10. Transcriptomic markers meet the real world: finding diagnostic signatures of corticosteroid treatment in commercial beef samples

Abstract

Abstract Background The use of growth-promoters in beef cattle, despite the EU ban, remains a frequent practice. The use of transcriptomic markers has already proposed to identify indirect evidence of anabolic hormone treatment. So far, such approach has been tested in experimentally treated animals. Here, for the first time commercial samples were analyzed. Results Quantitative determination of Dexamethasone (DEX) residues in the urine collected at the slaughterhouse was performed by Liquid Chromatography-Mass Spectrometry (LC-MS). DNA-microarray technology was used to obtain transcriptomic profiles of skeletal muscle in commercial samples and negative controls. LC-MS confirmed the presence of low level of DEX residues in the urine of the commercial samples suspect for histological classification. Principal Component Analysis (PCA) on microarray data identified two clusters of samples. One cluster included negative controls and a subset of commercial samples, while a second cluster included part of the specimens collected at the slaughterhouse together with positives for corticosteroid treatment based on thymus histology and LC-MS. Functional analysis of the differentially expressed genes (3961) between the two groups provided further evidence that animals clustering with positive samples might have been treated with corticosteroids. These suspect samples could be reliably classified with a specific classification tool (Prediction Analysis of Microarray) using just two genes. Conclusions Despite broad variation observed in gene expression profiles, the present study showed that DNA-microarrays can be used to find transcriptomic signatures of putative anabolic treatments and that gene expression markers could represent a useful screening tool.

Abstract

Pyrrolizidine alkaloids (PAs) are natural toxins, supposed to be converted to hepatotoxic metabolites in the liver of mammalians, and preventing the uptake of PAs is the only way to reduce the attendant health risks. A liquid chromatography coupled to mass spectrometry (LC-MS/MS) method was developed according to the Decision 2002/657/EC, to evaluate the contamination of Veneto honey by the following pyrrolizidine alkaloids: echimidine, heliotrine, lycopsamine, retrorsine, senecionine, and seneciphylline, as well as their N-oxides (pyrrolizidine alkaloid N-oxides (PANOs)). Before the solid-phase extraction, a reduction step with zinc dust in the presence of sulfuric acid was carried out for the reduction of PANOs to PAs in honey samples. For all of the analytes, the mean recovery ranged between 82.70 and 104.16 % and the limit of quantification (LOQ) was 0.25 μg kg−1, fully satisfying the LOQ of 1 μg kg−1 as requested by EFSA (2012). The validated method was applied for the analysis of 60 Veneto samples produced from local beekeepers. Among the samples, 17 % were positive for the PAs monitored, and their total PA contents ranged from 0.6 to 17.6 μg kg−1. In all of the samples, the contribution from the PANOs appeared to be negligible. The total PA contents found in the Veneto honey samples never exceeded the limit of 0.42 μg per day for an individual weighing 60 kg, established by the German Federal Institute of Risk Assessment (BfR) in 2011. These preliminary results provide useful advice to beekeepers for finding the most suitable location for beehives to achieve a substantial reduction of the PAs in honey.

Abstract

The pharmacokinetics of the extemporaneous combination of low doses of ketamine and propofol, known as 'ketofol', frequently used for emergency procedures in humans to achieve safe sedation and analgesia was studied in cats. The study was performed to assess propofol, ketamine and norketamine kinetics in six female cats that received ketamine and propofol (1:1 ratio) as a loading dose (2 mg/kg each, IV) followed by a continuous infusion (10 mg/kg/h each, IV, 25 min of length). Blood samples were collected during the infusion period and up to 24 h afterwards. Drug quantification was achieved by HPLC analysis using UV-visible detection for ketamine and fluorimetric detection for propofol. The pharmacokinetic parameters were deduced by a two-compartment bolus plus infusion model for propofol and ketamine and a monocompartmental model for norketamine. Additional data were derived by a noncompartmental analysis. Propofol and ketamine were quantifiable in most animals until 24 and 8 h after the end of infusion, respectively. Propofol showed a long elimination half-life (t(1/2λ2) 7.55 ± 9.86 h), whereas ketamine was characterized by shorter half-life (t(1/2λ2) 4 ± 3.4 h) owing to its rapid biotransformation into norketamine. The clinical significance of propofol's long elimination half-life and low clearance is negligible when the drug is administered as short-term and low-dosage infusion. The concurrent administration of ketamine and propofol in cats did not produce adverse effects although it was not possible to exclude interference in the metabolism.
