Während der industriellen Bearbeitung von Stahlblechen kommt es zu einer mechanischen Beanspruchung, die das Reißen des Stahlblechs zur Folge haben kann. Daher wird das Stahlblech vor oder während der Bearbeitung z.B. in Presswerken in der Automobilindustrie mit einem Umformungsöl versehen. Dabei muss die Ölschichtdicke auf dem Stahlblech online überwacht werden, denn bei zu geringen Ölbelegungen ist diese nicht effektiv genug und es kann zu einem Bänderriss kommen. Ist die Schicht zu dick kommt es vor allem in der Weiterverarbeitung des Stahls zu Problemen. Es bilden sich unter diesen Umständen z.B. beim Pressen „Ölbeulen“ aus. Die exakte Ölschichtdickenbestimmung stellt eine große Herausforderung in der Prozessanalytik dar. Der zu erfassende Ölbelegungsbereich liegt üblicherweise zwischen 0,8 g/m² und 2 g/m², was einer Schichtdicke zwischen 0,9 µm und 2,2 µm entspricht. Quantitative prozessanalytische Methoden für die Bestimmung der Ölbelegung müssen daher Bestimmungsgrenzen besitzen, die nicht über 0,8 g/m² liegen. Bisher wird die Analyse hauptsächlich mit Hilfe der Infrarot (IR)-Spektroskopie durchgeführt. Dazu wurden von verschiedenen Herstellern Messgeräte entwickeln, die sowohl kontinuierlich bei der Beölung im Stahlwerk als auch diskontinuierlich in Form von „Handgeräten“ bei der Weiterverarbeitung des Stahls eingesetzt werden können. Mit Hilfe der IR-spektroskopischen Messtechnik kann die Ölschichtdicke mit einer Verfahrensstandardabweichung von ca. 0,15 g/m² bestimmt werden, was für technische Zwecke ausreichend ist. Dieser Wert für die Verfahrensstandardabweichung wurde in der vorliegenden Arbeit als „technische Vorgabe“ auch für die fluorometrische Bestimmung übernommen. Die Beölung geschieht durch Beölungsmaschinen, die das erwärmte Öl in feinen Tröpfchen auf das Blech aufbringen. Dabei entstehen auf dem Stahlblech mikroskopische Öltröpfchen. Diese Öltröpfchen verteilen sich zu einer gleichmäßigen homogenen Schicht, nachdem das Öl bereits im Stahlwerk auf das Blech aufgebracht und anschließend zu einem Coil aufgerollt wurde. Beim Weiterverarbeiten wird das Coil aufgerollt und dabei kann die Ölbelegung wie beschrieben IR-spektrometrisch überwacht werden. Soll allerdings die Primär- oder die Sekundärbelegung mit Öl direkt überwacht werden, so bleiben die Tröpfchen während der Messung erhalten. Beim Vorliegen von Öltröpfchen wird mit der IR-Spektroskopie ein erheblicher Minderbefund ermittelt, sodass eine zuverlässige prozessanalytische Ölschichtdickenbestimmung in solchen Fällen unmöglich ist. Durch den Einsatz von Walzen ist es möglich die Tröpfchenbildung zu reduzieren. Allerdings ist dies mit einem großen Aufwand verbunden. Daher ist es wünschenswert die Ölbelegung auch beim Auftreten von Tröpfchen prozessanalytisch bestimmen zu können. Da es sich bei der Fluoreszenzspektroskopie um eine Emissions- und nicht wie bei der IR-Spektroskopie um eine Absorptionsmethode handelt, ist diese gegenüber Inhomogenitäten und damit möglicherweise auch Tröpfchenbildung robuster. Daher stellt sich die Frage, ob mit Hilfe der Fluoreszenzspektroskopie eine alternative Methode entwickelt werden kann, um die Ölbelegung quantitativ zu bestimmen. Die typischen in der Industrie verwendeten Öle zeigen eine gewisse Eigenfluoreszenz, die hierfür genutzt werden kann. Alternativ kann das Öl mit Fluoreszenzmarkern versehen werden. Sowohl mit natürlichen wie auch mit fluoreszenzmarkierten Ölen wurde daher überprüft, ob es möglich ist, die Ölbelegung mit Hilfe der Fluoreszenzspektroskopie in ausreichender Genauigkeit zu bestimmen und in wieweit neben der Art des Öls auch die Beschaffenheit der Oberfläche der Stahlbleche in die Kalibrierung eingeht. Unterschiedliche Oberflächen ergeben sich z.B. durch unterschiedliche Arten von Verzinkung oder Phosphatierung von Stahlblechen. Alle Untersuchungen wurden zunächst an homogenen Ölschichten durchgeführt. Zum Schluss der Arbeit erfolgten jedoch noch einige Experimente zum Thema Tröpfchenbildung. To protect the surfaces of steel sheets against aging during storage and mechanical