A new lucinoidean genus and species, Cerkesia contiae gen. et sp. nov., from the Kimmeridgian carbonate platform deposits of the Çerkes area (western Pontides, Turkey), is described and placed in the family Fimbriidae. Morphological comparisons reveal similarities with Fimbria, Cyclopellatia and Sphaera. A phylogenetic analysis supports the monophyly of the family. Similarities between Cerkesia and Fimbria determine inclusion of these genera in the same clade. The unusual features characterizing Cerkesia closely relate it to Cyclopellatia. Sphaera belongs to a different clade and its similarities with Cerkesia are owing to convergent and plesiomorphic characters. Cerkesia was a slow and shallow burrower. The morphology of the anterior adductor scar suggests a mode of life comparable to that of Fimbria but other features indicate adaptive strategies typical of the Lucinidae. The associated fossils and facies reveal that Cerkesia lived in soft and sandy substrates of quiet and protected areas of the carbonate platform under stressful environmental conditions. The appearance of Cerkesia in the western Pontides marks the eastward expansion of a clade that developed principally in the European seas during the Late Jurassic while the Sphaeriola-Sphaera clade was confined to the southernmost Tethyan margin.

Studies of Jurassic outcrops from Veneto, Central Italy and Sicily improved our knowledge on the gastropods of these areas, characterised by the presence, aside of archaic forms, of some modern ones. Noteworthy is the record from those outcrops of several species, which can be referred to the family Rissoidae (Rissooidea). Formerly, earliest rissoid records were only known from European outcrops of younger age. In the Italian records several species have been recognised belonging to at least two genera of Rissoidae, namely Rissocerithium and Zebinostoma. The presence of such a relatively diversified number of species, bears witness of a well established group. They should have had a common ancestor with the Rissoina-like rissoids of the continental margins. Their diversification can be related to the peculiar characters of the Intra-Tethyan communities.

Members of Ataphridae became a progressive group in one of the two types of Tethyan faunas. Some of the new forms, which evolved from ataphrid ancestors, can be classified within the family, whilst others show a transitional morphology towards the turbinid subfamily Colloniinae. Other finds suggest a close relationship between Ataphridae and Crossostomatidae.

The food waste reduction through an efficient recovery of its valuable building molecules has become an important topic with a positive effect on the economy and the environment. In this work, the revalorization of slaughterhouse calf fleshing meat through its enzymatic hydrolysis is proposed. The proteolytic activity of 11 enzymes was initially screened and the four most efficient enzymes (papain, trypsin, pancreatin, and bromelain) were selected. The molecular profiling of the different protein/peptide fractions by the Linear Trap Quadrupole-OrbiTrap technique showed compositional differences due to the specificity of the enzymes’ cleavage sites. In order to find a potential reuse of these hydrolysates, the analysis of antioxidant and, for the first time on fleshing meat hydrolysates, of anti-tyrosinase activities, was performed. Papain-digested samples were those showing the highest inhibition activity of tyrosinase enzyme (55.6%) as well as the highest antioxidant activity (3.52 g TEAC/L). In addition, the composition analysis of the lipid fraction was performed. The mono-unsaturated fatty acids resulted to be the most abundant lipid in all the samples with the exception of pancreatin-treated hydrolysates in which poly-unsaturated fatty acids were predominant. The present results seemed to support a possible valorization of isolated fractions from calf fleshing by-products, as food or feed ingredients, by the implementation of fraction isolation within the meat-processing pipeline.

In the past, wild edible alimurgic plants became an important alternative food source when poverty, wars or drought made it difficult to access crops. These plants were considered rich in highly nutritional compounds and also frequently used as food-medicine given their health-promoting properties. With the aim of improving our knowledge on the content of beneficial or detrimental compounds in relation with past local dietary and curative traditions, 12 wild food plant species were collected from two study areas selected for their very different degree of industrialization, urbanization, and conservation of local past traditions among the population: the Bologna province (Northern Italy) and the Middle Agri Valley (Southern Italy). Protein, polyphenol flavonoid and biogenic amine (both free and conjugated) contents and antioxidant activity of raw and boiled wild food plant extracts, and of cooking water were analyzed by means of spectrophotometric and high-performance liquid chromatography methods. The results demonstrated that most of the phenolic compounds were released in the cooking water which also showed the highest antioxidant activity. Seventeen different phenolic compounds were identified, of which the health-related luteolin, luteolin-7-glucoside and rutin were the most abundant (e.g., S. pratensis L. and C. intybus L.). On the other hand, biogenic amines were absent or present at very low levels in cooking water of those very same species (e.g., S. pratensis L., T. officinalis Weber, C. vesicaria subsp. taraxacifolia and C. intybus L.) of which traditionally a decoction is used for therapeutic purposes. Free and conjugated spermidine and spermine were generally the most abundant biogenic amines, while none of the known detrimental monoamines (e.g., histamine) was detected. In conclusion, the present results seem to support past local popular traditions which indicated beneficial medical properties of some wild edible plant, as well as of their cooking water.

