1. Insights into the role of hexa-bacterial consortium for bioremediation of soil contaminated with chlorantraniliprole

Abstract: Abstract This study evaluates the efficacy of discrete bacterial consortia in bioremediating sandy loamy soil deliberately contaminated with 20 mg/kg of chlorantraniliprole (CAP). It monitors alterations in total bacterial populations and CO2 emissions, tracking residual CAP levels through UV scanning and HPLC analysis. Six active bacterial degraders (four Bacillus strains (B. subtilis subsp. subtilis AZFS3, B. pumilus AZFS5, B. mojavensis AZFS15, and B. paramycoides AZFS18), one Alcaligenes strain (A. aquatilis KZFS11), and one Pseudomonas strain (P. aeruginosa KZFS4)) were used in single or combined preparations and grown on trypticase soy broth for 24 h at 30 °C before preparing the inoculants and adjusting the bacterial cell count to 107 CFU/dwt g soil. The bacterial consortia were added to the CAP-contaminated soil and incubated for 20 days at 30 °C. The di-, tetra-, and hexa-bacterial consortia recorded the highest levels of viable bacteria, reaching their peak after 3 to 11 days of incubation. Then, they declined to the minimum levels at the end of the 20 days, which coincided with their complete removal of CAP from the soil. At the end of the incubation period (20 days), the CAP was mainly biodegraded, scoring biodegradation rates of 90.05%, 93.65%, and 98.65% for T3, T4, and T5, respectively. This concurred with the highest average CO2 production. Based on the results of the HPLC analysis, the hexa-bacterial consortium T5 demonstrated the highest rate of CAP biodegradation (99.33%) after a 20-day incubation period, resulting in the lowest residual level of CAP in the soil (0.67%). Bioinformatic analysis predicted that the CAP biodegradation pathway reached CO2 and H2O. Under optimized conditions, the hexa-bacteria consortium is the most effective CAP biodegraded and is recommended as an eco-friendly treatment for eliminating CAP pollution in the field.

Abstract: Autoreclosure is an irreplaceable protective scheme that was initially engineered for power system stability enhancement especially in areas with high isokeraunic levels. Unsuccessful reclosure on transmission lines emanating from thermal power plants has been long identified as a potential hazard to the shafts of the turbine-generators in these power plants. Therefore, the principal focus of this work is to mitigate the shaft torsional oscillations resulting from unsuccessful reclosure. The main scope of this study is to utilize a battery energy storage system controlled via fuzzy logic controllers to mitigate shaft torsional oscillations resulting from unsuccessful reclosure. Additionally, a coordinated operation of fuzzy based dynamic braking resistor is considered to obtain a superior torsional torque profile. Local control input signal synthesized from the generator shaft speed is employed in this work for the proposed controllers. For testing the effectiveness of the proposed scheme, non-linear time-domain simulation study is performed on a single machine infinite bus and the IEEE 3-machine 9-bus system via MATLAB/Simulink-based modelling and simulation platform. Comparative simulation studies of the test systems after being subjected to unsuccessful reclosure of three-phase to ground fault conditions demonstrate the effectiveness of proposed scheme. From the time domain simulation results, the torsional torque profiles reach an excellent level due to the implementation of the proposed scheme.

3. Prediction of a positive surgical margin and biochemical recurrence after robot-assisted radical prostatectomy

Abstract: Abstract The positive surgical margin (PSM) and biochemical recurrence (BCR) are two main factors associated with poor oncotherapeutic outcomes after prostatectomy. This is an Asian population study based on a single-surgeon experience to deeply investigate the predictors for PSM and BCR. We retrospectively included 419 robot-assisted radical prostatectomy cases. The number of PSM cases was 126 (30.1%), stratified as 22 (12.2%) in stage T2 and 103 (43.6%) in stage T3. Preoperative prostate-specific antigen (PSA) > 10 ng/mL (p = 0.047; odds ratio [OR] 1.712), intraoperative blood loss > 200 mL (p = 0.006; OR 4.01), and postoperative pT3 stage (p 10 ng/mL (p 3 (p = 0.02; HR 1.964), and PSM (p = 0.027; HR 1.725) were four significant predictors for BCR in multivariable analysis. PSMs occurred mostly in the posterolateral regions (73.8%) which were associated with nerve-sparing procedures (p = 0.012) while apical PSMs were correlated intraoperative bleeding (p 3 (p = 0.002; HR 2.689) was a significant BCR predictor. These results indicate that PSA and pathological status are key factors influencing PSM and BCR.

