Sorghum is currently being introduced in the temperate regions of Europe. It is characterized by good digestibility and high biomass yields, which make it a useful crop for anaerobic digestion. In this study, six commercial sorghum varieties comprising four different cultivars of Sorghum bicolor L. Moench and two interspecific sorghum hybrids (Sorghum sudanense L. × S. bicolor L. Moench) were harvested on two different dates during the years 2016 and 2017 at two diverse soil‐climate sites in Germany. The fresh harvested material and silages were analyzed to examine the ensilability of the different varieties with contrasting maturity characteristics. Subsequently, methane production experiments were performed to determine the specific methane yield (SMY) of the samples. The sorghum fresh matter (FM) varied among the sorghum types, including the parameters total solids (TS; 22.69%–46.93%FM), water‐soluble carbohydrates (2.68%–11.38%TS), and nitrates (0%–0.35%TS). The excellent ensiling ability of all the sorghum types analyzed was confirmed by evaluating the fermentation profile (pH range of 3.7–4.6; dominant presence of lactic acid [LA]; acetic acid [AA] in the range of 0.70%–2.38%TS; insignificant amount of butyric acid). The SMY ranged between 231.25 and 321.31 Ln kg−1 VS and tended to decrease with the increasing harvest time and maturity. LA and AA were positively correlated with the SMY, while the neutral detergent fiber content was negatively correlated with it. The SMY—a key parameter reflecting the crop biomass quality for biogas production—was slightly higher for S. bicolor than for the sorghum hybrids. However, the results of this study confirmed that if the final purpose is biomethanation, the ensilability of different sorghum types imposes no restriction. Furthermore, different sorghum types offer a wide harvest window, which can be useful for cropping schemes, ensiling, and methane production.

Xylo-oligosaccharides are sugar oligomers with 2~7 xylose units considered non-digestible fibers that can be produced from biodegradable and low-cost biomass like wheat straw. An integrated approach consisting of hydrothermal pretreatment, alkaline treatment, enzymatic treatment and the combinations thereof was applied to overcome the recalcitrance structure of the wheat straw and allow selective fractioning into fermentable sugars and xylo-oligosaccharides. The hydrolysates and processed solids were chemically characterized by High-performance liquid chromatography and Ion chromatography, and the results were expressed as function of the severity factor and statistically interpreted. The concentration of fermentable sugars (glucose, xylose, arabinose) was the highest after the combination of alkaline and enzymatic treatment with xylanase (18 g/L sugars), while xylo-oligosaccharides (xylotriose and xylotetraose) were released in lower amounts (1.33 g/L) after the same treatment. Refining experiments were carried out to obtain a purified fraction by using anion and cation exchange chromatography. The polymer adsorber resin MN-502 showed efficient removal of salts, phenols and furan derivatives. However, the xylo-oligosaccharides yields were also slightly reduced. Although still requiring further optimization of the treatments to obtain higher purified oligomer yields, the results provide information on the production of xylo-oligosaccharides and fermentable sugars from wheat straw for potential use in food applications.

During the last years, a demand for regionally produced biogas feedstocks was created by government subsidies to biogas production in Germany—contrary to the trend of specialization of agricultural production towards global commodity markets. The question arose whether this trend could contribute to an increased cultivation of uncommon crops and diversification of cropping patterns, owing to comparably different and less restricted feedstock requirements. In the cooperative research project “EVA,” a multi-site experimental crop rotation field trial was conducted over 8 years at eight sites, representing the variety of soil-climatic conditions in Germany. The aim of the trial was to assess a variety of established and novel crops for anaerobic digestion. This paper presents the key findings of the trial. Special emphasis is given to biomass productivity and profitability. The chances for the approach “diversification of cropping patterns via energy cropping” are discussed. Results show that maize (average 4-year dry matter yield varied site-specific between 14.22 and 25.12 t ha−1) is clearly the most efficient crop for biogas production in Central Europe. Some cropping options for biogas feedstock production, such as winter triticale (whole crop, average yield of 6.71 to 15.17 t ha−1) or perennial fodder mixtures (average yield of 7.51 to 19.44 t ha−1) are feasible choices for farmers in some regions, which could contribute to diverse cropping systems.

