Abstract This article examined the effect of geographical (different climate conditions) and floral origins on some quality parameters of honey including the activity of diastase enzyme. Moreover, some non-quality parameters were investigated such as the pH, fructose, glucose, ratio of fructose/glucose and invertase. The honey samples were collected from Asir (cold climate) and Jazan (hot climate) regions at the southwestern part of Saudi Arabia. The geographical origin significantly affected the mean value moisture of the Acacia honey (p-value = 0.02), conductivity of the polyfloral honey (p-value = 0.03), sucrose of the Acacia honey (p-value = 0.02), diastase activity of the Acacia (p-value = 0.001), Ziziphus (p-value = 0.046) and polyfloral honey (p-value ≤ 0.001), fructose of the Acacia honey (p-value = 0.01), glucose of the Ziziphus honey (p-value = 0.03), fructose/ glucose ratio of the Ziziphus honey (p-value = 0.035), and invertase activity of the polyfloral honey (p-value ≤ 0.001). Regarding the effect of the floral origin of the honey from Asir region, the sucrose percentage of the Acacia honey was significantly more than that of the polyfloral honey (p- value = 0.003), the diastase activity of the Acacia honey was significantly more than its activity in the Ziziphus honey (p- value = 0.044), glucose percentage of the Ziziphus honey was significantly more the glucose percentage of the Acacia honey (p-value = 0.009) and the fructose/ glucose ratio of the Ziziphus honey was significantly more than that of the Acacia and polyforal honeys (p-value = 0.011 and p-value = 0.045, respectively). Concerning the significant effects of the floral origin on the quality parameters of the honey samples from Jazan region, the moisture of the Ziziphus honey was significantly increased when compared to the moisture of the Acacia honey (p-value = 0.038), the acidity of the polfloral honey was significantly more than the acidity of the Acacia honey (p-value = 0.049), the sum of fructose and glucose of the polyfloral honey was significantly increased compared to that of the Acacia honey (p-value = 0.015), the pH of the Ziziphus hiney was significantly more than the pH of the polyfloral honey (0.011) and the fructose of the polfloral honey was significantly more than that of the Acacia honey (p-value = 0.031). The effect of the geographical origin of the honey samples on their quality parameters depends on their floral origin and the effect of their floral origin differs according to their geographical origin. This article suggests considering collectively the geographical and floral origins effect when developing honey standards. However, the Codex standards for honey started considering this issue when it changed the standard concentration of HMF in honey from not more than 80–40 mg/Kg for honeys from cold climate and 80 mg/Kg for honeys from hot climates.

Background: Artificial sweeteners are low calorie polyhydric alcohols produced from carbohydrates or amino acids. Presence of artificial sweeteners in honey is not investigated before. Objective: this study measured the concentration of Xylitol and Sorbitol, the activity of glucose oxidase (GOX) and the concentration of glucose and acidity of honey samples from Asir region-Saudi Arabia. Furthermore, the values of pH and electric conductivity were determined.MethodsThe floral origin of the honey samples were confirmed through pollen analysis. The PH, electric conductivity (EC.), acidity, and glucose concentration were measured according to the methods of international honey commission. The glucose oxidase activity and the concentration of xylitol and sorbitol were determined according to the instructions of Megazyme International kits CAT.#: K-GLOX and CAT.#: K-SORB, respectively. The statistical package for social sciences (SPSS)-version 20 was applied to analyze the obtained results.Results.The floral origin analysis showed that the honey samples were Acacia (8) and Zizphus (4). The altitudes of the Acacia honey samples were 900 and 2000 m above sea level while the Zizphus honey samples were collected from 900 m above sea level. The studied quality parameters (Glucose, PH, Acidity, and EC.) were within the range of the CODEX standards and the Saudi Food and Drug Authority guide for honey. Irrespective of the altitude, the glucose oxidase activity in the Acacia honey samples was (4.7 ± 1.13) whereas its activity of the Zizphus honey was (5.0 ± 1.62). There was insignificant difference between the glucose activity of the Acacia and Zizphus honey samples (p-value = 0.7). The concentration of xylitol and sorbitol were not affected by the floral origin (p-value ≥ 0.17).Conclusions.The Acacia and Ziziphus honey are characterized by containing high amounts of sorbitol and trace xylitol content. The floral origin and year of harvest significantly affected pH, EC and Acidity of the honey samples.

Honey bee queen serves as a colony's centre hub, producing eggs and releasing pheromones to keep a colony together. Artificial insemination in Apis mellifera, trending research these days, is a consistent method for controlling mating in honey bees. Quality queen production is essential to maintaining a healthy bee colony. A review of comparative studies on superior quality honey bee (A. mellifera) queens production through nontraditional techniques and various factors affecting the instrumental insemination process has been compiled in this article. Several factors like; rearing conditions, stress, inseminator’s skills, food availability, mating age of queen, queen banking, temperature, sperms stored in the spermatheca, sperms quality and quantity, semen handling and storage, CO2 and nitrogen treatments, pheromones affect the performance of instrumentally inseminated queens. Moreover, different data collection showed that different methods used for queen treatment which have a significant effect on the performance of the queen. This review suggested that artificially inseminated queens could be more productive if experiments were done with proper attention and precision.