Not all the coffee produced goes to the roasting stage, because non-compliant green coffee beans are usually discarded by roasters and the silverskin of the coffee is usually removed and discarded. In the present work, non-compliant green coffee beans and coffee silverskins were fully characterized from a chemical point of view. In addition, enzyme-assisted extraction was applied to recover a fraction rich in proteins and polyphenols, tested for antimicrobial, antityrosinase, and antioxidant activities. Non-compliant green coffee beans showed higher amounts of polyphenols, flavanols, flavonoids, and caffeine than coffee silverskins (which were richer in tannins). The enzymatic extraction of non-compliant coffee green beans produced extracts with a good protein content and with a consistent quantity of polyphenols. The extract showed antioxidant, antityrosinase, and antimicrobial activity, thus representing a promising strategy to recover defective green coffee beans. The antioxidant and antimicrobial activity of coffee silver skins is lower than that of non-compliant coffee green beans extracts, while the antityrosinase activity is comparable.

The present work reports the enzymatic valorisation of the protein fraction of scotta, a dairy by-product representing the exhausted liquid residue of ricotta production. Scotta was subjected to ultra-filtration with membrane cut-offs from 500 to 4 kDa and the obtained protein-enriched fractions were used for the optimization of enzyme-based digestions aimed at producing potentially bioactive peptides. Nine different commercial proteases were tested and the best digestion conditions were selected based on protein yield, fraction bioactivity and foreseen scale up processing costs. Scale up of the 3% Pancreatin or 5% Papain processes was performed up to 2 L (37°C or 60°C respectively, 1 h incubation), and the digestion efficiency increased with the reaction volume as well as antioxidant activity (up to 60 gBSA eq/L and to 1.7 gAA eq/L). Retentate 1 digested fractions also showed, for the first time in dairy-based peptides, anti-tyrosinase activity, up to 0.14 gKA eq/L. Digested proteins were sub-fractionated by means of physical membrane separations and 30-10 kDa fraction from Papain treatment showed the highest antioxidant and anti-tyrosinase activities. The peptide sequence of the most bioactive fractions was achieved.

Unfolding turns immobilized cytochrome c into a His-His ligated form endowed with catalytic activity towards O(2), which is absent in the native protein. Dioxygen could be used by naturally occurring unfolded cytochrome c as a substrate for the production of partially reduced oxygen species (PROS) contributing to the cell oxidative stress.

The electron transfer (ET) process of progressively unfolded bovine cytochrome c immobilized on different self-assembled monolayers (SAMs) was investigated. Insight is gained on the role of the SAM surface on the functionality of the partially unfolded and non-native forms of the adsorbed protein. Direct electrochemical measurements were performed on cytochrome c adsorbed on mercaptopyridine (MP) and mixed 11-mercapto-1-undecanoic acid/11-mercapto-1-undecanol (MUA/MU) at varying temperature, in the presence of urea as unfolding agent. Under strongly unfolding conditions, a non-native form of cytochrome c , in which the methionine ligand is replaced by a histidine, was observed on both MP and MUA/MU SAMs. The E °′ of the native form, in which the haem is axially coordinated by methionine and histidine, slightly shifts to negative values upon increasing urea concentration. However, the non-native bis-histidinate species shows a much lower E °′ value (by approximately 0.4 V) which is by far enthalpic in origin and largely determined by axial ligand swapping. Analysis of the reduction enthalpies and entropies and of the ET rate constants indicate that the nature of the SAM (hydrophilic or anionic) results in changes in the conformational rearrangement of the cytochrome c under unfolding conditions.

We have studied the effect of urea-induced unfolding on the electron transfer process of yeast iso-1-cytochrome c and its mutant K72AK73AK79A adsorbed on electrodes coated by mixed 11-mercapto-1-undecanoic acid/11-mercapto-1-undecanol self-assembled monolayers. Electrochemical measurements, complemented by surface enhanced resonance Raman studies, indicate two distinct states of the adsorbed proteins that mainly differ with respect to the ligation pattern of the haem. The native state, in which the haem is axially coordinated by Met80 and His18, displays a reduction potential that slightly shifts to negative values with increasing urea concentration. At urea concentrations higher than 6 M, a second state prevails in which the Met80 ligand is replaced by an additional histidine residue. This structural change in the haem pocket is associated with an approximately 0.4 V shift of the reduction potential to negative values. These two states were found for both the wild-type protein and the mutant in which lysine residues 72, 73 and 79 had been substituted by alanines. The analysis of the reduction potentials, the reaction enthalpies and entropies as well as the rate constants indicates that these three lysine residues have an important effect on stabilising the protein structure in the adsorbed state and facilitating the electron transfer dynamics. © 2010 SBIC.

