6. Novel Matrix Hit and Run for Sampling Polytopes and Its GPU Implementation

Abstract

We propose and analyze a new Markov Chain Monte Carlo algorithm that generates a uniform sample over full and non-full dimensional polytopes. This algorithm, termed "Matrix Hit and Run" (MHAR), is a modification of the Hit and Run framework. For the regime $n^{1+\frac{1}{3}} \ll m$, MHAR has a lower asymptotic cost per sample in terms of soft-O notation ($\SO$) than do existing sampling algorithms after a \textit{warm start}. MHAR is designed to take advantage of matrix multiplication routines that require less computational and memory resources. Our tests show this implementation to be substantially faster than the \textit{hitandrun} R package, especially for higher dimensions. Finally, we provide a python library based on Pytorch and a Colab notebook with the implementation ready for deployment in architectures with GPU or just CPU.

Abstract

AbstractThe efficacy of therapeutics can be enhanced with their off-target effects being minimized through targeted delivery. In vaccination and cancer immunotherapy, targeting dendritic cells (DCs) and macrophages (Mφs) is crucial for the activation of the immune system. To this end, we have developed a set of carbohydrate-based ligands targeting C-type lectins (CTLs) present on the surface of DCs for receptor-mediated uptake. Branched structures functionalized with up to four mannose units were synthesized through CuI-catalysed click chemistry, and the products were further functionalized with the DBCO group to facilitate their conjugation with cargos of interest via Cu-free click chemistry. Thus, the ligands were conjugated with fluorescein azide, and the fluorescent conjugates were assessed for their internalization in CTL expressing immature DCs. The results showed that high-mannose branched structures were efficiently internalized through the endocytic pathway, with a positive qualitative correlation between branching degree of sugar-based ligands and internalization. As evidence for payload delivery, conjugating with a mannotriose ligand quantitatively increased the uptake of a fluorescently labeled oligonucleotide by immature DCs. Abstract

This study explores the bioconversion of diosgenin by Yarrowia lipolytica P01a, focusing on enhancing the antioxidant, antifungal, and herbicidal activities of the resulting extracts. The bioconversion process, involving glycosylation and hydroxylation, produced significant amounts of protodioscin and soyasaponin I. The extracts showed superior antioxidant activity, with up to 97.02% inhibition of ABTS· radicals and 33.30% inhibition of DPPH· radicals at 1000 mg L−1 of diosgenin. Antifungal assays revealed strong inhibitory effects against Botrytis cinerea, Alternaria sp., and Aspergillus niger, with maximum inhibition rates of 67.34%, 35.63%, and 65.53%, respectively. Additionally, the herbicidal activity of the bioconverted extracts was comparable to commercial herbicides, achieving 100% inhibition of seed germination in both monocotyledonous and dicotyledonous plants. These findings suggest that the Y. lipolytica P01a-mediated bioconversion of diosgenin could provide a sustainable and eco-friendly alternative for developing natural biofungicides and bioherbicides. Abstract

Cucumis sativus L. plants are subjected to stress during production, affecting their growth, development, and fruit quality due to abiotic factors. Thus, the aim of this research is to evaluate Pseudomonas paralactis, Sinorhizobium meliloti, and Acinetobacter radioresistens plant growth promoting rhizobacteria (PGPR) effect on C. sativus plants under three substrate moisture levels (100, 75, and 50%). A randomized complete block design (RCBD) was used with an A x B arrangement: (1) factor A (inoculant) with four levels; (2) PGPR and control (without bacterium); (3) factor B (substrate moisture content) with three levels. Plant height, root length, and fresh weight increased by inoculating PGPR (121%, 135%, and 134%, respectively); likewise, these variables increased with higher moisture content (177%, 204%, and 234%, respectively), while the effect of the interactions of the PGPR and the moisture content in the substrate showed statistical differences in plant height increasing of 197% and root length of 267%. On the other hand, the content of phenols, flavonoids, and antioxidant capacity was statistically different (p ≤ 0.05) in inoculated plants, with PGPR increasing by 117%, 126%, and 150% respectively. In the moisture content of the substrate, statistical differences were observed, with an increase in the flavonoid content (114%) and antioxidant capacity (116%). The assimilation of nitrogen was higher by 274% and phosphorus by 124% with the PGPR inoculation, the moisture content increased the nitrogen content in the plant (257%) and the phosphorus content in plant (135%), showing significant differences (p ≤ 0.05). However, the interaction of PGPR and moisture content only presented statistical differences in nitrogen assimilation. PGPR can be considered as an alternative to obtain vigorous cucumber seedlings. Abstract

