Background: The production of recombinant proteins in mammalian cell lines is one of the most important areas in biopharmaceutical industry. Viral transcriptional promoters are widely used to express recombinant proteins in mammalian cell lines. However, these promoters are susceptible to silencing, thus limiting protein productivity. Some CpG islands can avoid the silencing of housekeeping genes; for that reason, they have been used to increase the production of recombinant genes in cells of animal origin. In this study, we evaluated the CpG island of the promoter region of the v-actin gene of Cricetulus griseous (Chinese hamster), associated to the Cytomegalovirus (CMV) promoter, to increase recombinant antibodies production in Chinese Hamster Ovary (CHO) cells. Results: We focused on the non-coding region of CpG island, which we called RegCG. RegCG behaved as a promoter, whose transcriptional activity was mainly commanded by the CAAT and CArG boxes of the proximal promoter. However, the transcription started mainly at the intronic region before the proximal transcription start site. While the CMV promoter was initially more powerful than RegCG, the latter promoter was more resistant to silencing than the CMV promoter in stable cell lines, and its activity was improved when combined with the CMV promoter. Thereby, the chimeric promoter was able to maintain the expression of recombinant antibodies in stable clones for 40 days at an average level 4 times higher than the CMV promoter. Finally, the chimeric promoter showed compatibility with a genetic amplification system by induction with methotrexate in cells deficient in the dihydrofolate reductase gene. Conclusions: We have generated an efficient synthetic hybrid transcription promoter through the combination of RegCG with CMV, which, in stable cell lines, shows greater activity than when both promoters are used separately. Our chimeric promoter is compatible with a genetic amplification system in CHO DG44 cells and makes possible the generation of stable cell lines with high production of recombinant antibodies. We propose that this promoter can be a good alternative for the generation of clones expressing high amount of recombinant proteins, essential for industrial applications.

La pubertad es el período final del crecimiento y maduración del niño en el que se alcanza la talla final y la madurez psicosocial y reproductiva. Se inicia en el sistema nervioso central por factores neuroendocrinos que activan el eje hipotálamo-hipofisario-gonadal y sus mecanismos reguladores. Estos factores actúan bajo control genético e influencia ambiental. La pubertad precoz se define como la aparición progresiva de signos puberales a una edad por debajo de 2,5 DS de la media para una población determinada. Puede ser dependiente de gonadotropinas (central) o independiente de gonadotropinas (periférica). La mayoría de los casos son de origen idiopático, aunque también puede estar causada por enfermedades orgánicas de severidad variable. Por la repercusión desfavorable que tiene en la maduración del niño se recomienda tratar adecuadamente. Puberty refers to the final stage of children's growth and development in which adult height, reproductive and psychosocial maturity are reached. It is initiated in the central nervous system by neuroendocrine factors (under genetic control and environmental influence) that activate the hypothalamic- pituitary-gonadal axis and its regulatory mechanisms. Precocious puberty is defined as the progressive onset of pubertal signs at an age more than 2.5 SD below the mean of a population. It may be gonadotropin-dependent (central) or gonadotropin-independent (peripheral). Most of the cases are idiopathic, but occasionally the cause may be an underlying organic disease with variable severity. Treatment is recommended because of the negative effects it may have on the child's development.