Biogas production can cause environmental problems due to a biased alignment of one energy crop used as feedstock, e.g. maize in Germany. Diversification of crop rotations and resource-efficient management can be the key to sustainable crop management. Four crop rotations on eight sites across Germany were evaluated in terms of their resource efficiency (area use, energy and economic efficiency) in order to derive options. The analysis revealed high variation in all indicators under review, with a high variance explanation by the interaction between crop rotation and regional characteristics. Furthermore, results indicate that high area-specific methane yields do not equate to high energy efficiency. Crop management adaptation is a useful tool for optimizing resource efficiency.

Background In biogas plants, complex microbial communities produce methane and carbon dioxide by anaerobic digestion of biomass. For the characterization of the microbial functional networks, samples of 11 reactors were analyzed using a high-resolution metaproteomics pipeline. Results Examined methanogenesis archaeal communities were either mixotrophic or strictly hydrogenotrophic in syntrophy with bacterial acetate oxidizers. Mapping of identified metaproteins with process steps described by the Anaerobic Digestion Model 1 confirmed its main assumptions and also proposed some extensions such as syntrophic acetate oxidation or fermentation of alcohols. Results indicate that the microbial communities were shaped by syntrophy as well as competition and phage-host interactions causing cell lysis. For the families Bacillaceae, Enterobacteriaceae, and Clostridiaceae, the number of phages exceeded up to 20-fold the number of host cells. Conclusion Phage-induced cell lysis might slow down the conversion of substrates to biogas, though, it could support the growth of auxotrophic microbes by cycling of nutrients.

With today's growing focus on on-demand energy production, the dynamic operation of biogas plants is becoming more and more important. Detecting unacceptable load changes and so preventing process failures is vital. In the present contribution, we propose an operating scheme that includes the detection of disturbances and intervention. This is then evaluated systematically with the help of a mathematical model for step changes in the organic loading rate. We summarize the qualitative results for each diagnosis variable in an operation diagram, enabling an intuitive, comprehensive comparison. Of the diagnosis variables analyzed, we find that the most promising for identifying incipient process failures in biogas plants are the volatile fatty acids to total alkalinity ratio (VFA/TA), followed by the ratio of methane to carbon dioxide γ in the biogas. In addition, the combination of the online measured variables pH value and gas composition can reveal disturbances early on in a wide range of different conditions.

Methane production characteristics and chemical composition of 405 silages from 43 different crop species were examined using uniform laboratory methods, with the aim to characterise a wide range of crop feedstocks from energy crop rotations and to identify main parameters that influence biomass quality for biogas production. Methane formation was analysed from chopped and over 90days ensiled crop biomass in batch anaerobic digestion tests without further pre-treatment. Lignin content of crop biomass was found to be the most significant explanatory variable for specific methane yields while the methane content and methane production rates were mainly affected by the content of nitrogen-free extracts and neutral detergent fibre, respectively. The accumulation of butyric acid and alcohols during the ensiling process had significant impact on specific methane yields and methane contents of crop silages. It is proposed that products of silage fermentation should be considered when evaluating crop silages for biogas production.

In this study modeled full scale application of thermobarical hydrolysis of less degradable feedstock for biomethanation was assessed in terms of energy balance, greenhouse gas emissions, and economy. Data were provided whether the substitution of maize silage as feedstock for biogas production by pretreated cattle wastes is beneficial in full-scale application or not.A model device for thermobarical treatment has been suggested for and theoretically integrated in a biogas plant. The assessment considered the replacement of maize silage as feedstock with liquid and/or solid cattle waste (feces, litter, and feed residues from animal husbandry of high-performance dairy cattle, dry cows, and heifers). The integration of thermobarical pretreatment is beneficial for raw material with high contents of organic dry matter and ligno-cellulose: Solid cattle waste revealed very short payback times, e.g. 9months for energy, 3months for greenhouse gases, and 3years 3months for economic amortization, whereas, in contrast, liquid cattle waste did not perform positive replacement effects in this analysis.

