Abstract

We propose a method to test dependence or conditional dependence be- tween parts of the domain of functional data. The tests are based on permutation procedure that tests if suitable blocks of the covariance or precision matrix of ba- sis expansion coefficients are equal to zero. We show that the procedure is able to identify the true structure of conditional dependence.

Abstract

International audience; The GMO90+ project was designed to identify biomarkers of exposure or health effects in Wistar Han RCC rats exposed in their diet to 2 genetically modified plants (GMP) and assess additional information with the use of metabolomic and transcriptomic techniques. Rats were fed for 6-months with 8 maize-based diets at 33% that comprised either MON810 (11% and 33%) or NK603 grains (11% and 33% with or without glyphosate treatment) or their corresponding near-isogenic controls. Extensive chemical and targeted analyses undertaken to assess each diet demonstrated that they could be used for the feeding trial. Rats were necropsied after 3 and 6 months. Based on the Organization for Economic Cooperation and Development test guideline 408, the parameters tested showed a limited number of significant differences in pairwise comparisons, very few concerning GMP versus non-GMP. In such cases, no biological relevance could be established owing to the absence of difference in biologically linked variables, dose-response effects, or clinical disorders. No alteration of the reproduction function and kidney physiology was found. Metabolomics analyses on fluids (blood, urine) were performed after 3, 4.5, and 6 months. Transcriptomics analyses on organs (liver, kidney) were performed after 3 and 6 months. Again, among the significant differences in pairwise comparisons, no GMP effect was observed in contrast to that of maize variety and culture site. Indeed, based on transcriptomic and metabolomic data, we could differentiate MON- to NK-based diets. In conclusion, using this experimental design, no biomarkers of adverse health effect could be attributed to the consumption of GMP diets in comparison with the consumption of their near-isogenic non-GMP controls.

Abstract

In many application fields, data corresponds to curves. This work focuses on the analysis of spectrometric curves, composed of hundreds of ordered variables that corresponds to the absorbance values measured for each wavenumber. In this context, an automatic statistical procedure is developped, that aims at building a prediction model taking into account the heterogeneity of the observed data. More precisely, a diagnosis tool is built in order to predict a metabolic disease from spectrometric curves measured on a population composed of patients with differents profile. The procedure allows to select portions of curves relevant for the prediction and to build a partition of the data and a sparse predictive model simultaneously, using a mixture of penalized regressions suitable for functional data. In order to study the complexity of the data and of the application case, a method to better understand and display the interactions between variables is built. This method is based on the study of the covariance matrix structure, and aims to highlight the dependencies between blocks of variables. A medical example is used to present the method and results, and allows the use of specific visualization tools., Dans de nombreux domaines d’application, les données récoltées correspondent à des courbes. Ce travail se concentre sur l’analyse de courbes de spectrométrie, constituées de plusieurs centaines de variables ordonnées, correspondant chacune à une valeur d’absorbance associée aux nombres d’ondes mesurés. Dans ce contexte, une méthode de traitement statistique automatique est développée, avec pour objectif la construction d’un modèle de prédiction prenant en compte l’hétérogénéité des données observées. Plus particulièrement, un modèle de diagnostic d’une maladie métabolique est établi à partir de courbes mesurées sur des individus provenant d’une population constituée de profils de patients différents. La procédure développée permet de sélectionner l’information pertinente sous forme de portions de courbes discriminantes, puis de construire de façon simultanée une partition des données et un modèle de prédiction parcimonieux grâce à un mélange de régressions pénalisées adapté aux données fonctionnelles. Ces données étant complexes, tout comme le cas d’application étudié, une méthode permettant une meilleure compréhension et une meilleure visualisation des interactions entre les portions de courbes a par ailleurs été développée. Cette méthode se base sur l’étude de la structure des matrices de covariance, avec pour but de faire ressortir des blocs de dépendances entre intervalles de variables. Un cas d’application médicale est utilisé pour présenter la méthode et les résultats, et permet l’utilisation d’outils de visualisation spécifiques.

