The addition of silver nanoparticles (AgNPs) to biomedical textiles can be of great interest to protect the materials against microorganisms and prevent their spread. However, the human and environmental over‐exposure to AgNPs is leading to numerous concerns due to their toxicity. In this work, AgNPs were stabilized onto polyamide 6.6 fabrics (PA66) through atmospheric dielectric barrier discharge (DBD) plasma treatment and the use of chitosan (Ch) layers applied by spray. DBD plasma treatment revealed a crucial role in AgNPs adhesion (4.8 and 6.3 At%). A first layer of Ch decreased the AgNPs adhesion in both untreated and DBD plasma‐treated samples but treated samples show higher concentration (1.7 and 4.1 At%). The antibacterial activity was evaluated against Staphylococcus aureus and Escherichia coli after 2 and 24 h, showing a superior action in all samples with DBD plasma treatment after 24 h. The Ch in the first layers of the composites delayed the antimicrobial action of the samples but it also may enhance antimicrobial action. The obtained coatings will allow the development of novel and safe wound dressings with improved AgNPs deposition, controlled ions released and consequently, manage the antimicrobial performance and minimize the AgNPs side effects.

The GGGGCC (G(4)C(2)) repeat expansion mutation in the C9ORF72 gene is the most common genetic cause of frontotemporal dementia (FTD) and amyotrophic lateral sclerosis (ALS). Transcription of the repeat and formation of nuclear RNA foci, which sequester specific RNA-binding proteins, is one of the possible pathological mechanisms. Here, we show that (G(4)C(2))(n) repeat RNA predominantly associates with essential paraspeckle proteins SFPQ, NONO, RBM14, FUS and hnRNPH and colocalizes with known paraspeckle-associated RNA hLinc-p21. As formation of paraspeckles in motor neurons has been associated with early phases of ALS, we investigated the extent of similarity between paraspeckles and (G(4)C(2))(n) RNA foci. Overexpression of (G(4)C(2))(72) RNA results in their increased number and colocalization with SFPQ-stained nuclear bodies. These paraspeckle-like (G(4)C(2))(72) RNA foci form independently of the known paraspeckle scaffold, the long non-coding RNA NEAT1. Moreover, the knockdown of SFPQ protein in C9ORF72 expansion mutation-positive fibroblasts significantly reduces the number of (G(4)C(2))(n) RNA foci. In conclusion, (G(4)C(2))(n) RNA foci have characteristics of paraspeckles, which suggests that both RNA foci and paraspeckles play roles in FTD and ALS, and implies approaches for regulation of their formation.

Healthcare-associated infections (HCAI) are complications of healthcare that result in elevated patient morbidity and mortality. HCAI present a huge financial burden for patients, hospitals and insurers due to extended hospitalisation and associated care. According to the estimations, in the US alone, HCAI affects approximately 2 million patients annually, of whom approximately 90.000 patients die, with an estimated annual cost estimated to range from 28 billion to 45 billion US$. [1] European Union is facing the similar situation, the European Centre for Disease Prevention and control (ECDC) advice that approximately 4.1 million acute care patients acquire a HCAI annually, with 37.000 deaths directly attributed to HCAI. With increasing prevalence of HCAI across European countries and threatening development of antimicrobial resistance to widely used antibiotics, there is a recognised need for novel approach in battle against this healthcare burden [2]. One of the approaches involves a development and fabrication of materials with antimicrobial properties. Usually, these are coatings with integrated antibacterial agent that is responsible for the elimination of microorganisms that come into contact with active surface. There is a variety of different antibacterial compounds integrated in such coatings, such as different antibiotics, chemical compounds, peptides. Recently, metal nanoparticles (NPs) have been increasingly used in designing coatings with antibacterial properties due to their large surface-to-volume ration, physiochemical properties and biological multi-target mechanism of actions. Besides all beneficial properties of NPs their emergence of cytotoxicity is limiting their practical applications in human body. [3-4] To overcome this drawback it is important to design a new class of antibacterial coatings with firmly embedded NPs that allows controlled release of antimicrobial agent into the microenvironment. Atmospheric pressure plasma technology has shown a big promise as an alternative and cost-efficient method for deposition of coatings with antibacterial properties. This contribution explores the potential of plasma-assisted approach for fabrication of antibacterial coatings, containing different metal NPs on medical textiles. Plasma-assisted deposition of coatings was carried out with so-called ˝sandwich technique˝, where nanoparticles were embedded between two layers in order to tailor the desirable ion release and to prolong antibacterial effect of fabrics. Antibacterial effects of different nano-coatings were tested against G+ and G- bacterial species, Staphylococcus aureus and Escherichia coli, respectively. Besides antibacterial properties, potential cytotoxic effects were also studied. The study demonstrates that atmospheric pressure plasma can be an efficient technique for deposition of antibacterial coatings containing metal NPs. Medical textiles with plasma-assisted nano-coatings showed effective antibacterial properties. The choice of proper metal antimicrobial agent and optimal concentration of NPs should be considered in regards to potential cytotoxic effects when these materials would be used in medical environments., info:eu-repo/semantics/publishedVersion