The effects of air stress during storage, exposure to air at feed-out, and treatment with silage additives to enhance aerobic stability on methane production from maize silage were investigated at laboratory scale. Up to 17% of the methane potential of maize without additive was lost during seven days exposure to air on feed-out. Air stress during storage reduced aerobic stability and further increased methane losses. A chemical additive containing salts of benzoate and propionate, and inoculants containing heterofermentative lactic acid bacteria were effective to increase aerobic stability and resulted in up to 29% higher methane yields after exposure to air. Exclusion of air to the best possible extent and high aerobic stabilities should be primary objectives when ensiling biogas feedstocks.

In this study, microbiomes of 36 full-scale anaerobic digesters originated from 22 different biogas plants were compared by terminal restriction fragment length polymorphism (TRFLP) analysis. Regarding the differences in microbial community composition, a weighting of the environmental parameters could be derived from higher to lower importance as follows: (i) temperature, (ii) TAN and NH3 concentrations and conductivity, and (iii) the chemical composition of the supplied feedstocks. Biotic interactions between specific bacterial and archaeal community arrangements were revealed, whereby members of the phyla Bacteroidetes and Cloacimonetes combined with the archaeal genus Methanothrix dominated the conversion of homogeneous feedstocks, such as waste water sludge or industrial waste. As most of the detected TRFs were only found in a certain number of anaerobic digestion plants, each plant develops its unique microbiome. Sugar cane press mud pretreated by liquid hot water was separated into a liquid fraction and a solid fraction. These fractions were blended with vinasse in different proportions, according to a three-factor mixture design, to evaluate their synergetic and antagonistic effects on the methane yield. The biochemical methane potential was determined in batch assays under mesophilic conditions (37 ± 1 °C). The highest methane production rates were obtained for a mixture of 33% chemical oxygen demand (COD) vinasse and 67% COD liquid fraction of press mud and for sheer vinasse (73 and 84 N mL CH4 g–1 CODfed day–1). The maximum methane yield was found for the pure liquid fraction with a value of 369 N mL CH4 g–1 CODfed. For most mixtures examined, antagonistic effects were found, with significant differences when mixing vinasse and the liquid fraction.

Efficient utilization of lignocellulosic biomass requires pretreatment in order to liberate cellulose from lignin and disrupt its recalcitrant crystalline structure before effective enzymatic hydrolysis can take place. Three different pretreatment methods (pressure cooking with dilute alkali and dilute acid as well as alkaline extraction) to recover the xylooligosaccharides fraction from five different grass silage samples, whole crop rye silage and maize silage were compared. The predominant end products released were xylobiose, xylotetraose, xylopentaose and xylohexaose whereas the xylooligosaccharides release pattern differed with the substrate. Maximum values of xylooligosaccharides was found for grass silage 17.26 g/L, whole crop rye silage 3.06 g/L and for maize silage 5.77 g/L. Knowledge of fate and behavior of plant pathogens in the biogas production chain is limited and hampers the estimation and evaluation of the potential phytosanitary risk if digestate is spread on arable land as a fertilizer. Therefore, simulation is an appropriate tool to demonstrate the effects which influence the steady state of pathogen infected plant material in both digesters and digestate. Simple approaches of kinetics of inactivation and mass balances of infected material were carried out considering single-step as well as two-step digestion. The simulation revealed a very fast to fast reduction of infected material after a singular feeding, reaching a cutback to less than 1% of input within 4 days even for D90-values of 68 h. Steady state mass balances below input rate could be calculated with D90-values of less than 2 h at a continuous hourly feeding. At higher D90-values steady state mass balances exceed the input rate but are still clearly below the sum of input mass. Dilution further decreases mass balances to values 10(-5) to 10(-6) Mg m(-3) for first-step digestion and 10(-8) to 10(-9) for second-step.

