42. Effect of Ovariectomy on Circulating Calcitonin Levels in the Rat1

Abstract

The relationship between the reproductive system and calcitonin secretion was investigated in the rat. Plasma levels of calcitonin were measured in normal and ovariec-tomized rats at basal levels and during calcium challenge. Sensitivity of the normal and operated animals to exogenous calcitonin administration was also studied. Ovariectomy leads to a transient fall in both CT levels and in the secretion of the hormone in response to a calcium challenge. However, a greater sensitivity of ovariectomized animals to salmon calcitonin is maintained after the return to normal of both the basal level of the hormone and of its secretion in response to a calcium challenge. Estrogen substitutive therapy does not restore CT levels and antagonizes the action of exogenously administered CT in ovariectomized rats. These results suggest the existence of a direct or indirect effect of ovarian factors, other than estrogens, on CT secretion.

45. Influences of fixatives on the immunodetection of calcitonin in mouse "C" cells during pre- and post- natal development.

Abstract

The presence of calcitonin in fetal and postnatal mouse "C" cell was demonstrated by radioimmunoassays of fetal and adult thyroid extracts and by immunohistochemistry following the use of Bouin's fixative from 18 days postcoitum onwards. However, after fixation with formaldehyde no calcitonin could be detected in fetal or early postnatal samples, although it was still easily detectable in adult tissues treated with this fixative, raising the possibility that there may be age-dependent differences in the chemistry or physiological state of intracellular calcitonin. Experiments conducted with several other fixatives and the results of the radioimmunoassays suggest that the loss of calcitonin reactivity is not due to differences in the anti-genic sites of the molecule at the two stages in development. Neither can the positive detection of fetal material treated with Bouin's fixative be due to any action of this liquid. However, the inhibiting action of the aldehydes could be related to the preservation of the granule membranes in the fetal "C" cells: our ultrastructural studies show that the granules can be morphologically classified in dense-core and homogeneous matrix. Such granule membranes are abundant in the young and adult "C" cell and would be reactive, while the small dense-core granules are prevalent in the fetus and would be unreactive, confirming the hypothesis of a difference of structure of the granule membrane and not of calcitonin between fetal and late postnatal "C" cells.

46. Synthesis, molecular cloning, and restriction analysis of DNA complementary to vitamin D-dependent calcium-binding protein mRNA from rat duodenum.

Abstract

The mRNA coding for rat intestinal calcium-binding protein, a vitamin D3-induced protein (Mr 7500), has been partially purified from growing rat duodenum. Double-stranded DNA synthesized from the purified mRNA preparation was inserted into the PstI site of pBR322, using the oligo(dG-dC) tailing procedure. Clones containing DNA complementary to vitamin D-dependent calcium-binding protein mRNA were selected by differential colony hybridization with [32P] cDNA synthesized from enriched or low vitamin D-dependent calcium-binding protein mRNA preparations. Plasmid DNAs from the selected clones were each verified by both a solution hybrid-arrest assay and a filter hybrid-selection assay. Four recombinant clones showed identical endonuclease restriction maps and contained inserts ranging from 250 to 380 base pairs.

47. RESPONSE OF NORMAL, HYPOTHYROID AND HYPOTHALAMO-PITUITARY INSUFFICIENT CHILDREN TO SYNTHETIC THYROTROPHIN-RELEASING HORMONE

Abstract

Thyrotrophin-releasing hormone (TRH) was synthesized by the solid phase technique, administered to 13 children, and the time-course changes in the serum level of thyroid-stimulating hormone (TSH) assessed. In eight normal children, peak levels of TSH occurred 20 min after the injection, and circulating TSH remained significantly raised for 60 min. In three hypothyroid children, the increase in serum TSH was much greater than in normal children, suggesting the existence of large pituitary TSH stores. In two hypopituitary children with TSH deficiency, TSH reserves seemed normal. One of these patients had a craniopharyngioma; after operation, the increase in serum TSH was reduced. These results show that assay of serum TSH after administration of synthetic TRH provides a test which distinguishes pituitary from hypothalamic defects affecting TSH secretion.

Abstract

It was found in a study on 166 Negro women and also in personal experience that the less affluent patients do not relate the term family planning to birth control child spacing and family size. They think that family planning is only for married couples or that it refers to household budgeting. Also they believe it means counseling and assistance related to infertility or miscarriages. Therefore appropriate action must be taken to clarify the term by putting "birth control" side by side with the term "family planning" in clinics and directories.
