5. Research Opportunities From the Archived Neuroanatomic Slide Collections of the National Museum of Health and Medicine

Abstract

HUMAN BRAIN COLLECTIONS: YAKOVLEV-HALEEM COLLECTION: The Yakovlev-Haleem collection covers normal and pathological development of the brain as well as pathologic conditions based on the study of 1,200 human brain specimens collected between 1930 and 1994. Each of the brains are prepared in whole-mount serial histologic sections on glass slides. The collection consists of a total of more than 250,000 slides, with serially cut whole-brain celloidin-embedded sections that are stained alternately with Nissl and myelin preparations. Each specimen is accompanied by a case record that includes actuarial, clinical, and /or autopsy data. In addition to normative controls, pathological specimens include cerebrovascular disease, a wide variety of developmental disorders, results of postoperative neurosurgery (especially post frontal lobotomy cases) and also included are examples of various neuropathologic conditions including age-related neurodegenerative disorders. The collection was built by Dr. Paul Ivan Yakovlev (1894-1983), a neurologist/neuropathologist/neuroanatomist and esteemed member of the Harvard Medical School faculty. Yakovlev began the collection in 1930 while working at the Monson State Hospital and then moved to the Walter E. Fernald State School. In 1974 the entire collection from Harvard to the Armed Forces Institute of Pathology (AFIP), where it was managed by curator Mohamad Haleem. In 1994 it was transferred to the NMHM and renamed the Yakovlev-Haleem Collection. Of the collection, in 1972, Derek Denny-Brown, Professor of Neurology at the Harvard Medical School wrote: “There is no other collection of neuroanatomical material in serial sections consistently fixed, embedded, cut, stained and mounted to such exacting standards. Dr. Yakovlev early recognized that only in this way could truly accurate comparative measurements be made. In this respect the Yakovlev Collection is an irreplaceable reference material, patiently collected over a period of 42 years., RITM0032953, In past years, researchers created very large collections of serial whole mount histology brain slides. Many of these archival collections were donated to the US Government and are housed at the National Museum of Health and Medicine (NMHM), Silver Spring, MD. Currently, the only way for researchers to use these precious slides has been to travel to Maryland and physically examine them onsite. Nevertheless, over the years these collections have been widely used and a number of seminal publications have emerged from this unique resource. However, we are now proposing to digitize these entire collections and make them widely available through on-line internet access. The collections being considered for this project are as follows: 1) Yakovlev-Hakeem Collection consisting of 1200 serially cut human brain specimens (250,000+ slides, Nissl and myelin serial sections representing normal, and many pathologic entities). 2) Welker Comparative Neuroanatomy Collection: serial sections of 275 brains representing over 120 species (aardvark to zebra). 3) Starr Collection containing serial sections of brains of chimpanzees and gorillas. 3) Meyer Collection with serially sectioned slides from the brains of patients seen by psychiatrist Dr. Adolph Meyer who practiced at Johns Hopkins Hospital in the early part of the 20th century (including his clinical note), and the Blackburn Collection with neuropathology evaluations of St. Elizabeth’s Hospital cases dating back to 1884. Both the Meyer and Blackburn collections date from the pre-antibiotic era with numerous examples of unique pathologic entities, (e.g. various forms of neurosyphillis, catatonic schizophrenia, etc.). Examples of these holding will be displayed and we will provide an on-line demonstration of the opportunities available to researchers for dealing with this material in a digitized format.