Biogas production from grassland biomass harvested during landscape management may help to maintain species-rich grassland biotopes, but extensive management and late harvests often result in low-quality biomass. Biogas production from the vegetation of Alopecuretum pratensis, Molinietum caeruleae and Caricetum gracilis, three typical grassland biotopes in north German nature reserves, was investigated in relation to harvest date. In addition, the A. pratensis vegetation was investigated for ensiling and the application of bacterial silage additives. Results indicate that biogas production might be a reasonable utilization pathway for grassland biomass from landscape management if the first cut occurs up to late summer. Methane yields of grassland biomass decreased substantially with later harvest, from up to 309 lN kg−1 organic dry matter (ODM) in May to below 60 lN kg−1 ODM in February, in correlation with increasing crude fibre contents. Caricetum gracilis vegetation was the least suitable feedstock for biogas production. It showed a rapid decline in methane yields with later harvest and 25% lower methane yields compared with other types of grassland vegetation. Application of silage additives is recommended for adequate preservation of grassland biomass from landscape management by ensiling. Addition of homofermentative lactic acid bacteria improved acidification during ensiling if sufficient fermentable sugar was available. The use of inoculant and molasses enhanced methane yields by 3–55%. Additional carbohydrate source is necessary to ensure proper ensilage when grasses are harvested after late autumn.

Feedstock of anaerobic digestion infected with phytopathogens could enhance the risk of spreading those pathogens to uninfested field through digestate. The viability of Fusarium proliferatum, Fusarium verticillioides, Sclerotinia sclerotiorum, and Rhizoctonia solani was investigated in anaerobic digestion experiments using infected plant material of sorghum (Sorghum bicolor), sugar beet (Beta vulgaris subsp. vulgaris var. altissima), and potato (Solanum tuberosum L.). Results from lab-scale reactors were confirmed in full-scale biogas plants. Anaerobic digestion under mesophilic conditions (35–42 °C) reduced most of the phytopathogens of feedstocks investigated. Thus, S. sclerotiorum and R. solani lost their viability within 6 h. In the case of sorghum, however, Fusarium spp. infected feedstock required a maximum of 138 h for sanitation. Thus, the risk of spreading plant pathogens with the digestate can only be decreased when the feedstock would undergo an additional treatment before anaerobic digestion or of the resulting digestate.

Crop rotations are part of current agricultural practice, since they and their effects can contribute to a sustainable agricultural cropping system. However, in current Life Cycle Assessment (LCA) studies, crop rotation effects are insufficiently considered, since these effects are difficult to measure. LCA studies from crop production typically take only one vegetation period into account. As a result, the consideration of how the assessed crop is influenced by the previous crop (crop rotation effects) including: (1) nutrient carryover, (2) reduction in operational requirements and (3) different intensity and timing of farming activities, is outside of the system boundary. However, ignoring these effects may lead to incorrect interpretation of LCA results and consequently to poor agricultural management as well as poor policy decisions. A new LCA tool called the “Model for integrative Life Cycle Assessment in Agriculture (MiLA)” is presented in this work. MiLA has been developed to assess GHG emissions and cumulative energy demands (CED) of cropping systems by taking the characteristics of crop cultivation in rotation into account. This tool enables the user to analyze cropping systems at farm level in order to identify GHG mitigation options and energy-efficient cropping systems. The tool was applied to a case study, including two crop rotations in two different regions in Germany with the goal of demonstrating the effectiveness of this tool on LCA results. Results show that including crop rotation effects can influence the GHG emission result of the individual crop by − 34% up to + 99% and the CED by − 16 up to + 89%. Expanding the system boundary by taking the whole crop rotation into account as well as providing the results based on different functional units improves LCA of energy crop production and helps those making the assessment to draw a more realistic picture of the interactions between crops while increasing the reliability of the LCA results.

Summary Digestate, the semi-solid leftover after anaerobic digestion in biogas reactors, is frequently used as a crop fertiliser. The ability of seeds from five plant species (four weed species and one vegetable crop) to survive commercial biogas plants was tested, to assess the likelihood that these could be spread via digestate and to compare with results obtained from laboratory-scale reactors. Seeds were selected from among the best surviving species in experimental batch reactors in an earlier study. They were exposed for 1–9 days in two large-scale, commercial mesophilic biogas plants. The resulting exponential decay functions were estimated. The decimal reduction time, D, was estimated at 1.5 days for Abutilon theophrasti population D2003, 2 days for A. theophrasti population ES2008, 5.8 days for Malva neglecta, 4.7 and 19.7 days for Chenopodium album, and 1.2–9.1 days for Fallopia convolvulus. Given the differences in ranking of plant species, laboratory reactors are not necessarily a good model system for commercial reactors. Seeds could survive passage through biogas plants, although at (extremely) low numbers. If the objective is to eliminate all risks and dispose of all weed seeds, it will be necessary to either add extra sanitation steps, or to identify and manipulate the factors that are responsible for high seed mortality. However, it is currently unknown what these factors are.