Abstract: Abstract The work presented herein is focusing on mitigating sub-synchronous-resonance (SSR) oscillatory torque and speed responses developed in power network equipped with series capacitor compensation as an outcome of the network perturbations. The mitigation effect of a battery-energy-storage (BES) controlled via a fuzzy-logic-controller (FLC) is explored. It is also explored accompanied by a fuzzy-bases resistor brake controlled via FLC. The IEEE second benchmark system is selected as a test grid under the MATLAB™/Simulink simulation environment. The signal utilized for the propositioned controller is the alternator rotor speed deviation. The key conclusions of this investigation are the employment of a BES in the discharging mode alone might be utilized to alleviate the SSR relative speed, and torque oscillations, the BES employment accompanied with dynamic resistive brake is supplying faster-decaying rates for the SSR oscillations in observation with the BES employment only.

Abstract: Abstract Inter-area oscillations are, by far, the most detrimental oscillation category to the integrity of synchronously interconnected power systems. Inter-area oscillations are characterized by the inherent weak damping. The inherent poor damping associated with the inter-area oscillations leaves open wide probabilities for irrevocable widespread blackouts with the consequent eventual devastating outcomes measured in terms of the huge economic casualties and the possible human fatalities. The main purpose of this work is to mitigate inter-area power oscillations. This article explores the effectiveness of dual thyristor controlled braking resistor units with Interval Type-2 fuzzy-based centralized architecture for neutralizing the jeopardy of inter-area power oscillations in Kundur’s two-area test system using MATLAB™/Simulink environment. The effectiveness of the proposed scheme is examined by considering four case studies with different degrees of severity. The simulation results show that the proposed scheme is simple yet effective in treating the inter-area oscillations appropriately under the considered case studies.

6. Cross Cultivation on Homologous/Heterologous Plant-Based Culture Media Empowers Host-Specific and Real Time In Vitro Signature of Plant Microbiota

Abstract: Alliances of microbiota with plants are masked by the inability of in vitro cultivation of their bulk. Pure cultures piled in international centers originated from dissimilar environments/hosts. Reporting that plant root/leaf-based culture media support the organ-specific growth of microbiota, it was of interest to further investigate if a plant-based medium prepared from homologous (maize) supports specific/adapted microbiota compared to another prepared from heterologous plants (sunflower). The culture-independent community of maize phyllosphere was compared to communities cross-cultivated on plant broth-based media: CFU counts and taxa prevalence (PCR-DGGE; Illumina MiSeq amplicon sequencing). Similar to total maize phyllospheric microbiota, culture-dependent communities were overwhelmed by Proteobacteria (>94.3–98.3%); followed by Firmicutes (>1.3–3.7%), Bacteroidetes (>0.01–1.58%) and Actinobacteria (>0.06–0.34%). Differential in vitro growth on homologous versus heterologous plant-media enriched/restricted various taxa. In contrast, homologous cultivation over represented members of Proteobacteria (ca. > 98.0%), mainly Pseudomonadaceae and Moraxellaceae; heterologous cultivation and R2A enriched Firmicutes (ca. > 3.0%). The present strategy simulates/fingerprints the chemical composition of host plants to expand the culturomics of plant microbiota, advance real-time in vitro cultivation and lab-keeping of compatible plant microbiota, and identify preferential pairing of plant-microbe partners toward future synthetic community (SynComs) research and use in agriculture.

Abstract: Plants from the genus Astragalus are gaining attention for their pharmacological importance. However, the information available regarding the HPLC–MS/MS chemical profile of A. fruticosus is inadequate. In this study, we performed HPLC–MS/MS analysis using electrospray ionization (ESI) and atmospheric pressure chemical ionization (APCI). We tentatively identified 11 compounds in the A. fruticosus methanolic extract, including five flavonoidal and six saponin glycosides. The extract showed moderate antioxidant activity with 21.05% reduction in DPPH UV absorption. The preliminary cytotoxic screening against seven human cancer cell lines using 100 μg/mL extract showed prominent cytotoxic potential against colorectal cancer HCT–116 with 3.368% cell viability. It also showed moderate cytotoxic potential against prostate (DU–145), ovarian (SKOV–3) and lung (A–549) cancer cell lines with cell viability of 14.25%, 16.02% and 27.24%, respectively. The IC50 of the total extract against HCT–116 and DU–145 cell lines were 7.81 μg/mL and 40.79 μg/mL, respectively. The observed cytotoxicity of the total methanolic extract from the leaves against colorectal cancer might facilitate future investigations on cytotoxic agent(s) for disease management.