Se presentan los resultados resumidos de tres trabajos realizados para conocer el papel que juega el hongo Phytophthora cinnamomi en el desarrollo de plántulas de encina (Quercus ilex) y alcornoque (Quercus suber). Se realizaron dos prospecciones para detectar la presencia de P.cinnamomi en focos de seca en dehesas de encina y alcornoque de Extremadura en 1991-1992 y 1999-2000. El porcentaje de focos en los que se detectó la presencia de P. cinnamomi ha permanecido relativamente estable entre las dos prospecciones. También se determinó la tasa de crecimiento relativo para nueve aislados de P. cinnamomi a diferentes temperaturas, observando una elevada variabilidad tanto en la temperatura óptima de crecimiento como en el valor de la propia tasa diaria de crecimiento máxima. También se estudió la emergencia y supervivencia de plántulas de encina y alcornoque procedentes de dos dehesas ecológicamente diferentes, cultivadas en condiciones controladas en suelos de estas dehesas infectados naturalmente con P. cinnamomi y en estos mismos suelos previamente desinfectados en autoclave. Un factor limitante en los procesos de regeneración natural y en las repoblaciones por siembra directa de encinas y alcornoque puede ser su alta susceptibilidad a las infecciones por P. cinnamomi.Por último, se estudió durante dos años consecutivos, la supervivencia de las plantas de encina de una repoblación sobre un suelo naturalmente infectado con P. cinnamomi en la provincia de Badajoz (SO de España). Los resultados obtenidos se discuten teniendo en cuenta el déficit hídrico, los daños por P. cinnamomi, la competencia con otras especies y las posibles medidas sanitarias que amortigüen los daños en las repoblaciones con encinas._______________________________Three different studies were performed with the aim to determine the role of Phytophthora cinnamomi in the early development of seedlings of holm oak (Quercus ilex) and cork oak (Quercus suber). Two surveys, for the presence of Phytophthora cinnamomi in declining holm oak and cork oak stands in Extremadura (SW Spain) were performed in 1991-1992 and 1999-2000. The percentage of decline foci where P. cinnamomi is envolved has remained relatively stable in the lapse of time between the two surveys. Growth rates were determined for 9 P. cinnamomi isolates. Considerable variation in optimum growth temperatures and in daily growth rates occurred among isolates. The emergence and survival of pregerminated holm oak and cork oak acorns from two ecologically different dehesas (Mediterranean open woodlands) were studied in two soils from these stands naturally infected with P. cinnamomi, and in the same soils previously sterilized in the autoclave. The demonstrated high susceptibility of holm and cork oak young seedlings to P. cinnamomi could be a limiting factor in mediterranean opend woodlands (dehesas) not only in natural regenerationprocesses but also when reforestation by direct sowing is implemented. Finally, the viability of 1 year old holm oak seedlings in a soil naturally infected with P. cinnamomi was studied during two consecutive years in a plot located in SW Spain. Results obtained are discussed in the framework of seasonal water deficit, P. cinnamomi damage, weed competition and sanitation thechniques to be used in declined holm oak stands in Spain.

[Resumen] Este artículo tiene como objetivos, analizar las causas principales de las grandes quemaduras, de los pacientes ingresados en el Instituto del Quemado de Córdoba-Argentina, en el periodo de tiempo comprendido entre Enero de 2000 y Enero de 2001. Además de analizar la evolución de los autoinjertos, complicaciones más frecuentes, trastornos psiquiátricos asociados, morbimortalidad registrada, etc. presentamos una propuesta de tratamiento de fisioterapia adecuado al tipo y/o profundidad de la(s) quemadura(s). Para ello, se ha analizado una muestra de 100 historias clínicas, de pacientes ingresados en el Instituto del Quemado de Córdoba-Argentina, durante el periodo de tiempo antes señalado, y se ha realizado una revisión bibliográfica exhaustiva, que nos ha sido de utilidad para fundamenta la propuesta de tratamiento. [Abstract] The aims of this article is to analyze the mains reasons of the important burns in the patients being admitted to the Burned people Institute in Cordoba, Argentina between January of 2000 and 2001 year. We explain not only the progress of the skin graft, the more common complications, the associated psyquiatric problems, the registred morbility, etc... but also the plan of treatment suitable for the type and the deep of the burn. We have analyze a sample of 100 case histories related to admitted patients to the Burned people Institute in Cordoba, Argentina. After that, we have made a comprehensive bibliographic cheeking, which has been very useful to base the protocol of the treatment.