Shortening chopping length at harvest of biogas crops is a basic method of mechanical pretreatment that potentially affects subsequent conservation and biogas production processes. The objective of this study was to assess the influence of a wider range of particle size distributions achieved by laboratory chopping and of practice-oriented short and very short chopping lengths induced by forage harvesters, respectively, on ensiling parameters and methane formation. Shortening the cutting length enhanced lactic acid fermentation during ensiling and increased methane yield, although the effects of ensilage products on methane production partly overlaid the direct positive influence due to reduced particle size. A maximum increase in methane yield of 11 % to 13 % was obtained in the range of median particle lengths of 33 to 6 mm when taking storage losses into account. Thus, very short chopping lengths are beneficial for conservation and methane formation, yet full assessment necessitates further consideration of additional expenditures at harvest.

Reducing chopping length for biogas crop harvesting is a pretreatment method to support the processes of ensiling and methane formation, yet it also increases expenditures at harvest. To assess environmental performance and economic viability, the effects of reducing chopping lengths from common to very short settings on energy balance, greenhouse gas emissions and profitability were investigated. Assessment was based on data gained by monitoring harvesting and ensiling process chains in practice. Balancing results showed low overall effects of chopping length reduction on net energy yield and net greenhouse gas emissions. Shorter chopping length settings reduced net greenhouse gas emissions slightly while net energy yield and profit only increased significantly in 30 to 40 % of the farm-scale harvests investigated. Conditions promoting intensified chopping include the use of high-capacity harvesters, chopping of lignin-rich feedstocks, high subsidies for generated electricity and the utilisation of waste heat. Owing to numerous influencing factors, profitability cannot be guaranteed and so chopping lengths below 7 to 8 mm are not recommended.

Increased methane yields and enhanced ensilability are reasons in favour of very short cutting lengths when chopping crops for biogas utilisation, though this also leads to higher expenditure at harvest. The objective of this study was to evaluate the effects of very short chopping lengths in maize harvesting on capacities, labour time requirements and process costs of the harvest and ensiling chain. Based on data gathered in field experiments, a model calculation that allowed linkage of harvest and transport processes was applied to assess capacities and machinery performance. Results show a decrease in overall area capacities of harvest and transportation units by 4–24% at a nominal chopping length of 4–6 mm compared with common chopping lengths of 8–10 mm. Harvesting costs increased by 4–21%. The effect of lowering machinery chain performance was diminished with more powerful forage harvesters, smaller field sizes, and larger transport distances. Hence, considerable additional expenditures are to be assumed with shortening of chopping length. However, effects depend on particular field and machinery conditions. There is further need to take the additional benefit for storage and methane production into account before recommendations for optimal length of cut can be provided.

Continuous bio-methanization of different feedstocks (rye grain silage, maize silage, feed residue (mix of silages), solid cattle manure, and grass silage) was investigated in a long-term laboratory-scale experiment with and without enzyme application. Ten-liter reactors were operated simultaneously in a two-step digestion mode for the continuous production of biogas from different feedstocks over 354 days. One set of reactors was operated as main digester, while the second set was used for the second step. The daily input of feedstock was increased from an organic loading rate of 1 to 3 kg ODM·m–3·d–1. All digesters were run under stable conditions, indicated by the ratio of volatile fatty acids to the total inorganic carbon, ranging around 0.2 in the first step and 0.15 in the second step. Crops cultivated as feedstock for energy conversion processes (energy crops) could cover about 5% of the total energy demand of the European Union. Therefore, cultivation, harvest, processing, and finally utilisation of feedstock are required to be sustainable minimising environmental pressures and maximising environmental benefits. This contribution will give a comprehensive view on the environmental impacts of cultivating conventional energy crop species like cereals, rape seed and maize and of the hitherto uncommon species such as willow and poplar. Generalisations of environmental relevant parameters are deduced based on literature surveys as well as on results from own long-term experiments at Leibniz-Institut fur Agrartechnik institute. Effects of the ensiling process, storage periods of up to 1 year and several chemical and biological silage additives on biomethanation were assessed for maize, sorghum, forage rye and triticale with the aim to identify optimised conditions for silage production of crops used as feedstock in biogas plants. Ensiling, prolonged storage and biological silage additives showed positive effects on methane yield of up to 11%. These could be attributed to increases in organic acids and alcohols during ensiling. A regression model including acetic acid, butyric acid and ethanol accounts for 75-96% of the variation in methane yield. Storage periods of up to 1 year for properly ensiled crops could be possible without losses in methane production, considering the increase in methane yield and the losses of dry matter during this period. However, taking storage losses into account silage additives showed little effect on methane production.