stress in the manufacturing process e.g. in automobile industry, the surfaces are normally coated with oil films. It is necessary to control the coverage weight of the oil on the steel sheet to avoid damage due to oil layers which are too thin or thick. The determination of the exact oil coverage in process is a great challenge in process analysis. The used oil coverage is normally between 0.8 g/m² and 2 g/m² corresponding to an oil thickness between 0.9 µm and 2.2 µm. So the limit of quantification for the process analytical control of the coverage weight must not be above 0.8 g/m². Normally the oil layers are measured by use of infrared spectroscopy. For that purpose commercial instruments are available which control the coverage weight continuously on the steel sheets during processing. With use of the infrared technique the obtained analytical error is approximately 0.15 g/m² which is sufficient for technical purposes. So this value should also be achieved with fluorescence spectroscopy. When the oil is applied in the steel mill before the steel sheet is rolled up as a coil or if additional oil is added before further processing of the steel sheets, sometimes oil “droplets” emerge on the steel surface. In this case infrared spectroscopy fails because it shows results which are considerably below the real oil coverage. When rollers are used the droplet forming can be reduced but this is connected with more complexity and higher costs of the process. So it would be desirable to be able to determine the coverage weight exactly even when there are oil droplets on the steel surface. Fluorescence spectroscopy is, in contrast to infrared spectroscopy, not an absorption but an emission technique. Therefore fluorescence spectroscopy should be more robust against microscopic inhomogeneities by “droplet forming”. So, there is the question whether it is possible to apply fluorescence spectroscopy as an alternative method for determining the coverage weight of oil on steel sheets. The typical oils used in steel industry show weak fluorescence which can be used for oil coverage determination. To increase the native fluorescence, fluorescence markers can be added to the oil. In this work it was examined whether it is possible to apply fluorescence spectroscopy as an alternative method for determination of the coverage weight. The results for marked and non marked oils were compared. The influence of the type of oil used and the chemical and physical properties of the steel surface on the fluorescence signal was also examined. At first all experiments were performed with a homogenous oil layer. At the end some experiments with “oil droplets” were performed.

Bei den Alkylphenolethoxylaten (APEO) handelt es um Verbindungen, die aufgrund ihrer amphiphilen Molekülstruktur grenzflächenaktive Eigenschaften aufweisen und zur wirtschaftlich sehr bedeutenden Gruppe der nicht-ionischen Tenside gehören. Sie weisen in der Anwendung als Reinigungsmittel, Emulgatoren, Dispergiermittel, Lösungsvermittler oder Benetzungsmittel ein breites Spektrum auf. Sie werden z. B. in Wasch-, Reinigungs- und Desinfektionsmittel, als Textil- und Lederhilfsmittel, in der Kunststoffindustrie, bei der Farben- und Lackherstellung, als Hilfsmittel für die Zellstoff- und Papierindustrie, als Additiv von Schmierstoffen und Ölen, in Bauchemikalien und als Hilfsmittel in Agrochemikalien verwendet. Da APEO biologisch nicht immer vollständig abgebaut werden und da ihre Abbauprodukte, wie etwa Alkylphenole, Alkylphenolmono- und diethoxylate, Alkylphenolethoxycarboxylate und Carboxyalkylphenolethoxycarboxylate, teilweise persistent sind, eine hohe Neigung zur Bioakkumulation aufweisen und als Xenoöstrogene wirksam sind, wurde ihr Einsatz in einigen Staaten größtenteils verboten bzw. stark eingeschränkt. Kommen APEO in Pflanzenschutzmitteln zum Einsatz, so führt dies zu einer direkten Freisetzung in die Umwelt. Aus diesem Grund gibt es in Europa für die Verwendung in Pflanzenschutzmitteln einen Grenzwert von 0,1 % (w/w) für Nonylphenolethoxylat (NPEO). Bei