screening and diagnosis of otherwise generally healthy people. However, little is known about how health tests are promoted on social media, whether the information provided is accurate and balanced, and if there is transparency around conflicts of interest. ObjectiveThis study aims to understand and quantify how social media is being used to discuss or promote health tests with the potential for overdiagnosis or overuse to generally healthy people. MethodsContent analysis of social media posts on the anti-Mullerian hormone test, whole-body magnetic resonance imaging scan, multicancer early detection, testosterone test, and gut microbe test from influential international social media accounts on Instagram and TikTok. The 5 tests have been identified as having the following criteria: (1) there are evidence-based concerns about overdiagnosis or overuse, (2) there is evidence or concerns that the results of tests do not lead to improved health outcomes for generally healthy people and may cause harm or waste, and (3) the tests are being promoted on social media to generally healthy people. Background: This feeding clinic engaged parents as active participants in a group intervention. Parents' participated behind a two-way mirror. This novel approach to practice in this domain largely evolved from an integration of theory, service user feedback, service development and the local context. This review sought to understand how parents experienced being part of a reflecting team in this way. Methods: Semi-structured interviews were completed and analysed using thematic analysis. The researchers took a critical realist approach to the research, data analysis and report writing. Findings: Parents talked about benefiting from perspective taking that enabled them to see the child in context, which in part supported novel understandings of their child's feeding and their relationship to it. Three themes were identified: (1) "Connection and Distance" explored parents' experience of being behind the screen in terms of the dialectic potential that was created by being simultaneously close to and distanced from their children and the other parents; (2) "Playing with Techniques" describes the way parents valued learning in the group through discussion and working on techniques between sessions. In particular, they moved from looking for techniques to thinking about how and when to use "techniques" that is, second-order change; and (3) "Unexpected Gains" describes how parents came to new and unexpected insights about their understanding of their children, the presenting issues and their relationships to these issues. Conclusion: Parents' experiences suggest that there is merit in engaging parents in a reflecting team in a feeding clinic. This way of working with parents supports their engagement in change and positions them as active agents of this change. Parents taking up this position has the advantage of moving from passive consumers of health care to active and critical collaborators.

A series of C2-functionalied Pt (IV) glycoconjugates based on glucosamine have been synthesised, characterised and tested as anticancer agents on a series of different 2D and 3D cancer cell lines. The carbohydrate will act as a targeted delivery system to improve the selectivity, exploiting the Warburg Effect and the GLUTs receptors that are overexpressed in most of the cancer cells. The hydroxyl at C2 of the carbohydrates does not participate in hydrogen bonding with the GLUTs receptors, making C2 an attractive position for drug conjugation as seen in literature. In this study, we use the amino functionality at the C2 position in glucosamine and Copper-catalysed Azide-Alkyne Cycloaddition “click” (CuAAC) reaction to connect the prodrug Pt (IV) scaffold to the carbohydrate. We have investigated complexes with different linker lengths, as well as acetyl protected and free derivatives. To the best of our knowledge, this study represents the first series of Pt (IV) glucosamine-conjugates functionalised at C2.

ABSTRACTGram-negative bacteria have a thin peptidoglycan layer between the cytoplasmic and outer membranes protecting the cell from osmotic challenges. Hydrolases of this structure are needed to cleave bonds to allow the newly synthesized peptidoglycan strands to be inserted by synthases. These enzymes need to be tightly regulated and their activities coordinated to prevent cell lysis. To better understand this process in Escherichia coli, we probed the genetic interactions of mrcA (encodes PBP1A) and mrcB (encodes PBP1B) with genes encoding peptidoglycan amidases and endopeptidases in envelope stress conditions. Our extensive genetic interaction network analysis revealed relatively few combinations of hydrolase gene deletions with reduced fitness in the absence of PBP1A or PBP1B, showing that none of the amidases or endopeptidases is strictly required for the functioning of one of the class A PBPs. This illustrates the robustness of the peptidoglycan growth mechanism. However, we discovered that the fitness of ∆mrcB cells is significantly reduced under high salt stress and in vitro activity assays suggest that this phenotype is caused by a reduced peptidoglycan synthesis activity of PBP1A at high salt concentration.IMPORTANCEEscherichia coli and many other bacteria have a surprisingly high number of peptidoglycan hydrolases. These enzymes function in concert with synthases to facilitate the expansion of the peptidoglycan sacculus under a range of growth and stress conditions. The synthases PBP1A and PBP1B both contribute to peptidoglycan expansion during cell division and growth. Our genetic interaction analysis revealed that these two penicillin-binding proteins (PBPs) do not need specific amidases, endopeptidases, or lytic transglycosylases for function. We show that PBP1A and PBP1B do not work equally well when cells encounter high salt stress and demonstrate that PBP1A alone cannot provide sufficient PG synthesis activity under this condition. These results show how the two class A PBPs and peptidoglycan hydrolases govern cell envelope integrity in E. coli in response to environmental challenges and particularly highlight the importance of PBP1B in maintaining cell fitness under high salt conditions.

Administrative burdens are the frictions that people face in their encounters with public services, leading to meaningful costs that include learning, compliance, and psychological costs. We offer evidence that burdens are a key source and consequence of inequality, resulting in disparate outcomes in people’s access to basic rights. We also detail how these outcomes are patterned by targeting, federalism, bureaucratic pathologies, and the growing use of the private sector and tax system to deliver social welfare benefits. Throughout, we highlight recent and novel contributions, including empirical research in this double issue, that have helped clarify how and why administrative burdens shape inequality. Burdens have not received the political, policy, or research priority that is commensurate with their magnitude or impact on individuals. We conclude by arguing that we need a coherent language and framework to recognize and, where appropriate, reduce burdens across a wide array of policy domains.
