1. Impact of P2RX7 purinoceptor ablation on the morphometric, mechanical and tissue properties in the murine model of Duchenne muscular dystrophy

Abstract

Duchenne muscular dystrophy (DMD) is an inherited, lethal disorder characterised by progressive muscle degeneration and associated bone abnormalities. It has previously demonstrated that P2RX7, a purinergic receptor, contributed to the pathogenesis of DMD, and found that P2RX7 ablation alleviated the severity of the disease. In this work a dystrophic mdx mouse crossed with the global P2RX7 receptor has been used to generate a knockout mouse model (mdx/P2X7-/-), and compared its morphometric, mechanical and tissue properties against those of mdx,a mouse model without the dystrophin protein, as well as a wild type (WT) and aP2RX7 knockout (P2X7-/-). Micro-computed tomography (μCT), three-point bending testing, scanning electron microscopy (SEM) and nanoindentation were utilised in the study. The bones were analysed at approximately 4 weeks of age to examine the impact of P2RX7 ablation on the bone properties during the acute disease phase, before muscle wasting is fully developed. The results show that P2RX7 purinoceptor ablation has produced improvement or significant improvement in some of the morphological, mechanical and tissue properties of the dystrophic bones examined. Specifically, although the ablation produced smaller bones with significantly lower total cross-section area (Tt.Ar) and Second Moment of Area (SMA), significantly higher cortical bone area (Ct.Ar),cortical bone area ratio (Ct.Ar/Tt.Ar) and trabecular bone volume fraction (BV/TV)are found in the mdx/P2X7-/- mice than in any other types. Further, the mdx/P2X7-/- bones have relatively higher average flexural strength, work-to-fracture and significantly higher strain to failure compared with those of mdx, suggesting greater resistance to fracture. Indentation modulus, elasticity and creep are also significantly improved in the knockout cortical bones over those of mdx. These findings seem to suggest that specific pharmacological blockade of P2RX7 may improve dystrophic bones, with a potential for therapeutic application in the treatment of the disease.

Abstract

Students faced difficulties in adapting online distance learning (ODL) quickly during the pandemic COVID-19, which indirectly affects their attitude towards ODL. Aim: This study aims to assess the impacts of medical imaging students' (MIS) challenges with ODL on their attitude during the COVID-19. Methodology: A cross-sectional online survey was conducted on 202 MIS in Universiti Teknologi Mara (UiTM) Puncak Alam. Data were collected through google form and analysed using SPSS version 26. Results: Descriptive analysis results show that the majority (87.6%) of the MIS experienced moderate to high levels of challenges. The highest frequency of MIS reported a high level of challenges regarding curriculum content. There was not statistically significant in the overall mean total scores of attitudes towards ODL with regard to degree students experienced higher mean attitude compared to diploma students. The presence of the internet at home has a significant association with the challenges faced with ODL. Conclusion: MIS experiencing different levels of challenges with ODL have a negative attitude toward ODL. It is concluded that when the level of challenges increased, the attitude toward ODL tends to be negative. [ABSTRACT FROM AUTHOR]

10. Systematic Review on Multilevel Analysis of Radiation Effects on Bone Microarchitecture

Abstract

Introduction. Modern radiation therapy has become an effective method to treat and monitor tumour growth in cancer patients. It has proved to be a successful way to minimise mortality rates. However, the adverse effects of radiation have been historical evidence in the clinical environment involving diminishing the quality and density of bone and causing fragility fracture to the bone in the long run. This systematic review was aimed at identifying and evaluating the effects of irradiation on morphology and mechanical properties of murine model bone in previous publications. Methods. A systematic literature review was undertaken following the Preferred Reporting Items for Systemic Reviews and Meta-analysis (PRISMA) guidelines. A comprehensive literature search was performed using Scopus, Web of Science, and Science Direct databases (English only studies published between 2015 and 2020). The selected studies were evaluated according to three criteria: (1) criteria for study sample selection; (2) criteria for methodological procedures; and (3) criteria for detection and evaluation. Results. The initial search strategy identified 1408 related studies, 8 of were included based on inclusion and exclusion criteria. This review revealed an association between bone destruction and the magnitude of time and dose postirradiation. We agreed that the effect of radiation on bone morphology and strength primarily is a later stage event but noticeable in both low (1 Gy) and high dose (30 Gy) radiation. Trabecular and cortical bone microstructures were significantly altered at irradiation and contralateral sites. Besides, the mechanical strength was significantly impacted in both shorter and longer periods. Conclusion. Overall, the radiotherapy altered bone microstructures and substantially decreases bone mechanical properties. The alteration was related to quantity and the activity of the osteoblast and osteoclast. Early detection of those most at risk for radiation-induced bone alterations could lead to better prophylactic intervention decisions.

Abstract

Introduction: Hybrid Positron Emission Tomography with Computed Tomography (PET/CT) imaging is well established in the oncology setting. However, in the current cancer diagnostic imaging approach in Malaysia patients need separate CT and PET/CT examinations for morphologic and physiologic cancer staging, respectively, as PET/CT is unable to produce images with optimum diagnostic quality. Therefore, introducing contrast media into CT attenuation correction (CTAC) acquisition to increase image quality have raised concerns on heightened radiation exposure. This study aimed to verify the amount of external radiation exposure irradiated to the patient converging to the scanning protocol implemented in whole-body (WB) 18F-FDG-PET/CT examination. Methods: A retrospective study was conducted to determine the radiation dose delivered during CTAC acquisition of 18F-FDG-PET/CT examination at three hospitals providing PET/CT imaging services in Penang State. The implemented scanning protocols and parameters, and the effective dose received by the patient were analysed based on dose length product (DLP) and CTDIvol reported by the scanner. Results: CTAC of WB 18F-FDG-PET/CT imaging was executed as either non-contrast (NC-CTAC) or contrast-enhanced (CE-CTAC). CE-CTAC produced a 250% higher radiation dose compared to NCCTAC. The scanning parameter differences that significantly contributed to increased radiation dose were the tube current and pitch value. Conclusion: CE-CTAC delivered a higher radiation dose than NC-CTAC acquisition in WB 18F-FDG PET/CT imaging due to different mAs and pitch as scanning parameters. [ABSTRACT FROM AUTHOR]