Ten biogas plants across Europe were investigated for the concentrations of trace elements in their digestates. Many of these trace elements are important micro nutrients and act as microbial agents responsible for the anaerobic digestion of organic material. Great variations in concentrations from biogas plant to biogas plant were found covering a range of 1–2 orders of magnitude. No deficit of nutrients was detected in any of the biogas plants tested, but those plants with high inputs of energy crops and manure had lower values. Also biogas plants feeding high amounts of glycerol in addition to their agricultural feedstock generally showed low concentrations of micro nutrients. The highest concentrations of nutrients were detected at biogas plants fed by bleaching earth. Biogas plants fed with wastes like blood, kitchen and food waste also revealed higher concentrations of micro nutrients.

This chapter reviews the current knowledge on sanitation during anaerobic digestion and the hygienic status of digestate concerning a multitude of pathogens potentially compromising the health of humans, animals and plants. Physical, chemical and biological parameters influencing the efficiency of sanitation in anaerobic digestion are considered. The degree of germ reduction depends particularly on the resistance of the pathogen of concern, the processing conditions, the feedstock composition and the diligence of the operation management. Most scientific studies facing sanitation in biogas plants have provided data ascertaining reduction of pathogens by the biogas process. Some pathogens, however, are able to persist virtually unaffected due to the ability to build resistant permanent forms. As compared to the feedstock, the sanitary status of the digestate is thus improved or in the worst case, the sanitary quality remains almost unchanged. According to this, the spreading of digestate on agricultural area in accordance to current rules and best practice recommendations is considered to impose no additional risk for the health of humans, animals and plants.

Sugarcane press mud was pretreated by liquid hot water (LHW) at different temperatures (140-210 °C) and pre-treatment times (5-20 min) in order to assess the effects on the chemical oxygen demand (COD) solubilisation, inhibitors formation and methane yield. The experimental results showed that a high degree of biomass solubilisation was possible using LHW. Higher methane yields were obtained at lower severities (log(Ro) = 2.17-2.77) with (i) mild temperatures (140-150 °C) and long contact times (12.5 min, 20 min) or (ii) mild temperatures (175 °C) with short contact time (2 min). The highest increase in methane yield (up to 63%) compared to the untreated press mud was found at 150 °C for 20 min. At temperatures of 200 °C and 210 °C, low methane efficiency was attributed to the possible formation of refractory compounds through the Maillard reaction.

