24. Genomic Diversity of Streptomyces clavuligerus : Implications for Clavulanic Acid Biosynthesis and Industrial Hyperproduction.

Abstract

Streptomyces clavuligerus is a species used worldwide to industrially produce clavulanic acid (CA), a molecule that enhances antibiotic effectiveness against β-lactamase-producing bacterial strains. Despite its low inherent CA production, hyper-producing strains have been developed. However, genomic analyses specific to S. clavuligerus and CA biosynthesis are limited. Genomic variations that may influence CA yield were explored using S. clavuligerus strain genomes from diverse sources. Despite the slight differences obtained by similarity index calculation, pan-genome estimation revealed that only half of the genes identified were present in all strains. As expected, core genes were associated with primary metabolism, while the remaining genes were linked to secondary metabolism. Differences at the sequence level were more likely to be found in regions close to the tips of the linear chromosome. Wild-type strains preserved larger chromosomal and plasmid regions compared to industrial and/or hyper-producing strains; such a grouping pattern was also found through refined phylogenetic analyses. These results provide essential insights for the development of hyper-producing S. clavuligerus strains, attending to the critical demand for this antibiotic enhancer and contributing to future strategies for CA production optimization. Abstract

A new and threatened polypore species, Bondarzewia loguerciae, is described from the cloud forests of southern Brazil. It is characterized by single-pileate basidiomata that grow on dead branches and along living stems of standing trunks and present a context with dark lines and resinous tubes. When growing in axenic culture, this species also develops chlamydospores. We provide an illustrated morphological description and molecular analysis. Our specimens from Brazil form a monophyletic group among other species of the Southern Hemisphere. The conservation status of B. loguerciae is assessed and published as "Critically Endangered" based on the International Union for Conservation of Nature (IUCN) criteria. Abstract

Este trabajo propone el diseño de una microrred móvil bajo fuentes renovables de energía (solar-fotovoltaica y eólica), que brinda una solución energética autónoma y eficiente para una vivienda promedio de la Isla de Providencia, perteneciente al Archipiélago de San Andrés, Providencia y Santa Catalina, identificada como una Zona No Interconectada (ZNI) en Colombia. El problema que abarca esta investigación corresponde a la dependencia que tiene la isla con el diésel marino como combustible para las centrales de generación de energía eléctrica y los fletes involucrados para llevar dicho combustible desde la central de Cartagena, dada la imposibilidad de la isla para abastecerse del Sistema Interconectado Nacional; además del impacto ambiental que genera la quema del diésel. Sin embargo, la isla cuenta con un gran potencial solar y eólico.

40. Examining the Faculty Implementation of Intermediate Algebra for Statistics: An Evaluation Study

Abstract

This dissertation study examined the factors that influence the ability of math faculty at a local community college to implement a new intermediate algebra for statistics course designed to improve the math remediation pathway. A gap analysis framework was used to identify the knowledge, motivation and organizational influences found in the literature. The methodology used in this study included literature reviews, interviews, classroom observations, and document analysis. The data collected were used to determine if a gap for each influence was validated, not validated, or unable to validate based on the findings. According to the findings, two validated gaps tied to the organizational culture were found. Findings indicate that parts of the organizational culture do not believe that underprepared students can complete math coursework. It was also found that the organization does not support a standard approach to improve math remediation. The findings were used to develop recommendations specific to the validated gaps found at the college. Recommendations include that the college create a place where faculty can refer to references and resources on student success data, effective pedagogy, and instructional tools. It was also recommended that the college facilitate faculty discussions to share and review approaches to improving student outcomes in math remediation to ensure curriculum alignment and increase faculty buy-in on student success. Additionally, a training framework was used to develop an implementation and evaluation plan to train additional faculty or used as a model for other institutions attempting to adopt a similar math remediation intervention. Abstract

This study investigated the effects of Lacticaseibacillus rhamnosus HN001 supplementation on the architecture and gene expression in small intestinal tissues of piglets used as an animal model for infant humans. Twenty-four 10-d-old entire male piglets (4·3 (sd 0·59) kg body weight) were fed an infant formula (IF) (control) or IF supplemented with 1·3 × 105 (low dose) or 7·9 × 106 (high dose) colony-forming units HN001 per ml of reconstituted formula (n 8 piglets/treatment). After 24 d, piglets were euthanised. Samples were collected to analyse the histology and gene expression (RNAseq and qPCR) in the jejunal and ileal tissues, blood cytokine concentrations, and blood and faecal calprotectin concentrations. HN001 consumption altered (false discovery rate < 0·05) gene expression (RNAseq) in jejunal tissues but not in ileal tissues. The number of ileal goblet cells and crypt surface area increased quadratically (P < 0·05) as dietary HN001 levels increased, but no increase was observed in the jejunal tissues. Similarly, blood plasma concentrations of IL-10 and calprotectin increased linearly (P < 0·05) as dietary HN001 levels increased. In conclusion, supplementation of IF with HN001 affected the architecture and gene expression of small intestine tissue, blood cytokine concentration and frequencies, and blood calprotectin concentrations, indicating that HN001 modulated small intestinal tissue maturation and immunity in the piglet model. Abstract

We present the first results from the Cal\'an-Yale Deep Extragalactic Research (CYDER) survey. The main goal of this survey is to study serendipitous X-ray sources detected by Chandra in an intermediate flux range ($10^{-15}-10^{-12}$ ergs s$^{-1}$) that comprises most of the X-ray background. 267 X-ray sources spread over 5 archived fields were detected. The $\log N-\log S$ distribution obtained for this sample is consistent with the results of other surveys. Deep $V$ and $I$ images were taken of these fields in order to calculate X-ray-to-optical flux ratios. Identifications and redshifts were obtained for 106 sources using optical spectroscopy from 8-m class telescopes to reach the optically faintest sources, to the same level as deeper X-ray fields like the Chandra Deep Fields, showing that the nature of sources detected depends mostly on the optical limit for spectroscopy. In general, sources optically classified as obscured Active Galactic Nuclei (AGNs) have redder optical colors than unobscured AGN. A rough correlation between $f_X/f_{\rm opt}$ and hard X-ray luminosity was found for obscured AGN confirming the prediction by existing models that in obscured AGN the optical light is completely dominated by the host galaxy. The previously claimed decrease of the obscured to unobscured AGN ratio with increasing X-ray luminosity is observed. However, this correlation can be explained as a selection effect caused by the lower optical flux of obscured AGN. Comparison between the observed $N_H$ distribution and predictions by existing models shows that the sample appears complete up to $N_H<3\times 10^{22}$ cm$^{-2}$, while for more obscured sources incompleteness plays an important role in the observed obscured to unobscured AGN ratio., Comment: 47 pages, 19 figures, Astrophysical Journal Accepted