NPEO handelt es sich neben Octylphenolethoxylat (OPEO) um das bedeutendste APEO. Für ein APEO-Screening solcher Produkte, z. B. bezüglich der Einhaltung des o. g. Grenzwertes, bedarf es geeigneter analytischer Methoden. Die quantitative APEO-Bestimmung in Pestizidformulierungen ist insofern eine komplexe Aufgabenstellung, als dass zum einen die Ethoxymerenverteilung der als Formulierungshilfsmittel verwendeten APEO als auch die Beschaffenheit der Matrix aufgrund unterschiedlicher Formulierungsformen stark variieren kann. Unter Verwendung der Hochleistungsflüssigkeitschromatographie (HPLC) wurde eine Methode entwickelt, die eine Bestimmung von NPEO und OPEO in Pflanzenschutzmitteln und auch anderen Matrices ermöglicht. Es wurden für die unterschiedlichen Formulierungstypen (z. B. Pulver, Granulate, emulgierbare Konzentrate oder Suspensions-konzentrate) und in Abhängigkeit von den weiteren Beistoffen, wie etwa verschiedene Lösemittel, Tenside oder Öle, geeignete Methoden zur Probenvorbereitung (APEO-Extraktion, Matrixisolierung) entwickelt. Es wurde durch eine Kombination der Umkehrphasen-Flüssigkeitschromatographie (RPLC) an C18 und der Normalphasen-Flüssigkeitschromatographie (NPLC) an Aminopropylsilica unter Verwendung von UV-, Diodenarray- und Fluoreszenzdetektoren eine Methode entwickelt, die eine Identifizierung, Charakterisierung und Quantifizierung der APEO in Pestizidformulierungen und anderen Matrices ermöglicht. Mit Hilfe der RPLC ist eine Auftrennung der APEO in OPEO- und NPEO-Fraktionen möglich. Dafür wurden zwei unterschiedliche isokratische RPLC-Methoden entwickelt, die in Abhängigkeit von der Art der Probe zum Einsatz kamen. Außerdem wurde eine HPLC-Methode mit einer online Kopplung einer C8- mit einer C18-Säule verwendet. Die NPLC ermöglicht dann die Auftrennung der APEO-Fraktionen in ihre einzelnen Ethoxymere. Durch die Ermittlung der Ethoxymerenverteilung mittles NPLC wird die Bestimmung von mittleren Molmassen und mittleren Ethoxylierungsgraden ermöglicht. Eine universelle Kalibrierstrategie, bei der sowohl der APEO-Typ (z. B. OPEO oder NPEO) als auch die Ethoxymerenverteilung im verwendeten Standard und in der Probe berücksichtigt wird, diente zur Quantifizierung der APEO in den Pflanzenschutzmitteln und anderen Proben. In Abhängigkeit von der Matrix, also dem Formulierungstyp, sind unterschiedliche Probenvorbereitungsmethoden notwendig. Hierzu wurden unterschiedliche Methoden etabliert. Für die Isolierung von APEO aus Feststoffproben ist die Flüssig/Fest-Extraktion mit Acetonitril, Ethanol oder Methanol unter Verwendung von Schüttelmaschinen, Dispergiergeräten oder mit Hilfe von Ultraschall geeignet. APEO können aus wasserlöslichen oder aus in Wasser dispergierbaren Proben mit Hilfe der Wickbold-Methode durch Ausblasen mit N2 in Ethylacetat aus der wässerigen Phase extrahiert werden. Falls erforderlich, ist ein Clean-up der Extrakte mit Hilfe der Festphasenextraktion (SPE) an Silicagel möglich. Bei Proben, die in unpolaren Lösemitteln wie n-Hexan, Cyclohexan oder Dichlormethan löslich sind, kann mit Hilfe der Flüssig/Flüssig-Extraktion (30 %-ige Essigsäure / n-Hexan oder Acetonitril-Wasser 70:30 / n-Hexan) oder mittels SPE ein Entfernen von störenden Matrixbestandteilen erreicht werden. Alle oben beschriebenen Methoden wurden validiert. Mit Hilfe von Aufstockungsexperimenten wurde deren Richtigkeit überprüft und es konnte gezeigt werden, dass sie ebenfalls für Waschmittel und diverse Öle geeignet sind. Mit Hilfe dieser Methoden ließen sich in acht von zehn untersuchten Pflanzenschutzmitteln APEO in Gehalten von 0,4 bis 15 % (w/w) nachweisen. Diese enthielten zu drei Vierteln NPEO und zu einem Viertel OPEO. Die untersuchten Pflanzenschutzmittel waren mit APEO formuliert, die einen mittleren Ethoxylierungsgrad von 3,6 bis 9,6 aufwiesen. Alkylphenol ethoxylates (APEO) are surface-active agents on account of their amphiphilic molecular structure. APEO belong to the group of non-ionic surfactants of large economic relevance. They show a broad spectrum of different applications as