Abstract

Targeting regulatory T cell (Treg) infiltration is an emerging strategy for cancer immunotherapy. However, the efficacy of this strategy in advanced prostate cancer remains unclear. Here, we describe the therapeutic efficacy of this strategy in a canine model of advanced prostate cancer. We used dogs with naturally occurring prostate cancer to study the molecular mechanism underlying Treg infiltration into tumor tissues and the effect of anti-Treg treatment. We found that tumor-infiltrating Tregs were associated with poor prognosis in dogs bearing spontaneous prostate cancer. RNA sequencing and protein analyses showed that Treg infiltration was mediated by interaction between the tumor-producing chemokine, CCL17, and the receptor CCR4 expressed on Tregs. Dogs with advanced prostate cancer responded to mogamulizumab, a monoclonal antibody targeting CCR4, with improved survival and low incidence of clinically relevant adverse events. Exploratory analyses showed urinary CCL17 concentration and BRAFV595E mutation to be independently predictive of the response to mogamulizumab. Analysis of a publicly available transcriptomic dataset of human prostate cancer showed that the CCL17/CCR4 axis correlated with the Treg marker, Foxp3. In silico survival analyses showed that high expression of CCL17 was associated with poor prognosis. Immunohistochemistry confirmed that tumor-infiltrating Tregs expressed CCR4 in human patients with prostate cancer. These findings suggest that anti-Treg treatment through the blocking of CCR4 is a promising therapeutic approach for advanced prostate cancer.One Sentence SummaryTargeting regulatory T cell infiltration by CCR4 blockade induces objective responses and improves survival in a canine model of prostate cancer.

Abstract

Background: Infertility treatments (IFT) have been employed for couples having difficulty conceiving. Previous studies indicated that IFT result in the increased rates of the following conditions：multiple pregnancy, low-birth weight infants, preterm delivery, fetal growth restriction (FGR), and congenital anomalies. All these conditions increase infant morbidity. However, there are few studies examined the data from a single large neonatal intensive care unit (NICU) over a significant time period.Objectives: We attempt to determine the impact of IFT on NICU practice. We retrieved all data of 3,913 infants admitted to Jichi Medical University Hospital NICU for 11 years, then assessed the clinical features and various morbidities of infants.Results: Infants from IFT were approximately 10% of all admissions to NICU. We found a considerable amount of FGR in singletons from IFT, as well as CS, in pregnancies following IFT. Conclusion: There are increasing numbers of infants from IFT, and perinatal intensive care is indispensable for them.

37. Development of the Needle Insertion Robot for Percutaneous Vertebroplasty.

Abstract

Percutaneous Vertebroplasty (PVP) is an effective and less invasive medical treatment for vertebral osteoporotic compression fractures. However, this operative procedure is quite difficult because an arcus vertebra, which is narrow, is needled with accuracy, and an operator's hand is exposed to X-ray continuously. We have developed a needle insertion robot for Percutaneous Vertebroplasty. Its experimental evaluation on the basic performance of the system and needle insertion accuracy are presented. A needle insertion robot is developed for PVP. This robot can puncture with accuracy and an operator does not need to be exposed to X-ray. The mechanism of the robot is compact in size (350 mm × D 400 mm × H270 mm, weight: 15 kg) so that the robot system can be inserted in the space between C-arm and the patient on the operating table. The robot system is controlled by the surgical navigation system where the appropriate needle trajectory is planned based on pre-operative three-dimensional CT images. The needle holding part of the robot is X-ray lucent so that the needle insertion process can be monitored by fluoroscopy. The position of the needle during insertion process can be continuously monitored. In vitro evaluation of the system showed that average position and orientation errors were less than 1.0 mm and 1.0 degree respectively. Experimental results showed that the safety mechanism called mechanical fuse released the needle holding disk properly when excessive force was applied to the needle. These experimental results demonstrated that the developed system has the satisfactory basic performance as needle insertion robot for PVP. [ABSTRACT FROM AUTHOR]

Abstract

When opposing teeth with amalgam and gold restoration are in contact, current flows in the mouth at the instant the dissimilar metals touch. In this study, this condition was simulated by use of resistors and extracted human teeth with amalgam and MOD gold inlay restorations. When both teeth were in contact in a physiological saline solution, we measured current and electrical potential generated in each pulp chamber. Galvanic current generated in the tooth with amalgam was always larger (as much as 18.2 times at the instant of contact) than that in the tooth with gold. Electrical potential generated in the tooth with amalgam was always larger (as much as 9.7 times at the instant of contact) than that in the tooth with gold. It should be emphasized that the larger current generated in the tooth with amalgam was caused mainly by its larger electrical potential. These results correspond well with the clinical phenomenon of galvanic pain, which occurs in the tooth with amalgam rather than in the tooth with gold. [ABSTRACT FROM AUTHOR]

Abstract

Atopic dermatitis (AD) is very common in dogs, but its pathogenesis is not yet fully understood. It has been suggested that a Th2-dominant status may be associated with the occurrence of canine AD. IL-12 is thought to be important for the differentiation of Th1 cells. The IL-12 receptor β2 (IL-12Rβ2) gene is considered to play a critical role in signal transduction and is attracting attention as one of the causative genes of AD in humans. The purpose of this study was to investigate the relationship between IL-12Rβ2 gene expression and canine AD. The canine IL-12Rβ2 gene was cloned by RT-PCR and its nucleotide sequences were determined. Canine IL-12Rβ2 showed 76.8% homology at the amino acid level with human IL-12Rβ2, and its structural motifs were well conserved. cDNA with a 91 bp deletion including the transmembrane region was also cloned, which consequently produced a frame shift and an early stop codon. The deletion region corresponded to exon 14 of the human IL-12Rβ2 gene on chromosome 1. The expression of deleted canine IL-12Rβ2 mRNA in phytohemagglutinin-stimulated peripheral blood mononuclear cells was examined in seven healthy dogs and 11 AD dogs. Both deleted and intact mRNAs were expressed at constant ratios in healthy and AD dogs. The results indicate that the deletion of the transmembrane region is not associated with the occurrence of AD, and that the expression of the deleted mRNA may be constitutive and produced by alternative splicing.