Abstract: Plant microbiota have co-evolved with their associated plants in the entire holobiont, and their assemblages support diversity and productivity on our planet. Of importance is in vitro cultivation and identification of their hub taxa for possible core microbiome modification. Recently, we introduced the in situ-similis culturing strategy, based on the use of plant leaves as a platform for in vitro growth of plant microbiota. Here, the strategy is further extended by exploring plant organ compatible cultivation of plant microbiota when grown on corresponding leaf/root-based culture media. Pooling the advantages of MPN enrichment methodology together with natural plant-only-based culture media, the introduced method efficiently constructed a nutritional milieu governed by vegan nutrients of plant origin, i.e., leaf strips/root segments, immersed in plain semi-solid water agar. MPN estimates exceeded log 7.0 and 4.0 g−1 of endo-rhizosphere and endo-phyllosphere, respectively, of maize and sunflower; being proportionate to those obtained for standard culture media. With sunflower, PCR-DGGE analyses indicated divergence in community composition of cultivable endophytes primarily attributed to culture media, signaling a certain degree of plant organ affinity/compatibility. Based on 16S rRNA gene sequencing of bacterial isolates, 20 genera comprising 32 potential species were enriched; belonged to Bacteroidetes, Firmicutes, and Alpha-/Gammaproteobacteria. The described cultivation strategy furnished diversified nutritive platform in terms of homologous/heterologous plant organ-based medium and ambient/limited oxygenic cultivation procedure. Duly, cultivability extended to > 8 genera: Bosea, Brevundimonas, Chitinophaga, Pseudoxanthomonas, Sphingobacterium Caulobacter, Scandinavium, and Starkeya; the latter three genera were not yet reported for Sunflower, and possible unknown species or even one new putative genus. Thus, both potential members of the major microbiome and rare isolates of satellite microbiomes can be isolated using the presented method. It is a feasible addition to traditional cultivation methods to explore new potential resources of PGPB for future biotechnological applications.

Abstract: Improving cultivability of a wider range of bacterial and archaeal community members, living natively in natural environments and within plants, is a prerequisite to better understanding plant-microbiota interactions and their functions in such very complex systems. Sequencing, assembling, and annotation of pure microbial strain genomes provide higher quality data compared to environmental metagenome analyses, and can substantially improve gene and protein database information. Despite the comprehensive knowledge which already was gained using metagenomic and metatranscriptomic methods, there still exists a big gap in understanding in vivo microbial gene functioning in planta, since many differentially expressed genes or gene families are not yet annotated. Here, the progress in culturing procedures for plant microbiota depending on plant-based culture media, and their proficiency in obtaining single prokaryotic isolates of novel and rapidly increasing candidate phyla are reviewed. As well, the great success of culturomics of the human microbiota is considered with the main objective of encouraging microbiologists to continue minimizing the gap between the microbial richness in nature and the number of species in culture, for the benefit of both basic and applied microbiology. The clear message to fellow plant microbiologists is to apply plant-tailored culturomic techniques that might open up novel procedures to obtain not-yet-cultured organisms and extend the known plant microbiota repertoire to unprecedented levels. 10. 'In situ similis' Culturing of Plant Microbiota: A Novel Simulated Environmental Method Based on Plant Leaf Blades as Nutritional Pads

Abstract: High-throughput cultivation methods have recently been developed to accelerate the recovery of microorganisms reluctant to cultivation. They simulate in situ environmental conditions for the isolation of environmental microbiota through the exchange of growth substrates during cultivation. Here, we introduce leaf-based culture media adopting the concept of the plant being the master architect of the composition of its microbial community. Pre-physical treatments of sunflower plant leaves, namely punching, freezing, and/or autoclavation, allowed the diffusion of electrolytes and other nutrients to configure the leaf surface as a natural pad, i.e., creating an “in situ similis” environment suitable for the growth of rarely isolated microbiota. We used surface inoculation and membrane-filtration methods to assess the culturability of endophytic bacteria from the sunflower phyllosphere and rhizosphere. Both methods supported excellent colony-forming unit (CFU) development when compared to standard R2A medium, with a special affinity to support better growth of epiphytic and endophytic populations of the phyllosphere compared with the rhizosphere. A 16S rRNA gene analysis of >122 representative isolates indicated the cultivation of a diverse set of microorganisms by application of the new methods. It indicated the predominance of 13 genera of >30 potential species, belonging to Firmicutes, Proteobacteria, and Actinobacteria, and especially genera not commonly reported for sunflower, e.g., Rhizobium, Aureimonas, Sphingomonas, Paracoccus, Stenotrophomonas, Pantoea, Kosakonia, and Erwinia. The strategy successfully extended diversity and richness in the endophyllosphere compared to the endorhizosphere, while CFUs grown on the standard R2A medium mainly pertain to Firmicutes, especially Bacillus spp. MALDI-TOF MS analysis clustered the isolates according to their niche and potential functions, where the majority of isolates of the endorhizosphere were clustered away from those of the endophyllosphere. Isolates identified as Gammaproteobacteria and Alphaproteobacteria were distinguishably sub-clustered, which was in contrast to the heterogeneous isolates of Firmicutes (Bacillus spp.). In conclusion, leaf in situ similis cultivation is an effective strategy to support the future application of culturomics of plant microbiota. This is an effort to access novel isolates that are more adapted and competitive in their natural environments, especially those subjected to abiotic stresses like those prevailing in arid/semi-arid zones, and, consequently, to support the application of agro-biotechnologies, among other technologies, to improving agriculture in such zones.