cleaners, emulsifiers, dispersing agents, solubilizers and wetting agents. APEO are used in detergents, cleaning and disinfecting agents, as adjuvants in textile and leather production, during polymerisation processes, as dyeing aids, in varnishes, as adjuvants in pulp and paper manufacture, as additives in lubricants and oils, in construction chemicals, and as adjuvants in agrochemicals. The prohibition or limitation of APEO in some countries is a result of several studies showing that under some conditions APEO biodegradation is only poor and leads to persistent, bioaccumulable and xenoestrogenic metabolites, e.g. alkylphenols, alkylphenol mono- and diethoxylates, alkylphenol ethoxycarboxylates, and carboxyalkylphenol ethoxycarboxylates. The formulation of pesticides with APEO leads to a direct release of APEO into the environment. Therefore, a limiting value of 0.1 % (w/w) nonylphenol ethoxylate (NPEO) in pesticides was laid down in Europe. NPEO is the most important APEO beside OPEO. There is a demand for analytical methods suitable for an APEO screening in such products, e.g. with regard of above mentioned limiting value. The quantification of APEO in pesticides is a complex task because of a varying APEO ethoxymer distribution and diversity of matrix components in different formulation types. An analytical method suitable for the determination of NPEO and OPEO in pesticides and other matrices using high-performance liquid chromatography (HPLC) was established. With regard to the differences of the formulation types (e. g. powders, granulates, emulsifiable concentrates or suspension concentrates) and considering the presence of other adjuvants (solvents, surfactants or oils) in the pesticide different suitable sample preparation methods for APEO extraction or matrix isolation had been developed. Combining reversed phase liquid chromatography (RPLC) using a C18 column with normal phase liquid chromatography (NPLC) using an aminopropyl silica column an analytical method was developed for the APEO identification, characterization and quantification in pesticides and other matrices using UV, diode array and fluorescence detectors. By means of RPLC a separation of OPEO and NPEO is possible. Two different isocratic RPLC methods suitable for different sample types were established. Additionally, an online LC-LC coupling of a C8 column and a C18 column was used for this purpose. Then, NPLC enables ethoxymer separation of the different APEO fractions. The determination of the ethoxymer distribution by NPLC is used for the calculation of mean molecular masses and mean degrees of ethoxylation. Quantification of APEO in pesticides and other samples was performed by means of a universal calibration procedure considering APEO type (OPEO or NPEO) and ethoxymer distribution of the used standards and analysed samples. Depending on the matrix of the formulation different sample preparation methods are necessary. Therefore, various methods had been established. APEO can be extracted with acetonitrile, ethanol or methanol from solid samples by solid/liquid extraction using a shaker, a dispersing apparatus or ultrasonication. Solvent sublation according to the Wickbold procedure is suitable for APEO extraction of water soluble and in water suspended samples. If necessary clean-up of the extracts can be achieved by solid phase extraction (SPE) using silica. Are samples soluble in non-polar solvents like n-hexane, cyclohexane and dichloromethane interfering matrix compounds can be removed either by means of liquid/liquid extraction (30 % acetic acid / n-hexane or acetonitrile-water 70:30 / n-hexane) or by SPE using silica. The above mentioned methods had been validated, and via spiking experiments accuracy was proofed. It was shown that the methods are also suitable for detergents and various oils. In eight of ten analysed pesticides APEO were detected in a range from 0.4 to 15 % (w/w). 75 % of them contain NPEO, 25 % OPEO. The analysed pesticides had been formulated with APEO of an average degree of ethoxylation from 3.6 to 9.6.

