Abstract

The development of immune cell engagers (ICEs) can be limited by logistical and functional restrictions associated with fusion protein designs, thus limiting immune cell recruitment to solid tumors. Herein, a high affinity superantigen‐based multivalent ICE is developed for simultaneous activation and recruitment of NK and T cells for tumor treatment. Yeast library‐based directed evolution is adopted to identify superantigen variants possessing enhanced binding affinity to immunoreceptors expressed on human T cells and NK cells. High‐affinity superantigens exhibiting improved immune‐stimulatory activities are then incorporated into a superantigen‐based tri‐functional yeast‐display‐enhanced multivalent immune cell engager (STYMIE), which is functionalized with a nanobody, a Neo‐2/15 cytokine, and an Fc domain for tumor targeting, immune stimulation, and prolonged circulation, respectively. Intravenous administration of STYMIE enhances NK and T cell recruitment into solid tumors, leading to enhanced inhibition in multiple tumor models. Abstract

Many of the anomalous properties of water are amplified in the deeply supercooled region. Here we present neutron scattering measurements of the density of heavy water confined in a nanoporous silica matrix MCM-41-S (\approx15 {\AA} pore diameter), namely, the equation of state {\rho}(T,P), in a temperature-pressure range, from 300 K to 130 K and from 1 bar to 2900 bar, where bulk water will crystalize. A sudden change of slope in the otherwise continuous density profile (a "kink") is observed below a certain pressure Pc; however, this feature is absent above Pc. Instead, a hysteresis phenomenon in the density profiles between the warming and cooling scans becomes prominent above Pc. Hence, the data can be interpreted as a line of apparent 2nd-order phase transition at low pressures evolving into a line of 1st-order phase transition at high pressures. If so, the existence of a "tricritical point" at Pc \approx 1500 bar, Tc \approx 210 K becomes another possible scenario to explain the exceptionally rich phase behavior of low-temperature confined water., Comment: 15 pages, 4 figures

24. Observation of the Density Minimum in Deeply Supercooled Confined Water

Abstract

Small angle neutron scattering (SANS) is used to measure the density of heavy water contained in 1-D cylindrical pores of mesoporous silica material MCM-41-S-15, with pores of diameter of 15+-1 A. In these pores the homogenous nucleation process of bulk water at 235 K does not occur and the liquid can be supercooled down to at least 160 K. The analysis of SANS data allows us to determine the absolute value of the density of D2O as a function of temperature. We observe a density minimum at 210+-5 K with a value of 1.041+-0.003 g/cm3. We show that the results are consistent with the predictions of molecular dynamics simulations of supercooled bulk water. This is the first experimental report of the existence of the density minimum in supercooled water.

Abstract

By confining water in nanopores, so narrow that the liquid cannot freeze, it is possible to explore its properties well below its homogeneous nucleation temperature TH ~ 235 K. In particular, the dynamical parameters of water can be measured down to 180 K approaching the suggested glass transition temperature Tg ~ 165 K. Here we present experimental evidence, obtained from Nuclear Magnetic Resonance and Quasi-Elastic Neutron Scattering spectroscopies, of a well defined decoupling of transport properties (the self-diffusion coefficient and the average translational relaxation time), which implies the breakdown of the Stokes-Einstein relation. We further show that such a non-monotonic decoupling reflects the characteristics of the recently observed dynamic crossover, at about 225 K, between the two dynamical behaviors known as fragile and strong, which is a consequence of a change in the hydrogen bond structure of liquid water., Comment: 5 pages, 3 figures

26. Pressure Dependence of Fragile-to-Strong Transition and a Possible Second Critical Point in Supercooled Confined Water

Abstract

By confining water in nano-pores of silica glass, we can bypass the crystallization and study the pressure effect on the dynamical behavior in deeply supercooled state using neutron scattering. We observe a clear evidence of a cusp-like fragile-to-strong (F-S) dynamic transition. Here we show that the transition temperature decreases steadily with an increasing pressure, until it intersects the homogenous nucleation temperature line of bulk water at a pressure of 1600 bar. Above this pressure, it is no longer possible to discern the characteristic feature of the F-S transition. 35. Supported Au Nanoparticles on TiO2 for Visible Light Photocatalytic H2O2 Production: Effects of Au Particle Size and Density.

Abstract

Gold nanoparticles supported on titanium dioxide (Au NPs/TiO2) hold great potential as visible-light-driven photocatalysts through plasmon-induced charge separation (PICS). The photo-electrochemical properties of the Au NPs are intrinsically connected to their structure and interfacial interactions with the TiO2 support. Here, we present a highly efficient visible light photocatalytic system for H2O2 production via O2 reduction accomplished by tailoring the particle size and density of Au NPs on TiO2. Oleylamine-stabilized colloidal Au NPs with sub-10 nm particle diameters (d = 3.1, 5.8, and 7.8 nm) were synthesized and uniformly deposited on amine-functionalized TiO2 (P25) with tunable loading amounts (∼0.5–3.4 wt %) through an adsorption approach. These well-controlled Au NPs/TiO2 catalysts allowed us to separately investigate the catalytic effects arising from the Au particle size and particle density. Upon visible light irradiation (λ > 420 nm), increasing particle density of Au NPs proved advantageous for H2O2 formation, wherein injected hot electrons could reduce O2 on another Au NP through interparticle electron transfer. All sizes of the Au NPs/TiO2 catalysts achieved a steady-state H2O2 concentration exceeding 10 mM at elevated particle densities. Moreover, a size-dependent interfacial contact and charge transfer between Au NPs and TiO2 served to modulate the electronic structure of Au NPs, leading to a pronounced enhancement in the PICS efficiency of the smallest Au NP (d = 3.3 nm) with an outstanding apparent quantum yield (10.3%, λ = 528 nm) for visible light photocatalytic H2O2 production.
