9. Zinc Substituted Myoglobin−Albumin Fusion Protein: A Photosensitizer for Cancer Therapy.

Abstract

Myoglobin combined with human serum albumin (Mb‐HSA) can be produced using yeast Pichia pastoris as a host strain, with secretion into the culture medium. This Mb‐HSA fusion protein possesses identical O2 binding affinity to that of naked Mb. The Mb unit is reconstituted with a zinc(II) protoporphyrin IX, yielding (zinc substituted Mb)‐HSA, ZnMb‐HSA. The photophysical property and singlet O2 generation ability of ZnMb‐HSA are equivalent to those of ZnMb. In vitro cell experiments revealed that ZnMb‐HSA acts as a superior photosensitizer for photodynamic cancer therapy. It is noteworthy that ZnMb‐HSA shows long circulation lifetime in vivo. 10. A Study on Elderly Behaviour An Analysis of Exhibition Viewing at Suzhou Museum in China

Abstract

The responsibility of museum is to convey messages about their exhibit projects while meeting multiple visitor expectations. One way this can be achieved is through a deliberate design and planning that draw visitors in and engage them with interactive elements. However little research has been conducted about exhibit visiting in China, especially for elderly visitors. The purpose of this study was to make a reference, which allows designers and curators to cope with the complex planning and meet elderly’s needs. For a 4-week period in 2011, behavioral data of elderly were collected from 165 visitors who moved through the Suzhou Museum in China. It is predicted that both observed and reported data in this study would be related to elderly behavior areas which were emphasized in the visitor research and more effectively achieve the goal of convey messages.

Abstract

This study focuses on the efficacy and use of kansei (sensitivity) value creation for the construction of distinctive regional landscapes where man-made design elements construct a living space. This focus advances our objective of clarifying design relationships. Accordingly, we surveyed, analyzed and discussed the design elements of our regional case study, and looked at their efficacy and influence on the regional landscape and regional brand. As a result, we were able to draw up an index of ways to build project plans for landscape design elements that contribute to the construction of a regional brand.

Abstract

This project decided upon rules for designing river signs and also designed a case-study. In Japan, a great variety of signs are used at rivers and their surroundings. The information on these signs is diverse. Signs include such information as the name of the river; prohibitions, rules and recommended behavior at the river and its surroundings; and introductions to the history and culture of the river and its locality. The signs are haphazardly placed, and lack rules about the presentation of information. In our opinion, this is a primary cause of people’s difficulty with comprehending the signs, and causes harm to the river landscape. Therefore, this project concerns the design of signs that will be easily understood by the people and prevent damage to the river landscape.

Abstract

Herein, we report a heterogeneous TiO2-supported Re catalyst (Re/TiO2) that promotes various selective hydrogenation reactions, which includes the hydrogenation of esters to alcohols, the hydrogenation of amides to amines, and the N-methylation of amines, by using H-2 and CO2. Initially, Re/TiO2 was evaluated in the context of the selective hydrogenation of 3-phenylpropionic acid methyl ester to afford 3-phenylpropanol (p(H2)=5 MPa, T=180 degrees C), which revealed a superior performance over other catalysts that we tested in this study. In contrast to other typical heterogeneous catalysts, hydrogenation reactions with Re/TiO2 did not produce dearomatized byproducts. DFT studies suggested that the high selectivity for the formation of alcohols in favor of the hydrogenation of aromatic rings is ascribed to the higher affinity of Re towards the COOCH3 group than to the benzene ring. Moreover, Re/TiO2 showed a wide substrate scope for the hydrogenation reaction (19 examples). Subsequently, this Re/TiO2 catalyst was applied to the hydrogenation of amides, the N-methylation of amines, and the N-alkylation of amines with carboxylic acids or esters.

30. TiO2-Supported Re as a General and Chemoselective Heterogeneous Catalyst for Hydrogenation of Carboxylic Acids to Alcohols

Abstract

TiO2-supported Re, Re/TiO2, was found to promote selective hydrogenation of carboxylic acids having aromatic and aliphatic moieties to the corresponding alcohols. Re/TiO2 showed superior results compared to other transition-metal-loaded TiO2 and supported Re catalysts for selective hydrogenation of 3-phenylpropionic acid. 3phenylpropanol was produced in 97% yield under mild conditions (5 MPa H-2 at 140 degrees C). Contrary to typical heterogeneous catalysts, Re/TiO2 does not lead to the formation of dearomatized byproducts. The catalyst is recyclable and shows a wide substrate scope in the synthesis of alcohols (22 examples; up to 97% isolated yield).

