In this work we report the electrochemical, spectroscopical and spectro-electrochemical studies of a model complex [Cu ΙΙ (Bztpen)] 2+ , (Bztpen = (N-benzyl-N,N',N'-tris(pyridin-2-ylmethyl)ethylenediamine) in order to propose a methodology to evaluate the interaction of potential metal based anticancer agents during electron transfer processes, with transport proteins such as Bovine Serum Albumin (BSA). It was possible to establish a reversible electron transfer [Cu ΙΙ (Bztpen)] 2+ +1e → [Cu Ι (Bztpen)] + and a weak interaction energy between BSA and [Cu ΙΙ (Bztpen)] and [Cu Ι (Bztpen)] species, with no adsorption of protein over the electrode surface. Circular Dichroism (CD) Spectroelectrochemistry, not reported before, reveals no significant changes in BSA structure during the electron transfer [Cu ΙΙ (Bztpen)] 2+ + 1e → [Cu Ι (Bztpen)] + . A combined experimental and theoretical study of the biphenyl (BP) crystal is presented. The X-ray diffraction data collected at 100 K were subjected to Hirshfeld atom and multipole refinements of the electron density, ρ(r). A theoretical exploration of the potential energy surface (PES) of the crystal was also carried out. This investigation challenges the common assumption that the planar structure of BP in the phase I crystal is an average of two twisted configurations in a double-well potential. The theoretical computations provide compelling evidence that this structure corresponds to a minimum on the PES hence to a stable molecular arrangement. Consistently, the experiment showed no evidence of positional or dynamic disorder. The intramolecular hydrogen-hydrogen bonds detected are not repulsive. The topological analysis of the experimental and theoretical ρ(r) reveals that both the intra- and intermolecular H⋅⋅⋅H and the C-H⋅⋅⋅π contacts stabilize the BP crystal., (© 2021 Wiley-VCH GmbH.)

Compounds from the 2,2'-bipyridine molecular family were investigated for use as redox-active materials in organic flow batteries. For 156 2,2'-bipyridine derivatives reported in the academic literature, we calculated the redox potential, the pKa for the second deprotonation reaction, and the solubility in aqueous solutions. Using experimental data on a small subset of derivatives, we were able to calibrate our calculations. We find that functionalization with electron-withdrawing groups leads to an increase of the redox potential and to an increase of the molecular acidity (as expressed in a reduction of the pKa value for the second deprotonation step). Furthermore, calculations of solubility in water indicate that some of the studied derivatives have adequate solubility for flow battery applications. Based on an analysis of the phyisco-chemical properties of the 156 studied compounds, we down-select five molecules with carbonyl- and nitro-based functional groups, whose parameters are especially promising for potential applications as negative redox-active materials in organic flow batteries.

The highly stable nitrosyl iron(II) mononuclear complex [Fe(bztpen)(NO)](PF6 )2 (bztpen=N-benzyl-N,N',N'-tris(2-pyridylmethyl)ethylenediamine) displays an S=1/2↔S=3/2 spin crossover (SCO) behavior (T1/2 =370 K, ΔH=12.48 kJ mol(-1) , ΔS=33 J K(-1)  mol(-1) ) stemming from strong magnetic coupling between the NO radical (S=1/2) and thermally interconverted (S=0↔S=2) ferrous spin states. The crystal structure of this robust complex has been investigated in the temperature range 120-420 K affording a detailed picture of how the electronic distribution of the t2g -eg orbitals modulates the structure of the {FeNO}(7) bond, providing valuable magneto-structural and spectroscopic correlations and DFT analysis., (© 2016 WILEY-VCH Verlag GmbH & Co. The synthesis, crystal structures, magnetic behavior, and electron paramagnetic resonance studies of five new Fe(III) spin crossover (SCO) complexes are reported. The [Fe(III)N5O] coordination core is constituted of the pentadentate ligand bztpen (N5) and a series of alkoxide anions (ethoxide, propoxide, n-butoxide, isobutoxide, and ethylene glycoxide). The methoxide derivative previously reported by us is also reinvestigated. The six complexes crystallize in the orthorhombic Pbca space group and show similar molecular structures and crystal packing. The coordination octahedron is strongly distorted in both the high- and low-temperature structures. The structural changes upon spin conversion are consistent with those previously observed for [Fe(III)N4O2] SCO complexes of the Schiff base type, except for the Fe-O(alkoxide) bond distance, which shortens significantly in the high-spin state. Application of the Slichter-Drickamer thermodynamic model to the experimental SCO curves afforded reasonably good simulations with typical enthalpy and entropy variations ranging in the intervals ΔH = 6-13 kJ mol(-1) and ΔS = 40-50 J mol(-1) K(-1), respectively. The estimated values of the cooperativity parameter Γ, found in the interval 0-2.2 kJ mol(-1), were consistent with the nature of the SCO. Electron paramagnetic resonance spectroscopy confirmed the transformation between the high-spin and low-spin states, characterized by signals at g ≈ 4.47 and 2.10, respectively. Electrochemical analysis demonstrated the instability of the Fe(II) alkoxide derivatives in solution.