Abstract: The plant-based-sea water culture medium is introduced to in vitro cultivation and in situ recovery of the microbiome of halophytes. The ice plant (Mesembryanthemum crystallinum) was used, in the form of juice and/or dehydrated plant powder packed in teabags, to supplement the natural sea water. The resulting culture medium enjoys the combinations of plant materials as rich source of nutrients and sea water exercising the required salt stress. As such without any supplements, the culture medium was sufficient and efficient to support very good in vitro growth of halotolerant bacteria. It was also capable to recover their in situ culturable populations in the phyllosphere, ecto-rhizosphere and endo-rhizosphere of halophytes prevailing in Lake Mariout, Egypt. When related to the total bacterial numbers measured for Suaeda pruinosa roots by quantitative-PCR, the proposed culture medium increased culturability (15.3–19.5%) compared to the conventional chemically-synthetic culture medium supplemented with (11.2%) or without (3.8%) NaCl. Based on 16S rRNA gene sequencing, representative isolates of halotolerant bacteria prevailed on such culture medium were closely related to Bacillus spp., Halomonas spp., and Kocuria spp. Seed germination tests on 25–50% sea water agar indicated positive interaction of such bacterial isolates with the germination and seedlings’ growth of barley seeds.

Abstract: Plant microbiota support the diversity and productivity of plants. Thus, cultivation-dependent approaches are indispensable for in vitro manipulation of hub taxa. Despite recent advances in high-throughput methods, cultivability is lagging behind other environmental microbiomes, notably the human microbiome. As a plant-based culturing strategy, we developed culture media based on a broth of cooked aqueous mixtures of host plants. This improved the in vitro growth of representative isolates of plant microbiota and extended the in situ recovery of plant microbiota. With clover, 16S rRNA gene sequencing of representative isolates confirmed the predominance of Firmicutes, Alphaproteobacteria and Gammaproteobacteria, and less frequently Bacteroidetes and Actinobacteria. Whereas bovine-based culture media (modified R2A) confined the diversity to Firmicutes, the plant broth-based culture media revealed a wider scope of endophytes beyond rhizobia, i.e., multiple genera such as Chryseobacterium, Cronobacter, Kosakonia, Tsukamurella, and a potentially/presumptive novel species. Matrix-assisted laser desorption/ionization time-of-flight (MADI-TOF) analysis clustered isolates according to their plant niches, the endo-phyllosphere/endo-rhizosphere. We recommend the plant broth for simplicity, reproducibility and perdurable storage, supporting future culturomics applications, good laboratory practice (GLP) and good manufacturing practice (GMP). The strategy creates an “in-situ-similis” vegan nutritional matrix to analyze microbial diversity and reveal novel microbial resources pertinent to biotechnological and environmental applications.

Abstract: In an effort to axenically culture the previously uncultivable populations of the rhizobacteria of Lucerne (Medicago sativa L.), we propose plant-only teabags culture media to mimic the nutritional matrix available in the rhizosphere. Here, we show that culture media prepared from Lucerne powder teabags substantially increased the cultivability of Lucerne rhizobacteria compared with a standard nutrient agar, where we found that the cultivable populations significantly increased by up to 60% of the total bacterial numbers as estimated by Quantitative Real-time Polymerase Chain Reaction (qRT-PCR). Cluster analysis of 16S rDNA Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis (PCR-DGGE) of cultivable Colony-Forming Units (CFUs) revealed a more distinct composition and separation of bacterial populations recovered on the plant-only teabags culture media than those developed on a standard nutrient agar. Further, the new plant medium gave preference to the micro-symbiont Sinorhizobium meliloti, and succeeded in isolating a number of not-yet-cultured bacteria, most closely matched to Novosphingobium sp., Lysobacter sp. and Pedobacter sp. The present study may encourage other researchers to consider moving from the well-established standard culture media to the challenging new plant-only culture media. Such a move may reveal previously hidden members of rhizobacteria, and help to further explore their potential environmental impacts.

