Abstract

Existing procedures help express the fusion protein prepared by the coupling of the C-terminus of citrate synthase with the N-terminus of malate dehydrogenase in Escherichia coli. Gel filtration and Western blot analyses help determine the molecular weight of isolated, purified proteins. Kinetic studies of the fusion protein reveal less transient time for the coupled reaction sequence for fusion proteins in comparison with that of free enzymes. The fusion protein exhibits higher sequestering capacity toward oxaloacetate in comparison with that of systems with free enzymes.

Abstract

Simultaneous application of two chemically unique functional monomers, the feebly basic-2-vinylpyridine and the acidic and hydrogen bonding methacrylic acid, reveal noncovalent molecular imprinting in ethylene glycol dimethylacrylate based copolymers. Polymers synthesized with only one of these monomers, do not exhibit the enhanced recognition abilities of imprinted polymers.

Abstract

A novel and facile preparation method for spherical molecularly imprinted polymer (MIP) beads is presented. Two types of beads were synthesized and investigated: (i) silica-MIP composites were obtained by filling spherical, porous C-4-coated silica beads with print molecule and monomers followed by polymerisation; (ii) spherical molecularly imprinted polymer beads were acquired mirroring the silica particles in size, shape and pore structure by removing the silica matrix from the silica-MIP composites. With regard to their chromatographic properties and yield of the materials both types of particle were more advantageous compared to irregularly shaped traditional MIPs. Also the work-up time to obtain imprinted spherical particles is greatly reduced compared to traditional methods using polymer monoliths, which have to be ground, sieved and sedimented. Generally, the described method may open new possibilities for synthesis of novel types of imprinted polymer formats such as membranes, bulk polymers, films or in-situ columns using appropriate support or sacrificial materials., QC 20100525

50. Imprinted polymer based sensor system for herbicides using differential-pulse voltammetry on screen printed electrodes

Abstract

A sensor system for the herbicide 2,4-dichlorophenoxyacetic acid has been developed based on specific recognition of the analyte by a molecularly imprinted polymer and electrochemical detection using disposable screen-printed electrodes. The method involves a competitive binding step with a nonrelated electrochemically active probe. For batch binding assays, imprinted polymer particles are incubated in suspension with the analyte and the probe, followed by centrifugation and quantification of the unbound probe in die supernatant. Two different compounds, namely 2,4-dichlorophenol and homogentisic acid, were tested as potential electroactive probes. Both compounds could be conveniently detected by differential-pulse voltammetry on screen-printed, solvent-resistant three-electrode systems having carbon working electrodes. Whereas 2,4-dichlorophenol showed very high nonspecific binding to the polymer, homogentisic acid bound specifically to the imprinted sites and thus allowed calibration curves for the analyte in the micromolar range to be recorded. An integrated sensor was developed by coating the imprinted polymer particles directly onto the working electrode. Following incubation of the modified electrode in a solution containing the analyte and the probe, the bound fraction of the probe is quantified. This system provides a cheap, disposable sensor for rapid determination of environmentally relevant and other analytes.
