Het plaatsen van honingbijkasten op heidevelden heeft een negatief effect op het voorkomen van wilde bijen en andere bestuivende insecten op heide. Dit blijkt uit onderzoek van EIS Kenniscentrum Insecten van de afgelopen jaren op heidevelden in Oost-Nederland. Ook blijkt dat meer honingbijkasten meer negatieve invloed hebben.

The Netherlands is a country with a relatively high availability of fresh water sources. However, due to the drier summers and limited water storage capacity, drinking water winning from groundwater increasingly exceeds permitted levels in recent summer periods. Therefore, even for a country with an abundance of fresh water, drinking water production and distribution became a top priority. A redeeming feature of the Dutch drinking water is the excellent and well maintained conduit networks. Although the water losses of Dutch drinking water infrastructure are relatively low, maintaining this ageing infrastructure is costly. Reducing water loss by optimizing management of the distribution network therefore presents one of the most important responsibilities of the water companies.Prevention of water loss is not only important in reducing the loss of a valuable resource, bursts in drinking water conduits can also lead to contamination of the drinking water and damage to surrounding infrastructure. Conduit bursts are often caused by a combination of various factors, of which not all are easily identified. Although conduit bursts are complex processes, action can be taken to limit water losses. Traditionally, customers are used as surrogate sensors in order to facilitate reactive leakage control. Recent methods focus on more proactive leakage control strategies, concerned with preventing water loss by optimizing grid operation and management. These methods make use of techniques, such as timely replacement of ageing and damaged pipes, identification of weak assets, optimizing network pressures and valve operations.The information contained within the currently collected big data sets can be compared with an iceberg, where we only currently consider the tip visible above water, while not yet aware of the vast mass of information still hiding underwater. The value of this submerged information has become apparent to water distribution companies. A major challenge faced by, Nederland staat bekend als land met veel beschikbaar zoet water. Toch worden de zomers er droger (uitzondering 2021), met steeds verdere dalingen van het grondwaterpeil tot gevolg. Waterland of niet, drinkwaterwinning en distributie blijft ook voor Nederland van groot belang. Gelukkig is de Nederlandse drinkwaterinfrastructuur goed onderhouden en van hoge kwaliteit, waardoor het waterverlies laag wordt gehouden. Echter zijn veel verouderde leidingen binnenkort aan vervanging toe, waardoor de toch al hoge kosten van het netwerkonderhoud blijven toenemen. Hierdoor blijven optimaal netwerkgebruik en waterverliesminimalisatie hoog op de agenda staan van de Nederlandse drinkwaterbedrijven. Gesprongen of lekke leidingen zorgen niet alleen voor waterverlies, maar kunnen ook vervuiling van het drinkwater of schade aan omringende infrastructuur veroorzaken. De oorzaken van deze lekken zijn vaak veelvoudig en complex, wat detectie of preventie van waterlekken een lastig proces maakt. Traditioneel leunt een drinkwaterbedrijf op klanten om lekken te melden. Ook wordt er steeds meer ingezet of preventieve technieken met een focus op tijdig vervangen van verzwakte leidingen en een slimmer gestuurd netwerk qua e.g. waterdruk en sluitklep standen. De informatie die gevangen zit in de big data aan huidige (sensor)meetwaarden, kan vergeleken worden met een drijvende ijsschots, waarvan slecht het topje boven het water uitsteekt, terwijl de grote meerderheid zich voor ons onderwater verborgen houdt. Dat deze verborgen kolos aan informatie van waarde is, wordt door waterbedrijven niet meer weersproken. Echter blijft het een uitdaging de volledige ijsschots boven water te krijgen. Gelukkig bieden artificiële intelligentie en hydraulische modellen veelbelovende mogelijkheden om zo veel mogelijk informatie te onttrekken aan de huidige en groeiende collectie verzamelde data.