In dieser Arbeit werden zwei spektroskopische Techniken, die Nahinfrarot- (NIR-) und die Ramanspektroskopie, für fünf verschiedene Anwendungen vergleichend untersucht. Dazu kommen Applikationen zur Anwendung, bei denen verschiedene pharmazeutische Präparate sowohl hinsichtlich qualitativer als auch quantitativer Aspekte analysiert werden. Die beiden schnellen, zerstörungsfreien und dadurch kosteneffizienten spektroskopischen Methoden unterliegen dabei einer Gegenüberstellung mit den verhältnismäßig zeitaufwendigen, kostenintensiven und destruktiven chromatographischen Verfahren. Die Untersuchungen behandeln zunächst die Identitätsbestimmung weißer Dragees. Anhand von Produkten verschiedener Hersteller bzw. verschiedener Produktionsstandorte wird die Praktikabilität der beiden spektroskopischen Methoden gegenübergestellt. Bei der zweiten Anwendung, für die die NIR- und Ramanspektroskopie als zu vergleichende Methoden zum Einsatz kommen, handelt es sich um die Quantifizierung des Wirkstoffgehaltes in pharmazeutischen Wafern. In diesem Zusammenhang wird auch die Übertragung der Ergebnisse auf Marktpräparate überprüft. Ein dritter Anwendungsbereich befasst sich mit der Fragestellung, in wie weit die NIR- und Ramanspektroskopie in der Lage sind, den Nikotingehalt in transdermalen therapeutischen Systemen zu bestimmen. Auch hier wird die Übertragbarkeit der Untersuchungsmethoden und der Ergebnisse auf Marktpräparate beleuchtet. In einer weiteren Anwendung werden NIR- und Ramanspektroskopie als mögliche Techniken zur Gehaltsbestimmung des Wirkstoffs in Glucobay® -Tabletten geprüft. Dazu wird ein alternatives Vorgehen basierend auf der Vermessung von Mischungsreihen mit entsprechenden Hilfs- und Wirkstoffen gewählt. Die fünfte Anwendung umfasst abschließend die Untersuchung von Arzneimittelfälschungen mittels NIR- und Ramantechnik mit dem Ziel der schnellen und zerstörungsfreien Unterscheidung gefälschter Arzneimittel von Originalpräparaten. In diesem Zusammenhang erfolgt ein Vergleich der Leistungsfähigkeit zwischen tragbaren NIR- und Ramangeräten einerseits und Laborgeräten andererseits. Aufgrund der vorliegenden Ergebnisse läßt sich feststellen, dass sowohl die NIR- als auch die Ramanspektroskopie für eine Vielzahl der untersuchten Anwendungen geeignet ist, insbesondere auch für solche, bei denen diese beiden Techniken bisher nur im geringen Umfang oder noch gar nicht eingesetzt wurden. Die beiden Techniken können somit als effiziente Alternativen zu etablierten Analysemethoden in Betracht gezogen werden. This thesis compares two spectroscopic techniques, Near infrared (NIR) and Raman spectroscopy considering five different applications. Both qualitative and quantitative aspects of diverse pharmaceutical drug formulations are investigated. The two fast, non-destructive and therefore cost-effective spectroscopic techniques are thereby competing with relatively time-consuming, cost-intensive and destructive chromatographic methods. The first investigations in this work include the identification of white sugar coated tablets. For this purpose the feasibility of the two spectroscopic techniques is compared by examining pharmaceuticals produced by different manufacturers and at varying production sites, respectively. The second application opposing the NIR to the Raman spectroscopy focuses on the quantification of drug content in pharmaceutical wafers which are also called `thin strips´. Beyond that the transferability of the results to commercial products available on the market is investigated. The third application area deals with the question whether NIR and Raman spectroscopy are suitable to determine the content of Nicotine in pharmaceutical patches. Analogously, the transferability of the methods and results to market products is analyzed. Furthermore NIR- and Raman spectroscopy are tested concerning their feasibility to determine the drug content in Glucobay® tablets. For this purpose an alternative approach is chosen based on analysis of corresponding mixtures of excipients and drug content. Finally, the fifth application implies examinations of counterfeit drugs in order to rapidly and non destructively distinguish fake pharmaceuticals from original drug products. In this regard the capability of portable NIR and Raman devices is analyzed in contrast to laboratory instruments. By means of the five cases studies it is demonstrated that both NIR and Raman spectroscopy show their general suitability for a multitude of the examined applications especially for those for which these techniques were rarely or not yet applied. Therefore the two techniques can be considered as efficient alternatives to established analytical methods.