14. Bio-preparates support the productivity of potato plants grown under desert farming conditions of north Sinai: Five years of field trials

Abstract: Organic agriculture as well as good agricultural practices (GAPs) intrigues the concern of both consumers and producers of agricultural commodities. Bio-preparates of various rhizospheric microorganisms (RMOs) are potential sources of biological inputs supporting plant nutrition and health. The response of open-field potatoes to the application of RMO bio-preparates, the biofertilizer “Biofertile” and the bioagent “Biocontrol”, were experimented over 5 successive years under N-hunger of north Sinai desert soils. Both vegetative and tuber yields of a number of tested cultivars were significantly improved due to rhizobacterial treatments. In the majority of cases, the biofertilizer “Biofertile” did successfully supply ca. 50% of plant N requirements, as the yield of full N-fertilized plants was comparable to those received 50% N simultaneously with bio-preparates treatment. The magnitude of inoculation was cultivar-dependent; cvs. Valor and Oceania were among the most responsive ones. Bio-preparate introduction to the plant–soil system was successful via soaking of tubers and/or spraying the plant canopy. The “Biocontrol” formulation was supportive in controlling plant pathogens and significantly increased the fruit yields. The cumulative effect of both bio-preparates resulted in tuber yield increases of ca. 25% over control.

Abstract: More than 12 × 109 m3/year of Nile Delta drainage water is annually discharged into the Mediterranean Sea. El-Salam (peace) canal, having a mixture of such drainage water and the Nile water (1:1 ratio), crosses the Suez canal eastward to the deserts of north Sinai. The suitability of the canal water for agriculture is reported here. Representative samples were obtained during two successive years to follow effects of seasonal and spatial distribution, along the first 55 km course in north Sinai, on the water load of total bacteria, bacterial indicators of pollution, and chemical and heavy metals contents. In general, the canal water is acceptable for irrigation, with much concern directed towards the chemical contents of total salts (EC), Na and K, as well as the trace elements Cd and Fe. Extending the canal course further than 30 km significantly lowered the fecal pollution rate to the permissible levels of drinking water. Results strongly emphasize the need for effective pre-treatment of the used drainage water resources prior mixing with the Nile water.

16. The crude plant juices of desert plants as appropriate culture media for the cultivation of rhizospheric microorganisms

Abstract: The exclusive use of plant juices, not as a mere supplement to synthetic culture media, for culturing rhizospheric microorganisms (RMO) is introduced here. Juices were prepared from desert (Mesembryanthemum crystallinum L., Zygophyllum album L., Carpobrotus edulis L.) as well as cultivated (Trifolium alexandrinum L., Beta vulgaris L.) plants. Colonies of RMO (Azospirillum brasilense, Enterobacter agglomerans and Klebsiella pneumoniae) nicely developed on surface-inoculated agar plates prepared from crude and diluted juice of M. crystallinum (ice plant). Furthermore, hundreds of RMO colonies developed on various standard culture media were replicated (>90%) on agar plates of different plant juices. RMO cells grew nicely in liquid ice plant juice, with doubling times comparable to those grown in the reference culture medium. RMO populations resident in various host plants were able to develop on culture media prepared from homologous and heterologous juices. The application of a thin semi-solid overlay agar on the surfaces of inoculated agar plates significantly increased the recovery of micro-colonies on agar plates, particularly those prepared from plant juices.

18. Bio-inspired systems for treatment of diabetes

Abstract: There have been several technological advancements aimed at improving glycaemic control in people with Type 1 diabetes such as continuous glucose monitors (CGM) and continuous subcutaneous insulin infusion pumps (CSII). However, even with these advances, the millions of people living with diabetes struggle to keep their glucose levels within target range (4-8 mM). There are various reasons for this sub-optimal control of glucose levels which include lack of patient compliance, and most importantly, determining the amount of insulin needed to maintain glycaemic control is not a trivial task since glucose levels are affected by many different parameters. Since a healthy pancreas maintains optimal glycaemic control, a bio-inspired system that replicates the natural mechanisms of glucose sensing and physiological insulin secretion would be ideal for maintaining glycaemic control in people with Type 1 diabetes. Towards this aim, in this thesis a Bio-inspired Artificial Pancreas (BiAP) has been designed and implemented. It runs on dedicated hardware, and incorporates a bio-inspired controller that replicates the insulin-secreting physiology of the pancreatic β-cells. The controller runs on an embedded microchip and the system has been optimized for low power operation. In addition a glucagon controller is used with the aim of reducing hypoglycaemic episodes. The low-power, hand-held unit (BiAP) interfaces to a continuous glucose sensor (Medtronic Enlite or Dexcom G4) and sends the required insulin and glucagon dose information to two subcutaneous pumps (Roche Accu-Chek Combo). The BiAP has undergone over 250 hours of closed loop studies at the NIHR/Wellcome Trust clinical research facility at Hammersmith hospital. The results show that the BiAP is capable of achieving good control with both the single and dual hormone systems. Additionally, the hand-held unit achieves power efficient operation, lasting for 54 hours during full control. Furthermore, it was identified that one of the challenges to the successful realisation of an artificial pancreas system is sensor accuracy. Therefore, a bio-inspired methodology for improving glucose sensor performance is presented. Specifically, the noise shaping capability of gap junction coupling is utilised to synchronise β-cell behaviour to a varying glucose sensor input and reduce random noise. In addition, a CMOS chip was fabricated containing 4 β-cell circuits, which were then coupled off chip. These improvements will allow more accurate glucose sensing for use in an artificial pancreas.

