Vernacular architecture offers valuable technological and typological solutions rooted in knowledge systems that connect environmental, social, economic, and cultural contexts. This paper introduces the Heritage for People Platform, a collaborative digital tool developed under the VerSus+ project (Creative Europe Program), aimed at organising and disseminating vernacular architectural knowledge with a focus on sustainable practices that are both culturally and environmentally conscious. The platform builds upon lessons learned from similar projects, offering key improvements such as the adoption of a Case-Based Reasoning methodology, which organises examples by geographic location, materials, and intervention types. A structured classification system based on sustainability principles and strategies enables cross-disciplinary research and comparison. The design process was highly participatory, incorporating user feedback at every stage to ensure the platform is accessible to a wide range of users, including designers, scholars, craftspeople, and the general public. A cross-device, user-centered approach further broadens its accessibility. The platform provides a public, web-based geospatial repository where users can create, update, and explore a dynamic collection of artefacts, design models, people, and institutions from across the globe. This tool marks a significant advance in promoting and preserving both physical and intangible heritage, with strong educational and practical applications for sustainable architecture. The paper details the theoretical framework and real-world potential of the platform as a critical resource for promoting sustainability in the built environment.

Abstract Ultraviolet radiation (UVR) is a major environmental mutagen. In skin, UVR can initiate cancer through the induction of mutagenic DNA damage and promote its progression. An important cancer prevention mechanism is the regulated cell death (RCD), which can safely dispose of damaged cells. Apoptosis, a well-known RCD, is known to be activated by UVR, but part of the mechanism and proteins involved in UVR-induced apoptosis are still to be discovered. Receptor-interacting serine/threonine-protein kinase 3 (RIPK3) and mixed lineage kinase domain-like (MLKL) are two proteins involved in necroptosis, a form of RCD. Here, we have evaluated the implication of RIPK3 and MLKL in UVB-induced cell death in human diploid dermal fibroblasts. Our results show that RIPK3 and MLKL play opposite roles in UVB-induced cell death, in a necroptosis independent pathway. We showed that RIPK3 protects cells from UVB cell death, while MLKL sensitizes cells to UVB-induced apoptosis. Taken together these results are the first to show the implication of RIPK3 and MLKL in survival and apoptosis, respectively, bringing two new actors in UVB-induced cell death pathway.

Introdução: Os transtornos mentais relacionados ao trabalho representam a 2ª principal causa de doenças relacionadas ao trabalho e associam-se desde a exposição a agentes tóxicos até a estrutura hierárquica organizacional do trabalho, gerando diversos problemas psicossociais. Objetivo: Estabelecer relações entre os aspectos epidemiológicos das notificações por transtornos mentais relacionados ao trabalho no Brasil, na década de 2011 a 2020. Métodos: Trata-se de um estudo epidemiológico observacional do tipo ecológico, cujos dados foram levantados do Sistema de Informação de Agravos de Notificação (SINAN), disponíveis no Departamento de Informática do Sistema Único de Saúde (DATASUS), acessados por intermédio do TABNET. Resultados: Das 13.106 notificações de transtornos mentais relacionados ao trabalho na última década, o sexo feminino (63,07%) e a faixa etária de 30-49 anos (66,19%) prevaleceram. Além disso, predominaram os transtornos neuróticos, relacionados com o “stress” e somatoformes (50,48%), bem como 62,85% das notificações evoluem com incapacidade. Foram constatadas também correlações dos transtornos mentais com trabalhadores etilistas (6%) e tabagistas (4,73%). Conclusões: Portanto, diante do crescimento de afastamentos, absenteísmo, adversidades interpessoais e dependência de fármacos e outras substâncias consequentes dos transtornos mentais relacionados ao trabalho, faz-se necessário o reforço de intervenções assistenciais e preventivas direcionadas à saúde mental dos trabalhadores, visando a redução dos danos relacionados ao trabalho.

Since the early 2000s, the neoliberal social order, imposed in all aspects of Chilean social life, has begun to be disputed. Chile thus finds itself in a process of repoliticization concerning groups longtime excluded from traditional political process. This investigation covers different periods of mobilization and focus on the forms in which the radicalization of collective action is represented/performed. In this context, artistic practices will be analyzed as particular forms of radical action representation. This investigation deploys two central questions: how does art come about and become part of the different forms of collective action? Additionally, how have these artistic forms of expression evolved over the past 30 years? This work mobilizes a sociohistorical methodology and a visual ethnography. The study of audiovisual materials, interviews with young protesters and the observation of various direct actions during these movements, in an immersive activity, completes our methodological framework.