A complete study of the electronic density distribution in antineoplastic mixed chelate complexes of the type [Cu(N-N)(glycinate)H2 O]NO3 (N-N=2,2'-bipyridine (bpy) (1), 4,4'-dimethyl-bpy (2), 5,5'-dimethyl-2,2'-bipyridine (3), 1,10-phenanthroline (phen) (4), 4-methyl-phen (5); 5-methyl-phen (6); 4,7-dimethyl-phen (7), 5,6-dimethyl-phen (8), and 3,4,7,8-tetramethyl-phen (9)), a family known as Casiopeínas, was carried out with cyclic voltammetry, EPR, and computational methods. Crystal structures of 1⋅H2 O, 2⋅H2 O, 3⋅H2 O, 6⋅H2 O, and 8⋅H2 O show the variability in the geometries adopted by the copper compounds in the solid state. Experimental properties are described employing electronic descriptors obtained from computational methods. The main descriptors found were: The total electronic population of the metal ion (N(Cu)), delocalization of the metal ion electrons over the donor atoms (Δ(Cu)), atomic dipolar moment (μ(Cu)), and the atomic quadrupole moment (Q(Cu)). It was found that π-back-bonding is the principal factor that modulates the distribution of the electron density around the metal ion. The electronic descriptors obtained from the computational approach can be used as electronic descriptors of inorganic compounds that have shown antiproliferative activities instead the experimental data, aiding the rational design of good candidates of metal-based drugs., (© 2014 WILEY-VCH Verlag GmbH & Co. The aim of this work is to describe the specific recognition site between DNA and an anticancer copper complex by means of computational methods. Molecular dynamics were used to find the preferred site of binding between selected DNA chains and [Cu(2,2'-bipyridine)(acetylacetonate)(H(2)O)](+) (Cas). Full DFT optimizations of selected geometries extracted from simulations, followed by a topological analysis of electron density allowed us to define the specific interactions inside the recognition site. Cas links deoxyribose-phosphate by a coordination bond between metal and the phosphate group. There are several C-H···π, O···π(C) and O···π(N) interactions between the sugar group and the aromatic ligand of Cas. The results indicate that the adduct Cas-deoxyribose-phosphate may be an initial step toward the hydrolysis of DNA chains. Overall, this study provides insights into the initial step of the action mechanism of copper complexes as apoptosis-inducing agents and provides guidelines for the design of this kind of drugs.

A new synthetic pathway was reported to obtain N6 donor ligand 2,9-bis-(2',5'-diazahexanyl)-1,10-phenanthroline (L1) and its coordination compounds of essential divalent metal ions Mn, Fe, Co, Ni, Cu and Zn. Complete characterization of all compounds was done with the conventional techniques. Crystal structures of [NiL1](PF(6))(2) and [ZnL1](PF(6))(2)·H(2)O were also reported. Electrochemical studies have shown an active participation of the aromatic moiety of the ligand in redox reactions. The in vitro tests of the cytotoxic activity against human tumour cell lines HeLa (cervix) and CHP-212 (neuroblastoma) showed that all coordination compounds that involve redox active metal ions exhibit noteworthy antiproliferative activity, superior in all cases to cisplatin. [CuL1](2+) showed the lower IC(50) value in the HeLa cell line with 1.84 μM, meanwhile, [CoL1](2+) showed the lower value in neuroblastoma CHP-212 with IC(50) = 45.28 μM. None of these compounds were active against the SK-N-SH neuroblastoma cell line. In Entamoeba histolytica cultures, remarkable nanomolar IC(50) values were found for [NiL1](2+) and [MnL1](2+) with 60 nM and 80 nM respectively, improving the antiproliferative activity more than 1000 times compared with the first choice drug for clinical treatments of human amoebiasis, metronidazole. On the other hand, a free ligand does not show antiproliferative activity either on human tumor cell lines or on Entamoeba histolytica trophozoites, highlighting the role played by metal ions to produce cytotoxicity in tumor cells and protozoa systems.