Abstract

The U.S. Geological Survey (USGS) has long recognized the strategic importance and value of well-managed data assets as an integral component of scientific integrity and foundational to the advancement of scientific research, decision making, and public safety. The USGS investment in the science lifecycle, including collection of unbiased data assets, interpretation, peer review,' interpretive publications, and data release, ultimately contributes to the transparency and availability of science. Foundational Government directives and laws, such as the Foundations for Evidence-Based Policymaking Act of 2018 (Public Law 115-435, 132 Stat. 5529) as well as Executive Order 13642, "Making Open and Machine Readable the New Default for Government Information," provide a framework for addressing strategic data management. The USGS Data Strategy builds on that framework by outlining high-level goals and objectives that serve as a long-term, decadal guide toward achieving a broad, data-focused vision. Benefits of the USGS Data Strategy are many. The USGS will contribute to open science by increasing efficiencies in the consistent management of valuable data assets; driving innovation that results in modernized capabilities to ensure data are analysis ready; increasing data skills across the Bureau to enhance workforce data literacy; broadening capacity to understand and address needs of stakeholders; and measuring progress in producing findable, accessible, interoperable, and reusable (FAIR) data products. The major goals and objectives of the USGS Data Strategy promote maximizing the utility of USGS data based on stakeholder needs, promoting data innovation, coordinating common data practices, modernizing our USGS enterprise data architecture, and enhancing our data-centric culture. The goals and objectives in the strategy align with other Bureau strategic plans, guidance, and directives from the Department of the Interior and the Federal Government. This strategy is a key component to strengthen the Bureau's data ecosystem to ensure a relevant, long-term capacity that supports internal needs and achieves its scientific mission in the most efficient and effective manner. Abstract

This study examines the integration of CliftonStrengths, within the Department of Animal Science at Iowa State University. In previous semesters, CliftonStrengths was introduced to freshmen, but no further application of the assessment took place. Fall 2023 incoming students completed more intensive application following completion of the assessment. The objective of this study aims to determine if students’ soft skills are enhanced through awareness and application of their own strengths. This study surveyed new and current students, analyzing their perceptions towards CliftonStrengths and their experiences in group settings. Three surveys were administered at different time points during the fall 2023 semester, one current student survey (CSS) at the start of the semester, and two new student surveys for the beginning (pre-NSS) and end (post-NSS) of the semester. Surveys captured demographic data and responses on a 0 to 10 scale. Response rates were 42% from pre-NSS, 22% CSS, and 24% from post-NSS. The pre-NSS and CSS contained 46 and 37 first-generation students, respectively. The pre-NSS consisted of 120 freshmen and 17 transfer students, and the CSS consisted of 43 sophomores, 44 juniors, and 59 seniors. Survey questions were structured around the primary domains within CliftonStrengths; strategic thinking, relationship building, influencing, and executing. Analysis of the pre-NSS responses showed 61% of new students were not familiar with CliftonStrengths and 78% have not taken the assessment. The CSS showed that 66% of current students have taken the CliftonStrengths Assessment. New students that completed intensive application of CliftonStrengths reported a 2.8-point increase from pre-NSS (4.9 ± 2.96) to post-NSS (7.7 ± 2.51) in response to identifying the value of CliftonStrengths in everyday life. New students reported being more comfortable with strengths at the conclusion of this semester with an average response of 9 ± 1.4 on a 10-point scale. The responses to “I believe someone in my career path should know their strengths” had higher average response rates from post-NSS (7.7 ± 0.26) compared with CSS (6.9 ± 0.20). Student responses to “I believe knowing my Strengths helps me at my best” also had higher average response rates from post-NSS (8.3 ± 0.24) compared with CSS (7.4 ± 0.18). Students responded to “I feel confident that I can recognize strengths in others” with higher response rates in the post-NSS (7.1 ± 0.30) compared with CSS (6.1 ± 0.18). All questions asking about the ability to identify domain types of other individuals were higher in post- NSS responses compared with CSS. The post-NSS responses to understanding how to use different domains in a group were greater for strategic thinkers, relationship builders, and influencers. In conclusion, the intensive application of Clifton Strengths in the Department of Animal Science at Iowa State University has increased familiarity, confidence, and comfort with their strengths. Abstract

The purpose of this retrospective pilot study was to determine if aquatic therapy (AT) intervention concurrent with occupational therapy (OT) yielded greater functional gains in stroke patients than OT treatment alone. A retrospective chart review was conducted evaluating subjective data from an inpatient rehabilitation hospital in Morgantown, WV. Thirty-nine stroke survivor charts were examined. Patients who received OT with or without concurrent aquatic therapy were included. Differences were examined in the Functional Independence Measure (FIM) scores in patients who received (a) AT plus conventional OT or (b) conventional OT alone. Discharge bed/chair transfer (p = 0.009) and locomotion (p = 0.01) scores were higher for people who received OT and AT versus patients only receiving OT. Linear regression indicated AT treatment predicted discharge bed/chair transfer FIM score (p = 0.02), discharge locomotion (walking) FIM score (p = 0.002), discharge stairs FIM score (p = 0.04), and change in bed/chair transfer FIM score (p = 0.02). These findings indicate that AT, combined with OT, may predict success on specific FIM outcomes (e.g., bed/chair transfer FIM score, locomotion [walking] FIM score) in stroke patients. Abstract