Ultraviolet (UV) exposure and atmospheric pollution are both independently implicated in skin diseases such as cancer and premature aging. UVA wavelengths, which penetrate in the deep layers of the skin dermis, exert their toxicity mainly through chromophore photosensitization reactions. Benzo[a]pyrene (BaP), the most abundant polycyclic aromatic hydrocarbon originating from the incomplete combustion of organic matter, could act as a chromophore and absorb UVA. We and other groups have previously shown that BaP and UVA synergize their toxicity in skin cells, which leads to important oxidation. Even if mitochondria alterations have been related to premature skin aging and other skin disorders, no studies have focused on the synergy between UV exposure and pollution on mitochondria. Our study aims to investigate the combined effect of UVA and BaP specifically on mitochondria in order to assess the effect on mitochondrial membranes and the consequences on mitochondrial activity. We show that BaP has a strong affinity for mitochondria and that this affinity leads to an important induction of lipid peroxidation and membrane disruption when exposed to UVA. Co-exposure to UVA and BaP synergizes their toxicity to negatively impact mitochondrial membrane potential, mitochondrial metabolism and the mitochondrial network. Altogether, our results highlight the implication of mitochondria in the synergistic toxicity of pollution and UV exposure and the potential of this toxicity on skin integrity. The Historic Center (Chorá) of Patmos, located in one of the Dodecanese Islands of Greece, was inscribed in the World Heritage List in 1999. The morphological feature, the housing typologies and the building techniques that characterize the Chorá provide an authentic and astonishing expression of the stylistic and typological models of the vernacular architecture of the Greek islands. This paper presents part of the research carried out on the Chorá of Patmos, within the 3D Past project, funded by the European Creative Europe program. The first part illustrates the results of the research concerning the analysis of the material components of the architectural heritage of Patmos: the urban structure, the evolution of the main architectural typologies, the feature of the main building systems. The second part of the paper presents the application of the Heritage Impact Assessment (HIA), a tool for the management of site transformations and in particular for identifying, forecasting and evaluating the impact of potential development on the Outstanding Universal Value (OUV) of the properties. In the case of Patmos the application of HIA can provide an important contribution to manage changes and future transformations in order to preserve the cultural significance of the site and ensure its sustainable development.

Mine tailings are produced by mining activities and contain diverse heavy metal ions, which cause environmental problems and have negative impacts on ecosystems. Different microorganisms, including yeasts, play important roles in the absorption and/or adsorption of these heavy metal ions. This work aimed to analyze proteins synthesized by the yeast Yarrowia lipolytica AMJ6 (Yl-AMJ6), isolated from Andean mine tailings in Peru and subjected to stress conditions with common heavy metal ions. Yeast strains were isolated from high Andean water samples impacted by mine tailings from Yanamate (Pasco, Peru). Among all the isolated yeasts, the Yl-AMJ6 strain presented LC50 values of 1.06 mM, 1.42 mM, and 0.49 mM for the Cr+6, Cu+2, and Cd+2 ions, respectively. Proteomic analysis of theYl-AMJ6 strain under heavy metal stress showed that several proteins were up- or downregulated. Biological and functional analysis of these proteins showed that they were involved in the metabolism of proteins, nucleic acids, and carbohydrates; response to oxidative stress and protein folding; ATP synthesis and ion transport; membrane and cell wall; and cell division. The most prominent proteins that presented the greatest changes were related to the oxidative stress response and carbohydrate metabolism, suggesting the existence of a defense mechanism in these yeasts to resist the impact of environmental contamination by heavy metal ions.