Abstract: Nerve-sparing (NS) techniques could potentially increase positive surgical margins after robot-assisted radical prostatectomy (RARP). Nevertheless, the available studies have revealed ambiguous results among distinct groups. This study purposed to clarify the details of NS techniques to accurately estimate their influence on margin status.We studied RARPs performed by one surgeon from 2010 to 2018. Surgical margins were evaluated by the laterality and levels of NS techniques in site-specific prostate lobes. The multivariable analysis evaluated the effects of nerve-sparing procedures, combined with other covariate factors, on margin status.Overall, 419 RARPs involving 838 prostate lobes were analyzed. Notably, 181 patients (43.4%) had pT2-stage, and 236 (56.6%) had pT3-stage cancer. The PSM rates for patients who underwent unilateral, bilateral, and non NS procedures were 30.3%, 28.8%, and 50%, respectively ( p = 0.233) or in stratification by pT2 ( p = 0.584) and pT3 ( p = 0.116) stage. The posterolateral PSM rates among site-specific prostate lobes were 10.9%, 22.4%, and 18.9% for complete, partial, and non NS techniques, respectively ( p = 0.001). The partial NS group revealed a significant increase in PSM rate compared with the complete NS (OR 2.187, 95% CI: 1.19-4.03) and non NS (OR 2.237, 95% CI: 1.01-4.93) groups in site-specific prostate lobes.Partial NS procedures have a potential risk of increasing the positive surgical margins rate than complete and non NS procedures do. Therefore, correct case selection is required before performing partial NS techniques.

Abstract: Background: Guidelines for treatment of community acquired pneumonia suggests empirical therapy with combination of clarithromycin and ceftriaxone when infection with mycoplasma pneumoniae is considered. But there is limited evidence available. Aim: To ascertain effectiveness of ceftriaxone alone verses the combination of ceftriaxone and clarithromycin with respect to cure and length-of-stay and total hospital cost. Methodology: A randomized control trial with 128 patients of age 2 months to 5 years was conducted at Department of Paediatrics, Shaikh Zayed Hospital Lahore from 1st October 2017 to 30th September 2018. Patients selected according to selection criteria through non probability sampling technique and randomly divided into two groups. Antibiotics given according to study objectives, comparative analysis done, and chi square test applied. Results: The mean age of patients were 27.02±19.28 months in group A and 29.06±18.29 months in group B. In both groups most of the patients were under the age of 40 months. 72% cured in group A while 86% cured in group B. 80% patients stayed in hospital for 3 days in Group A while, 59% patients in group B. 69% patients had cost effectivity in group A while 84% in group B. Conclusion: The combination of Ceftriaxone and Clarithromycin was more advisable in treating pnemounia verses the use of ceftriaxone alone. Abstract: Capturing the diverse microbiota from healthy and/or stress resilient plants for further preservation and transfer to unproductive and pathogen overloaded soils, might be a tool to restore disturbed plant–microbe interactions. Here, we introduce Aswan Pink Clay as a low-cost technology for capturing and storing the living root microbiota. Clay chips were incorporated into the growth milieu of barley plants and developed under gnotobiotic conditions, to capture and host the rhizospheric microbiota. Afterward, it was tested by both a culture-independent (16S rRNA gene metabarcoding) and -dependent approach. Both methods revealed no significant differences between roots and adjacent clay chips in regard total abundance and structure of the present microbiota. Clay shaped as beads adequately supported the long-term preservation of viable pure isolates of typical rhizospheric microbes, i.e. Bacillus circulans, Klebsiella oxytoca, Sinorhizobium meliloti, and Saccharomyces sp., up to 11 months stored at −20°C, 4°C, and ambient temperature. The used clay chips and beads have the capacity to capture the root microbiota and to long-term preserve pure isolates. Hence, the developed approach is qualified to build on it a comprehensive strategy to transfer and store complex and living environmental microbiota of rhizosphere toward biotechnological application in sustainable plant production and environmental rehabilitation.

