Invertebrates are exceptionally diverse, but declining because of anthropogenic changes to their habitat, as exemplified by freshwater bivalves in Europe and North America. Much less information is available for African freshwater bivalves, especially for Unionidae, which comprise 9 genera and ~40 nominal species, many of which are endemic to African ancient lakes. The phylogenetic position of most of these genera and species remains uncertain, and their conservation status unassessed. Here, we present preliminary results of phylogenetic studies on the Unionidae of the East African Rift. We integrate a phylogenetic backbone based on four gene fragments with (1) sampling information to examine geographic patterns of diversity and with (2) geometric morphometrics of shell shape to examine the relation between morphological disparity and molecular diversity. African Unionidae apart from ‘Cafferia’ form a monophyletic clade, and the basal splits in this clade occur between the reciprocally monophyletic genera Pseudospatha and Grandidieria, both of which are currently endemic to Lake Tanganyika. Mweruella, Nyassunio and Prisodontopsis are also monophyletic in the preliminary analyses as is Nitia, although this latter taxon is nested within Coelatura, which highlights the need of systematic revisions. Biogeographic analyses indicate a statistically significant North-to-South colonization of the East African Rift by Coelatura sensu lato. Beyond deep phylogenetic splits among individual clades, limited molecular differentiation is observed within most clades, calling for population genetic studies. Background: A major goal in evolutionary biology is to understand the processes that shape the evolutionary trajectory of clades. The repeated and similar large-scale morphological evolutionary trends of distinct lineages suggest that adaptation by means of natural selection (functional constraints) is the major cause of parallel evolution, a very common phenomenon in extinct and extant lineages. However, parallel evolution can result from other processes, which are usually ignored or difficult to identify, such as developmental constraints. Hence, understanding the underlying processes of parallel evolution still requires further research. Results: Herein, we present a possible case of parallel evolution between two ammonoid lineages (Auguritidae and Pinacitidae) of Early-Middle Devonian age (405-395 Ma), which are extinct cephalopods with an external, chambered shell. In time and through phylogenetic order of appearance, both lineages display a morphological shift toward more involute coiling (i.e. more tightly coiled whorls), larger adult body size, more complex suture line (the folded walls separating the gas-filled buoyancy-chambers), and the development of an umbilical lid (a very peculiar extension of the lateral shell wall covering the umbilicus) in the most derived taxa. Increased involution toward shells with closed umbilicus has been demonstrated to reflect improved hydrodynamic properties of the shell and thus likely results from similar natural selection pressures. The peculiar umbilical lid might have also added to the improvement of the hydrodynamic properties of the shell. Finally, increasing complexity of suture lines likely results from covariation induced by trends of increasing adult size and whorl overlap given the morphogenetic properties of the suture. As one of the essential components of the fossil record, shells of molluscs provide crucial data for taxonomic, phylogenetic or evolutionary studies. The mollusc shell has often very few discrete morphological characters, and its most important character is its three-dimensional (3-D) geometry, which is well diversified, continuous and highly integrated. However, standard morphometric methods remain mostly two-dimensional (2-D) and do not account for the ontogenetic changes of the shell, which are preserved thanks to its accretionary mode of growth. This study proposes a new non-destructive method enabling acquisition of three-dimensional quantitative morphometric parameters that thoroughly describe the geometry of coiled mollusc shells throughout their ontogeny. First, digital three-dimensional data of a shell is acquired by means of microcomputed tomography, which produces a series of grey-scaled, two-dimensional images. Second, all these stacked images are processed to obtain a three-dimensional reconstruction of the shell, from which a centreline is extracted. Finally, the geometry of shell aperture through ontogeny is extracted by successive cross-sectioning of the shell with a succession of planes, each of these being perpendicular to this centreline. The resulting outlines of the successive apertures can be quantified by elliptic Fourier analysis. These geometric parameters, coupled with the displacement vector of the successive cross-sectioning planes, constitute an n-dimensional morphometric space, in which ontogenetic trajectories of different individuals and species can be compared. The approach proposed in this study provides a basis for the quantitative analysis of growth patterns within and across species.

This study documents ammonoids with a precise stratigraphic control at the middle/late Anisian (Pelsonian/Illyrian) boundary from a new locality in eastern Lombardy-Giudicarie (Monte Guglielmo) and from classical sections in Giudicarie. These ammonoid faunas allow revising the taxonomic interpretation of Ceratites cimeganus Mojsisovics 1882 and of the genus Paraceratites Hyatt 1900. Ceratites cimeganus is here assigned to the North American genus Rieppelites Monnet & Bucher 2005. In eastern Lombardy-Giudicarie, R. cimeganus is diagnostic of a distinct biochronological unit (cimeganus Zone) bracketed between the older Bulogites zoldianus Zone and the younger Judicarites euryomphalus–Paraceratites trinodosus zones. The recognition of this cimeganus Zone significantly improves worldwide correlation since it is recognized in several other Tethyan basins (Dolomites, Northern Calcareous Alps) as well as in North America (Nevada). These new data allow a redefinition of the middle/late Anisian boundary in the western Tethys, which is here intercalated between the zoldianus and cimeganus zones. This limit is marked by a clear ammonoid turnover (e.g. disappearance of Acrochordiceras and Balatonites, appearance of Rieppelites). Finally, the presence of sections including the cimeganus Zone in eastern Lombardy-Giudicarie allow the establishment of local gaps in sedimentation, which may reflect the regional and important transgression of the pelagic Prezzo Limestone over the shallow water platform carbonates of a “Camorelli-Dosso dei Morti barrier”, as also underlined by the spatial distribution of brachiopod lumachellas.

Aims: To demonstrate the presence of an active alpha-acetolactate decarboxylase in Streptococcus thermophilus and to investigate its physiological function. Methods and Results: Streptococcus thermophilus CNRZ385 contains a gene encoding an alpha-acetolactate decarboxylase. Comparison of the production of alpha-acetolactate and its decarboxylation products, by the parent strain and an alpha-acetolactate decarboxylase-deficient mutant, demonstrated the presence of a control of the pool of alpha-acetolactate by valine, leucine and isoleucine. This control occurs via an allosteric activation of the alpha-acetolactate decarboxylase. Cell-free extracts of S. thermophilus were not able to decarboxylate the isoleucine precursor alpha-acetohydroxybutyrate. Conclusions: These results strongly suggest that one of the physiological functions of the alpha-acetolactate decarboxylase in S. thermophilus is to regulate leucine and valine biosynthesis by diverting the flux of alpha-acetolactate towards acetoin when the branched-chain amino acids are present at a high concentration. Significance and Impact of the Study: Regulation of branched-chain amino acid biosynthesis by alpha-acetolactate decarboxylase may occur in several other micro-organisms and explain some of their growth properties.
