8. Assessing Falling Number Stability Increases the Genomic Prediction Ability of Pre-Harvest Sprouting Resistance in Common Winter Wheat.

Abstract

Pre-harvest sprouting (PHS) resistance is a complex trait, and many genes influencing the germination process of winter wheat have already been described. In the light of interannual climate variation, breeding for PHS resistance will remain mandatory for wheat breeders. Several tests and traits are used to assess PHS resistance, i.e., sprouting scores, germination index, and falling number (FN), but the variation of these traits is highly dependent on the weather conditions during field trials. Here, we present a method to assess falling number stability (FNS) employing an after-ripening period and the wetting of the kernels to improve trait variation and thus trait heritability. Different genome-based prediction scenarios within and across two subsequent seasons based on overall 400 breeding lines were applied to assess the predictive abilities of the different traits. Based on FNS, the genome-based prediction of the breeding values of wheat breeding material showed higher correlations across seasons ( r = 0.505 − 0.548 ) compared to those obtained for other traits for PHS assessment ( r = 0.216 − 0.501 ). By weighting PHS-associated quantitative trait loci (QTL) in the prediction model, the average predictive abilities for FNS increased from 0.585 to 0.648 within the season 2014/2015 and from 0.649 to 0.714 within the season 2015/2016. We found that markers in the Phs-A1 region on chromosome 4A had the highest effect on the predictive abilities for FNS, confirming the influence of this QTL in wheat breeding material, whereas the dwarfing genes Rht-B1 and Rht-D1 and the wheat–rye translocated chromosome T1RS.1BL exhibited effects, which are well-known, on FN per se exclusively. 14. Genetic analysis of Aegilops tauschii-derived seedling resistance to leaf rust in synthetic hexaploid wheat

Abstract

Seedling resistance to leaf rust available in the synthetic hexaploid wheat line Syn137 was characterised by means of cytogenetic and linkage mapping. Monosomic analysis located a single dominant gene for leaf rust resistance on chromosome 5D. Molecular mapping not only confirmed this location but also positioned the gene to the distal part of the long arm of chromosome 5D. A test of allelism showed that the gene, tentatively named LrSyn137, is independent but closely linked to Lr1. It appears that Syn137 is occasionally heterogeneous for Lr1since the analysis of the Lr1-specific marker RGA567-5 in the genetic mapping population indicated the presence of Lr1. Syn137 represents another source of genetic variation that can be useful for the diversification of leaf rust resistance in wheat cultivars.

Abstract

The rate at which wheat (Triticum aestivum L.) reaches anthesis and other developmental stages depends largely on the variation of three major factors: vernalization (Vrn), photoperiod (Ppd), and earliness per se (Eps). The objectives of this study were to characterize a set of genotypes for the presence of developmental genes and their potential association to adaptability and to identify diagnostic molecular markers for the vernalization gene Vrn-D1. Fifty-one genotypes were crossed with testers containing known Vrn and Ppd genes. To determine earliness per se and its interaction with temperature, genotypes were vernalized for 8 wk and placed in a 24-h photoperiod regime at two temperatures. To validate molecular markers for Vrn-D1, the cultivars were genotyped with closely linked SSR markers Xgwm292-5D and Xgwm212-5D. Segregation analysis on [F.sub.2] populations of field-grown plants showed that Vrn-D1 was present in 66% of the cultivars, while Vrn-A1, Vrn-B1, and Vrn4 were found in 41, 39, and 8% of the lines, respectively, either solely or in combination with other Vrn genes. A low percentage of cultivars appeared to have a constitutive genetic character for early flowering (Eps), while most of the cultivars may have temperature-sensitive genes that respond to local environmental changes. Vrn-D1 was reliably detected in 76.5% of the genotypes using the 215-bp allele of microsatellite marker Xgwm292-5D, whereas Xgwm212-5D did not reveal sufficient allelic variation within the tested wheats., THE DEVELOPMENT of broadly adapted, high yielding heat varieties has been one of CIMMYT's primary goals. 50. Allocation of the oat powdery mildew resistance gene Pm3to oat chromosome 1A

Abstract

Besides the mode of inheritance, the knowledge of the chromosome location and allelic relationships are the essentials towards a successful deployment and stacking of divergent disease resistance genes for a given pathogen in breeding programs. Powdery mildew of oats, to which 11 major resistance genes in the host Avena sativaL. have been characterized so far, is a prevalent fungal disease of the crop in Northwestern Europe. In the present study, the resistance gene Pm3was mapped by linkage analysis relative to molecular markers from oat consensus linkage group Mrg18 which was recently determined to represent oat chromosome 1A. Pm3was located at 67.7–72.6 cM on Mrg18 of the oat consensus map, a position at which also stem and crown rust resistance genes Pg13and Pc91and a large cluster of resistance gene analogs have been previously mapped. The closely linked marker GMI_ES03_c2277_336 was found to be useful for the prediction of Pm3in gene postulation studies. Although the major effect of the widespread gene got lost over time, the known genome location with associated markers will assist revealing in future genetic studies whether there is a possible residual effect of the gene contributing to adult plant resistance.
