4. Targeted Screening of the C9orf72 Gene in Bulgarian Amyotrophic Lateral Sclerosis Patients

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease, characterized by progressive degeneration of the upper and lower motor neurons, leading to muscle weakness, hypotrophy, swallowing and respiratory failure. The cause of ALS is not yet fully elucidated, but there are 35 associated genes and 2 gene loci with an unidentified gene. The most common are C9orf72, SOD1, TARDBP and FUS found in approximately 10% of patients. Variants in the C9orf72 gene are the main cause of fALS – 25-40% of cases (and a small percentage of sALS). The goal of the present study was to evaluate the significance of the C9orf72 hexanucleotide repeat expansion in Bulgarian patients with ALS, through the means of in house and triplet repeat-primed PCR assay (TP-PCR). From 171 patients diagnosed with ALS and included in the current study, we have identified the repeat expansion with more than 145 GGGGCC repeats in 7 (4,1%). Short expansions or borderline values (24 to 30 repeats) were not detected. Due to absence of sufficient data, we have established an ALS-focused research for the association of the C9orf72 gene in clinically well-characterized Bulgarian ALS patients. Published data show variable percentage ratios for genetically verified cases (4-40%), which is mainly due to small sample counts and sALS-fALS ratios. Our patients’ group also contains sALS and fALS cases, which explains the low percentage of genetic verification. The obtained results enrich the worldwide database and shed light onto genetically characterized Bulgarian ALS patients. Affected patients and their families can receive adequate medical-genetic consultation and prenatal diagnostic testing.

NARS1-related neurodevelopmental disorder is caused by pathogenic variants in the NARS1 gene. The aim of the present study was to determine the genetic reason for the epilepsy and microcephaly, observed in the tested patient. Using whole exome sequencing two novel heterozygous variants c.676G>C, p.Val226Leu and c.986G>A, p.Arg329Gln in the NARS1 gene were detected. The obtained results from the molecular-genetic analysis can enrich the spectrum of epilepsy related variants and can explain the genetic etiology of a severe epileptic disease.

Introduction: Thyroid carcinoma is the most common endocrine cancer. Some somatic mutations in genes (BRAF, NRAS and TERT) involved in key signaling pathways and genome stability have been recently identified to play an important role in its development. Very little research has been done on their frequency and clinical relevance in Bulgarian patients with papillary thyroid cancer (PTC). This study is focused on investigating somatic mutation frequency in Bulgarian patients with PTC and their association with clinicopathologic features.

The process of neoangiogenesis is one of the classic hallmarks of a cancer. Its intricate mechanisms have long been one of the major domains in cancer research and a hope for a therapeutic breakthrough. Last decade a new subgroup of non-coding RNA molecules was reported called microRNAs. Literally hundreds of new molecules in this class are being uncovered as pivotal regulators in virtually all intracellular processes. The aim of this study is to classify and review those microRNA molecules that have a role in the processes of tumor angiogenesis and map their places in the regulatory framework of the classical proangiogenic genes and their canonical cascades.

9. Altruistic Surrogacy – Ethical Issues and Demographic Differences in Public Opinion

This is the first study of the attitude of Bulgarian people towards the allowing of altruistic surrogacy which is prohibited in Bulgaria. This study used an online survey which was active during one year (July 2010-June 2011) and which was answered by 951 respondents between 18-65 years of age. The majority of them (87%) are young people between 18-43 years. The respondents are men and women with secondary, university and medical university education from the capital and the countryside. They have different marital status. The data were treated with statistical package SPSS 16. The link between the demographic characteristics (gender, age, education, marital status and place of residence) and the answers of the respondents was identified. The majority of the respondents (73%) think that the altruistic surrogacy must be allowed in Bulgaria and the main supporters are the women and the residents in the countryside – married or living with partner. Only 38% of the respondents mostly divorced middle aged persons accept the access of same sex couples to surrogacy. The majority of the respondents (53%) fear that the surrogacy may transform poor women into incubators for babies. This opinion is shared by the men, by the youngest and the oldest respondents and by the unmarried persons.

10. The Donation of Gametes and the Anonimity of the Donors

A study of the public opinion in Bulgaria in regard to the possibility of the adult children, created by gamete donation, to learn who their biological parents are was made. This investigation was an on-line survey. The participants in the study are individuals between 18 and 65 years of age - Internet users. The survey was included into different web sites and this enlarged the number of respondents (up to 994) and 85% of them were persons in fertile age - from 18 to 43 years. The answers of the respondents in relation of the demographic features - gender, age, education, family status and place of living were studied. The data were calculated with the special statistical product SPSS 16. A critical level of significance 0.05 was used. Results: Almost half (47%) of the respondents agree that the children born from gamete donors have the right to know at adulthood their biological parents, 35% disagree and 18% have no opinion. The demographic features influencing the answers of the question are the gender, the education and the family status of the respondents (p < 0.05). Most of the men (60%) consider that the children have the right to know their biological parents while only 44% of the women approve. The highest support of the idea about contact between the donors and their genetic off springs show the people with secondary education (56%) and the most skeptical are the respondents with high non-medical education (40%). The family status influences the opinion of the respondents - the approval of the married and unmarried is 38% and 60%, respectively.