The secretion of α-hemolysin by uropathogenic Escherichia coli (UPEC) is commonly associated with the severity of urinary tract infections, which makes it a predictor of poor prognosis among patients. Accordingly, this toxin has become a target for diagnostic tests and therapeutic interventions. However, there are several obstacles associated with the process of α-hemolysin purification, therefore limiting its utilization in scientific investigations. In order to overcome the problems associated with α-hemolysin expression, after in silico prediction, a 20.48 kDa soluble α-hemolysin recombinant denoted rHlyA was constructed. This recombinant is composed by a 182 amino acid sequence localized in the aa542–723 region of the toxin molecule. The antigenic determinants of the rHlyA were estimated by bioinformatics analysis taking into consideration the tertiary form of the toxin, epitope analysis tools, and solubility inference. The results indicated that rHlyA has three antigenic domains localized in the aa555–565, aa600–610, and aa674–717 regions. Functional investigation of rHlyA demonstrated that it has hemolytic activity against sheep red cells, but no cytotoxic effect against epithelial bladder cells. In summary, the results obtained in this study indicate that rHlyA is a soluble recombinant protein that can be used as a tool in studies that aim to understand the mechanisms involved in the hemolytic and cytotoxic activities of α-hemolysin produced by UPEC. In addition, rHlyA can be applied to generate monoclonal and/or polyclonal antibodies that can be utilized in the development of diagnostic tests and therapeutic interventions.

Cancer is characterized by the development of abnormal cells that divide in an uncontrolled way and may spread into other tissues where they may infiltrate and destroy normal body tissue. Several previous reports have described biochemical anti-tumorigenic properties of crude snake venom or its components, including their capability of inhibiting cell proliferation and promoting cell death. However, to the best of our knowledge, there is no work describing cancer cell proteomic changes following treatment with snake venoms. In this work we describe the quantitative changes in proteomics of MCF7 and MDA-MB-231 breast tumor cell lines following treatment with Bothrops jararaca snake venom, as well as the functional implications of the proteomic changes. Cell lines were treated with sub-toxic doses at either 0.63 μg/mL (low) or 2.5 μg/mL (high) of B. jararaca venom for 24 h, conditions that cause no cell death per se. Proteomics analysis was conducted on a nano-scale liquid chromatography coupled on-line with mass spectrometry (nLC-MS/MS). More than 1000 proteins were identified and evaluated from each cell line treated with either the low or high dose of the snake venom. Protein profiling upon venom treatment showed differential expression of several proteins related to cancer cell metabolism, immune response, and inflammation. Among the identified proteins we highlight histone H3, SNX3, HEL-S-156an, MTCH2, RPS, MCC2, IGF2BP1, and GSTM3. These data suggest that sub-toxic doses of B. jararaca venom have potential to modulate cancer-development related protein targets in cancer cells. This work illustrates a novel biochemical strategy to identify therapeutic targets against cancer cell growth and survival.

Este artículo invita a pensar el rol de las emociones en la participación y la radicalización política de jóvenes estudiantes chilenos provenientes de barrios populares. Este estudio etnográfico fue realizado durante el proceso de contesta­ción estudiantil de los años 2011 y 2012 en Santiago de Chile. El argumento propuesto señala que el paso a una acción riesgosa en una carrera militante está fuertemente ligado a las dinámicas emocionales que se presentan en cada actor. Lo que queremos analizar es cómo se construye y cómo se organiza esta dinámica emocional, es decir, las formas a través de las cuales las emociones se despliegan, desaparecen o se transforman durante un proceso de contestación.

Abstract Introduction One of the main issues in juvenile systemic lupus erythematosus (JSLE) patients is infection, such as tuberculosis (TB). Of note, SLE patients are susceptible to pulmonary and extrapulmonary TB. However, to our knowledge, this contagious disease was rarely reported in pediatric lupus population, particularly diffuse or miliary TB. Therefore, from January 1983 to December 2011, 5,635 patients were followed-up at our Pediatric Rheumatology Unit and 285 (5%) of them met the American College of Rheumatology classification criteria for SLE. Case reports Four (1.4%) of our JSLE patients had disseminated TB and were described herein. All of them were female gender, received BCG vaccination and did not have a history of TB household contact. The median of current age at TB diagnosis and the period between JSLE and TB diagnosis were 17 years old (range 14-20) and 5.5 years (range 2-7), respectively. All patients developed miliary TB during the course of the disease. The median of SLE Disease Activity Index 2000 (SLEDAI-2K) was 4 (2-16) and the patients were treated with immunosuppressive agents (glucocorticoid, azathioprine and/or intravenous cyclophosphamide). Two of them presented sepsis and TB diagnosis was only established at autopsy, especially with lungs, central nervous system and abdominal involvements. Anti-TB therapy (isoniazid, rifampicin and pyrazinamide) was indicated in the other two TB cases, however they deceased. Discussion Miliary TB is a rare and severe opportunist infection in pediatric lupus population. This study reinforces the importance of routine searches for TB in JSLE patients.