The development of new multifunctional coatings with antimicrobial properties has a special interest in several applications for pharmaceutical and medical products. This work reports on the deposition of antimicrobial coatings based on silver nanoparticles (Ag NPs) embedded in an organosilicon film onto woven and nonwoven textiles. The Ag nanoparticles admixed with hexamethyldisiloxane (HMDSO) vapours are introduced by means of an atomizer system in the remote discharge of an atmospheric pressure plasma source operating in argon. Non-woven textile materials with antimicrobial properties are of high demands for applications ranging from medical dressing to everyday cleaning products. We proposed plasma assisted route to engineer polymer films and non-woven fabrics with antimicrobial nano-composite coatings. Atmospheric pressure plasma is used for deposition of nano-composites. Nano-particles Ag, Cu and ZnO are tested as antimicrobial agents with average nano-particles size of about 50 nm. Nanoparticles are incorporated in between two layers of organosilicon film with impregnation process or alternatively through aerosol injection step. Incorporation is made in “sandwich-like structure” where top layer coating (barrier) of 5 - 50 nm thickness is used for precise control of metal ions release and reservation layer of about 200 nm is used for NPs load. The effect of the barrier coating on surface concentration of nano-particles is determined with XPS method. It is shown that increase of the thickness of the barrier layer from 5 to 25 nm leads to decrease of Ag content from 0.24 to 0.08 at. %. Similar effect is found for nano-particles of Cu and ZnO. Antibacterial efficiency of the samples against P. aeruginosa ATCC 9027 and S. aureus M u50 bacteria shows that all treated samples exhibit higher antibacterial efficiency against S. aureus. The antibacterial efficiency of AgNPs and CuNPs is above 90% which is practically interested for medical application while ZnONPs shows lower antibacterial efficiency. A fully portable atmospheric pressure air plasma system is reported to be suitable for the microbial decontamination of both surfaces and liquids. The device operates in quiescent air, and includes an integrated battery which is charged from a solar cell and weighs less than 750 g, making it highly amenable for a wide variety of applications beyond the laboratory. Using particle imaging velocimetry to visualise air flows around the device, the geometric configuration of the plasma generating electrodes was enhanced to induce a gas flow on the order of 0.5 m s-1 directed towards a sample placed downstream, thus improving the transport of plasma generated reactive species to the sample. The microbial decontamination efficiency of the system was assessed using potable water samples inoculated with common waterborne organisms Escherichia coli and Pseudomonas fluorescens. The reduction in the number of microorganisms was found to be in the range of 2-8 log and was strongly dependent on the plasma generation conditions.

Calcium phosphate (CaP) ceramics are of interest in bone substitution due to their good biocompatibility and bioresorbability. Currently certain CaPs in the market are loaded with antibiotics in order to prevent infections but further control is needed over antibiotic release patterns. Cold plasmas have emerged as a useful means of modifying the interactions with drugs through surface modification of polymer materials. In this work we explore the possibility of using atmospheric pressure plasmas as a tool for the surface modification of these CaP materials with newly populated bonds and charges, with views on enabling higher loading and controlled drug release. Herein the surface modification of ß-tricalcium phosphate ceramics is investigated using an atmospheric pressure helium plasma jet as a tool for tuning the controlled release of the antibiotic doxycycline hyclate, employed as a drug model. The surface chemistry is tailored mainly by plasma jet surface interaction with an increasing O/C ratio without changes in the topography as well as by build-up of surface charges. With this surface tailoring it is demonstrated that the atmospheric plasma jet is a new promising tool that leads to the design of a control for drug release from bioceramic matrices.

Fused in sarcoma (FUS) and TAR DNA-binding protein 43 (TDP-43) are RNA-binding proteins pathogenetically linked to amyotrophic lateral sclerosis (ALS) and frontotemporal lobar degeneration (FTLD), but it is not known if they regulate the same transcripts. We addressed this question using crosslinking and immunoprecipitation (iCLIP) in mouse brain, which showed that FUS binds along the whole length of the nascent RNA with limited sequence specificity to GGU and related motifs. A saw-tooth binding pattern in long genes demonstrated that FUS remains bound to pre-mRNAs until splicing is completed. Analysis of FUS(-/-) brain demonstrated a role for FUS in alternative splicing, with increased crosslinking of FUS in introns around the repressed exons. We did not observe a significant overlap in the RNA binding sites or the exons regulated by FUS and TDP-43. Nevertheless, we found that both proteins regulate genes that function in neuronal development.

