13. Using Ants, Animal Behavior & the Learning Cycle to Investigate Scientific Processes

Abstract

The behavior of animals is an intrinsically fascinating topic for students from a wide array of backgrounds. We describe a learning experience using animal behavior that we created for middle school students as part of a graduate-student outreach program, Graduate Partners in Science Education, at Arizona State University in collaboration with a K-8 public school. This activity capitalizes on the interest that animal behavior can generate to introduce and reinforce student understanding of the scientific method. Specifically, our activity highlights the general utility of the scientific method and uses this method to examine ant social behavior, with emphasis on generating and testing hypotheses. Furthermore, this activity introduces the idea of animal societies and encourages students to apply the concepts they learn to other species, including humans. By collecting ants locally, from schoolyards or nearby habitats, this experience situates learning in the context of students' own communities. We also provide optional assessment materials that teachers can use to assess learning objectives and standard mastery.

Abstract

Chromosome evolution presents an enigma in the mega-diverse Lepidoptera. Most species exhibit constrained chromosome evolution with nearly identical haploid chromosome counts and chromosome-level gene collinearity among species more than 140 million years divergent. However, a few species possess radically inflated chromosomal counts due to extensive fission and fusion events. To address this enigma of constraint in the face of an exceptional ability to change, we investigated an unprecedented reorganization of the standard lepidopteran chromosome structure in the green-veined white butterfly (Pieris napi). We find that gene content in P. napi has been extensively rearranged in large collinear blocks, which until now have been masked by a haploid chromosome number close to the lepidopteran average. We observe that ancient chromosome ends have been maintained and collinear blocks are enriched for functionally related genes suggesting both a mechanism and a possible role for selection in determining the boundaries of these genome-wide rearrangements.

Abstract

Complex signaling traits such as pheromone profiles can play an important role in the early stages of reproductive isolation between populations. These signals can diverge along multiple trait axes, and signal receivers are often sensitive to subtle differences in signal properties. In the Lepidoptera, prior research has highlighted that natural selection can drive rapid chemical signal divergence, for instance via mate recognition to maintain species boundaries. Much less is known about the occurrence of such changes for predominantly sexually selected chemical signals, such as those released by many male lepidopterans. We evaluated the divergence in male and female wing volatile profiles between two recently isolated subspecies of the pierid butterfly Pieris rapae Linnaeus (Lepidoptera: Pieridae): P. rapae rapae and P. rapae crucivora. In laboratory settings, these subspecies exhibit strong premating isolation, with females rejecting males of the opposite subspecies despite the fact that males direct equivalent courtship effort toward females of either subspecies. Using gas chromatography–mass spectrometry, we analyzed the volatile chemical profiles of individual males and females of each subspecies. We find that males of each subspecies differ in their wing volatile profiles, including quantitative differences in a male sex pheromone, ferrulactone. In contrast, female wing volatiles profiles have diverged significantly less. These sexspecific patterns suggest that male chemical profiles may play a role in the observed premating isolation between these two subspecies, providing support for future investigations of sexually selected chemical traits in population divergence.

Abstract

Reproductive traits are some of the most rapidly evolving traits in the natural world. However, research to date has focused heavily on male reproductive traits such as male genitalia, leaving female reproductive traits understudied. As a result, we know little about female reproductive adaptations or how they may have co-evolved with male reproductive traits. Butterflies provide an excellent system to study reproductive interactions and physiology from the female perspective. Female butterflies have a specialized reproductive organ called the bursa copulatrix that appears to be co-evolving with specific male traits. The bursa accepts and actively digests the complex male ejaculate, called the spermatophore. Our research focused on the proteases present in the bursa of the Cabbage White butterfly, Pieris rapae. Using a general colorimetric assay, we discovered high levels of overall protein digestion within the bursa. Using transcriptomic and proteomic approaches, we identified nine putative proteases from two main functional classes that may play a role in bursal digestion of the spermatophore. Using a zymogram approach, we conducted an in vitro protein digestion assay to measure the activity of specific bursal proteases. Finally, we investigated functionality of each protease by disrupting specific modes of protease action using targeted protease inhibitors and quantifying subsequent loss of activity. Our findings offer new insights into female bursal adaptations, and provide groundwork for future studies of co-evolution between the bursa and spermatophore.
