An(IV) clusters are multiple metallic centers molecules, were actinide atoms are linked by oxo or hydroxo groups. Those species are formed by condensation reactions with water. They are stabilized thanks to organic or inorganic ligands. If the structures of the clusters are well known at solid state, this is not the case in solution. Hexameric cluster with an octahedral shape core is the main structure detected in solution. This structure is stabilized by ligand with carboxylate functions. Information exists for Th(IV), U(IV) and Np(IV) hexanuclear in solution, but few studies are reported for Pu(IV). Thermodynamic and kinetic data on the formation and the stability of the cluster are scarce. Consequently, clusters are not taken into account for speciation diagram. The goal of this work is to study the plutonium clusters behavior in solution with a simple carboxylate ligand, acetate.In order to detect all potentially formed, plutonium with acetate species solutions were first characterized by Vis-NIR absorption spectroscopy. Large variations of pH and acetate concentrations were used and eight different species are identified : three aquo cations (Pu3+, Pu4+, PuO22+) and five plutonium complexes with acetate. Those five acetate complexes were characterized by coupling experimental (Vis-NIR and EXAFS spectroscopy and ESI-MS spectrometry) and theoretical approach through quantum chemistry. As a result, Pu6O4(OH)4(AcO)12(H2O)12 hexameric cluster has been identified, the missing bloc in the An(IV) series with formate, acetate ligands. The four other complexes are are attributed to plutonium acetate monomeric complexes. Species of Pu(IV) with acetate being described, their fractions in solution are evaluated and reported on a speciation diagram on the pL axis.; Les clusters d’An(IV) sont des molécules à plusieurs centres métalliques reliés entre eux par des groupes oxo et/ou hydroxo en solution aqueuse (le ceur). Ces espèces se forment en présence d’eau par des réactions de condensation, et sont stabilisées par des ligands organiques ou inorganiques (en surface). Si les structures des clusters sont souvent décrites à l’état solide, elles le sont beaucoup moins en solution. La plupart des clusters observés en solution sont des clusters hexamèriques à géométrie octaédrique, complexés à des ligands à fonction carboxylate. Si quelques données existent pour les clusters hexamériques de Th(IV), U(IV) et de Np(IV) en solution, très peu d’informations sont rapportées sur le Pu(IV). Les données thermodynamiques et cinétiques sur la formation des clusters en solution sont rares. Il en résulte que les clusters ne sont pas pris en compte dans les modèles de spéciation. L’objectif de la thèse est l’étude en solution des clusters de Pu(IV) en présence d’un ligand carboxylate simple, le ligand acétate.Afin d’identifier l’ensemble des espèces susceptibles de se former en solution, le Pu(IV) en présence d’acétate a d’abord été caractérisé par spectroscopie d’absorption Vis-NIR. Huit espèces différentes ont pu être identifiées par spectroscopie Vis-NIR : trois cations aquo (Pu3+, Pu4+ et PuO22+) et cinq espèces d’acétate de Pu(IV). Ces cinq espèces ont ensuite été caractérisées en couplant des approches expérimentales (Vis-NIR, EXAFS et ESI-MS) et théorique par calcul de chimie quantique (DFT). Le cluster hexamérique octaédrique Pu6O4(OH)4(AcO)12(H2O)12 a été identifié en solution. Cette espèce était la structure manquante des clusters hexamèrique An6O6(OH)4(AcO)12 des An(IV). Les quatre autres espèces qui l’accompagnent sont proposées comme étant des complexes « classiques » monomériques. Connaissant les espèces d’acétates de Pu(IV) formées en solution, leurs fractions ont pu être évaluées par déconvolution et reportées sur un diagramme de distribution des espèces en pL.

The selectivity of L-lysine acylation by lauric acid catalysed by Candida antarctica lipase B (CALB) was investigated combining experimental and theoretical methodologies. Experiments showed the near-exclusive acylation of lysine $\epsilon$-amino group; only traces of product resulting from the acylation of lysine $\alpha$-amino group were observed fleetingly. Molecular modelling simulations were performed aiming to understand the molecular rules for selectivity. Flexible docking simulations combined with structural investigations into lysine/CALB binding modes also suggested the preferential acylation of lysine $\epsilon$-amino group without, however, excluding the acylation of the lysine $\alpha$-amino group. Electrostatic interaction energy between lysine and the residues covering the catalytic cavity was calculated in order to understand the discrimination between the two lysine amino groups. The results suggests that the proximity of the carboxylate group hinders the binding of the substrate in configurations enabling the N$\alpha$-acylation. Key interactions with the polar region covering the catalytic triad were identified and a plausible explanation for selectivity was proposed.