Many real world phenomena are better represented by non-precise data rather than by single-valued data. In fact, non-precise data represent two sources of variability: the natural phenomena variability and the variability or uncertainty induced by measurement errors or determined by specific experimental conditions. The latter variability source is named imprecision. When there are information about the imprecision distribution the fuzzy data coding is used to represent the imprecision. However, in many cases imprecise data are natively defined only by the minimum and maximum values. Technical specifications, stock-market daily prices, survey data are some examples of such kind of data. In these cases, interval data represent a good data coding to take into account the imprecision. This paper aims at describing multiple imprecise data by means of a suitable Principal Component Analysis that is based on specific interval data coding taking into account both sources of variation.

The aim of the paper is to analyze, for both men and women, the role played by value orientation and social condition in the choice of the various forms of partnership (marriage and cohabitation) in some European countries that represent the varying diffusion of cohabitation and are characterized by different ideational dimensions. The data analyzed using classification trees were obtained from the European Fertility and Family Surveys; the trees, identified separately for men and women in each country, were compared using a modified version of a recently proposed proximity measure, aimed at evaluating whether decisional processes differ between genders and whether they are specific to the nations examined.

In this paper the problem of cell classification using gene expression data is addressed. One of the main features of this kind of data is the very large number of variables (genes), relative to the number of observations (cells). This condition makes most of the standard statistical methods for classification difficult to employ. The proposed solution consists of building classification rules on subsets of genes showing a behavior across the cells that differs most from that of all the other ones. This variable selection procedure is based on suitable linear transformations of the observed data: a strategy resorting to independent component analysis is explored. Our proposal is compared with the nearest shrunken centroid method (Tibshirani et al. (2002)) on three publicly available data sets.

In condition of uncertainty regarding a random phenomenon, particularly in decision making and risk analysis, valuable information may be provided by “experts” in terms of subjective probability distributions for a random variable. In this paper, some ideas about this topic are discussed.

This paper describes an application of Caged (Cluster Analysis of Gene Expression Dynamics) to a data set of gene expression temporal profiles from Saccaromicys Cerevisiae. The goal of the analysis is to identify groups of genes with similar temporal patterns of expression during the cell cycle. We show that Caged can group gene expression temporal profiles into meaningful clusters and identifies novel genes with a putative role in the cell cycle.

Synthetic indices are useful tools used to summarize multivariate information by means of a single value. In the environmental framework these indices are frequently proposed both for measuring pollution and for measuring the associated risks. The drawback of using synthetic indices is, however, that some important information about their components is lost. In this paper we propose to save and interpret these components by transforming them into compositional data. The interpretation of compositions is performed conditionally on the meteorology. In particular, much attention is directed towards explaining the absence of specific pollutants in the index determination.

Centrosymmetric matrices have been recently studied on an algebraic point of view: properties like the existence of the inverse, the expression of the determinant and the eigenspaces characterisation in the case of square matrices have been object of interest. The theoretical results obtained for this class of matrices find applications in many fields of statistics. In this study, we introduce two classes of centrosymmetric matrices that are used in probability calculus and time series analysis, namely, the transition matrices for the classification of states of periodic Markov chains and the smoothing matrices for signal extraction problems.

Methods for comparing and combining classification trees based on proximity measures have been proposed in the last few years. These methods could be used to analyse a set of trees obtained from independent data sets or from resampling methods like bootstrap or cross validation applied to the same training sample. In this paper we consider, as an alternative to the pruning techniques, a modified version of a consensus algorithm we have previously proposed that combines trees obtained by bootstrap samples. This consensus algorithm is based on a dissimilarity measure recently proposed. Experimental results are provided to illustrate, in two real data sets, the performances of the proposed consensus method.

Agro-waste reduction and reuse are among the current main social challenges. In this perspective, the present research was aimed at the complete valorisation of Garganega grape pomace by recovering bioactive phenol extracts and by testing the solid fibre extract residues in composite formulation for packaging applications. The pomace was derived from white wine production, therefore, respect to red pomace, it was promptly removed from must after pressing, and its exploitation can be particularly interesting and valuable as still rich in active compounds. Phenol extracts were obtained both via solvent-based and pressurised liquid extractions and their phytochemical compositions were compared in terms of total amount of phenols, flavonoids, flavanols, anthocyanins, hydroxycinnamic acids, and reducing sugars. Antioxidant activity and detailed phenol profiles were also achieved. The highest phenol yield was obtained via solvent-based extraction with 75% acetone (v/v), solid/liquid ratio 1:5, 2h incubation at 50°C (77.9 gGAeq/kgDW). The fibrous solid residue of the extraction was characterized via thermogravimetric analysis and used for composite preparation by melt mixing with the renewable and biodegradable PHBV polymer through a green approach (solvent-less process). The composites resulted thermally stable at high temperatures, showing initial degradation processes only at temperatures higher than 250°C. Differential scanning calorimetry analyses were carried out to study melting and crystallization phenomena, while mechanical properties were investigated by tensile tests. The materials finally showed properties similar to those of the matrix. The bio-composites can be considered as an alternative to plain PHBV, since they are less expensive and eco-friendlier thanks to a reduced polymeric content, and they could represent a suitable way for full agro-waste exploitation.