Artificial queen rearing has changed the beekeeping business in contrast to natural queen replacement, as it provides a newly mated queen into a bee colony, reduces the time between eggs not being laid, and increases the production of young bees in a colony. This study was conducted to investigate the larval acceptance rate grafted in queen cups made from different materials and to find out whether the various materials used for queen cups were suitable for the acceptance of grafted larvae in cell builder colonies. The evaluated materials included fresh bee wax (T1), old bee wax (T2), uncapping bee wax (T3), pure paraffin (T4), 50% paraffin + 50% old bee wax (T5), and brown plastic queen cups (T6) as a control. Results indicated that T3 was the material that most increased the larval acceptance (5.2 ± 0.37), followed by T4 (2.4 ± 0.5), T2 (2.2 ± 0.58), T5 (1.8 ± 0.37), T1 (1.6 ± 0.4), and minimum larval acceptance was in the T6 (1.0 ± 0.4) respectively during the spring season of the year 2020–2021. Similar findings were reported of larval acceptance rate during the spring season of 2021–2022. In addition, the wax material that least affected larval acceptance was the fresh comb bee wax. However, all materials used were coupled with a more larval acceptance rate than the control treatment during both spring seasons of 2020–2021 and 2021–2022. The larval acceptance rate was statistically significant in T3 as a compared to other queen cell cup materials during the both spring seasons (p = 0.001). These findings imply that using different types of bee wax for preparing queen cell cups during larvae grafting, particularly uncapping bee wax, may stimulate and promote grafted larvae acceptance during the queen rearing process.

The antibacterial, anticancer, and wound-healing effects of honey can vary according to the type, geographical region, honey bee species, and source of the flowers. Nanotechnology is an innovative and emerging field of science with an enormous potential role in medical, cosmetics, and industrial usages globally. Metal nanoparticles that derived from silver and range between 1 nm and 100 nm in size are called silver nanoparticles (AgNPs). Much advanced research AgNPs has been conducted due to their potential antibacterial and anticancer activity, chemical stability, and ease of synthesis. The purpose of the present study was to explore the physicochemical properties of honey and the potential to use forest honey to synthesize AgNPs as well as to appraise the nanoparticles’ antimicrobial and anticancer effects. Here, we used three different percentages of forest honey (20%, 40%, and 80%) as biogenic mediators to synthesize AgNPs at room temperature. The development of AgNPs was confirmed by color change (to the naked eye) and ultraviolet-visible spectroscopy studies, respectively. The absorbance peak obtained between 464 to 4720 nm validated both the surface plasmon resonance (SPR) band and the formation of AgNPs. Regarding the sugar profile, the contents of maltose and glucose were lower than the content of fructose. In addition, the results showed that the SPR band of AgNPs increased as the percentage of forest honey increased due to the elevation of the concentration of the bio-reducing agent. A bacterial growth kinetic assay indicated the strong antibacterial efficacy of honey with silver nanoparticles against each tested bacterial strain. Honey with nanotherapy was the most effective against hepatocellular carcinoma (HepG2) and colon cancer (HCT 116) cells, with IC50s of 23.9 and 27.4 µg/mL, respectively, while being less effective against breast adenocarcinoma cells (MCF-7), with an IC50 of 32.5 µg/mL.