Amphibian biology is intricate, and there are many inter-related factors that need to be understood before establishing successful Conservation Breeding Programs (CBPs). Nutritional needs of amphibians are highly integrated with disease and their husbandry needs, and the diversity of developmental stages, natural habitats, and feeding strategies result in many different recommendations for proper care and feeding. This review identifies several areas where there is substantial room for improvement in maintaining healthy ex situ amphibian populations specifically in the areas of obtaining and utilizing natural history data for both amphibians and their dietary items, achieving more appropriate environmental parameters, understanding stress and hormone production, and promoting better physical and population health. Using a scientific or research framework to answer questions about disease, nutrition, husbandry, genetics, and endocrinology of ex situ amphibians will improve specialists' understanding of the needs of these species. In general, there is a lack of baseline data and comparative information for most basic aspects of amphibian biology as well as standardized laboratory approaches. Instituting a formalized research approach in multiple scientific disciplines will be beneficial not only to the management of current ex situ populations, but also in moving forward with future conservation and reintroduction projects. This overview of gaps in knowledge concerning ex situ amphibian care should serve as a foundation for much needed future research in these areas.

Abstract

Animal managers in zoological facilities are faced with unique challenges when feeding large collections of species. Few nutrient requirements of exotic species are known or established; therefore, diet formulation is a series of scientific extrapolations, mingled with an understanding of natural history of individual species. Goals of nutrition programs in zoos also include supporting behavioral diversity. This leads to the use of a wide variety of animal-based ingredients within zoo nutrition programs to support species across multiple taxa inclusive of amphibians, reptiles, avians, mammals and fish. Commonly used rendered protein ingredients such as fish meal and other meat meals are commonly used in extruded and processed diets developed for exotic species. For example, a commercially marketed extruded diet designed for cranes includes fish meal, porcine meat and bone meal and dried whey. An extruded diet for small exotic felids includes chicken meal, poultry by-product meal and fish meal. While these are commonly accepted animalbased ingredients across different animal industries, other animal protein ingredients are well-accepted in zoos and yet fail to reach general acceptance in other industries including pet food. Some of these common animal-based ingredients used in various zoo diets include insects, whole prey (rodents), raw meat (inclusive of horse and pork) and raw organ meats. These ingredients are often met with reluctance, misunderstanding, or dated information when applied to other animal industries. One example of dated information is the fear of using raw pork in relation to the risk of trichinella. The zoo nutrition industry may provide benefit to other animal industries when exploring opportunities to safely utilize and include novel animal-based ingredients in diet formulations. 25. Study of Water Adsorption in Poly(N-isopropylacrylamide) Thin Films Using Fluorescence Emission of 3-Hydroxyflavone Probes

Abstract

The noncontact measurement of water uptake in microscale (1−100 μm), thermoresponsive poly(N-isopropylacrylamide) thin films is challenging. We assessed the efficacy of three different 3-hydroxyflavone (3-HF)-based fluorophores to monitor water uptake in pNIPAM thin films close to the lower critical solution temperature (LCST) at 25 and 37 °C. These 3-HF fluorophores undergo excited-state intramolecular proton transfer, yielding emission from normal (N*) and tautomeric (T*) excited-state forms. The emission intensity ratio, log(IN*/IT*), and N* band position are environmentally sensitive. Water adsorption in pNIPAM thin films follows a nonlinear, two-phase process: at low relative humidity, the adsorbed water disrupts polymer−fluorophore hydrogen bonding, yielding small changes in log(IN*/IT*) ratios and overall intensity; at higher relative humidity, these intensity parameters (but not fluorescence lifetime) change dramatically, indicating a larger change in local polarity. These probes are thus sensitive indicators of water uptake in pNIPAM.