Abstract

The GMO90+ project was designed to identify biomarkers of exposure or health effects in Wistar Han RCC rats exposed in their diet to 2 genetically modified plants (GMP) and assess additional information with the use of metabolomic and transcriptomic techniques. Rats were fed for 6-months with 8 maize-based diets at 33% that comprised either MON810 (11% and 33%) or NK603 grains (11% and 33% with or without glyphosate treatment) or their corresponding near-isogenic controls. Extensive chemical and targeted analyses undertaken to assess each diet demonstrated that they could be used for the feeding trial. Rats were necropsied after 3 and 6 months. Based on the Organization for Economic Cooperation and Development test guideline 408, the parameters tested showed a limited number of significant differences in pairwise comparisons, very few concerning GMP versus non-GMP. In such cases, no biological relevance could be established owing to the absence of difference in biologically linked variables, dose-response effects, or clinical disorders. No alteration of the reproduction function and kidney physiology was found. Metabolomics analyses on fluids (blood, urine) were performed after 3, 4.5, and 6 months. Transcriptomics analyses on organs (liver, kidney) were performed after 3 and 6 months. Again, among the significant differences in pairwise comparisons, no GMP effect was observed in contrast to that of maize variety and culture site. Indeed, based on transcriptomic and metabolomic data, we could differentiate MON- to NK-based diets. In conclusion, using this experimental design, no biomarkers of adverse health effect could be attributed to the consumption of GMP diets in comparison with the consumption of their near-isogenic non-GMP controls. Abstract

Introduction: Depression is a frequent pathology with severe consequences on a humane and social plane. Its diagnosis, based on international diagnostic criteria, could be improved by the use of diagnostic tools. These diagnostic tools are still under-used in general practice. The objective of the study was to assess the limitations of these tools, so they can be considered when used in general medicine. Methods: Qualitative study by semi-directive interviews of a reasoned sample of general practitioners. Transcripts were coded by an independent double linear encoding until saturation. An inductive analysis was used on the linear encoding. A consensual axial and systematic encoding was obtained. It was organised according to a heuristic scheme. Results: Saturation was obtained in 9 interviews. 65 axial codes were obtained. Three themes were found. The tools were unknown (lack of training). They were judged useless in practice. They were considered maladjusted in their indications and shape. Discussion: These barriers, explained by specificities of practice within general medicine, showed two main axes for the improvement of the use of one tool. The first one would be to promote this tool in medical studies and to the patients. The second would be to give a particular attention to the ergonomics of the tool: clear, rapid in its use, accessible in general medicine practice, flexible, its objectives should be broad and well defined. Conclusion: The promotion of a new diagnostic tool for depression should be adapted to the general medicine practice.; Introduction : La dépression est une pathologie fréquente et lourde de conséquences sur le plan humain et sociétal. Trois thèmes ont été trouvés. Les outils étaient méconnus (carence de formation). Ils étaient jugés inutiles en pratique. Ils étaient jugés inadaptés dans leur indication et dans leur forme. Discussion : Ces barrières, justifiées par les spécificités de l'exercice pratique du médecin généraliste, mettaient en avant deux axes pour améliorer l'utilisation d'un outil. D'une part, il serait à promouvoir en formation médicale et auprès du patient. Abstract

Body mass index is a known breast cancer risk factor due to, among other mechanisms, adipose-derived hormones. We developed a method for steroid hormone profiling in adipose tissue to evaluate healthy tissue around the tumor and define new biomarkers for cancer development. A semi-automated sample preparation method based on gel permeation chromatography and subsequent derivatization with trimethylsilyl (TMS) is presented. Progestagens and androgens were determined by GC-EI-MS/MS (LOQ 0.5 to 10 ng/g lipids). For estrogen measurement, a highly sensitive GC-APCI-MS/MS method was developed to reach the required lower limits of detection (0.05 to 0.1 ng/g lipids in matrix, 100-200 fg on column for pure standards). The combination of the two methods allows the screening of 27 androgens and progestagens and 4 estrogens from a single sample. Good accuracies and repeatabilities were achieved for each compound class at their respective limit of detection. The method was applied to determine steroid hormone profiles in adipose tissue of 51 patients, collected both at proximity and distant to the tumor. Out of the 31 tested steroid hormones, 14 compounds were detected in human samples. Pregnenolone, 17-hydroxypregnenolone, dehydroepiandrosterone (DHEA), and androstendione accounted together for 80% of the observed steroid hormone profiles, whereas the estrogens accounted for only 1%. These profiles did not differ based on sampling location, except for ß-estradiol; steroid hormone conversions from androgens to estrogens that potentially take place in adipose or tumoral tissue might not be detectable due a factor 100 difference in concentration of for example DHEA and ß-estradiol. Abstract