[ESP] Los tratamientos insecticidas, formando parte o no de programas de control integrado, siguen siendo actualmente imprescindibles para el control de Helicoverpa armigera sobre resistencia a insecticidas en España es prácticamente inexistente, a pesar de que la plaga ostente a nivel mundial una probada historia. Intentando paliar esta situación, durante las campañas 1995-1998 se investigó la resistencia a insecticidas de H. armigera en dos de las comunidades españolas de mayor transcendencia hortícola, Extremadura y Murcia. Se efectuaron en laboratorio bioensayos toxicológicos sobre la progenie de poblaciones de campo (18 extremeñas y 8 murcianas) colectadas sobre diversos cultivos o con trampas de luz. Se probaron hasta 20 materias activas (2 clorados, 4 carbamatos, 7 fosforados y 7 piretroides) mediante bioensayos tópicos sobre orugas de tercer estadio. Por cada población e insecticida se trataron al menos 120 orugas, con 3 (eventualmente 2) repeticiones de 10 orugas por dilución y un mínimo de 5 (eventualmente 4) diluciones, obteniendo un rango de mortalidad en torno al 0-100%. Los datos se analizaron mediante análisis Probit, estimando entre otros parámetros la DL50 y el factor de resistencia (FR = DL50 población campo / DL50S población susceptible). Se utilizó una estimación toxicológica técnica de la DL50S, calculada en función de la dosis máxima recomendada de cada insecticida. El propoxur no fue efectivo en ninguna población a dosis tan elevadas como las ensayadas (hasta 8 (Ag/oruga L3). En Extremadura se detectaron resistencias moderadas (FR = 5x-10x) a endosulfán, metamidofos, triclorfón, monocrotofos y fenvalerato; elevadas (10x-20x) a carbaril, fenitrotión, metil-azinfós, cipermetrín, ciflutrín y lambda-cihalotrín; y muy elevadas (>20x) a lindano y deltametrín. En Murcia, la situación fue aún más adversa, detectándose resistencias moderadas (FR = 5x-10x) a clorpirifos, fenitrotión, triclorfón, monocrotofos, bifentrín, ciflutrín, permetrín y fenvalerato; elevadas (10x-20x) a endosulfán y lambda-cihalotrín; y muy elevadas (>20x) a lindano, carbaril, metil-azinfós, cipermetrín y deltametrín. Los resultados indican que H. armigera presenta actualmente en España resistencias preocupantes a varios insecticidas que, en el peor de los casos, pueden derivar en fallos de control en campo. [ENG] Insecticide treatments, included or not in IPM programs, are currently indispensable for the control of Helicoverpa armígera Hübner, particularly in processing tomato crops. However, information about insecticide resistance of H. armígera in Spain is practically absent, in spite of the fact that this pest presents at the world level a proven history. To mitigate this undesirable situation, the insecticide resistance status of H. armígera was investigated during 1995-1998 in two of the most important horticultural regions of Spain, Extremadura (SW) and Murcia (SE). Toxicological bioassays were completed in the laboratory on the Fl offspring of field-derived populations (18 from Extremadura and 8 from Murcia) collected from several crops (specially tomato) or with light traps. Until 20 insecticides (2 organochlorines, 4 carbamates, 7 organophosphorous and 7 pyrethroids) were tested per population. Appropriate serial dilutions were prepared and 2 fj.1 of solution were applied to the thoracic dorsum of each third instar larvae. At least 120 larvae of each population were treated per insecticide, usually 3 replicates of 10 larvae at each of 4-5 or more insecticide concentrations, within a 0-100% mortality range. Dose-mortality regressions and LD50 were computed by Probit analysis and the resistance factors estimated (RF = LD50 of collected population / LD50S of susceptible population). A technical- toxicological estimation of the LD50S was used, as a function of the maximal dose recommended for each insecticide. Propoxur was not effective against H. armígera despite the high doses bioassayed (0.5 to 8 u.g/3rd instar larvae). In Extremadura, moderate insecticide resistance (RF = 5x-10x) was detected to endosulfan, methamidofos, trichlorphon, monocrotophos and fenvalerate; high resistance (10x-20x) to carbaryl, fenitrothion, azinphos-methyl, cypermethrin, cyfluthrin and lambda-cyhalothrin; and very high resistance (>20x) to lindane and deltamethrin. In Murcia, the situation was yet more adverse, being detected moderate resistance (RF = 5x-10x) to chlorpyrifos, fenitrothion, trichlorphon, monocrotophos, bifenthrin, cyfluthrin, permethrin and fenvalerate; high resistance (10x-20x) to endosulfan and lambda-cyhalothrin; and very high resistance (>20x) to lindane, carbaryl, azinphosmethyl, cypermethrin and deltamethrin. The results indicate that in Spain H. armígera currently presents worrying resistances to an array of insecticides that, at worst, could determine field control failures. Agradecemos a los Drs. J. M. [ESP] Con el objetivo de caracterizar las poblaciones de Pythium spp. de suelos agrícolas y forestales de Cáceres se muestrearon un total de 18 suelos localizados en esta provincia: 14 suelos agrícolas (6 de plantaciones de cerezo, 2 de espárrago, 4 de tabaco y 2 de hortícolas) y 4 forestales (2 de pinar y 2 de robledal). El género Pythium estuvo presente en 17 de las 18 muestras de suelo estudiadas (excepto en una correspondiente a un suelo de cultivo de tabaco). Las densidades de población, estimadas según el método de las Tasas de Contaminación, oscilaron entre 0,9 y 14,1 