El estudio realizado a lo largo de estos capítulos se ha centrado en conectar dentro del ámbito español las prácticas y los contextos de las artes visuales con aquellas de la danza contemporánea, fundamentalmente durante los años setenta y ochenta. Además de atender a los aspectos formales y conceptuales de ambas disciplinas, se ha llevado a cabo un análisis de la situación cultural y política en las que se desarrollaron. Para ello, hemos tratado de comprender el papel que jugaron los E.E.U.U. en el orden geopolítico mundial y el impacto cultural y artístico del modelo norteamericano en España. Las prácticas que maduraron a lo largo de los sesenta, como la performance, Fluxus, o las experimentaciones de la Judson Church Dance Theatre, plantaron la semilla para el posterior florecimiento de buena parte de las expresiones artísticas españolas. Con esto, tratamos de asentar la influencia de los estos movimientos en el conjunto de circunstancias del contexto español. Nos hemos acercado a figuras como Merce Cunningham y Anna Halprin, entre otras, cuando las creaciones se solapaban sin perder autonomía. El desarrollo de la danza contemporánea y las artes visuales de los años 70 y 80 desplegó una gran confluencia de planteamientos artísticos en los que se ha dejado constancia del papel de las mujeres, sobre todo en la transformación de los estereotipos culturales y el cuestionamiento de los modelos heredados. Esto nos ha llevado a esbozar un horizonte paralelo al de las artes visuales en aras de comprender cómo iban apareciendo nuevas disidencias a las disciplinas reguladas por el régimen. El deseo de aperturismo nos permite poner en evidencia ciertas actitudes desafiantes a partir de dinámicas grupales y estrategias colaborativas. Así, revisamos un conjunto de obras en las que el cuerpo se presentaba en estado de fragmentación y confusión. Abstract

The humic substances are an alternative to reduce excessive doses of fertilizers in cultivated species. The objective was to evaluate the effect of levels of electrical conductivity (EC) of the nutrient solution and doses of humic acids (HA) on the yield and quality of chili fruits (Capsicum annuum L.). The design was completely randomized with a bifactorial arrangement with 12 treatments and 15 replications. The factor one was the levels of EC of nutrient solutions (1.0, 1.5 and 2.0 dS·m -1); the factor two was the doses of HA (0, 2.5, 3.75, and 5.0 mL·L-1). The yield, length, equatorial diameter, epicarp thickness, firmness, weight, and number of fruits were evaluated. The nutraceutical quality was also evaluated (total phenols, capsaicin, antioxidant capacity and total flavonoids). The results showed that the yield, length, equatorial diameter, weight, and number of fruits showed differences between EC, while, yield, length, thickness of the pericarp, weight and number of fruits showed differences for HA. The fruit firmness showed differences in the interaction EC×HA. Most of the variables showed higher values in 1.0 dS·m -1 of EC and 2.5 of HA; the firmness was higher in 2.0 dS·m-1 of EC and 3.75 mL·L-1 of HA. The content of phenols, flavonoids, antioxidants, and capsaicin showed differences between EC, while flavonoids did not show differences for HA. The four variables showed differences in the interaction of the factors, with increases of 1.0 and 1.5 dS·m -1 of EC and 5.0 mL·L-1 of HA. The HA are an alternative to increase the yield of chili fruits and their nutraceutical quality, Las sustancias húmicas son una alternativa para reducir el exceso de fertilizantes. El objetivo fue evaluar la conductividad eléctrica (CE) de la solución nutritiva (SN) y ácidos húmicos (AH) en el rendimiento y calidad de frutos Capsicum annuum L. El diseño fue completamente al azar con arreglo bifactorial con 12 tratamientos y 15 repeticiones. El factor uno fue SN con niveles de CE (1,0, 1,5 y 2,0 dS·m -1); el factor dos fue dosis de AH (0, 2,5, 3,75 y 5,0 mL·L-1). Las variables evaluadas fueron, rendimiento, longitud, diámetro ecuatorial, espesor de epicarpio, firmeza, peso y número de frutos. La firmeza de fruto mostró diferenci as para CE×AH. La mayoría de las variables mostraron valores superiores en 1,0 dS·m -1 de CE y 2,5 de AH; la firmeza fue mayor en 2,0 dS·m-1 de CE y 3,75 mL·L-1 de AH. El contenido de fenoles, flavonoides, antioxidantes y capsaicina, mostraron diferencias para CE, mientras que, flavonoides no mostró diferencias para AH. Abstract

Arbuscular mycorrhizal fungi, plant-growth-promoting bacteria (PGPB) and vermicompost constitute important environmental and economic resources for improving the production and quality of tomato fruits. The present research aims to determine the single and combined effect of Glomus fasciculatum (Gf) fungus, Azotobacter chroococcum (Azot), PGPB and vermicompost leachate (VL) organic fertilizer on the yield and quality of tomato fruit. Thus, an open-field experiment was established with seven treatments, a control and three replicates. Total soluble solids, vitamin C, acidity, fruit mass and fruit diameter were evaluated as fruit quality variables; the yield was recorded and estimated in tons per hectare−1. The results showed that Gf, Azot and VL were effective in promoting tomato yield and fruit quality. As a trend, the triple combination (Gf + Azot + VL) evidently obtained the highest values of total soluble solids, vitamin C and fruit acidity. The range of improvement concerning the fruit size was 66.6% (single treatment) compared to 78.5% (triple combination). The maximum yield of 54.5 t/ha−1 was recorded for the Gf + Azot + VL combination. Therefore, G. fasciculatum, A. chroococcum and VL are considered useful as organic alternatives for open-field tomato biofertilization programs in tropical countries. [ABSTRACT FROM AUTHOR]
