Pearl millet prolamin, a non gluten protein called pennisetin, was used to formulate microparticles by phase separation, using glacial acetic acid (10%) and aqueous ethanol (70%). Optical microscopy (OM) and scanning electron microscopy (SEM) showed that microparticles produced by aqueous ethanol (ME) were spherical with smooth surface and their average diameter was 13 ± 2 µm, whereas those prepared with glacial acetic acid (MA) were irregularly shaped with tough surface and high diameters ranging from 22.87 to 119.25 µm. ME morphology was highly affected by antisolvent concentration and mixing speed. Thermogravimetric analysis (TGA) indicated that microparticles began losing weight at 60°C probably due to water evaporation, then a second step of degradation occurred at higher temperature 318, 324°C and 308°C for ME, MA and pennisetin powder, respectively. The differential scanning calorimetry (DSC) showed that ME were more heat stable than MA and the original pennisetin powder. Fourier transform infrared (FTIR) spectra showed an increasing formation of ß-sheet structure in ME and MA indicating tendency to protein aggregation during microparticle formation. Interstellar water ice is mainly amorphous, but the nature of its morphology still remains poorly known. The experimental study described in this work focuses on how relevant changes of the ice morphology result from atomic hydrogen exposure and subsequent recombination. We show that there is an exponential decrease in the porosity of the amorphous water ice sample following hydrogen-atom irradiation. These and other laboratory results lead us to suggest that water ice in space is almost certainly amorphous and non-porous (compact). [ABSTRACT FROM AUTHOR]

We present a combined theoretical and experimental study of the adsorption of two pairs of organic isomers, (i) acetic acid AA (CH3COOH) and methyl formate MF (HCOOCH3), and (ii) ethanol EtOH (CH3CH2OH) and dimethyl ether DME (CH3OCH3), onto crystalline water ice surfaces at low temperatures. Both approaches show that, for each pair, the most stable isomer from a thermodynamical point of view, i.e. AA and EtOH, is also the one which is the more tightly bound to the water ice surface compared to the less stable isomers (MF and DME). This finding, which can be explained by the ability of AA or EtOH to efficiently interact with the ice surface via hydrogen bondings, may have important consequences in an astrophysical context, since it could explain why the most stable isomer is not the most abundant observed in the interstellar gas phase. The effect of wet and dry milling on the functional properties of whole sorghum grain flour (SF) and extracted kafirin were assessed. White sorghum landrace was used to prepare two SFs by wet milling (SF1) or dry milling (SF2) and to extract their respective kafirins SK1 and SK2. Protein contents of SK1 and SK2 were 90.07 and 94.23%. Wet milling of SF allowed increasing the oil binding capacity, the least gelling concentration and the instant foam capacity and decreasing the water binding capacity and foam stability. The emulsifying activity index of SF1 and SF2 were in the same range, while emulsion stability was two time higher in SF2. Functional properties of SK1 and SK2 showed an appropriate water binding capacity of 2.20 ± 0.10 and 1.82 ± 0.22 (g water/g dry mater), respectively. Both SK1 and SK2 showed higher oil binding capacity than SF1 and SF2 with no gel and foam formation. The wet milling improved water and oil binding capacities of SK1 by 17 and 5%. The emulsifying activity indexes were approximately similar for SK1 and SK2 with emulsion stability exceeding 60%. Fourier transform infrared spectroscopy of SK1 and SK2 showed that wet milling induced a decrease of α-helical structure and an increase of intermolecular β-sheet and β-turns and no change in the anti parallel β-sheet. This study showed that wet milling could allow extracting kafirin with preserving the most important functional properties of SF and kafirin and could constitute an interesting method for protein recovery and starch isolation., (© Association of Food Scientists & Technologists (India) 2019.)

This work aims to evaluate starch digestion in whole sorghum grains. Nine sorghum cultivars were sampled from the Sahara of Algeria. The structural characteristics of sorghum grains were measured. Total starch (TS) varied between 67.67% and 74.82%, digestible starch (DS) between 64.34% and 69.70%, and resistant starch (RS) ranged from 2.55% to 7.98%. The kinetic of starch digestion displayed first-order model. For all sorghum cultivars, starch were digested with different extents, DS at infinite time (D ∞) ranged from 52.58 to 102.13 g/100 g dry starch, while the hydrolysis index (HI) ranged from 41.55% to 76.93% and high average glycemic index (GIavg) ranged from 65.97 to 94.14. The results showed that there are differences in grain quality of Algerian sorghum cultivars. The starch fractions have acceptable nutritional value with good in vitro digestibility characteristics suitable for human health and nutrition.

The morphology of water ice in the interstellar medium is still an open question. Although accretion of gaseous water could not be the only possible origin of the observed icy mantles covering dust grains in cold molecular clouds, it is well known that water accreted from the gas phase on surfaces kept at 10 K forms ice films that exhibit a very high porosity. It is also known that in the dark clouds H(2) formation occurs on the icy surface of dust grains and that part of the energy (4.48 eV) released when adsorbed atoms react to form H(2) is deposited in the ice. The experimental study described in the present work focuses on how relevant changes of the ice morphology result from atomic hydrogen exposure and subsequent recombination. Using the temperature-programmed desorption (TPD) technique and a method of inversion analysis of TPD spectra, we show that there is an exponential decrease in the porosity of the amorphous water ice sample following D-atom irradiation. This decrease is inversely proportional to the thickness of the ice and has a value of ϕ(0) = 2 × 10(16) D-atoms cm(-2) per layer of H(2)O. We also use a model which confirms that the binding sites on the porous ice are destroyed regardless of their energy depth, and that the reduction of the porosity corresponds in fact to a reduction of the effective area. This reduction appears to be compatible with the fraction of D(2) formation energy transferred to the porous ice network. Under interstellar conditions, this effect is likely to be efficient and, together with other compaction processes, provides a good argument to believe that interstellar ice is amorphous and non-porous., (This journal is © the Owner Societies 2011)

Nuclear spin conversion (NSC) of ortho- to para-H(2) and para- to ortho-D(2) has been investigated on an amorphous solid water (ASW) surface at 10 K, in the presence of co-adsorbed O(2). The dynamics of the nuclear spin conversion could be revealed by combination of resonance enhanced multiphoton ionization spectroscopy (REMPI) with temperature programmed desorption (TPD) experiments. The conversion rates are consistent with a diffusion of molecular hydrogen inducing a nuclear spin conversion enhanced in the vicinity of molecular oxygen. The conversion times were found to increase with decreasing O(2) and H(2) coverage. Finally, on oxygen free ASW surface, the extremely long conversion characteristic times measured showed that such surface is not an efficient catalyst for NSC, in contradiction with hypothesis commonly made for interstellar medium.

An XIP (xylanase inhibiting protein) type xylanase inhibitor was purified from Algerian pearl millet ( Pennisetum glaucum L.) grains and characterized for the first time. Cation exchange and affinity chromatography with immobilized Trichoderma longibrachiatum glycoside hydrolase (GH) family 11 xylanase resulted in electrophoretically pure protein with a molecular mass of 27-29 kDa and a pI value of 6.7. The experimentally determined N-terminal amino acid sequence of the purified XIP protein is 87.5%, identical to that of sorghum ( Sorghum bicolor L.) XIP and 79.2% identical to that of wheat ( Triticum aestivum L.) XIP-I. The biochemical properties of pearl millet XIP are comparable to those described earlier for sorghum XIP, except for the higher specific activity toward a T. longibrachiatum GH family 11 xylanase. On the basis of immunoblot neither TAXI nor TLXI type xylanase inhibitors were detected in pearl millet grains.
