Among beetles, the rice weevil Sitophilus oryzae is one of the most important pests causing extensive damage to cereal in fields and to stored grains. S. oryzae has an intracellular symbiotic relationship (endosymbiosis) with the Gram-negative bacterium Sodalis pierantonius and is a valuable model to decipher host-symbiont molecular interactions. We sequenced the Sitophilus oryzae genome using a combination of short and long reads to produce the best assembly for a Curculionidae species to date. We show that S. oryzae has undergone successive bursts of transposable element (TE) amplification, representing 72% of the genome. In addition, we show that many TE families are transcriptionally active, and changes in their expression are associated with insect endosymbiotic state. S. oryzae has undergone a high gene expansion rate, when compared to other beetles. Reconstruction of host-symbiont metabolic networks revealed that, despite its recent association with cereal weevils (30 Kyear), S. pierantonius relies on the host for several amino acids and nucleotides to survive and to produce vitamins and essential amino-acids required for insect development and cuticle biosynthesis. In addition to being an agricultural pest and a valuable endosymbiotic system, S. oryzae can be a remarkable model for studying TE evolution and regulation, along with the impact of TEs on eukaryotic genomes.

The design of doubly-curved systems called for innovative modelling techniques to evaluate the final geometry of the structure. They can simulate the behaviour of beams under large displacements. Even though it is never analysed, the deformation path is can be critical in such complex configurations. This paper analyses the potential of digital technologies to investigate the behaviour of beams under bending-torsion coupling in large deformation. In a first part, we will present the setup for an experiment, based on the use of robotic arms. The versatility of robotic arms allows to access complex configurations or loading paths, while controlling the test either in displacements or in forces. In a second part, an enhanced photogrammetry procedure is introduced. The aim is to retrieve the geometry of the beam even in the case of complex 3D shapes. The acquisition yields a point cloud representation of the beam. Finally, we propose a workflow to make the interaction with robotic arm, simple and efficient. The novel experimental setup aims at making possible the study of beams with complex form, under ever more complex force.

Most endogenous viruses, an important proportion of eukaryote genomes, are doomed to slowly decay. Little is known, however, on how they evolve when they confer a benefit to their host. Bracoviruses are essential for the parasitism success of parasitoid wasps, whose genomes they integrated ~103 million years ago. Here we show, from the assembly of a parasitoid wasp genome, for the first time at a chromosomal scale, that symbiotic bracovirus genes spread to and colonized all the chromosomes. Moreover, large viral clusters are stably maintained suggesting strong evolutionary constraints. Genomic comparison with another wasps revealed that this organization was already established ~53 mya. Transcriptomic analyses highlight temporal synchronization of viral gene expression, leading to particle production. Immune genes are not induced, however, indicating the virus is not perceived as foreign by the wasp. This recognition suggests that no conflicts remain between symbiotic partners when benefits to them converge.

International audience; Phytophagous insects use volatile organic compounds (VOC) emitted by plants to orient towards their hosts. In lepidopteran pests, crop damages are caused by larval stages-the caterpillars-that feed extensively on leaves or other plant tissues. However, larval host plant choice has been poorly studied, and it is generally admitted that caterpillars feed on the plant where the female laid the eggs. The mobility of caterpillars has been generally overlooked even though several studies showed that they can orient towards odors and change host plant. Recently, a large number of odorant receptors (ORs) tuned to plant volatiles have been characterized in the model pest moth Spodoptera littoralis (Noctuidae). In the present work, we identified nine of these deorphanized ORs as expressed in S. littoralis caterpillars. In order to understand whether these ORs are involved in host searching, we tested the behavioral significance of their ligands using a larval two-choice assay. This OR-guided approach led to the identification of nine plant volatiles, namely 1-hexanol, benzyl alcohol, acetophenone, benzaldehyde, (Z)3-hexenol, (E)2-hexenol, indole, DMNT and (Z)3-hexenyl acetate, which are active on S. littoralis caterpillar behavior, increasing our knowledge on larval olfactory abilities. To further explore the link between OR activation and behavioral output induced by plant volatiles we used a modeling approach, thereby allowing identification of some ORs whose activation is related to caterpillar attraction. These ORs may be promising targets for future plant protection strategies.

International audience; This paper presents the design and fabrication of an innovative structure, spanning 6.5m with a structural thickness of 12cm. The structural system derives from nexorades, also called reciprocal frames by some authors, obtained by a novel form-finding method based upon translation of the edges. It also allows for the covering with planar facets if the geometry before transformation is covered with planar facets. This property is used for the pavilion, covered with planar timber panels used as bracing elements. The benefit of this structural system, called shell-nexorade hybrid is discussed through a comparative study between the braced and un-braced nexorade (the stiffness is multiplied by ten adding only 30% mass). Finally, the fabrication process, achieved through robotized milling of the beams, is shown, illustrating the potential of pluri-disciplinary approach for conceptual design.

Olfaction plays a dominant role in the mate-finding and host selection behaviors of the codling moth (Cydia pomonella), an important pest of apple, pear and walnut orchards. Antennal transcriptome analysis (Bengtsson et al. 2012, Walker et al. 2016) revealed a number of abundantly expressed genes related to the moth olfactory system, including those encoding the olfactory receptors (ORs) CpomOR1, CpomOR3 and CpomOR6a, which belong to the putative pheromone receptor (PR) lineage, and the co-receptor (CpomOrco). Using heterologous expression, in both human embryonic kidney (HEK293T) cells and in Drosophila olfactory sensory neurons, coupled with calcium imaging and electrophysiological recording, respectively, we characterize the basic physiological and pharmacological properties of these receptors and demonstrate that they form functional ionotropic receptor channels.