International audience; Quantification of outputs of antibacterial agents from fish farms is necessary to assess their environmental impact. In effluents, antibacterial agents are dissolved or associated with mineral or organic materials, the heaviest of which quickly settle at the exit of the fish farm. Oxolinic acid (OA) is one of the quinolones the most frequently used in turbot farms. For the first time, procedures are described for determination of dissolved, suspended, and settled forms of OA by reversed-phase high-performance liquid chromatography. Chromatography was performed on a 5 mu m C-18 cartridge, with a mixture of acetonitrile and aqueous orthophosphoric acid solution as mobile phase, and fluorescence detection. OA was easily and completely isolated by solid-liquid extraction. The method is selective, accurate, and precise; limits of quantitation were 0.005 mu g mL(-1), 0.025 mu g per filter, and 0.025 mu g g(-1), respectively for dissolved, suspended, and settled forms of OA. These methods enabled initial assessment of the amounts of dissolved, suspended, and settled forms of OA (72, 23, and 2%, respectively) of the antibacterial agent administered.

Abstract

Humans are significantly exposed to phthalates via food packaging, cosmetics and medical devices such as tubings and catheters. Testicular Leydig cells (LCs) are suggested to be among the main targets of phthalate toxicity in the body. However, their sensitivity to phthalates is species-dependent. This paper describes the response of the LCs from different species (mouse, rat and human) to phthalate exposure in different experimental paradigms (in vivo, ex vivo and in vitro), with particular focus on mechanisms of phthalate action on LC steroidogenesis. A comprehensive analysis of the impact of phthalate diesters and phthalate monoesters on LCs in different stages of their development is presented and possible mechanisms of phthalates action are discussed. Finally novel, not yet fully elucidated sites of action of phthalate monoesters on the backdoor pathway of 5α-dihydrotestosterone biosynthesis in immature mouse LCs and their effects on steroidogenesis and redox state in adult mouse LCs are reported. 40. Expansion of interventional endoscopy and day-case procedures: A nationwide longitudinal study of gastrointestinal endoscopy in France.

Background: An accurate assessment of the evolution of GI endoscopy volumes is warranted to identify long-term trends and to help anticipate training, infrastructure and human resource needs. The main objective of this longitudinal study was to evaluate the evolution of GI endoscopy in France., Methods: This retrospective study consisted of a cross-sectional analysis repeated each year from 2008 to 2018 using data from a national health database related to hospital admissions. All day-case and hospital stays presenting at least one of the 119 GI endoscopy procedures were extracted., Results: This study showed an increase in day-case and hospital stays including a GI endoscopy procedure of 18.4 %. In addition, day-case endoscopy increased from 67.8 % to 76.9 % of hospital admissions. There was a 19.6 % increase in lower GI endoscopy, with in particular a 247 % increase in endoscopic mucosal resection. Abstract

Masculinization of the external genitalia in humans is dependent on formation of 5α-dihydrotestosterone (DHT) through both the canonical androgenic pathway and an alternative (backdoor) pathway. The fetal testes are essential for canonical androgen production, but little is known about the synthesis of backdoor androgens, despite their known critical role in masculinization. In this study, we have measured plasma and tissue levels of endogenous steroids in second trimester human fetuses using multidimensional and high-resolution mass spectrometry. Results show that androsterone is the principal backdoor androgen in the male fetal circulation and that DHT is undetectable (<1 ng/mL), while in female fetuses, there are significantly lower levels of androsterone and testosterone. In the male, intermediates in the backdoor pathway are found primarily in the placenta and fetal liver, with significant androsterone levels also in the fetal adrenal. Backdoor intermediates, including androsterone, are only present at very low levels in the fetal testes. This is consistent with transcript levels of enzymes involved in the alternate pathway (steroid 5α-reductase type 1 [SRD5A1], aldo-keto reductase type 1C2 [AKR1C2], aldo-keto reductase type 1C4 [AKR1C4], cytochrome P450 17A1 [CYP17A1]), as measured by quantitative PCR (qPCR). These data identify androsterone as the predominant backdoor androgen in the human fetus and show that circulating levels are sex dependent, but also that there is little de novo synthesis in the testis. Instead, the data indicate that placental progesterone acts as substrate for synthesis of backdoor androgens, which occurs across several tissues. Masculinization of the human fetus depends, therefore, on testosterone and androsterone synthesis by both the fetal testes and nongonadal tissues, leading to DHT formation at the genital tubercle.
