Paleoenvironmental archives in East Antarctica have revealed significant changes during the Holocene, marked by ice sheet retreat leading to the isolation of submarine basins. These basins offer valuable insights into past climate, glaciology, and oceanography shifts that impact sedimentary processes. In this study, environmental magnetism and biogeochemical proxies to investigate the Mid-to-Late-Holocene transitions in Pup Lagoon, a coastal isolation basin is presented. Our findings reveal distinct stratigraphic zones reflecting shifts from marine to lacustrine environments. The results reveal predominant mechanical weathering in the Stornes region, producing coarse-grained "soft" ferrimagnetic minerals. Notably, a period of warm oceanographic conditions between 6000 and 4722 cal. yr BP was characterized by mixed magnetic grain sizes and ultrafine superparamagnetic grains, indicating relatively oxic open waters in the basin. Subsequent shifts to reducing conditions coincide with persistent marine sea ice cover from 4722 to 2634 cal. yr BP, favoring the retention of coarse-grained ferrimagnets. Finer magnetic grain sizes between 2634 and 2185 cal. yr BP was attributed to the increased freshwater inputs associated with the Mid-Holocene Hypsithermal. Further, diagenetic changes under persistent sea ice cover between 2185 and 1970 cal. yr BP led to the selective dissolution of fine-grained ferrimagnets. Transitioning to freshwater isolated basin conditions between 1970 and 588 cal. yr BP, fine ferrimagnet precipitation indicate oxic to suboxic conditions alongside drier conditions. Biogenic productivity increased post-isolation, which was reflected in increased (Total Organic Carbon) TOC and (Total Nitrogen) TN percentages. Additionally, the presence of greigite in the isolated phase sediment indicates reducing conditions owing to organic matter decomposition. Notably, χfd% exhibits an inverse trend to sea ice concentration, potentially indicating anoxic-dysoxic conditions due to the presence of sea ice. These observations align with broader regional sea ice concentration changes, emphasizing the interconnected behavior of local and regional factors shaping Antarctic coastal environments. This study discusses the metal concentrations and sedimentological data from Lake L55, a dried, land-locked lake from Schirmacher Oasis, East Antarctica. Toxic metals in sediments are a major threat to the environment. Twenty-five surface sediment samples were analyzed for the presence of fifteen metals. The sedimentological data indicates that the sediments are predominantly sandy with low concentrations of silt and clay. The chemical weathering index for the sediments reflects low degree of weathering. Negligible concentrations of organic content were present in the sediments owing to the cold and dry conditions of the study area. The environmental pollution indices indicate elevated concentrations of Ba, Zn, As and Pb in the sediments. Barium could be supplied by the weathering of the quartzofeldspathic catchment rocks, while Zn, As and Pb could be supplied from the fuel burning for power generation and logistical activities, paint residue, and battery waste. The sediment quality guidelines were calculated for the sediments, and they indicate that the sediments have a 21% risk of being toxic. The pollution load index, potential ecological risk index (PERI) and toxic risk index (TRI) values suggest low toxic risk in the sediments of the lake. [ABSTRACT FROM AUTHOR]

The continued scaling of genetic perturbation technologies combined with high-dimensional assays such as cellular microscopy and RNA-sequencing has enabled genome-scale reverse-genetics experiments that go beyond single-endpoint measurements of growth or lethality. Datasets emerging from these experiments can be combined to construct perturbative "maps of biology", in which readouts from various manipulations (e.g., CRISPR-Cas9 knockout, CRISPRi knockdown, compound treatment) are placed in unified, relatable embedding spaces allowing for the generation of genome-scale sets of pairwise comparisons. These maps of biology capture known biological relationships and uncover new associations which can be used for downstream discovery tasks. Construction of these maps involves many technical choices in both experimental and computational protocols, motivating the design of benchmark procedures to evaluate map quality in a systematic, unbiased manner. Here, we (1) establish a standardized terminology for the steps involved in perturbative map building, (2) introduce key classes of benchmarks to assess the quality of such maps, (3) construct 18 maps from four genome-scale datasets employing different cell types, perturbation technologies, and data readout modalities, (4) generate benchmark metrics for the constructed maps and investigate the reasons for performance variations, and (5) demonstrate utility of these maps to discover new biology by suggesting roles for two largely uncharacterized genes. Author summary: Due to the rapid advancements in genetic perturbation, laboratory robotics, sequencing, and computer vision, more researchers are now generating datasets that capture cellular responses to genetic perturbations. These datasets can be powerful discovery tools for examining known biological relationships and revealing new associations in an unbiased manner when paired with a computational pipeline that can assemble the data into a digestible format. However, the challenge arises from the variety of cellular models, assay designs, terminologies, codebases, and analysis methods involved. In this work we define a unified framework for building and benchmarking perturbative maps, benchmark four different datasets assembled into 18 different maps, explore the impact of different design decisions, and demonstrate how these maps can be used to elucidate gene functions. Our goal is to facilitate comparisons across various technologies and methods by introducing a shared language for the field. The open-source codebase, capable of incorporating