Casiopeínas® are copper complexes with the general formula [Cu(N-N)(N-O)]NO(3) and [Cu(N-N)(O-O)]NO(3) where N-N denotes a substituted bipyridine or phenanthroline, N-O indicates α-aminoacidate or peptide and O-O represents acetylacetonate or salicylaldehyde. This family of compounds has been evaluated in vitro and in vivo showing cytotoxic, genotoxic, and antineoplastic activity. The action mechanism is still not completely elucidated, but the possibility exists that these compounds interact with DNA by intercalation due to the aromatic moiety. In this work we found, using the properties of the electron density of a π-complex model base-Casiopeína®-base, that the stacking mechanism between Casiopeínas® and DNA bases is due to an electron density deficiency of the ligand of the Casiopeína® which is compensated for by an electron transfer from adenines by a π-π interaction.

In addition to a previous report, the reaction of tigogenin acetate with ICl in refluxing CHCl(3) produced the hitherto unknown 23R-iodotigogenin acetate, bearing an axial iodine atom at C-23 and its already reported 23S-epimer. The same treatment of sarsasapogenin acetate led to a single diasteromer characterized as 23S-iodosarsasapogenin acetate. A full characterization of the obtained compound including (1)H, (13)C NMR, MS and X-ray diffraction is provided.

Benzimidazole 2-carbamates, such as albendazole (ABZ) and mebendazole (MBZ), used for the treatment of helmintic infections, have low aqueous solubility and poor bioavailability, both of which lead to high interindividual variability when used for human systemic helmintiosis; therefore, it is necessary to search for new anthelmintics with better biopharmaceutical properties. In the present study the solubility, pKa, logP and apparent permeability in the Caco-2 cells system of four novel anthelmintic (1H-benzimidazol-5(6)-yl)carboxamide derivatives (compounds 1-4) with a 2-methylthyo group were evaluated. Also the pharmacokinetic parameters of compound 1 which in previous studies showed activity similar to ABZ against T. spirallis, was evaluated in BALB/c mice, as a representative molecule of the series. The novel anthelmintics, showed better solubility than ABZ in aqueous acid pH and in organic solvents. The logP, P(app) and Caco-2 data indicate that the 4 derivatives are highly permeable drugs, but it is possible that an efflux system could be involved in the transport of these compounds. Plasma levels of compound 1 and its sulfoxide (compound 5) were high after the first 5 min. This fact strongly suggests that compound 1 is rapidly metabolized in the small intestine. On the other hand, the sulfone metabolite (compound 6) levels were lower than those of compound 5. The half life and mean residence time (MRT) of compound 1 and its main metabolites indicate that their elimination is very rapid. More studies in mammalian species are necessary in order to understand the pharmacokinetic behavior of these novel compounds.

The structural and solution characterization of novel Fe(II) compounds of the general formula [Fe(bztpen)X]PF6 and [Fe(bztpen)CH3CN](PF6)2 is presented, where bztpen is the pentadentate ligand N-benzyl-N,N',N'-tris(2-methylpyridyl)ethylenediamine and X- is a monodentate ligand. All complexes were characterized in solution and in the solid state, employing the usual techniques and single-crystal X-ray diffraction. The results obtained are discussed in terms of the existing information for some previously reported analogous compounds to arrive at a rationalization regarding the influence of a variation in the coordination environment of all compounds and to evaluate their relative stability. The observed magnetic response in the solid state is paramagnetic in the entire temperature range for the Cl-, Br-, I-, OCN-, and SCN- derivatives, while the N(CN)2-, CH3CN, and CN- derivatives are diamagnetic. The diamagnetic character of these last two compounds is confirmed in acetonitrile solution, while a spin transition step is observed for the N(CN)2- derivative. Diffraction data for all compounds as hexafluorophosphates shows that the I-, Br-, and OCN- derivatives crystallize in the orthorhombic space group Pbca, while the CN-, SCN-, and CH3CN compounds crystallize in the triclinic space group P. Average bond lengths and the trigonal distortion parameter can be correlated to the observed magnetic susceptibility depending on the coordinated monodentate ligand. Solution measurements of electronic properties for the compounds follow the trend established by the spectrochemical series. The relative stability of the Fe(II) complexes can be established in terms of the percentage of dissociation from the voltammetry and conductivity results, which are consistent with those obtained spectrophotometrically, mainly, the larger stability for the CN- derivative and the lower for the I- derivative. The redox potential and percentage of dissociation values allow for the estimation of the relative stability constants for the Fe(II) and Fe(III) complexes.