Vascular plants employ an extensive vascular system, known as the xylem, to transport water from their roots to their leaves. Xylem vessels have thick secondary cell wall reinforcements that help them withstand the pressures generated during water transport. These cell wall reinforcements are deposited in a variety of intricate patterns, depending on the type of xylem. Protoxylem has a ringed or spiral cell wall pattern, to allow further elongation, while metaxylem has a pattern of regularly spaced gaps for radial transport in an otherwise solid wall. These patterns are determined by underlying patterns of cortical microtubules, which guide the deposition of cell wall material. Establishing the microtubule pattern, in turn, requires localised activity so-called Rho of Plant (ROP) proteins. ROPs are a type of small GTPase, molecular switches with an active and an inactive form. Microtubules and small GTPases also interact in other important processes, including the development of complex cell shapes. In this thesis, I aim to improve our understanding of the way in which ROPs and microtubules generate the xylem patterns using dynamic models. Chapters 2 and 3 focus on ROP patterning, whereas chapters 4 and 5 deal with microtubule dynamics.Xylem patterning requires the formation of spotted (metaxylem) or banded (protoxylem) ROP patterns. The molecular properties of ROPs and other small GTPases make them particularly suited to spontaneously form patterns through so-called reaction-diffusion mechanisms. Many models for GTPase patterning exist, but most of them only yield a pattern with just one single stable cluster. Initially, these models may give rise to multiple clusters, but in the long run these clusters compete with each other until only one remains. The study of genetically identical cells frequently reveals that substantial variation exists between the expression levels of molecules, such as mRNAs and proteins, with respect to time in individual cells and across a population. This is due to intrinsic noise arising from the random timing of biochemical reactions in the gene regulatory network. An additional source of noise in biological systems comes from interactions of unknown molecular components with the network, which are collectively termed extrinsic noise. Mathematical modelling has been used to understand the factors that contribute to the resulting stochasticity in the gene expression dynamics of cells. As many molecular species are present in small numbers, we require a stochastic description of these biochemical systems. Chapter 1 gives a general introduction into the sources of gene expression noise and non-genetic cell-to-cell variability. Throughout Chapters 2-5, we investigate cell-to-cell variability in a wide range of biological systems through the integration of experimental approaches and mathematical modelling.In Chapter 2, we derive a novel approximate method to obtain closed-form expressions for the means, variances, and power spectra of the molecular species within a chemical reaction network with intrinsic and slowly changing extrinsic noise. We do this by extending the conventional linear-noise approximation (LNA) to include systems where extrinsic noise manifests itself as fluctuations in the reaction rates. These extrinsic fluctuations are characterised by a longer timescale compared to the typically fast intrinsic reaction processes. Plant resilience is an important part of sustainable crop protection. In this project, natural resilience to powdery mildew,an economically important disease problem in greenhouse horticulture, was investigated. Plant resilience is based on morphological properties such as leaf hairs or wax layers on the leaf. In addition, plants are rich in plant compounds ,especially secondary metabolites that are involved in the defense of plants against diseases and pests. By comparing susceptible and less susceptible plants of the same species, metabolite patterns, involved in plant resilience, can be identified. Gerbera and rose were used as model crops for this research. In gerbera, 4 compounds have been found that are related to resistance to mildew in both young and old leaves: gerberin, gerberinside, parasorboside and5-hydroxyhexanolzuur. For this purpose, a total of 50 different mini and standard varieties of different origins were tested. A predictive model, based on compounds, has been created and validated. With an accuracy of 72%, it ispossible to predict the resilience of a gerbera variety to mildew on the basis of a metabolite profile. Because no chemical differences have been observed for roses, it was decided to look at morphological differences. For this purpose, the composition and strength of the wax layer of young and old leaves was investigated. The young leaves of mildew susceptible varieties showed a more extensive wax layer compared to the mildew susceptible varieties. Subsequently,tests were carried out to strengthen the wax layer of susceptible rose varieties. This has been done by means of UV-B treatments. Compared to the untreated control, which showed strong mildew infestation, the UV-B treated plants were free of mildew. The spraying with wax was also successful: spraying with 2 % beeswax (animal wax) or with 2 %carnauba wax (plant wax) led to significantly less mildew compared to the untreated control., Plantweerbaarheid vormt een belangrijk onderdeel van duurzame gewasbescherming. In dit project is de natuurlijke weerbaarheid tegen echte meeldauw, een economisch belangrijk ziekteprobleem in de glastuinbouw, onderzocht. Plantweerbaarheid is gebaseerd op morfologische eigenschappen zoals bladharen of waslagen op het blad. Daarnaast zijn planten rijk aan inhoudsstoffen, vooral aan secundaire metabolieten, welke betrokken zijn bij de afweer van planten tegen ziekten en plagen. Vervolgens zijn er proeven uitgevoerd om de waslaag van vatbare rozenrassen te versterken. Dit is middels UV-B behandelingen gedaan. In vergelijking met de onbehandelde controle, welke sterke meeldauw aantasting liet zien, waren de UV-B behandelde planten vrij van meeldauw.