N-acylation of amino acids or peptides results in bioactive and/or functional molecules showing increased bioavailability, hydrophobicity and stability. Acylated amino acids have been broadly described as being a kind of surfactant with great surface chemistry properties, interesting biological activities, weak toxicity and low environmental impact. Acylation of amino acids or peptides is being performed chemically at industrial scale. It creates constraints in term of reaction selectivity, environmental safety and cost of polluted wastewater treatment. Enzymatic catalysis is an alternative to chemical acylation reaction. Several enzyme/solvent pairs have already been described in the literature. Their performance are however somewhat limited. The objective of this thesis work was thus to improve the capacity of acylation processes at different scales. At the molecular scale, a study was performed using Candida antarctica’s (CALB) lipase B. Molecular modeling was used to create a methodology coupling docking simulation and interaction calculus that would allow for a better understanding of CALB regioselectivity during lysine acylation by different fatty acids. Studies were also conducted at the reaction level, especially by searching for new aminoacylase-type of biocatalysts in Streptomyces ambofaciens raw extract. Regioselectivity and performance of these enzyme’s catalytic reactions were compared to those of CALB. Results brought into light a promising potential from S. ambofaciens’ aminoacylases in synthesizing acylated amino acids/peptides. Indeed, on top of their ability to catalyse acylation reaction in aqueous solution, these enzymes have a different regioselectivity compared to CALB’s. Regioselectivity targeting N-terminal groups is a rarely researched phenomenon allowing acylation to be performed without modifying amino acids or peptides lateral chains and hence their functionality. In the last part part of this work, studies at process scale were performed. Aminoacylase were first immobilized on mesoporous silicates and several immobilisation methods were compared. Using physisorption, a method for the immobilisation of S. ambofaciens’ aminoacylases was developed to reach a conserved specific activity during 3 cycles. Finally, intensification of acylation reaction was examined in microwave or microstructured reactors. First, an experimental set up was performed in an heated reactor using microwaves irradiation. This kind of reactor was demonstrated as being adapted to acylation reaction using a commercial immobilized form of CALB (Novozym435®) as catalyst in organic solvent. The microwave reactor was however not suited for free S. ambofaciens aminoacylase in aqueous solution. For that latter reaction, intensification had to be approached through other aspects of the process. Hydrodynamic appeared indeed as an important aspect for this reaction occurring in a biphasic medium composed of fatty acids and aqueous solution. A microstructured microfluidic reactor was hence tested. Conversion yield were increased with this system. This study demonstrated how mixing quality was an important factor for acylation reaction and could be a way to intensify the enzymatic process at larger scale, Approche multi-échelle pour l’étude de la réaction de N-acylation enzymatique d’acides aminés La réaction de N-acylation d’acides aminés ou de peptides permet l’obtention de dérivés de ces molécules présentant des propriétés bioactives et/ou techno-fonctionnelles, avec une biodisponibilité, une hydrophobie et une stabilité accrue. Les acides aminés acylés ont été largement décrits comme constituant une classe d'agents tensioactifs avec d'excellentes propriétés de surface, des activités biologiques intéressantes, un faible potentiel de toxicité et un faible impact environnemental. Actuellement réalisée de manière chimique à l’échelle industrielle, l’acylation de ces acides aminés ou peptides présente des contraintes en termes de sélectivité réactionnelle et d’innocuité vis-à-vis de l’environnement ainsi qu’en termes de coût de retraitement des effluents polluants. Une alternative à cette voie chimique est l’utilisation d’enzymes capables de catalyser ces réactions d’acylation. Dans la littérature, différents couples d’enzymes et de solvants ont déjà été décrits. Néanmoins, les performances réactionnelles de ces systèmes demeurent parfois