Objectives: This article investigated the effect of plant origin, storage and altitude on the triglycerides, cholesterol and some physicochemical properties of honey samples. Methods: Nineteen honey samples (9 Ziziphus and 10 Acacia) were involved in this study representing two floral origins and seasons and four altitudes. The plant origin of the honey samples was confirmed microscopically and their quality parameters were analyzed according to the International Honey Commission harmonized methods of honey analysis. The lipid parameters were analyzed using spectrophotometric- enzymatic methods. The SPSS version 20 was used for the statistical analysis of the obtained data. Beeswax is a natural product that is primarily produced by honey bees of the genus Apis. It has many uses in various kinds of industries, including pharmacy and medicine. This study investigated the effect of storage and floral origin on some physicochemical properties of four beeswax samples. The floral origin of the beeswax samples was determined microscopically and the investigated physical properties were the melting point, color, surface characteristics and thermal behavior. The studied chemical constituents were the acid value, ester value, saponification value and the ester/acid ratio. The FT-IR, SEM, EDX, XRD and TGF techniques were applied to meet the objectives of this study. The physical properties of the beeswax were affected by the storage period and floral origin. The melting point of the beeswax samples significantly increased with the increase in the storage time, from 61.5 ± 2.12 °C for the 3 month sample to 74.5 ± 3.54 °C for the 2 year stored sample (p-value = 0.027). The acid values of the 3 month, 6 month, 1 year and 2 years stored samples were 19.57 ± 0.95, 22.95 ± 1.91, 27 ± 1.91 and 34.42 ± 0.95 mgKOH/g, respectively. The increase in the acid value was significant (p-value = 0.002). The ester values of the studied beeswax samples significantly increased with the increase in storage time as follows: 46.57 ± 2.86 mgKOH/g for the 3 month stored sample, 66.14 ± 3.82 mgKOH/g for the 6 month stored sample, 89.77 ± 0.95 mgKOH/g for the one year stored sample and 97.19 ± 1.91 mgKOH/g for the 2 year stored sample (p-value ≤ 0.001). Similarly, the saponification value and the carbon percentages increased with the increase in storage time. Unlike the results of the chemical components, the oxygen percentage decreased with the increase in storage time as follows: 11.24% (3 month), 10.31% (6 month), 7.97% (one year) and 6.74% (two year). The storage and floral origin of beeswax significantly affected its physicochemical properties in a way that qualify it to act as a phase changing material in the thermal storage energy technology.

Propolis (bee glue) is a complex, phyto-based resinous material obtained from beehives. Its chemical and biological properties vary with respect to bee species, type of plants, geographical location, and climate of a particular area. This study was planned with the aim of determining the chemical composition and to investigate various properties (against oxidants and microbes) of different extracts of Saudi propolis collected from Arabian honey bee (Apis mellifera jemenitica) colonies headed by young queens. Chemical analysis of propolis extracts with different solvents, i.e., ethyl acetate (Eac), methanol (Met), butanol (BuT), and hexane (Hex) was done through colorimetry for the total phenolic content (TPC) and total flavonoid content (TFC) evaluation. For separation and extensive characterization of the Met extract, chromatography and 1H NMR were deployed. Six different microorganisms were selected to analyze the Saudi-propolis-based extract’s antimicrobial nature by measuring zones of inhibition (ZOI) and minimum inhibitory concentration (MIC). Molecular docking was done by utilizing AutodDock, and sketching of ligands was performed through Marvin Chem Sketch (MCS), and the resultant data after 2D and 3D clean were stored in .mol format. The highest TFC (96.65 mg quercetin equivalents (QE)/g of propolis) and TPC (325 mg gallic acid equivalents (GAE)/g of propolis) were noted for Met. Six familiar compounds were isolated, and recognition was done with NMR. Met extract showed the greatest 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) free radical scavenging activity and Ferric Reducing Antioxidant Power (FRAP). Met showed max microbial activity against Staphylococcus aureus (ZOI = 18.67 mm, MIC = 0.625 mg/mL), whereas the minimum was observed in Hex against E. coli (ZOI = 6.33 mm, MIC = 2.50 mg/mL). Furthermore, the molecular docking process established the biological activity of separated compounds against HCK (Hematopoietic cell kinase) and Gyrase B of S. aureus. Moreover, the stability of protein–ligand complexes was further established through molecular dynamic simulation studies, which showed that the receptor–ligand complexes were quite stable. Results of this research will pave the way for further consolidated analysis of propolis obtained from Arabian honey bees (A. m. jemenitica).

The activity of honey enzymes are affected by floral and geographical origins, climate conditions, honeybee species, health and nutrition. This article investigated the effect of floral and geographical origins on the activity of glucose oxidase (GOx) and catalase (CAT) enzymes in honey samples from the southwestern region of Saudi Arabia. Moreover, the moisture, total sugars, pH and conductivity were measured as quality parameters. The floral origin of the honey samples was determined microscopically while the quality parameters were measured according to the methods of international honey commission. The activity of the honey enzyme was determined following the instructions of the Megazyme International kits. The obtained results were statistically analyzed by the statistical Package for Social Sciences (SPSS, v.20). The GOx activity of the Acacia, Ziziphus and polyfloral honey samples of the Asir region were (5.19 ± 2.33 U/g), (4.01 ± 1.17 U/g) and (5.69 ± 1.67 U/g), respectively. The Acacia, Ziziphus and polyfloral honey samples from the Jazan region had GOx activities of (6.85 ± 0.47 U/g), (10.48 ± 9.22 U/g) and (5.31 ± 2.7 U/g), respectively. The geographical origin significantly affected the GOx activity of Ziziphus honey (p-value = 0.005) and the GOx activity of the Ziziphus honey was significantly more than that of the polyfloral honey of the Jazan region (p-value = 0.009). With regard to the CAT activity in Asir region honey samples, the mean values of the Acacia, Ziziphus and polyfloral honeys were (2.89 ± 1.08 U/g), (3.58 ± 1.59 U/g) and (2.84 ± 1.24 U/g), respectively. The mean values of the CAT activity in the Jazan honey samples were Acacia (4.35 ± 1.01 U/g), Ziziphus (3.94 ± 0.04 U/g) and polyfloral (3.43 ± 0.67 U/g). The geographical origin significantly affected the CAT activity in Acacia honey (p-value = 0.014). The geographical and floral origins had significant effects on the activity of the honey GOx and CAT enzymes.