Abstract

Anticancer agent doxorubicin‐loaded liposomes (DoxLs) were drawn spontaneously into nanotubes comprising multilayers of polyelectrolytes and gold nanoparticles (PAuNTs). We describe a unique structure of PAuNTs incorporating DoxLs (DoxL‐PAuNT). The number of DoxLs adsorbed on the tube interior surface was ascertained as 2.4×103. Heating stimuli engendered gel‐liquid crystal phase transition of DoxL in the capillary, subsequently inducing efficient release of Dox without structural changes of the tubule or liposome. Abstract

In an attempt to obtain effective guidelines for planning community events, this study examined the structure of rating special events held in communities by taking up “Minoshima Project – Let’s Go Out to Enjoy”, an experiment conducted by the Japanese Ministry of Land, Infrastructure and Transport in Minoshima, Hakata-ku, Fukuoka. Analysis of visitors’ answers given a questionnaire survey revealed that planning and implementing projects that would enhance six items of evaluation (factor evaluation), “a. events are entertaining”, “b. events are useful to boosts economy in the district”, “e. events are fashionable” and “f. events enhance the unique character of the district”, would improve the value of individual projects, and heighten the comprehensive evaluation of the event in community (MINOSHIMA Project). Study and analysis of community events and examination of evaluation items (factor evaluation) are believed to delineate the evaluation structure of community events.

Haemoglobin (Hb)–albumin (HSA) trimers were synthesized using five distinct Hb variants in which the structures were genetically and chemically tuned as an artificial O2 carrier and used as a red blood cell (RBC) substitute. The trimers were found to have moderately low O2 affinity (p50 = 23–34 Torr, 37 °C) and high co-operativity, yielding a maximum O2 transport efficiency 1.8-fold higher than that of human RBCs. Abstract

We describe synthesis of a symmetric protein triad comprising a genetically fused recombinant hemoglobin(di-a/Cß120) center and human serum albumin sides [rHb(di-a/Cß120)–HSA2]. This trimer is not cleaved at low concentration. It shows adequate resistance to haptoglobin binding as an artificial O2carrier used as a red blood cell (RBC) substitute. Also, O2affinity of the trimer was modulated to the appropriate value, similarly to that of RBCs, by complexation of inositol hexaphosphate and a Kß108 mutation into the Hb core.A genetically fused recombinant hemoglobin [rHb(di-a/Cß120)] was coupled with two human serum albumins (HSAs), yielding a rHb(di-a/Cß120)–HSA2trimer as an artificial O2carrier. The di-a fusion prevents cleavage of the Hb core and haptoglobin binding. O2affinity can be tuned to the appropriate value by complexation of an inositol hexaphosphate and a Kß108 mutation into the Hb core.

Abstract

We describe the synthesis and O2 affinity of genetically engineered human adult haemoglobin (rHbA) wrapped covalently with recombinant human serum albumins (rHSAs) as an artificial O2 carrier used for a completely synthetic red blood cell (RBC) substitute. Wild-type rHbA [rHbA(wt)] expressed in yeast species Pichia pastoris shows an identical amino acid sequence and three-dimensional structure to those of native HbA. It is particularly interesting that two orientations of the prosthetic haem group in rHbA(wt) were aligned by gentle heating in the natural form. Covalent wrapping of rHbA(wt) with three rHSAs conferred a core–shell structured haemoglobin–albumin cluster: rHbA(wt)–rHSA3. Three variant clusters containing an rHbA mutant core were also created: Leu-β28 → Phe, Leu-β28 → Trp, and Leu-β28 → Tyr/His-β63 → Gln. Replacement of Leu-β28 with Trp decreased the distal space in the haem pocket, thereby yielding a cluster with moderately low O2 affinity which is nearly the same as that of human RBC.