Abstract: Turbine generator shaft torsional oscillations is an interdisciplinary power system dynamic problem as it involves mechanical and electrical engineering. Torsional oscillations occur in the mechanical for electrical reasons. Torsional oscillations cause fatigue life expenditure of the mechanical shaft system. There have been great motivations to mitigate the shaft torsional oscillations especially when unrestricted high speed reclosure (HSR) is utilized on the overhead transmission lines emanating from a generation station. Mitigation of torsional oscillation compromises between the use of HSR and preserving the mechanical integrity of the involved turbine generator set. Therefore, braking resistor (BR) controlled by fuzzy logic controller is presented in this paper as a low cost, reliable mean for torsional oscillations mitigation. BR was first utilized for the system transient stability enhancement. It serves as an extra load capable of dissipating extra generated power in case of system severe faults close to a generation station consequently prevents generator pole slipping conditions. IEEE 3 machine 9 bus system is adopted in this paper to test the effects of BR on shaft torsional oscillations mitigation in interconnected power system. Comparative simulation studies between the unsuccessful reclosure with and without fuzzy controlled BR prove the effectiveness of the scheme for mitigation of torsional oscillations significantly.

This paper introduces fuzzy logic controller of Type-2 to organize the interventions of novel resistor bake model for inter-area oscillation stabilization in Kundur's two-area benchmark test power system via MATLAB/Simulink-based modelling and simulation environment with the help of Interval Type-2 fuzzy logic controller toolbox. The propositioned controller uses the relative kinetic energy deviation between the two areas as an input control signal. Three case studies, including small, mild, and severe disturbances should elucidate the effectuality of the propositioned scheme. The obtained comparative time-domain simulation results should prove that the suggested stabilization regime is a reliable mean for treating the issue of inter-area power oscillations.

Abstract: This work describes how to utilize the electrochemical technique to determine labetalol hydrochloride (Lab) in pure form and combined pharmaceutical formulation for quality control purposes. Four membrane sensors were developed using two plasticizers, dioctyl phthalate with 2-hydroxypropyl-β-cyclodextrin and ammonium reineckate (RNC) for sensors 1a and 2a, and tributyl phthalate with 2-hydroxypropyl-β-cyclodextrin and ammonium reineckate for sensors 1b and 2b as ionophores in polyvinyl chloride (PVC) matrix. Fast response and stable Nernstian slopes of 59.60, 57.58, 53.00 and 55.00 mV/decade for sensors 1a, 2a, 1b, and 2b, respectively, were obtained by developed sensors within a concentration range 10–4 M–10–2 M over pH range 2.00–5.10. Developed sensors showed good selectivity for Lab in pure form, in the presence of co-administered drugs, many of interfering ions, and excipients present in pharmaceutical formulation. No remarkable difference was detected upon the statistical comparison between the results of proposed sensors and the official method.

Abstract: Inter-area oscillations are, by far, the most detrimental to the synchronous integrity of interconnected power systems. This detriment comes from their wide frequency spectrum and the large numbers of the participant generators. The inherent poor damping associated with the inter-area oscillations leaves open wide probabilities for irrevocable widespread blackouts with the consequent eventual devastating outcomes measured in terms of the huge economic casualties and the possible human fatalities. This article explores the influences of the Interval Type-2 fuzzy logic-based strategized dynamic braking interventions of dual brake models, namely Thyristor Controlled Braking Resistors (TCBRs), for neutralizing the jeopardy of negatively damped inter-area power oscillations in Kundur’s two-area test system, using MATLAB™/Simulink environment. The relative inner generator's speed deviation is employed in this work as a control signal to the proposed controller. The effectiveness of the proposed scheme is authenticated by considering four case studies with different severity degrees. By analyzing the performance repercussions due to four disturbances, without the implementation of the proposed scheme, the unstable nature of the system responses is clearly noticed. With the implementation of the proposed scheme, the system oscillatory behavior is stabilized in an appropriate manner. The performed comparative non-linear time-domain simulation results emphasize the great potential of the proposed scheme in mitigation of inter-area power oscillations according to the considered disturbances. The proposed scheme is simple yet effective in treating the inter-area oscillations appropriately under the considered case studies.