Alle organischen und anorganischen Verbindungen meisten charakteristische Infrarotspektren. Daher IR-Spektroskopie kann als ein effektives Werkzeug für die Qualitätskontrolle verwendet werden. Mittleren und Nahen Infrarot mit moderner Instrumentierung und Zubehör wie Diamant-ATR-und diffuser Reflexion Zubehör liefern Spektren, die gut reproduzierbar machen es möglich, auch kleinere spektrale Abweichungen durch Verunreinigungen verursacht werden. In dieser Arbeit wird der Fokus auf Qualitätskontrolle in der chemischen Verfahrenstechnik Umgebungen, in denen die Reinheit der Materialien zu bestimmen mehr oder weniger automatisch. Die Korrelation zwischen dem Spektrum eines potentiell kontaminierten Probe und der Referenz-Spektrum des entsprechenden reinen Verbindung kann als Maß für die Reinheit in Bezug auf den Korrelationskoeffizienten r verwendet. R ist durch Regression der Probe Spektrums auf dem Referenzspektrum erhalten. Eine Simulation zeigte, dass es vorteilhaft, r zu verwandeln ist Fisher's z-Koeffizienten, da z ist viel besser als r geeignet für die Erkennung kleiner spektrale Abweichungen durch Verunreinigungen verursacht. Auf dieser Grundlage zwei spektrale Reinheit Parameter SPR_1 und SPR_2 wurden durch Multiplikation der Korrelationskoeffizient bzw. erhalten. die normierte z-Koeffizienten mit 100. Als zweite Möglichkeit für die Entdeckung Verunreinigungen eine Methode der dynamischen Differenz-Spektroskopie wurde entwickelt, um den die Differenz zwischen dem Spektrum der Probe und der Referenz-und die entsprechende Differenz Faktor werden automatisch berechnet. Als Parameter spektrale Reinheit der erhaltenen Differenz Faktor selbst, SPR_3 und alternativ das Integral der Differenz Spektrum, SPR_4, verwendet werden. Die Methoden, die auf lineare Regression basiert erwies sich wirksamer und Nachweisgrenzen von Verunreinigungen nach unten bis 0,002 g/100 g für Flüssigkeiten und 0,03 g/100 g für feste Stoffe konnte erreicht werden kann. Des Weiteren konnte gezeigt werden, dass durch Verwendung eines Quantum Cascade Laser Spektrometer statt eines FT-IR-Instrument noch niedrigere Nachweisgrenzen erreicht werden kann. All organic and most inorganic compounds show characteristic Infrared spectra. Therefore IR Spectroscopy can be used as an effective tool for quality control. Middle and Near Infrared with modern instrumentation and accessories like Diamond ATR and Diffuse Reflectance accessories, provide spectra which are highly reproducible making it possible to detect even minor spectral deviations caused by impurities. In this thesis the focus is on quality control in chemical process environments where the purity of materials has to be determined more or less automatically. The correlation between the spectrum of a potentially contaminated sample and the reference spectrum of the corresponding pure compound can be used as measure for purity in terms of the correlation coefficient r. r is obtained by regressing the sample spectrum on the reference spectrum. A simulation study showed that it is advantageous to transform r to Fisher's z-coefficient because z is much better suited than r for detecting small spectral deviations caused by impurities. On this basis two spectral purity parameters SPR_1 and SPR_2 were obtained by multiplying the correlation coefficient resp. the normalized z-coefficient with 100. As a second way for discovering impurities a method of dynamic difference spectroscopy was developed, by which the difference between the spectrum of the sample and the reference and the corresponding difference factor are calculated automatically. As spectral purity parameters the obtained difference factor itself, SPR_3, and alternatively the integral of the difference spectrum, SPR_4, are used. The methods based on linear regression proved to be more effectual and detection limits of impurities down to 0.002 g/100 g for liquids and 0.03 g/100 g for solids could be achieved. Further it could be shown that by using a Quantum Cascade Laser spectrometer instead of a FT-IR instrument still lower limits of detection can be attained.