Biogas production from energy crops and biodegradable waste is one of the major sources for renewable energies in Germany. Within a biogas plant (BGP) a complex microbial community converts biomass to biogas. Unfortunately, disturbances of the biogas process occur occasionally and cause economic losses of varying extent. Besides technical failures the microbial community itself is commonly assumed as a reason for process instability. To improve the performance and efficiency of BGP, a deeper knowledge of the composition and the metabolic state of the microbial community is required and biomarkers for monitoring of process deviations or even the prediction of process failures have to be identified. Previous work based on 2D-electrophoresis demonstrated that the analysis of the metaproteome is well suited to provide insights into the apparent metabolism of the microbial communities. Using SDS-PAGE with subsequent mass spectrometry, stable protein patterns were evaluated for a number of anaerobic digesters. Furthermore, it was shown that severe changes in process parameters such as acidification resulted in significant modifications of the metaproteome. Monitoring of changing protein patterns derived from anaerobic digesters, however, is still a challenge due to the high complexity of the metaproteome. In this study, different combinations of separation techniques to reduce the complexity of proteomic BGP samples were compared with respect to the subsequent identification of proteins by tandem mass spectrometry (MS/MS): (i) 1D: proteins were tryptically digested and the resulting peptides were separated by reversed phase chromatography prior to MS/MS. (ii) 2D: proteins were separated by GeLC-MS/MS according to proteins molecular weights before tryptic digestion, (iii) 3D: proteins were separated by gel-free fractionation using isoelectric focusing (IEF) conducted before GeLC-MS/MS. For this study, a comparison of two anaerobic digesters operated at mesophilic and at thermophilic conditions was conducted. The addition of further separation dimensions before protein identification increased the number of identified proteins. On the other hand additional fractionation steps increased the experimental work load and the time required for LC-MS/MS measurement. The high resolution of the 3D-approach enabled the detection of approximately 750 to 1650 proteins covering the main pathways of hydrolysis, acidogenesis, acetogenesis and methanogenesis. Methanosarcinales dominated in the mesophilic BGP, whereas Methanomicrobiales were highly abundant in the thermophilic BGP. Pathway analysis confirmed the taxonomic results and revealed that the acetoclastic methanogenesis occurred preferentially at mesophilic conditions, whereas exclusively hydrogenotrophic methanogenesis was detected in thermophilic BGP. However, for the identification of process biomarkers by comprehensive screening of BGP it will be indispensable to find a balance between the experimental efforts and analytical resolution.

Lab-scale experiments were conducted to assess the impact of thermobarical treatment of cattle waste on anaerobic digestion. Treatment was at temperatures of 140–220 °C in 20 K steps for a 5-min duration. Methane yields could be increased by up to 58% at a treatment temperature of 180 °C. At 220 °C the abundance of inhibitors and other non-digestible substances led to lower methane yields than those obtained from untreated material. In an extended analysis it could be demonstrated that there is a functional correlation between the methane yields after 30 days and the formation rate and methane yield in the acceleration phase. It could be proved in a regression of these correlation values that the optimum treatment temperature is 164 °C and that the minimum treatment temperature should be above 115 °C.

In biogas plants agricultural waste and energy crops are converted by complex microbial communities to methane for the production of renewable energy. In Germany, this process is widely applied namely in context of agricultural production systems. However, process disturbances, are one of the major causes for economic losses. In addition, the conversion of biomass, in particular of cellulose, is in most cases incomplete and, hence, insufficient. Besides technical aspects, a more profound characterization concerning the functionality of the microbial communities involved would strongly support the improvement of yield and stability in biogas production. To monitor these communities on the functional level, metaproteome analysis was applied in this study to full-scale agricultural biogas plants. Proteins were extracted directly from sludge for separation by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and subsequent identification with mass spectrometry. Protein profiles obtained with SDS-PAGE were specific for different biogas plants and often stable for several months. Differences of protein profiles were visualized by clustering, which allowed not only the discrimination between mesophilic and thermophilic operated biogas plants but also the detection of process disturbances such as acidification. In particular, acidification of a biogas plant was detected in advance by disappearance of major bands in SDS-PAGE. Identification of proteins from SDS-PAGE gels revealed that methyl CoM reductase, which is responsible for the release of methane during methanogenesis, from the order Methanosarcinales was significantly decreased. Hence, it is assumed that this enzyme might be a promising candidate to serve as a predictive biomarker for acidification.

Landscape management grass is generally harvested late, resulting in unfavorable composition for many utilization purposes. This study explores various technical concepts of biogas production and their economic viability. The Lower Oder Valley National Park is taken here as an example. This National Park in North-East Germany comprises large grassland areas with conservation-related restrictions on management. The concepts of biogas production and use considered are: (1) decentralized digestion and use of biogas at five autonomous combined heat and power (CHP) units, (2) decentralized digestion and delivery of the biogas to a centralized CHP unit, (3) decentralized digestion, upgrading of the biogas and feeding into the natural gas grid, and (4) one central biogas plant with centralized CHP unit. Annual costs and revenues of biogas production were calculated for each alternative. Biogas production from landscape management grass meets the conservational demands of late cutting periods and under certain circumstances shows a profit.