Abstract: Summary Background Congenital anomalies are the fifth leading cause of mortality in children younger than 5 years globally. Many gastrointestinal congenital anomalies are fatal without timely access to neonatal surgical care, but few studies have been done on these conditions in low-income and middle-income countries (LMICs). We compared outcomes of the seven most common gastrointestinal congenital anomalies in low-income, middle-income, and high-income countries globally, and identified factors associated with mortality. Methods We did a multicentre, international prospective cohort study of patients younger than 16 years, presenting to hospital for the first time with oesophageal atresia, congenital diaphragmatic hernia, intestinal atresia, gastroschisis, exomphalos, anorectal malformation, and Hirschsprung's disease. Recruitment was of consecutive patients for a minimum of 1 month between October, 2018, and April, 2019. We collected data on patient demographics, clinical status, interventions, and outcomes using the REDCap platform. Patients were followed up for 30 days after primary intervention, or 30 days after admission if they did not receive an intervention. The primary outcome was all-cause, in-hospital mortality for all conditions combined and each condition individually, stratified by country income status. We did a complete case analysis. Findings We included 3849 patients with 3975 study conditions (560 with oesophageal atresia, 448 with congenital diaphragmatic hernia, 681 with intestinal atresia, 453 with gastroschisis, 325 with exomphalos, 991 with anorectal malformation, and 517 with Hirschsprung's disease) from 264 hospitals (89 in high-income countries, 166 in middle-income countries, and nine in low-income countries) in 74 countries. Of the 3849 patients, 2231 (58·0%) were male. Median gestational age at birth was 38 weeks (IQR 36–39) and median bodyweight at presentation was 2·8 kg (2·3–3·3). Abstract: Plant microbiota have co-evolved with their associated plants in the entire holobiont, and their assemblages support diversity, health and productivity on our planet. Of importance is the in vitro cultivation and identification of their hub taxa for possible core microbiome modification. Therefore, we recently introduced the in situ-similis culturing strategy, based on the use of plant leaves as platform for in vitro growth of plant microbiota. Here, we augment the strategy by the incorporation of the Most Probable Number (MPN) method, based on the use of leaf strips and root segments immersed in plain semi-solid water agar, as growth milieu. Cultivability of bacterial endophytes of sunflower plants (Helianthus annuus L.) was monitored in terms of MPN population estimates, community composition through PCR-DGGE analysis and 16S rRNA gene sequencing of representative isolates. The method efficiently enriched the plant endophytes as MPN estimates exceeded log 7.0 g-1 leaf/root. Results of PCR-DGGE analyses indicated divergence in community composition of cultivable bacterial endophytes primarily attributed to the type of culture media, signaling a certain degree of plant-organ specificity and compatibility. Based on 16S rRNA gene sequencing of representative bacterial isolates, 20 genera comprising 32 species were enriched in leaf strips- and root segments-based media. They belonged to the three phyla Bacteroidetes, Firmicutes and Proteobacteria, especially Alpha- and Gammaproteobacteria. The MPN strategy furnished a diversified nutritive platform in terms of homologous/heterologous and ambient/limited oxygenic cultivation. As a result, cultivability was extended to more 8 genera - Bosea, Brevundimonas, Caulobacter, Chitinophaga, Pseudoxanthomonas, Scandinavium, Sphingobacterium, Starkeya - among them three genera, namely Caulobacter, Scandinavium and Starkeya, which were not yet reported for Sunflower, even with the use of metagenomics sequencing methods. Possible unknown species or even one new putative genus were enriched. Thus, not only potential members of the major microbiome but also the rare isolates of satellite microbiomes can be isolated using the presented in situ similis MPN-cultivation strategy. It is a feasible addition to traditional cultivation methods to probe deeper for both the major and rare taxa and fill the many cultivation gaps within plant microbiota.

Abstract: Background:Hydrochlorothiazide (HCTZ) is potent diuretic that is used alone or in combination with other drugs such as labetalol (Lab) (mixtures Ι) or nebivolol (Neb) (mixtures ΙΙ) to control moderate to sever hypertension.Introduction:This paper demonstrates the establishment of different validated spectrophotometric and chemometric methods for simultaneous estimation of these mixtures in pure form and pharmaceutical formulations in the presence of HCTZ related impurities in quality control laboratories.Methods:(A) Derivative method (D3) of Lab and HCTZ and its related impurities at 245.3nm and 278.5nm respectively, (D1) of Neb and HCTZ at 294.2nm and 282.2nm, respectively. (B) First derivative of ratio spectra method (DD 1) of Lab at 244.3nm, HCTZ at 261.2nm and 275.4nm, while at 294nm for Neb and 269.4nm for HCTZ. (C) Ratio difference method which depends on measuring the distinction between the amplitudes of ratio spectra at 240nm and 288.3nm for Lab and at 270.1nm and 277.4nm for HCTZ for mixture Ι while at 290.4nm and 299.2nm for Neb and at 232.2nm and 254nm for HCTZ for mixture ΙΙ. (D) Mean centering of ratio spectra (MC) and (E) partial least squares regression (PLS) and principal component regression (PCR).Results:These methods were applied over concentration ranges of 10-100 µg/ml, 10-75 µg/ml and 2.5- 25 µg/ml of Lab, Neb and HCTZ, respectively. Methods were validated according to ICH guidelines and statistical comparison of results of reported and proposed methods revealed no difference.Conclusion:The methods were successfully used for the frequent analysis of selected mixtures in quality control laboratories.
