Abstract

Dmrt1 is a highly conserved transcription factor, which is critically involved in regulation of gonad development of vertebrates. In medaka, a duplicate of dmrt1—acting as master sex-determining gene—has a tightly timely and spatially controlled gonadal expression pattern. In addition to transcriptional regulation, a sequence motif in the 3′ UTR (D3U-box) mediates transcript stability of dmrt1 mRNAs from medaka and other vertebrates. We show here that in medaka, two RNA-binding proteins with antagonizing properties target this D3U-box, promoting either RNA stabilization in germ cells or degradation in the soma. The D3U-box is also conserved in other germ-cell transcripts, making them responsive to the same RNA binding proteins. The evolutionary conservation of the D3U-box motif within dmrt1 genes of metazoans—together with preserved expression patterns of the targeting RNA binding proteins in subsets of germ cells—suggest that this new mechanism for controlling RNA stability is not restricted to fishes but might also apply to other vertebrates., In medaka a duplicate of dmrt1 acts as master sex-determining gene, with tightly controlled expression pattern in the gonads. Abstract

International audience; Sex-determination relies on the proper control of a hierarchically structured multilayered network of genes. The genes at the top orchestrate complex transcriptional and post-transcriptional regulations. Despite such critical function they appear to be dispensable in evolutionary terms and can be quickly replaced with the emergence of new lineages. Our analysis shows that concomitantly to the acquisition of a dominant position within the sex-determining network, the medaka master male determiner dmrt1bY was subjected to a profound rearrangement of its regulatory landscape. Requiring a complete rewiring of the regulatory network, this evolutionary innovation was partly brought about by exaptation of a “ready-to-use” pre-existing cis-regulatory element contributed by a transposable element (TE), called Izanagi. This element acts as a silencer, which recruits both proteins Dmrt1bY and Dmrt1a to turn-off the dmrt1bY gene after it has fulfilled its function as the primary male SD gene. Exceeding this simple feedback regulation we now report that TE-mediated transcriptional rewiring can reach an unexpected level of complexity. We find that another TE, Rex1, has jumped into Izanagi. Through disruption of Izanagi, Rex1 immobilized this TE and fixed the Dmrt1-mediated down-regulation. Moreover, Rex1 brought in a preformed regulatory element for Sox5, which critically functions in establishing the temporal and cell-type specific expression pattern of dmrt1bY. Although Sox5 has not been implicated in sex-determination previously we show its critical involvement in medaka male development through regulation of primordial germ cell numbers. Disruption of sox5 leads to XX female-to-male sex-reversal, underpinning the importance of the Rex1 TE for the establishment of a new sex-determining mechanism in medaka.

Abstract

Abstract Background In lower vertebrates, steroid-induced oocyte maturation is considered to involve membrane-bound progestin receptors. Two totally distinct classes of putative membrane-bound progestin receptors have been reported in vertebrates. A first class of receptors, now termed progesterone membrane receptor component (PGMRC; subtypes 1 and 2) has been studied since 1996 but never studied in a fish species nor in the oocyte of any animal species. A second class of receptors, termed membrane progestin receptors (mPR; subtypes alpha, beta and gamma), was recently described in vertebrates and implicated in the progestin-initiated induction of oocyte maturation in fish. Methods In the present study, we report the characterization of the full coding sequence of rainbow trout PGMRC1 and mPR beta cDNAs, their tissue distribution, their ovarian expression profiles during oogenesis, their hormonal regulation in the full grown ovary and the in situ localization of PGMRC1 mRNA in the ovary. Results Our results clearly show, for the first time in any animal species, that rainbow trout PGMRC1 mRNA is present in the oocyte and has a strong expression in ovarian tissue. In addition, we show that both mPR beta and PGMRC1, two members of distinct membrane-bound progestin receptor classes, exhibit highly similar ovarian expression profiles during the reproductive cycle with maximum levels during vitellogenesis and a down-expression during late vitellogenesis. In addition, the mRNA abundance of both genes is not increased after in vitro hormonal stimulation of full grown follicles by maturation inducing hormones. Conclusion Together, our findings suggest that PGMRC1 is a new possible participant in the progestin-induced oocyte maturation in fish. However, its participation in the process of oocyte maturation, which remains to be confirmed, would occur at post-transcriptional levels.

National audience; The histological features, steroidogenic activities (aromatase, I I P-hydroxylase) and gene expressions (cyp19A1, cyp11b1, amh, ddx4, gcl, fst) of trout embryonic gonads have been compared before and after 7 days of in vitro culture at 12 degrees C in three different media. MCDB 105 gave the best results and was used to analyse the effect of 50 nM 17 beta-estradiol on testicular cyp11b1 expression. A significant six-fold decrease has been observed for cyp11b1, while amh, ddx4 and gcl did not change.

Abstract

National audience; Sexual maturation rate at the age of 2 years was significantly lower in a group of rainbow trout (Oncorhynchus mykiss) under continuous light from the end of the vesicle resorption stage than in the control. This inhibition was almost total in females, while sexual maturation rate was reduced by half in males. The same phenomenon has been observed in a group of females maintained under constant illumination from the first day of 0+ July to the first day of 1+ July. The analysis of the physiological parameters has shown that the exogenous vitellogenesis did not start during the 1 + year. Moreover, it was confirmed that the continuous light exposure has a beneficial effect on the growth rate of the treated fish in comparison with control.; Le taux de maturation sexuelle à 2 ans dans un lot de truites arc-en-ciel (Oncorhynchus mykiss) soumises à un éclairement constant depuis la fin du stade de la résorption de la vésicule a été significativement inférieur à celui du lot témoin. Cette inhibition était presque totale chez les femelles, alors que chez les mâles le taux de maturation diminuait de moitié. Le même phénomène a été observé dans un lot de femelles soumises à une période d'éclairement continu du 1er juillet 0+ au 1er juillet 1+. L'analyse des paramètres physiologiques a montré, chez ces femelles, que la vitellogénèse exogène ne démarrait pas pendant l'année 1+. De plus, il a été confirmé qu'un éclairement continu a un effet favorable sur la croissance des poissons traités, comparée à celle du témoin.

Abstract

50-kg pigs fed a purified wheat starch and casein diet were fitted with a gastric cannula. Gastric emptying was studied by checking the residual gastric contents at 30 min and 1, 2, 4 and 7 hrs after feeding. Five of the 25 pigs were used as controls; 5 were submitted to total vagal deafferentation including removal of the left nodose ganglion, right truncal vagotomy at the diaphragm and resection of the interconnecting branches between the two vagi on their intrathoracic course; 5 were submitted to total contralateral vagal deafferentation; 5 only underwent left truncal vagotomy and resection of the connectives; 5 were submitted to the contralateral operation. Total vagal deafferentation, suppressing bilateral afferences originating below the diaphragm, resulted in much higher residual gastric contents at all times studied after the meal. Significant gastric retention was also observed after unilateral truncal vagotomy as compared to the controls. Nevertheless, this retention was significantly lower than after total deafferentation. The reduced emptying rate after total vagal deafferentation was a result of the loss of all afferences, while the simultaneous reduction of efferences was of little importance. The mechanisms responsible for gastric retention have been discussed in relation with the sustained antral hyperactivity previously found in deafferented pigs and with the various controls of gastric emptying. Several factors may be involved, including both motor and secretory changes. The prolonged postprandial activity of the gastric antrum seemed to be a consequence of gastric retention.