new methods, aims to be a resource for researchers developing laboratory or computational methodology. While we caution against definitive recommendations due to numerous variables at play, we hope to stimulate studies directly comparing methods under controlled conditions. Metal contamination in the Arctic region has increased over the years despite its remote and isolated location. Thus, to evaluate the bioavailable fractions of various metals and their effects on sediment-associated biota, the surface sediments from the fjords were analyzed for bulk concentration of metals and their speciation in different fractions. Metals concentrations were higher in the inner fjord region and decreased towards the outer fjord, supported by the terrigenous influence (TI%) calculated. Cr and Pb showed higher excess values attributed to their additional source other than the catchment rocks. So, to assess the metal-related ecological risk, the bulk concentration of metals was compared with Arctic sediment quality guidelines (ASQGs). Cr, Cd, and Pb concentrations were high, indicating potential adverse biological effects in the study. To avoid the risk of overestimation, metal speciation was conducted, showing that overall metal concentrations were higher in the residual fraction; however, higher concentrations of Mn in labile phases pose a moderate risk to the sediment-associated biota. Additionally, the population density of foraminifera in the sediments was calculated to assess the influence of bioavailable metal on benthic foraminifera. It was found that the presence of metals in bioavailable fractions affected the abundance of the foraminifera. However, no morphological abnormalities were observed in the species. The analysis of water samples collected during the MD218 CROTALE expedition (February-March, 2019), and the 4th Indian Southern Ocean Expedition (SOE) (January-February, 2010) (re-visited samples) resulted in the occurrence of a new coccolithophore species belonging to the genus Alisphaera, and is named Alisphaera crostae sp. nov. This new species appears monomorphic and shows distinctive morphological differences in terms of its coccolith size, flange structure, and nodules, compared to other known Alisphaera species. In this study, morphological differences between A. crostae and other Alisphaera species are illustrated and compared. We suggest that the genus Alisphaera may have a higher diversity and may possess a wider distribution than previously evidenced, which requires more careful observation [ABSTRACT FROM AUTHOR]

The optical characteristics of coastal Antarctic waters exhibit complexity due to the dynamic hydrography influenced by meltwater intrusion, which alters nutrient levels, thermohaline structure, and optically active substances (OAS) regimes. Studies on bio-optical variability and its implications on phytoplankton productivity (PP) are scanty in coastal polar regions. On this backdrop, timeseries measurements (72 h at 6 h intervals) of bio-optical properties such as phytoplankton biomass (chlorophyll-a), absorption (aph), and total suspended matter (TSM) concurrently with PP were measured to understand their interplay and variability in relation to the ambient physicochemical settings in the undersampled Prydz Bay, coastal Antarctica. Our findings revealed thermohaline stratification within the bay, likely attributed to the inflow of less saline meltwater from nearby glaciers and minimal wind activity. The consistent presence of subsurface chlorophyll maximum (SCM) beneath the stratified layer underscored the light-acclimatization response of shade-adapted phytoplankton. Surface waters exhibited higher TSM compared to deeper layers, indicating glacial melt influence, while the depth of the sunlit layer remained relatively stable, suggesting limited watermassmovement and/or variability in OAS at the study site. An inverse relation between chlorophyll-a and chlorophyll-a-specific phytoplankton light absorption (a*ph(λ)) manifested 'pigment package effect' within the prevailing phytoplankton community, implying reduced light-absorption efficiency and consequent lower PP. Compared to chlorophyll-a, the phytoplankton light absorption (aph(l)) emerged as a better proxy for explaining PP variability. Nutrient availability was not limiting, which was conducive to micro (large) phytoplankton growth. Classification of phytoplankton size classes (micro, nano, and pico) based on the B/R ratio (aph at Blue (443 nm)/Red (676 nm) region) confirmed the dominance of larger (micro) phytoplankton that aremore susceptible to package effect, thus have implications on reduced PP potential of this polar marine ecosystem. Background Mutations present in emerging SARS-CoV-2 variants permit evasion of neutralization with prototype vaccines. A novel Omicron BA.1 subvariant–specific vaccine (NVX-CoV2515) was tested alone or as a bivalent preparation with the prototype vaccine (NVX-CoV2373) to assess antibody responses to SARS-CoV-2. Methods Participants aged 18 to 64 years immunized with 3 doses of prototype mRNA vaccines were randomized 1:1:1 to receive a single dose of NVX-CoV2515, NVX-CoV2373, or the bivalent mixture in a phase 3 study investigating heterologous boosting with SARS-CoV-2 recombinant spike protein vaccines. Immunogenicity was measured 14 and 28 days after vaccination for the SARS-CoV-2 Omicron BA.1 sublineage and ancestral strain. Safety profiles of vaccines were assessed. Results Of participants who received trial vaccine (N = 829), those administered NVX-CoV2515 (n = 286) demonstrated a superior neutralizing antibody response to BA.1 vs NVX-CoV2373 (n = 274) at day 14 (geometric mean titer ratio, 1.6; 95% CI, 1.33–2.03). Seroresponse rates were 73.4% (91/124; 95% CI, 64.7–80.9) for NVX-CoV2515 vs 50.9% (59/116; 95% CI, 41.4–60.3) for NVX-CoV2373. All formulations were similarly well tolerated. Conclusions NVX-CoV2515 elicited a superior neutralizing antibody response against the Omicron BA.1 subvariant as compared with NVX-CoV2373 when administered as a fourth dose.