Daher IR-Spektroskopie kann als ein effektives Werkzeug für die Qualitätskontrolle verwendet werden. Mittleren und Nahen Infrarot mit moderner Instrumentierung und Zubehör wie Diamant-ATR-und diffuser Reflexion Zubehör liefern Spektren, die gut reproduzierbar machen es möglich, auch kleinere spektrale Abweichungen durch Verunreinigungen verursacht werden. In dieser Arbeit wird der Fokus auf Qualitätskontrolle in der chemischen Verfahrenstechnik Umgebungen, in denen die Reinheit der Materialien zu bestimmen mehr oder weniger automatisch. Die Korrelation zwischen dem Spektrum eines potentiell kontaminierten Probe und der Referenz-Spektrum des entsprechenden reinen Verbindung kann als Maß für die Reinheit in Bezug auf den Korrelationskoeffizienten r verwendet. R ist durch Regression der Probe Spektrums auf dem Referenzspektrum erhalten. Eine Simulation zeigte, dass es vorteilhaft, r zu verwandeln ist Fisher's z-Koeffizienten, da z ist viel besser als r geeignet für die Erkennung kleiner spektrale Abweichungen durch Verunreinigungen verursacht. Auf dieser Grundlage zwei spektrale Reinheit Parameter SPR_1 und SPR_2 wurden durch Multiplikation der Korrelationskoeffizient bzw. erhalten. die normierte z-Koeffizienten mit 100. Als zweite Möglichkeit für die Entdeckung Verunreinigungen eine Methode der dynamischen Differenz-Spektroskopie wurde entwickelt, um den die Differenz zwischen dem Spektrum der Probe und der Referenz-und die entsprechende Differenz Faktor werden automatisch berechnet. Als Parameter spektrale Reinheit der erhaltenen Differenz Faktor selbst, SPR_3 und alternativ das Integral der Differenz Spektrum, SPR_4, verwendet werden. Die Methoden, die auf lineare Regression basiert erwies sich wirksamer und Nachweisgrenzen von Verunreinigungen nach unten bis 0,002 g/100 g für Flüssigkeiten und 0,03 g/100 g für feste Stoffe konnte erreicht werden kann. Des Weiteren konnte gezeigt werden, dass durch Verwendung eines Quantum Cascade Laser Spektrometer statt eines FT-IR-Instrument noch niedrigere Nachweisgrenzen erreicht werden kann.

Es wurden PVC-Sorten unterschiedlicher Herkunft und diverse bei der PVC-Verarbeitung verwendete Additive infrarotspektroskopisch untersucht und eine Methode für die Identitätskontrolle entwickelt. Als Messtechniken wurden im Mittleren Infrarot die Diffuse-Reflexions- und die Diamant-ATR-Technik und im Nahen Infrarot die Reflexions-, die Transmissions- und die Transflexionstechnik verwendet. Nach der Untersuchung der unterschiedlichen Stoffklassen mit dem Mittleren und dem Nahen Infrarot auf Basis einer Clusteranalyse mit Hilfe der PCA-Methode wurde das Mittlere Infrarot für die Entwicklung der Identitätskontrollmodelle ausgewählt. Für das Erstellen derselben wurde die Compare-Methode verwendet, wobei mit Hilfe der Korrelationskoeffizienten die Ähnlichkeit zwischen den Spektren berechnet wurde. Als Ergebnis stellte sich heraus, dass sich die spektrometrische Identitätskontrolle im Mittleren Infrarot auf die bei der PVC-Verarbeitung auftretenden Produkte erfolgreich anwenden lässt. Dabei wurde das im Hinblick auf die Probenpräparation relativ einfache Verfahren der Diamant-ATR-Technik verwendet. Darüber hinaus kam in dieser Arbeit für die Identitätskontrolle von PVC-Pulvern erstmals das Verfahren der diffusen Reflexion im Mittleren Infrarot zur Anwendung. PVC of different origin and various additives used for PVC processing have been examined by means of infrared spectroscopy and a method suitable for identity control has been developed. As measurement techniques diffuse reflection and Diamond ATR have been employed for the Middle Infrared (MIR); reflection, transmission and transflection have been used for the Near Infrared (NIR) respectively. Results of a cluster analysis based on a Principal Component Analysis (PCA) pointed towards the MIR as most suitable spectral region compared to the NIR for the development of an identity control scheme. The so called Compare Method has been used to develop the identity control scheme, whereby the similarity between a sample and a reference spectrum was determined by calculating the correlation coefficient. Our spectroscopically based identity control scheme can be successfully applied to all products applied in PVC processing, like plasticizers, inorganic additives and bonding agents. As sample preparation the relatively simple Diamond ATR technique has been used. Furthermore, diffuse reflection in the MIR has been applied for the first time for the identity control of PVC powders.