Honey is known for its content of biomolecules, such as enzymes. The enzymes of honey originate from bees, plant nectars, secretions or excretions of plant-sucking insects, or from microorganisms such as yeasts. Honey can be characterized by enzyme-catalyzed and non-enzymatic reactions. Notable examples of enzyme-catalyzed reactions are the production of hydrogen peroxide through glucose oxidase activity and the conversion of hydrogen peroxide to water and oxygen by catalase enzymes. Production of hydroxymethylfurfural (HMF) from glucose or fructose is an example of non-enzymatic reactions in honey.

Objective This study investigated immunohistochemical staining results of two cytoskeletal proteins (vimentin and cytokeratin-18) and two extracellular matrix proteins (fibronectin-1 and laminin-1 receptor) in different stages of triple negative breast cancer. Methods Forty triple negative cancerous breast tissues from patients diagnosed as stage 2A (15), 2B (nine), 3A (10), 3B (four), and 3C (two) were included in this study and were compared with 42 normal breast tissues. Immunohistochemistry results were statistically analyzed using the t-test percent of the StatPac program. Results The percentages of positive staining in cancerous tissues for all of the studied parameters were significantly greater than their percentages in normal tissues, except for vimentin. All cancerous tissues from patients diagnosed as stage 3A, 3B, and 3C were positive for both fibronectin-1 and laminin-1 receptor. Conclusion Fibronectin-1 and laminin-1 receptor are promising markers for stage 3 triple negative breast cancer.

The physiochemical properties of six Acacia honey samples taken from three different altitudes in the Asir region were determined. The means of all the studied parameters were within the international standards and were comparable to previous studies (0.11 ± 0.08% for ash, 10.93 ± 1.97% for water, 3.56 ± 0.19 for pH, 38.63 ± 17.17 meq/kg for acidity, 659.51 ± 324.98 μS/cm for conductivity, and 1.45 ± 0.02 for specific gravity). Comparison of the mean values of the parameters at the different altitudes revealed gradual increases with increasing altitude, except for ash and specific gravity. Significant differences were seen in ash and water percentages, acidity, and conductivity. While the mean values of the studied physiochemical properties of the Acacia honey samples were within the ranges of international standards, the honey produced at high altitudes exhibited variable physiochemical properties.

Smoking is well known to be correlated with cardiovascular abnormalities, in particular atherosclerosis and heart diseases. This article investigates the effect and relationship of smoking tobacco on the thickness of the intima–media (IMT) belonging to the common carotid arteries (CCAs), and also blood concentration of the lipid profile (LP), mainly the total cholesterol, High Density Lipoprotein (HDL), Low Density Lipoprotein (LDL) and also triglycerides. Nineteen male tobacco smokers and thirty-five healthy male non-smoking Saudi participants were involved in this study after obtaining their informed consent. An ultrasound and a spectrophotometer were used to determine the IMTs and lipid parameters, respectively. The thicknesses of the smokers’ right (RCA) and left carotid (LCA) arteries (0.72 and 0.7 mm, respectively) were significantly greater than the thicknesses of the arteries of the non-smokers (0.58 and 0.62 mm, respectively) (p-value = 0.005 and 0.04). Insignificant differences between the means of the other parameters in the two groups were studied. Smoking is a risk factor for stroke, because it significantly increases the IMTs of both the right and left carotid arteries.

One of the major duties of any lecturer is to adopt different teaching methods in order to obtain excellent student performance. This study was conducted to investigate the performance of students in a seminar based examination compared to a lecture and practical sessions based examination. The basic course of biochemistry, taught to 35 students of chemistry and biology of the faculty of science at King Khalid University, was conducted following three teaching methods; lectures, practical sessions and seminar based teaching. Vitamins were taught through seminars prepared and presented by the students while the amino acids, protein and enzymes course was delivered through lectures and practical sessions. The final exam of the course included questions testing the knowledge of the students concerning the seminar and lecture based information. Regarding the results of the seminar based questions; 15 (42.9%) passed the exam and the range of the marks was (25- 90) while the students who passed the lecture and practical sessions based questions were 17 (48.6%) and the range of the marks was (35- 80). However, the pass mark was 60%. Although the highest marks were scored in the seminar based questions, the general performance of the students in the seminar based questions was poor compared to the lecture and practical sessions based questions.
