Abstract

Background: Podocalyxin (PODXL) is a highly sialylated adhesion glycoprotein that plays an important role in podocyte's physiology. Recently, missense and nonsense dominant variants in the PODXL gene have been associated with focal segmental glomerulosclerosis (FSGS), a leading cause of nephrotic syndrome and kidney failure. Their histologic description, however, was superficial or absent. Methods: We performed exome sequencing on a three-generation family affected by an atypical glomerular nephropathy and characterized the disease by light and electron microscopy. Results: The disease was characterized by FSGS features and glomerular basement membrane duplication. Six family members displayed chronic proteinuria, ranging from mild manifestations without renal failure, to severe forms with end-stage renal disease. Exome sequencing of affected twin sisters, their affected mother, healthy father, and healthy maternal uncle revealed a new nonsense variant cosegregating with the disease (c.1453C>T, NM_001018111) in the PODXL gene, which is known to be expressed in the kidney and to cause nephropathy when mutated. The variant is predicted to lead to a premature stop codon (p.Q485*) that results in the loss of the intracytoplasmic tail of the protein. Conclusion: This is the first description of a peculiar association combining a PODXL stop-gain variant and both FSGS and membranoproliferative glomerulonephritis features, described by light and electron microscopy.

Abstract

Item does not contain fulltext, Recurrence of primary membranous nephropathy after transplantation occurs in up to 44% of patients and is driven by PLA2R antibody. Here, we asked whether genetic determinants could improve risk prediction. First, we sequenced PLA2R1 and HLA-D loci in 248 patients with primary membranous nephropathy and identified two independent single nucleotide polymorphisms (SNPs) at risk for primary membranous nephropathy at each locus. These were rs9271188 (intergenic between HLA-DRB1 and HLA-DQA1,) and rs9275086 (intergenic between HLA-DQB1 and HLA-DQA2) at the HLA-D locus along with rs6726925 and rs13018963 at the PLA2R1 locus. Then we investigated whether primary membranous nephropathy at-risk variants were associated with recurrence in a retrospective cohort of 105 donor-recipient pairs and a replication cohort of 40 pairs. Seven SNPs located between HLA-DRB1 and HLA-DQA1 in linkage disequilibrium with rs9271188, and three SNPs in the PLA2R1 region predicted recurrence when presented by the donor, but not when presented by the recipient. The two SNPs in the HLA-D region most strongly associated with recurrence (rs9271705 and rs9271550) were confirmed in the replication cohort. A genetic risk score based on the two best predictors at each locus (rs9271705, rs9271550, rs17830558, and rs3828323) identified a group of patients with high risk of recurrence. Thus, our results suggest that the graft contributes to recurrence of primary membranous nephropathy through the disease susceptibility HLA-D and PLA2R1 SNPs in an autoimmune milieu. Further studies are needed before implementation of genetic testing for these in donor selection.

Abstract

A new procedure for dynamic analysis of complex structures, based on the fictitious-mass component mode synthesis method, is presented. Normal modes of separate components are calculated by finite-element analysis with the interface coordinates loaded with fictitious masses that generate local boundary deformations in the low-frequency modes. The original fictitious-mass method is extended to include three types of component interconnections: displacement constraints, connection elements, and structural links. The connection elements allow the introduction of springs and dampers between the interface points without adding structural degrees of freedom. The structural links facilitate the inclusion the discrete finite-element representation of typically small components in the coupling equations. This allows a convenient treatment of loose elements and the introduction of nonlinear effects and parametric studies in subsequent analyses. The new procedure is demonstrated with the structural model of a typical vehicle with four major substructures and a relatively large number of interface coordinates. High accuracy is obtained in calculating the natural frequencies and modes of the assembled structure and the separate components with the fictitious masses removed. Dynamic response analysis of the vehicle travelling over a rough road, performed by modal coupling, is in excellent agreement with that performed for the full model. [DOI: 10.1115/1.2202156]

Abstract

Control laws are designed for the alleviation of dynamic gust loads on a wind-tunnel model of a transport aircraft using wing-mounted control surfaces. Three different control surfaces are used: the symmetrically actuated main ailerons, special underwing forward-positioned control surfaces at about 0.8 of the wingspan, and special wing-tip forward-positioned control surfaces. The 5.3-m-span cable-mounted wind-tunnel model was constructed at the TsAGI laboratories and is being tested as there part of the 3AS 5th Framework European Commission research project. The length of a one-minus-cosine vertical gust velocity profile is tuned to yield maximal wing-root bending moment. All the control laws are based on simple low-pass filters for easy and robust application in the wind tunnel. Each is based on single input of a wing-tip accelerometer, which is shown to react sufficiently before the peak of the wing-root bending moment. All the three control means are shown to alleviate the extreme wing-root moments at intermediate design flight velocities by 9-16% and the wing-tip accelerations by 26-33%. The wing-tip control surface is found to be most effective, whereas the existing aileron least effective at these speeds. The existing ailerons become even less effective at higher speeds due to reduction in their aeroelastic effectiveness. They are also shown to be most problematic in creating aeroservoelastic instability when the gust-alleviation loop is closed. The effects of the designed control laws on the statistical response of the wing loads to continuous gusts are shown to be similar to their effects on discrete gusts.

Abstract

Frequency-domain and time-domain approaches to dynamic response analysis of aeroservoelastic systems to atmospheric gust excitations are presented. The discrete and continuous gust inputs are defined in either time-domain or stochastic terms. The various options are formulated in a way that accommodates linear control systems of the most general form. The frequency-domain approach is based on the interpolation of generalized aerodynamic force coefficient matrices and the application of Fourier transforms for time-domain solutions. The time-domain approach uses state-space formulation that requires the frequency-dependent aerodynamic coefficients to be approximated by rational functions of the Laplace variable. Once constructed, the state-space equations of motion are more suitable for time simulations and for the interaction with control design algorithms. However, there is some accuracy loss because of the rational approximation. The spiral nature in the complex plane of the gust-related aerodynamic terms is discussed, and means are provided for dealing with the associated numerical difficulties. A hybrid formulation that does not require the rational approximation of the gust coefficients is also presented for optional use in discrete gust response analysis. The various methods were utilized in the ZAERO software and applied to a generic transport aircraft model.

Abstract

A new modal coupling technique for efficient flutter and aeroservoelastic analyses of aircraft with multiple external-store configurations is presented. The aircraft is represented by a set of free-free normal modes obtained with large fictitious masses loading the interface coordinates to yield high-accuracy subsequent coupling results. Each store is represented by its own vibration modes obtained with a subset of statically determined interface coordinates clamped to the ground, and the remaining coordinates (if any) are loaded with large fictitious masses. The technique is formulated in a way that facilitates its application using standard options of common commercially available software packages. A finite element code is used to construct the aircraft and stores modal databases. A linear unsteady aerodynamics code is used to read the modal information, calculate the aerodynamic databases, construct and solve the aircraft-store coupling equations, and perform frequency-domain and state-space flutter analyses with or without control system effects. The process allows analyzing numerous store configurations with previously stored structural and aerodynamic databases, with the store aerodynamics taken into account. The method accuracy and efficiency are demonstrated with a model of the Euro Fighter EF-2000 with four wing store stations.