The mononuclear diamagnetic compound {Fe(bztpen)[N(CN)2]}(PF6)CH3OH (1) (bztpen = N-benzyl-N,N',N'-tris(2-pyridylmethyl)ethylenediamine) has been synthesized and its crystal structure studied. Complex 1 can be considered to be the formal precursor of two new dinuclear, dicyanamide-bridged iron(II) complexes with the generic formula {[(Fe(bztpen)]2[mu-N(CN)2]}(PF6)3 x n H2O (n = 1 (2) or 0 (3)), which have been characterized in the solid state and in solution. In all three complexes, the iron atoms have a distorted [FeN6] octahedral coordination defined by a bztpen ligand and a terminal (1) or a bridging dicyanamide ligand (2 and 3). In the solid state, 2 and 3 can be considered to be molecular isomers that differ by the relative position of the phenyl ring of the two {Fe(bztpen)[N(CN)2]}+ halves (cis and trans, respectively). Depending on the texture of the sample, 2 exhibits paramagnetic behavior or displays a very incomplete spin transition at atmospheric pressure. Complex 3 undergoes a gradual two-step spin transition with no observed hysteresis in the solid state. Both steps are approximately 100 K wide, centered at approximately 200 K and approximately 350 K, with a plateau of approximately 80 K separating the transitions. The crystal structure of 3 has been determined in steps of approximately 50 K between 400 K and 90 K, which provides a fascinating insight into the structural behavior of the complex and the nature of the spin transition. Order-disorder transitions occur in the dicyanamide bridge and the PF6(-) ions simultaneously, with the spin-crossover behavior suggesting that these transitions may trigger the two-step character. In solution, 2 and 3 display very similar continuous spin conversions. Electrochemical studies of 2 and 3 show that the voltammograms are typical of dimeric systems with electronic coupling of the metals through the dicyanamide ligand.

Reaction of dichlorotris(triphenylphosphine) ruthenium(II) [RuCl(2)(PPh(3))(3)] with 1,8-bis(2-pyridyl)-3,6-dithiaoctane (pdto), a (N(2)S(2)) tetradentate donor, yields a new compound [Ru(pdto)(PPh(3))Cl]Cl (1), which has been fully characterized. (1)H and (31)P NMR studies of 1 in acetonitrile at several temperatures show the substitution of both coordinated chloride and triphenylphosphine with two molecules of acetonitrile, as confirmed by the isolation of the complex [Ru(pdto)(CH(3)CN)(2)]Cl(2) (2). Cyclic voltammetric and spectroelectrochemical techniques allowed us to determine the electrochemical behavior of compound 1. The substitution of the chloride and triphenylphosphine by acetonitrile molecules in the Ru(II) coordination sphere of compound 1 was also established by electrochemical studies. The easy substitution of this complex led us to use it as starting material to synthesize the substituted phenanthroline coordination compounds with (pdto) and ruthenium(II), [Ru(pdto)(4,7-diphenyl-1,10-phenanthroline)]Cl(2).4H(2)O (3), [Ru(pdto)(1,10-phenanthroline)]Cl(2).5H(2)O (4), [Ru(pdto)(5,6-dimethyl-1,10-phenanthroline)]Cl(2).5H(2)O (5), [Ru(pdto)(4,7-dimethyl-1,10-phenanthroline)]Cl(2).3H(2)O (6), and [Ru(pdto)(3,4,7,8-tetramethyl-1,10-phenanthroline)]Cl(2).4H(2)O (7). These compounds were fully characterized, and the crystal structure of 4 was obtained. Cyclic voltammetric and spectroelectrochemical techniques allowed us to determine their electrochemical behavior. The electrochemical oxidation processes in these compounds are related to the oxidation of ionic chlorides, and to the reversible transformation from Ru(II) to Ru(III). On the other hand, a single reduction process is associated to the reduction of the substituted phenanthroline in the coordination compound. The E(1/2) (phen/phen(-)) and E(1/2) (Ru(II)/Ru(III)) for the compounds (3-7) were evaluated, and, as expected, the modification of the substituted 1,10-phenanthrolines in the complexes also modifies the redox potentials. Correlations of both electrochemical potentials with pK(a) of the free 1,10-phenathrolines, lambda(max) MLCT transition band, and chemical shifts of phenanthrolines in these complexes were found, possibly as a consequence of the change in the electron density of the Ru(II) and the coordinated phenanthroline.

Studies of complexation equilibria of the antibiotic anions nalidixate and cinoxacinate with [Cu(phen)]2+ and [Cu(bipy)]2+ are reported. These studies indicate that the stability of this type of complex is strongly related to the metal environment. A correlation between the stability constants, determined here, with the sigma donation character of the ligand is proposed. This study shows that the stability constant for the reaction between the quinolones and the moiety [Cu(N-N)]2+ is dependent on the coordinate diamine to the metal ion. This is in agreement with previous studies where other physical properties as their electronic absorption spectra in the visible region, display similar behavior. These results suggest that inside the living cells, a possible interaction with some metal ion will be strongly controlled by the type of ligand bound to the cation.