PURPOSE: To document the pattern of enhancement and morphologic changes on MR images that occur in the intervertebral disk and adjacent vertebral bodies after diskectomy and to correlate the presence of intervertebral disk enhancement with the preoperative and postoperative clinical findings. METHODS: Preoperatively, and at 3 months and 6 months after surgery, 94 adults who had first-time surgery for a herniated lumbar intervertebral disk that was associated with radiculopathy, expressed as leg symptoms or signs (with or without lower back pain), were asked to respond to a questionnaire regarding pain, were given serial physical examinations, and were examined with contrast-enhanced MR imaging. The measures of clinical outcome that were evaluated included the straight leg raise sign, radicular pain, and lower back pain. Type of disk herniation, intervertebral disk enhancement, disk space height, and degenerative end-plate changes were also assessed. RESULTS: Of the 94 patients evaluated, 19 (20%) had postoperative intervertebral disk enhancement that was not present on the preoperative study. The pattern of enhancement was remarkably consistent, with 18 of the cases showing linear enhancement within the intervertebral disk, manifested as two thin bands paralleling the end plates. End-plate enhancement was present in 7 (37%) of the 19 patients with disk enhancement. There were no significant associations between disk enhancement and specific clinical symptoms before or after surgery. CONCLUSION: Our group of asymptomatic postoperative patients had anular enhancement (curette site), disk enhancement, and vertebral end-plate enhancement on MR images without evidence of disk space infection. This finding points out the need to understand asymptomatic postoperative changes that are sequelae of surgery and not necessarily indicators of infection.

PURPOSE: To compare conventional T2-weighted spin-echo imaging with a rapid acquisition relaxation enhanced (RARE) technique in the routine evaluation of lumbar degenerative disk disease. METHODS: Thirty consecutive patients referred for evaluation of the lumbar spine for suspected degenerative disk disease were evaluated with sagittal and axial T1-weighted spin-echo, conventional T2-weighted spin-echo, and T2-weighted RARE "turbo spin-echo" sequences (4000/93/2 (repetition time/echo time/excitations), 192 x 256, echo train length of 8). Conventional T2-weighted and RARE images were evaluated independently by two neuroradiologists for image quality, presence of artifacts, cerebrospinal fluid signal intensity, extradural interface conspicuity, intradural nerve root conspicuity, soft-tissue detail, and signal intensity of normal and degenerated intervertebral disks. RESULTS: Both readers rated the cerebrospinal fluid signal higher, the extradural interface conspicuity higher, and the nerve root detail greater on the turbo spin-echo than on conventional spin-echo images. Neither reader had a significant difference in ranking "normal" or "degenerated" disk signal on the two sequences. Both readers rated soft-tissue detail higher for conventional than for turbo spin-echo. CONCLUSION: RARE sequences can replace conventional T2-weighted spin-echo sagittal studies for degenerative lumbar disk disease.

PURPOSE: To investigate an observed preferential T2-shortening effect seen in patients with intracerebral metastases from adenocarcinoma of the GI tract. METHODS: Ten patients with intracerebral metastases from adenocarcinoma of the GI tract were evaluated with MR imaging with CT and histopathologic correlation. RESULTS: Nine of 10 patients demonstrated intracranial masses with decreased signal intensity on T2-weighted images. T1-weighted images showed decreased or isointense signal in all cases. Special histologic staining was performed in four of 10 cases, all of which were positive for increased protein (mucin) with no evidence of blood products or calcium. CONCLUSION: We propose that the observed preferential T2-shortening effect is secondary to slowing of rotational and translational proton motion caused by increased protein concentration in the form of mucin.

One-hundred-fifteen patients thought to have intracranial disease were studied with various two-dimensional short repetition time, partial-flip-angle gradient-echo techniques: at 1.0 T, fast low-angle short (FLASH) at 10 degrees and 30 degrees and fast imaging with steady-state precession (FISP) at 60 degrees; at 1.5 T, FLASH 10 degrees, 30 degrees, and 60 degrees, FISP 60 degrees, and contrast-enhanced fast steady state at 60 degrees. These sequences were compared with a T2-weighted spin-echo sequence to determine the relative sensitivities of these techniques in detecting parenchymal lesions. Except for hemorrhagic lesions a substantial number of abnormalities were either not visible or poorly seen on the partial-flip-angle gradient-echo sequences. Minimizing echo time (6-9 msec) to decrease magnetic susceptibility effects did not improve lesion detection. Current usage of two-dimensional gradient-echo techniques for lesion detection is limited, except when calcification or hemorrhage is involved. Their utility may be expanded via the incorporation of three-dimensional techniques and IV contrast agents.
