Abstract

Simple Summary: Fourier Transform Infrared Spectroscopy (FTIR) is a rapid, cost-effective, and routinely used tool for milk analysis that can be easily applied to the classification of valuable dairy products such as Podolica milk. In the work herein presented, we applied machine learning to rapidly classify the FTIR datasets of milk from different bovine breeds. We were able to successfully recognize non-Podolica milk with 86% sensitivity and 100% specificity, demonstrating that the combination of these tools might be used in the future for the rapid classification of milk from different bovine breeds. The Podolica cattle breed is widespread in southern Italy, and its productivity is characterized by low yields and an extraordinary quality of milk and meats. Most of the milk produced is transformed into "Caciocavallo Podolico" cheese, which is made with 100% Podolica milk. Fourier Transform Infrared Spectroscopy (FTIR) is the technique that, in this research work, was applied together with machine learning to discriminate 100% Podolica milk from contamination of other Calabrian cattle breeds. The analysis on the test set produced a misclassification percentage of 6.7%. Among the 15 non-Podolica samples in the test set, 2 were misclassified and recognized as Podolica milk even though the milk was from other species. The correct classification rate improved to 100% when the same method was applied to the recognition of Podolica and Pezzata Rossa milk produced by the same farm. Furthermore, this technique was tested for the recognition of Podolica milk mixed with milk from other bovine species. The multivariate model and the respective confusion matrices obtained showed that all the 14 Podolica samples (test set) mixed with 40% non-Podolica milk were correctly classified. In addition, Pezzata Rossa milk produced by the same farm was detected as a contaminant in Podolica milk from the same farm down to concentrations as little as 5% with a 100% correct classification rate in the test set. The method described yielded higher accuracy values when applied to the discrimination of milks from different breeds belonging to the same farm. One of the reasons for this phenomenon could be linked to the elimination of the environmental variable. Abstract

Dietary particle size is one of the most relevant factors influencing rumen function and the selection of the most palatable components of the total mixed ration (TMR) by cows. The aim of this study was to evaluate the particle size distribution (PSD), homogeneity, sorting level, physically effective NDF (peNDF) content of TMRs, and production performances in Calabrian commercial dairy herds in southern Italy. The research was conducted in 13 farms, including 8 with hay-based TMR and 5 with silage-based TMR. All herds delivered fresh feed once a day. At each farm, the TMRs were examined with the Penn State Particle Separator (PSPS) to determine PSD, homogeneity, and feed sorting at two time points (i.e., at fresh feed delivery = T0 and 24 h after feed delivery = Tf). None of the diets that were evaluated met the recommended PSD, showing an excess of long fraction, very short fraction, or both. The homogeneity was good except for three diets, but particle selection raised some concerns in 85% of the farms due to the preferential consumption of the very short fraction by the cows, with rejection of the long fractions. All the diets analyzed met the Penn State University recommended neutral detergent fiber (NDF) values (>28%) except for one farm's diet. In three of the TMRs observed, however, the content of peNDF > 8 mm was less than 15%. NDF and peNDF > 8 mm values of TMRs showed statistically significant correlations with milk fat content. Simple Summary: Preweaning nutrition plays a crucial role in the growth of dairy calves, determining their future productivity and overall health. Researchers are still studying the most appropriate dietary strategies in the preweaning and early post-weaning periods. In particular, the effect of different levels of forage inclusion in the diet of young cattle is not yet clear. Our study evaluated the effects of starter restriction and ad libitum inclusion of the total mixed ration on intake, growth performance, rumination time, and health status of Holstein dairy calves during preweaning and in the immediate post-weaning period. The results obtained from the present study indicate that providing TMR (high fiber content, with or without concentrate restriction) does not impair growth performance and health of calves, and at the same time it leads to a greater time spent for ruminating when calves are weaned. The latter indicates a better rumen development and likely a better rumen fermentation efficiency in post-weaning. The aim of the current study was to evaluate the effect of the starter restriction and of the ad libitum TMR (total mixed ration) inclusion on intake, growth performance, rumination time (RT), and health condition of Holstein dairy calves during weaning. We randomly assigned thirty female Holstein calves (with an average weight of 38.5 ± 1.96 kg at birth) to one of three treatments. From 21 days of age, the calves were fed one of three treatments as follows: a control diet (CTR) with an ad libitum calf starter but without TMR; Treatment 1 diet (TRT1) with both an ad libitum calf starter and ad libitum TMR; Treatment 2 diet (TRT2) with ad libitum TMR and a restricted amount of a calf starter (50% of the intake recorder in the control group day by day). Calves in the TRT2 group, between 56 and 63 days of age, had a lower body weight (80.1; 79.5; 75.6 kg for the CTR, TRT1, and TRT2 groups, respectively) compared with CTR and TRT1 calves. This outcome is ascribed to the average daily gain (0.759; 0.913; 0.508 kg/day for the CTR, TRT1, and TRT2 groups, respectively), resulting also in TRT2 being lower than CTR or TRT1 calves. The inclusion of ad libitum TMR increased the rumination time, especially after weaning (15.28 min/h, 18.38 min/h, and 18.95 min/h for the CTR, TRT1, and TRT2 groups, respectively). Concerning the rumen metabolism and inflammometabolic response, overall, no differences were observed between the three dietary treatments. In conclusion, the results indicated that a TMR could partially replace a calf starter in weaning dairy calves, since neither growth performance nor health status were impaired. Abstract