limitées. L’objectif de cette thèse a donc été l’amélioration du procédé d’acylation par une approche à différentes échelles. À l’échelle moléculaire, une étude a été réalisée avec la lipase B de Candida antarctica (CALB). Une approche de modélisation moléculaire a été utilisée afin de mettre au point une méthodologie associant des simulations de docking et des calculs d’interaction permettant d’améliorer la compréhension et permettre la prédiction de la régiosélectivité de CALB lors de l’acylation de la lysine par différents acides gras. Des études ont également été conduites à l’échelle réactionnelle, notamment avec la recherche de nouveaux biocatalyseurs de type aminoacylases dans l’extrait brut de Streptomyces ambofaciens. La régiosélectivité et les performances de la réaction catalysée par ces enzymes ont été comparés à celles de CALB. Les résultats ont mis en évidence un potentiel très prometteur des aminoacylases de S. ambofaciens concernant la synthèse d’acide aminés/peptides acylés. En effet, en plus de leur aptitude à réaliser la réaction d’acylation en milieu aqueux, ces enzymes possèdent une régio-sélectivité qui diffère de celle de CALB. Cette régio-sélectivité orientée vers les groupements N-terminaux est un atout très peu décrit à ce jour, car elle permet d’acyler ces molécules sans modifier les chaînes latérales des acides aminés ou des peptides et donc leurs fonctionnalités. Dans la dernière partie de ces travaux, des études à l’échelle procédé ont été menées. Tout d’abord, l’immobilisation des aminoacylases sur des matériaux mésoporeux silicatés a été réalisée et différentes méthodes d’immobilisation ont pu être comparées. Cette étude a permis de proposer une méthode d’immobilisation des aminoacylases de S. ambofaciens par physisorption, permettant de conserver l’activité spécifique pendant au moins 3 cycles. Puis, dans une dernière partie, l’intensification de la réaction d’acylation en réacteur micro-ondes ou microstructurés a été abordée. Les expérimentations réalisées dans un réacteur chauffé par irradiation micro-onde ont montré que ce type de réacteur était adapté à la réaction d’acylation catalysé par CALB sous sa forme immobilisée commerciale (Novozym435®) en solvant organique, ce qui n’est pas le cas avec des aminoacylases de S. ambofaciens libres, en milieux aqueux. Pour cette réaction, d’autres méthodes d’intensification ont été envisagées, notamment en réacteur microstructuré de type microfluidique. L’efficacité du mélange étant primordiale notamment en milieu biphasique, celle-ci a pu être améliorée avec un taux de conversion supérieur dans ce réacteur comparativement à un réacteur classique agité mécaniquement

The modelisation of energetic performance (Heat of formation and specific impulse) of new highly nitrogen compounds based on tetrazene bones led to the selection of new focuses for space propulsion application in order to replace hydrazines (MMH and UDMH). In one hand, study on experimental faisability and isolation of new compounds showed the sensibility of azobistetrazole derivatives to impact and friction test (BAM choc et BAM friction) and in another hand, tetrazoline and tetrazolidine derivatives were not syntheses by catalytic hydrogenation or classical reduction of tetrazole derivatives. In this study, we achieved to synthesis new alkyltetrazenes by direct azidonation of 1,1',4,4'-tetramethyl-2-tetrazene (TMTZ): it was the 1,4-di(azidomethyl)-1',4'-dimethyl-2-tetrazene (DADMTZ) and the 1-azidomethyl-1',4,4' trimethyl-2-tetrazene (MATMTZ). Synthesis optimization led to the optimal conditions for total and exclusive conversion of TMTZ in mono- or di-azido derivatives. Thermal stability was studied at different temperature and the structure of the di-azido derivatives was confirmed by NOESY experiment in association with modelisation, La modélisation des performances énergétiques (enthalpie de formation et impulsion spécifique) de nouveaux composés fortement azotés basés sur le squelette du tétrazène (N-N=N-N) a permis de sélectionner de nouvelles cibles pour la propulsion spatiale dans le but de remplacer les hydrazines (MMH ou UDMH) dans les systèmes à biergols stockables. L'étude expérimentale de la faisabilité de synthèse et d'isolement de ces nouveaux composés a mis en évidence le caractère très sensible des dérivés azobistétrazolés vis-à-vis des stimuli mécaniques (BAM choc et BAM friction) et