A yeast-like fungus producing a large amount of extracellular polysaccharide was isolated from the biogas reactors at North Wyke Research, UK (latitude, 50°46′29″N; longitude, 3°55′23″W). This strain was identified as Aureobasidium pullulans based on nucleotide sequence of the D1/D2 domain of the large-subunit (LSU) rRNA gene. Extracellular polysaccharide or pullulan, produced from this new strain of A. pullulans in shake flasks and a stirred tank fermenter was investigated. A maximum polysaccharide concentration of 40.1 g l −1 , and productivity of 12.5 g l −1 per day was obtained in a batch culture from stirred tank fermenter. The optimal cultivation conditions for pullulan production for this strain in 250 ml shake flask containing 100 ml of sucrose medium were observed at 28 °C and with 200 rpm. The effect of different sugars and nitrogen sources on pullulan production was investigated. It was found that more pullulan was produced when the strain was grown in the medium supplemented with NaNO 3 than when it was cultivated in the medium supplemented with C5 and C6 sugars and other nitrogen sources.

Currently biomass contributes to 69 106 tons of oil equivalents (MtOE) or 4% of the total energy consumption in Europe. According to the European Union (EU) Biomass Action Plan biofuels shall contribute 150 MtOE to the total energy consumption in 2010. This share shall increase to 20% or 220 MtOE in 2020. Approximately half of it will be derived from arable land, i.e. 23 MtOE woody biomass (short rotation coppice [SRC]) and 88 MtOE herbaceous biomass. This would comprise 15% of the arable land and hence, the question arises whether this energy resource can be exploited sustainably or not. It is assumed that using energy crops as resource increases biodiversity and farmers sources of income. In this chapter sustainability will be assessed in relation to energy balance, greenhouse gas (GHG) reduction and emission of other air pollutants.

The aim of this review is to summarize current knowledge on suitability and sustainability of grassland biomass for combustion. In the first section grassland management for solid biofuel as well as information on harvest, postharvest and firing technology are described. An extensive grassland management system with one late cut and low level of fertilization is favored for grass as a solid biofuel. The grass harvest usually involves drying in the field and clearing with conventional farm machinery. Pelleting or briquetting improves the biofuel quality. Grass combustion is possible as stand-alone biomass-firing or co-firing with other fuels. Firing herbaceous biomass requires various specific adaptations of the different combustion technologies. In the second section economic and environmental aspects are discussed. Costs for biomass supply mainly depend on yields and harvesting technologies, while combustion costs are influenced by the size and technical design of the plant. Market prices for grass and possible subsidies for land use are crucial for profitability. Regarding biogeochemical cycles a specific feature of combustion is the fact that none of the biomass carbon and nitrogen removed at harvest is available for return to the grassland. These exports can be compensated for by fixation from the air given legumes in the vegetation and sufficient biomass production. Greenhouse gas emissions can be considerably reduced by grass combustion. Solid biofuel production has a potential for predominantly positive impacts on biodiversity due to the extensive grassland management.

Grassland biomass is suitable in numerous ways for producing energy. It is well established as feedstock for biogas production. The aim of this review is to summarize current knowledge on suitability and sustainability of grassland biomass for anaerobic digestion. In the first section grassland management for biogas feedstock as well as specifics of harvest, postharvest and digestion technology are described. Methane yields from grass are influenced by many factors. While the effects of some parameters such as grass species, cutting period and management intensity can be regarded as well known, other parameters such as preservation and processing still need investigation. In the second section economic aspects and environmental impacts are discussed. Profitability can be achieved depending on grass silage supply costs and the concept of anaerobic digestion and energy use. Grassland biomass for biogas production competes with other feedstock and other forms of grassland use, in particular animal husbandry. In developed countries a growing production of milk and meat is achieved with decreasing ruminant numbers, resulting in an increasing amount of surplus grassland with a remarkable bioenergy potential. In emerging and developing countries a rapidly rising demand for and production of milk and meat induce growing pressure on grasslands, so that their use for animal feed presumably will take priority over use for bioenergy. Grasslands provide a variety of essential environmental benefits such as carbon storage, habitat function, preservation of ground and surface water quality. When producing biogas from grassland these benefits will remain or even grow, providing appropriate grassland management is implemented. In particular, greenhouse gas emissions can be considerably reduced.