26. Ouantitative, Non-Contact Measurements of the Physicochemical Properties of Thermoresponsive Polymers using Fluorescence Methods

Abstract

In biomedical science and engineering the use of microscale thin polymer films is widespread. These polymer films are often formed on complex geometries which can provide problems for in-situ analysis and the accurate measurement of multiple physiochemical properties of thin polymer films is critical in understanding the polymer function and performance. When undertaking physicochemical characterisation of thermoresponsive polymers we must consider polymer structure and also the environment created by a polymer when it is used as a thin film. In addition, we need to account for the infiltration of solvents and other factors which may mediate the polymer behaviour. Water uptake in a polymer film can lead to significant changes in the physiochemical properties. This in turn can affect mechanical properties and may lead to such problems, in medical devices, as reduced adhesion, pronounced physical and chemical aging and swelling and expansion, compromising the intended function of the polymer and hindering biocompatibility. Thus there is a need for a non-contact, non-destructive method of analysis of these polymer films. Fluorescence spectroscopy offers a potential non-contact method for the in-situ analysis of thin polymer films. In this work we demonstrate the use of the dual-band emission, 3-hydroxyflavone based fluorophores for monitoring the hydration process in thin (~ 10 m) films of poly(N-isopropylacrylamide) (PNIPAm), and poly(NIPAM-co-ethylpyrrolidone-methacrylate) (poly(NIPAM-co- EPM) copolymers. The fluorescence of 3-HF probes is very sensitive to changes in local polarity and hydrogen bonding. This is observed as shifts and changes in the relative intensity of the two emission bands (N* and T*), which originate from an excited state intramolecular proton transfer (ESIPT) process. The spectroscopic parameters of these probes can thus be correlated with the physiochemical properties of the probe microenvironment. We incorporated the probes into films of the thermoresponsive polymers and studied their fluorescence emission behaviour at different levels of humidity. Analysis of the spectral data showed that with increasing humidity, the ratio of two emission bands (log(IN*/IT*)), for all ii fluorophores increases. Changes were also observed in position of two emission bands; as humidity increases the N* band shifts to the red with respect to the T* band. It was also observed that the spectral intensity varied with increasing humidity. When measurements were made above and below the lower critical solution temperature (LCST) , it was clear that the films were still absorbing water above the LCST. Fluorescence measurements also revealed that these probes are also very sensitive to the subtle differences in copolymer composition as the fraction of EPM is increased. Results indicate a more equilibrated hydration process with the incorporation of EPM. Polarity measurements performed (using the solvatochromic Reichardt¿s betaine dye) revealed that increasing water content in the polymer films resulted in an increase in polarity (which would be expected due to the highly polar nature of water). However, little or no variation in band positions or ET(30) parameter indicated that polarity did not vary with changes in chemical composition, with the exception of PEPm films where a red shift of the absorption band was noted indicating a decrease in polarity. However a non-linear response of band position with increasing RH for PEPm indicated that solvent sorting was taking place.

Interest in preparing pet foods at home has continued to build momentum over the last decade. Websites, blogs, and social media groups praise the benefits of feeding raw and home-prepared diets. Consistent research has demonstrated the beneficial effects of raw diets on macronutrient digestibility (primarily fat and protein); however, research is lacking on overall health impacts. While it is important to acknowledge the lack of substantial research, owner choice and compliance also should be considered. With well-established nutrient requirements for dogs and cats available through AAFCO and the National Research Council (NRC), it is relatively uncomplicated to meet requirements when typical processing methods are not required (extrusion or canning). Although it is not complicated to meet the requirements with home-based fresh or lightly cooked ingredients, care must be taken to properly ensure that vitamin and mineral requirements are met. In review of over 100 homemade diets submitted for evaluation, 0% met all vitamin and mineral requirements for dogs or cats. All diets were supplemented with some form of calcium. Approximately 25% of submitted diets utilized bones as the source of calcium. Bones can be problematic depending on type, particularly for digestion, broken pieces that can cause intestinal blockages, and broken teeth. None of the diets were properly balanced for copper, zinc and iron. It is likely, this triad of minerals has been overlooked in diet formulations. It is critical to note that improper balance of these minerals could play a significant role in oxidative stress and aging. Iodine is typically met through additions of kelp; however, variations in quality can result in over or under-supplementation having detrimental effects on the thyroid. Vitamins E and D also are often undersupplemented in these diets. Educating owners on nutrient requirements and ratios, and providing proper supplementation, can assure a quality and balanced diet.