électrostatiques (ES) d'une part, et l'impossibilité d'accéder à des dérivés tétrazolines ou tétrazolidines par hydrogénation catalytique ou par réduction du cycle tétrazole, d'autre part. Cette étude a également permis l'obtention de dérivés mono- et di-azoturés d'alkyltétrazènes par une ix-azoturation directe du 1,1',4,4'-tétraméthyl-2-tétrazène (TMTZ) : il s'agit du 1-azidométhyl-1',4,4'-triméthyl-2-tétrazène (MATMTZ) et du 1,4-di(azidométhyl)-1',4'- diméthyl-2-tétrazène (1,4-DADMTZ), respectivement. L'optimisation de la synthèse a permis de définir les conditions optimales pour une conversion totale et exclusive du TMTZ, en dérivé mono- ou di-azoturé, dont la stabilité thermique a été étudiée à différentes températures. La structure du dérivé di-azoturé a été confirmée par des analyses NOESY en association avec des calculs de distances interatomiques. In the current context of oil resources rarefaction, the development of biobased polymers is of major importance. The present work focused on the development of a biobased amorphous polyphthalamide, based on furan-2,5-dicarboxylic acid. The first part of the study was devoted to the development of a molecular modelling protocol that could calculate the glass transition temperature of polyphthalamides with accuracy. In order to do this, model polyphthalamides based on isophthalic, terephthalic acid and hexamethylene diamine were synthesized and characterized as well as simulated. By comparison between simulated and measured Tg, the protocol was validated. In a second part of the study, this protocol was applied to FDCA based polyphthalamides. These PPA were also synthesized. It was found that the PA 6-F undertakes a decarboxylation, preventing it from reaching high a molar mass. It was also found that the mechanical and thermal properties decrease as the amount of FDCA in the copolymers increases., Dans le contexte actuel de raréfaction des ressources fossiles, le développement des polymères biosourcés est d'une grande importance. Le travail de cette thèse consiste donc en la synthèse et le développement d'un polyphtalamide amorphe à base d'acide furan-2,5-oique. Dans un premier temps, un protocole de modélisation moléculaire permettant de calculer la Tg de polyphthalamides a été développé. Pour cela, des PPA modèles (basés sur un mélange d'acides isophtaliques et téréphtaliques ainsi que l'hexaméthylène diamine) ont été synthétisés et caractérisés. En comparant les Tg obtenues par modélisation et par mesure en DSC, le protocole de modélisation a été validé. Dans un deuxième temps, ce protocole a été appliqué à des PPA à base de FDCA. Ces polymères ont également été synthétisés et caractérisés. Il en ressort que le PA 6-F subit une importante décarboxylation durant sa synthèse, l’empêchant d'atteindre une masse molaire importante. Il a aussi été montré que l'ajout de FDCA dans des copolyphthalamides diminuait leurs propriétés thermiques et mécaniques.

Monogalactosyldiacylglycerol (MGDG), the major lipid of plant and algal plastids, is synthesized by MGD (or MGDG synthase), a dimeric and membrane-bound glycosyltransferase of the plastid envelope that catalyzes the transfer of a galactosyl group from a UDP-galactose donor onto a diacylglycerol acceptor. Although this enzyme is essential for biogenesis, and therefore an interesting target for herbicide design, no structural information is available. MGD monomers share sequence similarity with MURG, a bacterial glycosyltransferase catalyzing the transfer of N-acetyl-glucosamine on Lipid 1. Using the x-ray structure of Escherichia coli MURG as a template, we computed a model for the fold of Spinacia oleracea MGD. This structural prediction was supported by site-directed mutagenesis analyses. The predicted monomer architecture is a double Rossmann fold. The binding site for UDP-galactose was predicted in the cleft separating the two Rossmann folds. Two short segments of MGD (〈β2–α2〉 and 〈β6–β7〉 loops) have no counterparts in MURG, and their structure could not be determined. Combining the obtained model with phylogenetic and biochemical information, we collected evidence supporting the 〈β2–α2〉 loop in the N-domain as likely to be involved in diacylglycerol binding. Additionally, the monotopic insertion of MGD in one membrane leaflet of the plastid envelope occurs very likely at the level of hydrophobic amino acids of the N-terminal domain.