Colostrum of ruminants is the first secretion of the mammary gland produced after birth. It is a complex biological fluid that ensures the immune support of newborns in early life. There is a growing interest in human consumption of colostrum as a nutraceutical due to its nutrients, antibodies, vitamins, immune factors, and growth factor contents. This review illustrates the applications of colostrum as an emerging food focusing on its nutraceutical aspect and the importance of colostrum as a functional food. Emphasis is also posed on the use of colostrum for its potential probiotic properties as well as an immunomodulator, antioxidant, and anticancer activities. We focused on the latest technology adopted in colostrum quality evaluation and explored the effects of treatments on bioactive compounds. In conclusion, soon, colostrum-based supplements may play a complementary/adjuvant role in the prevention and treatment of different diseases as well as for pharmaceutical purposes or as an everyday food supplement. Abstract

The authors would like to issue an erratum for this article [1], and would like to declare the following competing interests which we inadvertently failed to include in our original publication. The authors would like to apologise for this omission. In addition, a detailed method for co-ultramicronization process is reported: Co-ultramicronization process was performed in a jet-mill equipment endowed with a chamber of 300 mm in diameter which operates with “spiral technology” and was driven by compressed air at 10-12 bars. The crashing is determined by the high number of collisions that occurs among particles, as a result of the high level of kinetic – not mechanical - energy. This process was effective not only in reducing the products particle size, but also in modifying their crystalline structure. Observations by scanning electron microscopy (SEM) shows an intimate intermixing of the two components of the composite, while analysis throw differential scanning calorimetry (DSC) and X-ray diffraction (XRD) have documented the transformation in a new crystalline form different from the original two, definable with “a higher energy content form.” The composite shows the following particle size distribution: 96 %

Abstract

Aim of the present study was to evaluate the potential beneficial and/or adverse effects of Capsicum annuum L. (cv. Fiesta) extracts at two stage of ripening (immature and mature), and at two dosages (low and high) by evaluation of biochemical profile and oxidative status in CD-1 mice. The extracts were daily administered to mice by oral gavage for 20 days. At the end of the trial, the animals were euthanatized and blood was collected. Evidence of liver damage (increase of AST, ALT and bilirubin) in the group receiving the higher dosage of immature peppers extract were observed. Even if no adverse effects were seen at the lower doses, also no signs of beneficial effects in term of health status, biochemical profile and oxidative status were detected. These results are in contrast with in vitro findings and raise doubts about the possible use of Capsicum annuum L. (cv. Fiesta) as a nutritional supplement. [ABSTRACT FROM AUTHOR]

Abstract

Background: Osteoarthritis (OA) is a common progressive joint disease in dogs and cats. The goal of OA treatment is to reduce inflammation, minimize pain, and maintain joint function. Currently, non-steroidal anti-inflammatory drugs (e.g., meloxicam) are the cornerstone of treatment for OA pain, but side effects with long-term use pose important challenges to veterinary practitioners when dealing with OA pain. Palmitoylethanolamide (PEA) is a naturally-occurring fatty acid amide, locally produced on demand by tissues in response to stress. PEA endogenous levels change during inflammatory and painful conditions, including OA, i.e., they are typically increased during acute conditions and decreased in chronic inflammation. Systemic treatment with PEA has anti-inflammatory and pain-relieving effects in several disorders, yet data are lacking in OA. Here we tested a new composite, i.e., PEA co-ultramicronized with the natural antioxidant quercetin (PEA-Q), administered orally in two different rat models of inflammatory and OA pain, namely carrageenan paw oedema and sodium monoiodoacetate (MIA)-induced OA. Oral treatment with meloxicam was used as benchmark. Results: PEA-Q decreased inflammatory and hyperalgesic responses induced by carrageenan injection, as shown by: (i) paw oedema reduction, (ii) decreased severity in histological inflammatory score, (iii) reduced activity of myeloperoxidase, i.e., a marker of inflammatory cell infiltration, and (iv) decreased thermal hyperalgesia. Overall PEA-Q showed superior effects compared to meloxicam. In MIA-treated animals, PEA-Q exerted the following effects: (i) reduced mechanical allodynia and improved locomotor function, (ii) protected cartilage against MIA-induced histological damage, and (iii) counteracted the increased serum concentration of tumor necrosis factor alpha, interleukin 1 beta, metalloproteases 1, 3, 9 and nerve growth factor. Abstract

Background: Osteoarthritis (OA) is a degenerative joint disease produced by a cascade of events that can ultimately lead to joint damage. The aim of this study was to evaluate the effect of adelmidrol, a synthetic palmitoylethanolamide analogue, combined with hyaluronic acid on pain severity and modulation of the inflammatory response in a rat model of monosodium iodoacetate (MIA)-induced osteoarthritis. Methods: OA was induced by intra-articular injection of MIA in the knee joint. On day 21 post-MIA administration, the knee joint was analyzed. Rats subjected to OA were treated by intra-articular injection of adelmidrol in combination with sodium hyaluronate at different doses and time points after MIA induction. Limb nociception was assessed by the paw withdrawal latency and threshold measurement. Samples were examined macroscopically, histologically, and by immunohistochemistry. Results: At day 21 post-MIA injection, the MIA + solvent and MIA + 1.0% sodium hyaluronate groups showed irregularities and fibrillation in the surface layer, a decrease in blood cells and multilayering in transition and radial zones, no pannus formation, and modified Mankin scores significantly higher than sham knees. The combination of hyaluronic acid and adelmidrol dose-dependently (adelmidrol 0.6% + 1.0% sodium hyaluronate and adelmidrol 2% + 1.0% sodium hyaluronate) reduced the histological alterations induced by MIA. Moreover, degeneration of articular cartilage, mast cell infiltration, and pro-inflammatory cytokine and chemokine plasma levels were significantly downregulated by treatment with a combination of hyaluronic acid and adelmidrol at the above doses. Conclusions: Our results clearly demonstrate that the combination of hyaluronic acid and adelmidrol improves the signs of OA induced by MIA.