propágulos/g de suelo en las muestras en las que se aisló Pythium spp. El género Phytophthora De Bary no se detectó en ninguna de las muestras. Se estudiaron taxonómicamente un total de 73 aislamientos de Pythium spp. procedentes de 16 de las muestras analizadas: 46 se identificaron como P. mamillatum Meurs, 9 como P. irregulare Buisman, 1 como P. torulosum Coker y Patterson y 1 como P. dissimile Vaartaja; 16 de los aislamientos no pudieron ser identificados específicamente, debido a la imposibilidad de observar las estructuras necesarias para ello. Mediante inoculaciones sobre pepino se comprobó el poder patógeno de 55 aislamientos de Pythium spp.: 29 de los 30 aislamientos de P. mamillatum inoculados presentaron índices de patogeneicidad (IP) del 100%, independientemente del tipo de suelo, agrícola o forestal, del que procedían. Los 7 aislamientos de P. irregulare, procedentes de suelos agrícolas, presentaron IP del 100%. Los aislamientos de P. torulosum y P. dissimile, ambos de un suelo de espárrago, presentaron IP bajos (10% y 17,5% respectivamente). Los 16 aislamientos no identificados específicamente, procedentes de suelos agrícolas y forestales, presentaron un comportamiento patogénico heterogéneo con IP desde 0% hasta 100%, y es probable que este grupo de aislamientos estuviera formado por varias especies de Pythium. [ENG] In order to characterize Pythium spp. populations of agricultural and forest soils of Cáceres (South-Western Spain), 18 soils were sampled: 14 agricultural soils (6 of cherry tree orchards, 2 of asparagus, 4 of tobacco and 2 of horticultural crops) and 4 of forest soils (2 of pine and 2 of oak). The genus Pythium was recovered from 17 of the 18 soil samples studied (except from one sample of tobacco soil). Population densities varied between 0,9 and 14,1 propagules/g of soil in the soil samples where Pythium spp. was present. The genus Phytophthora De Bary was not recovered from any of the sampled soils. Seventy-three Pythium spp. isolates from 16 of the analyzed soil samples were studied: 46 were identified as P. mamillatum Meurs, 9 as P. irregulare Buisman, 1 as P. torulosum Coker y Patterson and 1 as P. dissimile Vaartaja; 16 isolates could not be specifically identified because the structures necessary for taxonomy were not observed. Pathogenicity of 55 Pythium spp. isolates were studied inoculating cucumber plants: 29 of the 30 P. mamillatum isolates showed pathogenicity indexes (PI) of 100%, regardless of the soil which they were isolated from. The 7 P. irregulare isolates, recovered from agricultural soils, showed PI of 100%. The P. torulosum and P. dissimile isolates, both recovered from an asparagus soil, showed low PI (10% and 17,5% respectively). The 16 isolates unidentified specifically, recovered from agricultural and forest soils, showed variable PI from 0% to 100%, and probably a number of Pythium species were present in this group of isolates.

[ESP] En un estudio de dos años sobre los hongos entomopatógenos que atacan a las larvas del trips del trigo, Haplothrips tritici (Kurdjumov), durante el período de diapusa en el suelo, se encontró que Beauveria bassiana (Balsamo) Vuillemin representa un factor de mortalidad importante, aunque con variaciones acusadas entre años. En los bioensayos de patogenicidad realizados se detectaron diferencias de patogenicidad entre aislados. Se detectaron varios depredadores potenciales de larvas diapausantes, dos especies de carábidos, Paradromius linearis Olivier y Microlestes sp. y varias especies de arañas. Sobre las espigas de trigo se encontraron larvas y adultos de Aeolothrips, que depredan sobre los huevos y las larvas I de H. tritici. Las larvas II no son presas de Aeolothrips ya que se defienden con éxito, llegando a invertirse los papeles, depredando H. triltritici sobre Aeolothrips. En las espigas se recogieron larvas de Chrysopa y sírfidos con síntomas de haberse alimentado sobre larvas de H. tritici, según se pudo comprobar en laboratorio. En cambio, las mariquitas (Coccinella septempunctata L.) no se alimentaron de larvas de H. tritici en cautividad. [ENG] A two year sampling of pathogens and predators of Haplothrips tritici was carried out in Santa Olalla (Toledo) during the years 1994-96. Among the winter mortality factors was found the attack of the fungus Beauveria bassiana, there being differences of pathogenicity detected between isolates. Its incidence was higher in a dry year than in a wet year. Several potential predators of overwintering larvae were detected, two species of Carabidae, Paradromius linearis and Microlestes sp., and several species of spiders. On the spikes larvae and adults of Aeolothrips were found which, according to laboratory observations, prey upon eggs and larvae I of H. tritici. The larvae II are not Aeolothrips preys since they defended themselves with success, the predator-prey role being inverted, H. tritici depredating upon Aeolothrips larvae. In the spikes larvae of Chrysopa and Syrphidae were collected showing symptoms of feeding upon larvae of H. tritici, according to laboratory observations. Coccinella septempunctata did not feed upon larvae of H. tritici.