Duisburg, Essen, Univ., Diss., 2005 Pharmaceutical adjuvants or excipients are important ingredients of medicaments. These substances are used as support and for building the matrix for the active ingredients. In this way, the active ingredients will be realised in accurately and defined way at the specific site of action. In this study, the most important pharmaceutical adjuvants are characterised and analysed via near infrared-spectroscopic methods. The powdery samples were measured by means of diffuse reflectance with six NIR-spectrometer. The obtained NIR-spectra were used to build a near infrared spectra library based on SpecInfo systems of the firm Chemical Concepts. Furthermore, discrimination analysis were carried out, while reference spectra were compared with the database for performing an identity control.

Spectroscopical procedures are well proven over years analysing solids, fluids and gaseous substances because of their high selectivity, sensitivity and their short response time. They allow continuous and non-destructive measurements wherefore the nondispersive spectroscopy (NDIR) is well locatet in the field of industrial gas analysis. In nondispersive spectrometers which were exclusively used for gas analysis no monochromator or modulator is used but selective detektors. Those detectors, nearly all of them are opto-pneumatical, are filled with the gas which should be measured. In the context of my work it was tried for the first time to analyse a mixture of two gaseous compounts with only one opto-pneumatical detector. For the two choosen mixtures of CO/CO2 und SO2/H2O it was tried to develop a new analysis method for each component while taking spactal data from databases, simulation tools and chemometrical, i.e. mathematical-statistical models. Therefore not only the intensity of the signals were used but the phase of the signals, too. Taking pure gas data calculative mixtures has been build. Afterwards they have been analysed by computer sumulated calculations. Based on these raw data calibration models were build and validated by pure gas and mixtures. In the available work the proof could successfully be made that also the NDIR spectroscopy in combination with chemometric can be used. The application possibilities cover the building of quantitative calibration models for the gas components which can be determined, as well as the determination of the filling gas condition of the detector. Thus could be pointed out in the form of simulation calculations for the first time, how by consideration of signal phases and amplitudes a genuine two-component analysis with a non-dispersive procedure can be accomplished.

Due to legal requests in future the identity control of products will not only be required in pharmaceutical industry but also in chemical industry. Therefore the demands on the analytical methods are very high. The company Merck realized an identity control tool by combining NIR and Raman spectroscopy. This Tool is suited for identification of a broad range of organic and inorganic substances. In this work different chemometric methods and data pretreatments were tested. The NIR and the Raman library have been optimized and the validation results visualized. Alternatives as binary coding and wavelet transformation, which lead to data compression at the same time, were tested

In most cases chemometric methods (PCA, PCR, PLS) are used for data modelling when NIR-spectrometry is applied for analytical purposes. This work is about the application of the wavelet-transform as a tool in chemometrics for analysing NIR-spectra. Beside aspects of data pretreatment, like smoothing of NIR-spectra, and data compression the focus was set on NIR-analysis using wavelet-transformed NIR-spectra. A new method for building multivariate calibration models based on wavelet-coefficients selected by a genetic algorithm is introduced. In addition the results obtained by calibrating wavelet-coefficients using PCR and PLS are descriped. Furthermore the identification of post consumer plastic waste by wavelet-transformed NIR-spectra and an artificial neural network is demonstrated. Two new special types of wavelets which allow analysing finitely extended signals (like spectra) without introducing artifacts near the boundaries were applied

In der vorliegenden Dissertation werden anodische Oxidschichten auf Aluminium mit Hilfe der FT-IR-Spektrometrie charakterisiert. Hierbei werden die Methoden der gerichteten Reflexion und der FT-IR-Ellipsometrie eingesetzt. Der Schwerpunkt der Untersuchungen liegt auf der physikalisch-chemischen Charakterisierung der anodischen Schichten. Es wird ein optisches Modell entwickelt auf dessen Basis die optischen Konstanten, der Brechungsindex n und der Absorptionskoeffizient k, der anodischen Schichten ermittelt werden. In the present thesis anodic layers on Aluminium are characterised by FT-IR-Spectrometry. FT-IR-Ellipsometry and specular reflection and are used. The main emphasis is put on the physical-chemical characterisation of the anodic layers. An optical model is developed. On the basis of this model the optical constants, the refractive index n and the absorption coefficient k, are determined.