In this paper, we analytically compute all master integrals for one of the two non-planar integral families for five-particle massless scattering at two loops. We first derive an integral basis of 73 integrals with constant leading singularities. We then construct the system of differential equations satisfied by them, and find that it is in canonical form. The solution space is in agreement with a recent conjecture for the non-planar pentagon alphabet. We fix the boundary constants of the differential equations by exploiting constraints from the absence of unphysical singularities. The solution of the differential equations in the Euclidean region is expressed in terms of iterated integrals. We present the findings of a Whole Exome Sequencing in a 2-year-old boy, conceived via In VitroFertilization with donor sperm, who suffers from an undiagnosed neurological syndrome. The following heterozygous variant in the EPHA4gene was identified and classified as likely pathogenic: c.1655_1656, p.(Ser552CysfsTer23). Subsequent segregation analysis showed that the variant was not inherited from the mother and the sperm donor is not accessible for genetic testing. The presented results can further expand upon the genetic variants considered when diagnosing complex neurological syndromes and shows the importance of access to biological samples from donor banks in genetically ambiguous cases.

- Abstract
The transferability and clinical value of genetic risk scores (GRSs) across populations remain limited due to an imbalance in genetic studies across ancestrally diverse populations. Here we conducted a multi-ancestry genome-wide association study of 156,319 prostate cancer cases and 788,443 controls of European, African, Asian and Hispanic men, reflecting a 57% increase in the number of non-European cases over previous prostate cancer genome-wide association studies. We identified 187 novel risk variants for prostate cancer, increasing the total number of risk variants to 451. An externally replicated multi-ancestry GRS was associated with risk that ranged from 1.8 (per standard deviation) in African ancestry men to 2.2 in European ancestry men. The GRS was associated with a greater risk of aggressive versus non-aggressive disease in men of African ancestry (P = 0.03). - Abstract
Prostate cancer is a highly heritable disease with large disparities in incidence rates across ancestry populations. We conducted a multiancestry meta-analysis of prostate cancer genome-wide association studies (107,247 cases and 127,006 controls) and identified 86 new genetic risk variants independently associated with prostate cancer risk, bringing the total to 269 known risk variants. The top genetic risk score (GRS) decile was associated with odds ratios that ranged from 5.06 (95% confidence interval (CI), 4.84-5.29) for men of European ancestry to 3.74 (95% CI, 3.36-4.17) for men of African ancestry. Men of African ancestry were estimated to have a mean GRS that was 2.18-times higher (95% CI, 2.14-2.22), and men of East Asian ancestry 0.73-times lower (95% CI, 0.71-0.76), than men of European ancestry. These findings support the role of germline variation contributing to population differences in prostate cancer risk, with the GRS offering an approach for personalized risk prediction.Copyright © 2021, The Author(s), under exclusive licence to Springer Nature America, Inc.

- Abstract
Prostate cancer is a highly heritable disease with large disparities in incidence rates across ancestry populations. We conducted a multiancestry meta-analysis of prostate cancer genome-wide association studies (107,247 cases and 127,006 controls) and identified 86 new genetic risk variants independently associated with prostate cancer risk, bringing the total to 269 known risk variants. The top genetic risk score (GRS) decile was associated with odds ratios that ranged from 5.06 (95% confidence interval (CI), 4.84-5.29) for men of European ancestry to 3.74 (95% CI, 3.36-4.17) for men of African ancestry. Men of African ancestry were estimated to have a mean GRS that was 2.18-times higher (95% CI, 2.14-2.22), and men of East Asian ancestry 0.73-times lower (95% CI, 0.71-0.76), than men of European ancestry. These findings support the role of germline variation contributing to population differences in prostate cancer risk, with the GRS offering an approach for personalized risk prediction.

- Abstract
Prostate cancer is a highly heritable disease with large disparities in incidence rates across ancestry populations. We conducted a multiancestry meta-analysis of prostate cancer genome-wide association studies (107,247 cases and 127,006 controls) and identified 86 new genetic risk variants independently associated with prostate cancer risk, bringing the total to 269 known risk variants. The top genetic risk score (GRS) decile was associated with odds ratios that ranged from 5.06 (95% confidence interval (CI), 4.84-5.29) for men of European ancestry to 3.74 (95% CI, 3.36-4.17) for men of African ancestry. Men of African ancestry were estimated to have a mean GRS that was 2.18-times higher (95% CI, 2.14-2.22), and men of East Asian ancestry 0.73-times lower (95% CI, 0.71-0.76), than men of European ancestry. These findings support the role of germline variation contributing to population differences in prostate cancer risk, with the GRS offering an approach for personalized risk prediction.

We introduce a novel harmonic superspace for 3dN=6 superconformal field theories that is tailor made for the study of correlation functions of BPS operators. We calculate a host of two- and three-point functions in full generality and put strong constraints on the form of four-point functions of some selected BPS multiplets. For the four-point function of 12-BPS operators we obtain the associated Ward identities by imposing the absence of harmonic singularities. The latter imply the existence of a solvable subsector in which the correlator becomes topological. This mechanism can be explained by cohomological reduction with respect to a special nilpotent supercharge.
